Solid tumor growth and metastasis depends on angiogenesis.Anti-angiogenesis cancer therapy research has become one of the hot topics.Studies have shown that many kinds of Chinese medicine have an effectiveness of anti-tumor angiogenesis. It is one of the most important mechanisms to anti-tumor Chinese medicine.Chinese medicine on the anti-tumor angiogenesis research in recent years will be reviewed.

Objective To study the changes of SUR1-regulated NCCa-ATP channels in core area during focal cerebral ischemia and reperfusion in rats, and to explore the therapeutic window of focal cerebral ischemia. Methods Adult male Wistar rats were rendered to undergo 120 minutes of the left middle cerebral artery occlusion (MCAO) by the intraluminal thread technique before reperfusion. The core area of the left hemisphere cortex ischemia/reperfusion at different times (reperfusion:R3 h, 6 h, 8 h, 12 h and 24 h) was taken to be tested the level of mRNA and protein expression of sulfonylurea receptor 1 (SUR1) using RT-PCR and Western-blot techniques. SUR1 of the ischemic brain microvascular endothelial cells were observed by immunofluorescence double staining at the peak expressing time point of SUR1. Results We found the up-regulation of SUR1 mRNA and SUR1 protein in ischemic infarcts tissues for R3 h, 6 h, 8 h and 12 h, peaked at R8 h, compared with the sham-operation group ( <0.05) . However, SUR1 expression increased was observed at R12 h by double immunofluorescence in microvascular endothelium. Conclusion SUR1-regulated NCCa-ATP channel may take part in cerebral ischemic damage during focal cerebral ischemia and reperfusion. The expressions of SUR1 mRNA and SUR1 protein were up-regulated in ischemic infarcts tissues. The data suggested that the best time to apply SUR1 inhibitor is within 8-12 hours after ischemia and reperfusion.

[Objective]To investigate the effect of transforming growth factor ?1(TGF-?1) genes transfection on the proliferation and differentiation of chondrocyte by adenovirus vector,and to observe the quality in repair cartilage defect through constructing tissue-engineered cartilage.[Method]Using replicat ion defective adenovirus Adeno-X~(TM) as a carrier,Articular chondrocytes were transferred by high titers level recombinant adenovirus taking TGF-?1 genes.Ultromicrostructure,prolife ration anddifferentiation were observed by light and electron microscope,flow cytometry and so on.Induce expression of exogenous genes coding protein and cartilage cell phenotype were detected by Immunocytochemistry and Northern blot.Gross observation,histology and effect classification were used to evaluate results in repair cartilage defect by combi nation of infected chondrocytes and bone matrix gelatin(BMG).[Result]Immunocytochemistric staining showedthe expression of exogenous gene coding proteins after infection,and procollagen Ⅱ mRNA in cells was detected increasingly by Northern blot.TGF-?1 stimulated proliferation of chondrocytes in primary cultured,and resulted in recruitment of articular chondrocytes into S-G2/M phase.Chondrocytes seeded onto BMG showed high level of proliferation.Transplantation in vivo showed: cartilage defect was repaired satisfactorily by Combination of BMG and infected chondrocytes,and the structure of repair tissue got close to normal articular cartilage.[Conclusion] Articular chondrocytes prolifera tion could be stimulated by adenovirus vector taking TGF-?1 genes,which could be used to constructed tissue-engineered cartilage,and applied in repairing joint cartilage defect.

Objective To investigate the effect of intrathecal gastrodin on skin cancer pain in mice.Methods Thirty-two female Balb/c mice,aged 8 weeks,weighing 20-25 g,were randomly divided into 4 groups (n=8 each) using a random number table:sham operation group (group S),skin cancer pain group (group SCP),gastrodin group (group G),and artificial cerebrospinal fluid (ACSF) control group (group ASCF).Skin cancer pain was produced by injecting phosphate buffer solution 20 μl containing about 2 ×105 4T1 breast cancer cells into the plantar surface of the left hindpaw.At 14th day after inoculation of cancer cells,ASCF 5 μl was injected intrathecally in S and ACSF groups,and gastrodin 150 μg/kg (5 μl) was injected intrathecally in group G.Before inoculation,at 30 min before intrathecal injection,and at 15,30,60,90,120 and 150 min after intrathecal injection,the thermal paw withdrawal latency (TWL) was measured.The expression of acid-sensing ion channel (ASIC)-3 mRNA in the spinal dorsal horn was detected using the real-time reverse transcriptase polymerase chain reaction after the last measurement of the pain threshold.Results Compared with group S,the TWL was significantly decreased at each time point before and after intrathecal injection in SCP,ACSF and G groups,and the expression of ASIC-3 mRNA in the spinal dorsal horn was significantly down-regulated in group G (P＜0.05).Compared with group SCP,the TWL was significantly increased at each time point after intrathecal injection,and the expression of ASIC-3 mRNA in the spinal dorsal horn was significantly down-regulated in group G (P＜0.05),and no significant change was found in the parameters mentioned above in group ACSF (P＞0.05).Conclusion Intrathecal gastrodin can reduce skin cancer pain and down-regulate ASIC-3 expression in the spinal dorsal horn which is helpful in maintaining the analgesic effect in mice.

Objective To study the effect of somatostatin analogue (SMS201-995,SMS) on apoptosis of human cholangiocarcinoma SK-ChA-1 cells and apoptosis-regulated gene bcl-2 and bax. Methods The changes of apoptosis and bcl-2/bax gene expression were detected by DNA gel electrophoresis, flowcytometric analysis and immunohistochemistry, in situ hybridization methods in cholangiocarcinoma SK-ChA-1 cells after treated with SMS(100?ng/ml) for 6, 12 and 24 hours.Results The characteristic ladder was not detected by agarose gel electrophoresis after treated with SMS(100?ng/ml) for 6 hours. When the cells were treated for 12 and 24 hours, the ladder of DNA electrophoresis was observed. While the cells were treated with SMS for 6, 12 and 24 hours, the percentage of annexin V-positive cells were (22?5)% ,(39?7)% and(58?10)%. Besides,SMS inhibited bcl-2 protein expression and increased bax protein and mRNA expression. Conclusion SMS can induce SK-ChA-1 cell apoptosis mainly with regulation of bcl-2 and bax gene expression.

Objective To investigate the expression and significance of transforming growth factor-beta1(TGF-? 1),transforming growth factor-beta receptor typeⅠ(TGF-? RⅠ) in human ovarian carcinoma. Methods Inmmunohistochemical SP method was used to detect the expression of TGF-? 1 and TGF-? RI in 85 cases of benign and malignant ovarian tumors. Results The positive rate of TGF-? 1 in benign and malignant ovarian tumors was significantly higher than that in the normal ovarian tissues(P
