1.Clinical Observation on Integrated Traditional Chinese and Western Medicine in Treating Diabetic Nephropathy

Objective To evaluate the curative effect of integrated traditional Chinese and western medicine in treating diabetic nephropathy. Methods 114 cases of type 2 diabetes were randomly separated into experiment group and control group. Members in the experiment group were treated by integrated traditional Chinese and western medicine and Members in the control group were treated with western medicine alone for 8 weeks' course separately. At the end of therapeutic courses, levels of 24 h total proteinuria, serum creatinine, blood-fasting sugar were evaluated. Results In the experiment group, 8 cases got outstanding effects(11.1%), 50 cases got improvement(69.4%), and 14 cases got no effect (19.4%), with a total effective rate of 80.6%. In the control group, 0 cases got significant effect, 24 cases got improvement(33.3%), and 48 cases got no effect(66.7%), with a total effective rate of 33.3%. There was significant difference in total effective rate between the two groups (P<0.05), that the total effective rate of the experiment group was significantly higher than that of the control group. Conclusion Integrated traditional Chinese and western medicine therapy can be more effective in treating diabetic nephropathy.
2.Effect of Repeated Weight Attached Swimming on Renal Ultrastrueture and Function in Rats

The alterations of ultrastrueture in rat kidney were observed under transmission electron microscopy, results showed that both glomerular filtration membranes and renal tubules had changed immediately after repeated loaded swimming and there were consistency between the recovery of renal ultrastructures and that of urinary protein and excration rate.
3.Inhibition of dedifferentiation of human articular chondrocytes by transfection with recombinant TGF-?_2 in vitro

Objective In cartilage tissue engineering,it was a difficult problem to keep chongdro-cytes phenotype for long time.The aim was made to investigate the dedifferentiation of human articular chon-drocytes,and to verify the inhibition to dedifferentiation by transfection with recombinant pcDNA3.1(+)/TGF-? 2 in vitro.Methods Free chondrocytes were isolated from healthy articular cartilage harvested from6adult patients underwent operation because of other condition.The recombinant pcDNA3.1(+)/TGF-? 2 con-structed in advance was transferred by liposome mediated transfection to monolayer cultured articular chon-drocytes.The first,sixth and ninth generation chondrocytes being transfected and non-transfected were as-sayed by histological examination of HE and Safranine-O staining to observe their morphological changes.RT-PCR,ELISA,immunohistochemistry analysis and prime hybridization were performed to these cells to i-dentify the expression and biosynthesis of TGF-? 2 and cartilage-associated genes and proteins.Results The non-transfected articular chondrocytes in monolayer culture demonstrated the tendency of dedifferentia-tion,and lost the characteristics of their phenotype progressively in subculture.The expression of TGF-? 2 ,collagen typeⅡand proteoglycan in non-transfected cells were decreased,but expression of collagen typeⅠincreased gradually with time going on.On the contrary,as for the transfected chondrocytes,the trans-ferred gene was expressed in cells of all examined generations,and the expression levels were higher than that of non-transfected cell of same generations.The transfected chondrocytes maintained their phenotype and characteristics of articular cartilage.They expressed and produced collagen typeⅡand proteoglycan in a higher level,and collagen typeⅠin a lower level than that of non-transfected cells.Conclusion Human articular chondrocytes in monolayer culture have the tendency of dedifferentiation.The results suggested that chondrocytes tranfected by pcDNA3.1(+)/TGF-? 2 could produce endogenous TGF-? 2 .Transfection of pcD-NA3.1(+)/TGF-? 2 is able to provide transient and long lasting expression of TGF-? 2 in chondrocytes,and may be a feasible method to inhibit dedifferentiation of articular chondrocytes in vitro.
4.Determination of Protocatechuic Acid in Pterospermum heterophyllum by RP-HPLC

OBJECTIVE:To establish an HPLC method for determination of protocatechuic acid in Pterospermum heterophyllum.METHODS: The separation was performed on Agilent TC C18(250 mm?4.6 mm,5.0 ?m)column.Moblie phase consisted of acetonitrile-0.1% glacial acetic acid solution(pH 3.16,10 ∶ 90)with flow rate of 1.0 mL?min-1 and detection wavelength of 260 nm.The column temperature was set at 35℃ and injection volume was 10 ?L.RESULTS:The linear range of protocatechuic acid was 0.007 76~0.310 4 ?g(r=0.999 7) with an average recovery of 97.68% (RSD=1.51%,n=6).CONCLUSION:The method is simple,accurate and reproducible for the quality control of P.heterophyllum.
5.Fanconi anemia protein and acute myeloid leukemia

Fanconi anemia (FA) is an rare autosomal recessive inherited disease which manifests progressive marrow failure, congenital bone malformation, high risk to cancers and so on. Chromatin of FA cells display auto-instability and high hypersensitivity to interstrand DNA cross-links such as mitomycin C. As normally FA develop into acute myeloid leukemia easily, it has been regarded as pre-leukemia state. Till now 11 FA genes have been found and play a role in sustaining stability of gene groups through the same mechanism. As an active connecting protein, FANCF protein play an important part in correct FA complex formation. Which makes FANCD2 single ubiquitin. Ubiquitin FANCD2 induces chromatin and BRCA1 interact, and repair injured DNA. FA gene defect makes gene group instable and increases the risk of chromatin collapse, which finally leads to acute myeloid leukemia and myelodysplastic syndrome.
6.Clinical effect and safety of Guipi pills combined with levothyroxine sodium in hypothyroidism

Objective To evaluate the efficacy and safety of levothyroxine sodium combined with Guipi pills in the treatment of hypothyroidism . Methods A total of 114 cases from August 2013 to November 2015 in Lanxi Traditional Chinese Medicine Hospital with hypothyroidism were selected and randomly divided into control group and experiment group, 57 cases in each group.The patients in the both groups were given conventional treatment and symptomatic treatment.Patients in the control group were given levothyroxine sodium tablets 25 μg orally once a day, two weeks after continuous treatment according to the patient’s condition to 50μg.Patients in the experiment group were given levothyroxine sodium tablets 25μg orally once a day combined with Guipi pills 1.2g, three times a day.Two groups of patients a cycle of 28days, a total of 6 cycles of treatment.The clinical efficacy, thyroid hormone, lipid metabolic indexes and adverse drug reactions were compared between two groups.Results After treatment, the total effective rate of the control group was 78.95%, significantly lower than 92.98% of experimental group (P <0.05).The level of serum TSH, TC and TG were significantly decreased in two groups after treatment, and the experimental group was significantly lower than the control group(P<0.05); the levels of FT3 and FT4 were significantly increased, and the experimental group was significantly higher than the control group ( P <0.05 ).The incidence of adverse reactions was 10.53% in the control group and 5.26% in the experimental group, the difference was not statistically significant. Conclusion Levothyroxine sodium combined with Guipi pills in the treatment of hypothyrosis was effective with high safety.
7.Clinical trial on the effect of leflunomide in treating refractory nephropathy syndrome

Objective To investigate the effect of leflunomide and prednisone in treating refractory nephropathy syndrome.Methods The 60 refractory nephropathy patients in our hospital from Oct,2000 to May,2003 were divided into two groups at random.Paients in test group received leflunomide and prednisone.Patients in control group received mycophenolate mofetil and prednisone.Clinical data were observed in the 2nd,4th,6th,8th,12th,16th,20th,24th and 28th week.Results After receiving leflunomide therapy in the test group,the proteinuria was decreased significantly(P0.05),but rate of showing efficacy in 12th week in Leflunomide group was higher than mycophenolate mofetil group(P
8.Clinical Observation on Therapeutic Effects of Treatment of Diabetic Peripheral Neuropathy with Integrative Traditional Chinese and Western Medicine

Objective To evaluate the clinical efficacy of Dan-Hong Injection and Methycobal combined therapy in treating diabetic peripheral neuropathy(DPN).Methods 72 cases of peripheral neuropathy and type 2 diabetes(conformed to ADA standard published in 1999)were randomly separated into an experimem group and a control group.Members in the experiment group were treated with Dan-Hong injection 20ml/day i.v.and Methycobal 500ug/day i.m.Members in the control group were treated with Methycobal only.At the end of 2 weeks'therapeutic courses,motor nerve conducdon velocity (MNCV)and sural nerve conduction velocity(SNCV)were evaluated.Results The overall effective rate was 92.1% in the experiment group and 67.6% in the control group,showing significant difference between the two groups(P<0.01).The post-therapeutic values of both SNCV and MNCV were significantly different from those of previous treatment.(P<0.01)The post-therapeutic values of SNCV and MNCV of the experiment group were also significantly different from those of the control group(P<0.05).No evidently relative adverse effects were observed in both groups.Conclusions Combined therapy of Dan-Hong injection and Methycobal was more effective in controlling the clinical activity of diabetic peripheral neuropathy.
9.Protective effects of carvedilol on oxidative stress injury induced by hydrogen peroxide in vascular endothelial cells

Aim To observe the influence of carvedilol on the injury and expression of intercellular adhesion molecule-1 induced by hydrogen peroxide in ECV-304 cells and investigate the anti-atherosclerotic effect of carvedilol.Methods: The viability of ECV-304 cells was detected by MTT assay.Morphological changes of ECV-304 cells were observed under converse microscope.The level of lactate dehydrogenase released to the extracellular medium,the intracellular superoxide dismutase activity and the extracellular and intracellular Malondialdelyde level were determined using automatic biochemistry analyser.The expression of ICAM-1 in protein level and mRNA level was detected with flow cytometric technique and RT-PCR.Results Pretreated with carvidilol(1.0?10~(-5)~1.0?10~(-9)mol?L~(-1)) for 24 h,the cell survival rate was increased significantly in a concentration-dependent manner.Pre-incubation for 24 h with carvedilol results in a significant concentration-dependent decline of LDH release from hydrogen peroxide(1.0?10~(-6)mol?L~(-1))injured cells.While ECV-304 cells were pre-incubated with carvedilol,the level of MDA decreased and the activity of SOD increased significantly.Carvedilol produced a concentration-dependent inhibition of the expression of ICAM-1 protein and mRNA in hydrogen peroxide injured ECV-304 cells in a similar manner.Conclusion: These experiments demonstrated that carvedilol was able to protect ECV-304 cells from the oxidative stress injury and inhibit ICAM-1expression in ECV-304 cells induced by hydrogen peroxide.Therefore,we can consider that carvedilol maintains and improves the function of endothelium damaged by hydrogen peroxide from many aspects,which does indicate extensive antioxidant effects on the hydrogen peroxide-injured vascular endothelial cells and suggest promising effects in atherogenesis process.
10.Optimization of Microwave Assisted Extraction Technology of Icariin from Epimedii Folium

OBJECTIVE: To optimize the microwave assisted extraction technology of icariin from Epimedii Folium. METHODS: The microwave assisted extraction technology of icariin from Epimedii Folium was optimized by L9(34)orthogonal design using extraction temperature, extraction time, alcohol concentration,ratio of solid and liquid with the content of icariin as index. RESULTS: The optimal extraction process of icariin was as follows: extraction temperature of 60 ℃,extraction time of 6 min, 30% alcohol and ratio of solid and liquid was 1 ∶ 20. CONCLUSION: The extraction technology is high efficiency and requires short period for extraction of icariin from Epimedii Folium with low cost.
