1.Simultaneous Determination of 5 Active Ingredients in Begonia Grandis Dry from Different Habitats by UP-LC

Objective:To establish a method for the determination of protocatechuic acid, protocatechuic, catechin, epicatechin and rutin in Begonia grandis Dry. Methods:The 5 active ingredients in the samples were extracted by 70% methanol and analyzed by ultra performance liquid chromatography (UPLC). Chromatographic separation was performed on a Waters ACQUITY HSS T3(2. 1 mm × 100 mm, 1. 8 μm) column with gradient elution, and the mobile phase was solvent A: acetonitrile and solvent B:0. 2% glacial acetic acid (0-12 min:A:5→25 and B:95→75;12-30 min:A:25→50 and B:75→45). The effluent was monitored by a VWD detector set at 280 nm. The flow rate was set at 0. 2ml/min and the injection volume was 1μl. The column temperature was maintained at 30℃. Results:The calibration curves were all linear (r≥0. 99) over the test ranges. The average recovery was 96. 92%-99. 75%(n=6). Conclusion:The established method can rapidly obtain an accurate and reproducible result, which can be used to control the quality of Begonia grandis Dry.
2.Simultaneous Determination of 3 Ingredients in Shilintong Tablet by HPLC

OBJECTIVE:To establish a method for simultaneous determination of vicexin 2,vicexin 1 and schaftoside in Shilin-tong tablet. METHODS:HPLC was performed on the column of Phenomenex C18 with mobile phase of methanol-0.1% phosphoric acid (gradient elution)at the flow rate of 0.8 ml/min,the detection wavelength was 334 nm,the column temperature was 25 ℃, and the injection volume was 10 μl. RESULTS:The linear range was 0.31-3.10 μg for vicexin 2 (r=0.9999),0.28-2.80 μg for vicexin 1(r=0.9999)and 0.39-3.90 μg for schaftoside(r=0.9999),respectively;RSDs of precision,stability and reproducibili-ty tests were lower than 3.0%;recoveries were 95.2%-100.4%(RSD=1.6%,n=9),96.3%-100.8%(RSD=1.4%,n=9) and 95.7%-100.9%(RSD=1.7%,n=9),respectively. CONCLUSIONS:The method is simple,sensitive ,accurate,reliable and repro-ducible,and can be used for the quality control of Shilintong tablet.
3.Therapeutic Observation of Acupuncture-moxibustion for Urinary Retention after Anorectal Operation

Objective To observe the clinical efficacy of acupuncture-moxibustion in treating urinary retention after anorectal operation.Method Forty patients with urinary retention after anorectal operation were randomly allocated to a treatment group and a control group, 20 cases each. The treatment group was intervened by acupuncture -moxibustion, and the control group by oral administration of Pyridostigmine bromide tablets. For the two groups, 10 d treatments were taken as a course. Two treatment courses later, the residual urine volume and spontaneous micturition were measured to see the changes, and the clinical efficacies were also compared. Result The total effective rate was 90.0% in the treatment group versus 70.0% in the control group, and the between-group difference was statistically significant (P<0.05). The residual urine volume was significantly changed after the treatment in both groups (P<0.05). After the intervention, the residual urine volume in the treatment group was significantly different from that in the control group (P<0.05). After the treatment, the mean spontaneous micturition time was (16.52±3.18)min in the treatment group, versus (41.37±5.63)min in the control group, and the between-group difference was statistically significant (P<0.05).Conclusion Acupuncture-moxibustion is an effective approach in treating urinary retention after anorectal operation, since it can reduce the residual urine volume and shorten the spontaneous micturition duration.
5.Impact of mice apoptosis antigen of lung cancer induced by paclitaxel on function of dendritic cells

Objective To investigate the in vitro effect of low dosage of paclitaxel on normal murine bone marrow-derived dendritic cells(mDCs)and its role in reactivating tumor-pulsed DCs. Methods The concentration of paclitaxel which could induce 30% apoptosis of 3LL cell lines was figured out.mDCs were generated from murine bone marrow precursors.Cell culture insert system was used and four groups were divided as following: mDC,mDC+3LL,mDC+ low dose of paclitaxel,and mDC+3LL with 30% apoptosis induced by low dose of paclitaxel.The phenotypes,chemoattractive function to MIP1? and MIP-3?,and viability in activating allogeneic T cell proliferation of DCs in the four groups were analysed. Results Paclitaxel of 50 nmol/L could induce 30% apoptosis of 3LL,and had protective effects on DCs.It could stimulate the maturation of mDCs by up-regulating the phenotypes of CD11cCD80,CD11cCD86,CD11cCD40 and CD11cCDIab,and could enhance the chemoattractive function to chemokine MIP-3?.Compared with those cocultured with 3LL,DCs pulsed with apoptosis antigen of 3LL cell which was induced by 50 nmol/L paclitaxl up-regulated the phenotype of CD11cCD40,enhanced the the chemoattractive function to MIP1? and MIP-3?,and activated the proliferation of T cells. Conclusion Paclitaxel of 50 nmol/L can stimulate the maturation of DC,and can partially recover the phenotype and function of tumor-pulsed DC.

9. Effect of silencing Survivin gene expression by RNA interference on the radiosensitivity of cervical carcinoma cell line SiHa

Objective: To explore the influence of silencing Survivin expression by RNA interference on the radiosensitivity of cervical carcinoma SiHa cells. Methods: The recombinant eukaryotic expression plasmid pSurvivin-shRNA was constructed and transfected into SiHa cells. The mRNA and protein expressions of Survivin were determined by RT-PCR and Western blotting, respectively. The activity of caspase-3 was measured by kinase activity determination method. The apoptotic rate was determined by flow cytometry. The changes of radiosensitivity were explored by colony formation test. Results: Compared with SiHa cells transfected with pNeg-shRNA (SiHa/ pNeg-shRNA cells) and untransfected SiHa cells, the expressions of survivin in the cells transfected with pSurvivin-shRNA (Si-Ha/ pSurvivin-shRNA cells) were inhibited significantly at both mRNA and protein levels. The activity of caspase-3 in SiHa/pSurvivin-shRNA cells was significantly enhanced, and the D405 was 1.34 ± 0.05 (P < 0.05). The apoptotic rate of SiHa/pSurvivin-shRNA cells was (5.13 ± 0.81)% and (11.27 ± 1.89)% after 24 hand 48 h X-ray irradiation at 6 MV and 8 Gy, respectively, which was significantly higher than SiHa/pNeg-shRNA cells and untransfected cells (P < 0.05). The colony formation ability of SiHa/pSurvivin-shRNA cells was markedly decreased after irradiation (P < 0.05), which indicated that the sensitivity of SiHa/pSurvivin-shRNA cells was enhanced. Conclusion: The inhibition of Survivin by RNAi could enhance the radiosensitivity of SiHa cells by increasing caspase-3 activity and inducing apoptosis.
10. Relation between ionization and the expressions of HIF-1α and VEGF in anoxia condition

Objective: To investigate the effect of ionization on the expression of HIF-1 α and VEGF in anoxia condition, in order to search an effective method for improving the radiosensitivity. Methods: HepG2 hepatoma carcinoma cells were divided into four groups: the control group; the hypoxia group; the radiation group; the hypoxia and radiation group. The cell apoptosis was detected by fluorescence microscope. The cell viability was analyzed by MTY method. And we measured the expressions of HIF-1 α mRNA and VEGF mRNA by RT-PCR. Results: Cell apoptosis: no apoptosis was observed in the control group; few apoptosis occurred in the hypoxia group; bulk apoptosis was observed in the radiation group; less apoptosis occurred in the hypoxia and radiation group than that in the hypoxia group. The order of surviving fraction was the control group > the hypoxic group > the hypoxia plus radiation group > the radiation group; The order of the expression of HIF-1 α mRNA: the hypoxia plus radiation group > the hypoxic group > the radiation group, no significant difference was observed between the radiation group and the control group; The order of the expression of VEGF mRNA: the hypoxia plus radiation group > the hypoxic group > the radiation group > the control group. Conclusion: The expression of HIF-1α induced by hypoxia can prevent hepatoma cells from the damage of radiation. The HIF-1 α decreases the radiosensitivity through inducing VEGF expression in the HepG2 cell line.
