OBJECTIVE:To study the stability of the mixture of cefradine for injection and ornidazole sodium chloride injection.METHODS:HPLC method was adopted in which the Nova-pak C 18 was taken as the chromatographic column,methanol-water(35∶65)was taken as the mobile phase with detection wavelength at240nm.The contents were determined after mixing of cefradine for injection and ornidazole sodium chloride injection under the room temperature within8hours,and the appearance of the solution was observed and its pH value was determined.RESULTS:No significant differences were found in terms of the appearance,pH value and the contents of the mixed solution.CONCLUSION:The mixture of ornidazole for injection and cefradine sodium chloride injection can be used under room temperature within8hours of mixing.

4. Effect of silencing Survivin gene expression by RNA interference on the radiosensitivity of cervical carcinoma cell line SiHa

Objective: To explore the influence of silencing Survivin expression by RNA interference on the radiosensitivity of cervical carcinoma SiHa cells. Methods: The recombinant eukaryotic expression plasmid pSurvivin-shRNA was constructed and transfected into SiHa cells. The mRNA and protein expressions of Survivin were determined by RT-PCR and Western blotting, respectively. The activity of caspase-3 was measured by kinase activity determination method. The apoptotic rate was determined by flow cytometry. The changes of radiosensitivity were explored by colony formation test. Results: Compared with SiHa cells transfected with pNeg-shRNA (SiHa/ pNeg-shRNA cells) and untransfected SiHa cells, the expressions of survivin in the cells transfected with pSurvivin-shRNA (Si-Ha/ pSurvivin-shRNA cells) were inhibited significantly at both mRNA and protein levels. The activity of caspase-3 in SiHa/pSurvivin-shRNA cells was significantly enhanced, and the D405 was 1.34 ± 0.05 (P < 0.05). The apoptotic rate of SiHa/pSurvivin-shRNA cells was (5.13 ± 0.81)% and (11.27 ± 1.89)% after 24 hand 48 h X-ray irradiation at 6 MV and 8 Gy, respectively, which was significantly higher than SiHa/pNeg-shRNA cells and untransfected cells (P < 0.05). The colony formation ability of SiHa/pSurvivin-shRNA cells was markedly decreased after irradiation (P < 0.05), which indicated that the sensitivity of SiHa/pSurvivin-shRNA cells was enhanced. Conclusion: The inhibition of Survivin by RNAi could enhance the radiosensitivity of SiHa cells by increasing caspase-3 activity and inducing apoptosis.
5.Changes of survivin mRNA and protein expression during paclitaxel treatment in breast cancer cells.

In order to investigate the role of antiapoptosis gene, survivin in the resistance to palcitaxel, the expression of survivin mRNA and protein in the process of paclitaxel treatment in breast cancer cell line MCF-7 was detected MCF-7 cells were incubated with paclitaxel at different concentrations. The growth inhibition rate of MCF-7 was investigated by tetrazolium bromide (MTT) colorimetry. The change of apoptosis was detected by Annexin-V/PI methods. The changes in the expression of survivin mRNA and protein were studied by reverse transcription polymerase chain reaction (RT-PCR) and Western-blot assay respectively. The growth inhibition rate of MCF-7 was increased in a concentration-and time-dependent manner. Paclitaxel of higher concentration could effectively induce apoptosis in MCF-7 cells after 48 h, while the expression of survivin was increased at early time (within 6 h) and decreased after 24 h regardless of treatment concentrations of paclitaxel. It suggested that tumor cells might evade the paclitaxel-induced cell cycle arrest and apoptosis by increasing the level of survivin at early treatment time.

7.Transfusion transmitted virus infection in the neonates

Objective To investigate the relationship between transfusion transmitted virus (TTV)infection and neonatal hyperbilirubinemia,the effect of TTV infection on the liver function, and analyse the feature of nucleotide sequences in TTV ORF1.Methods Serum TTV DNA,which were from 58 neonates with high direct bilirubin(DB,including 5 with hepatitis Syndrome),92 ones with high indirect bilirubin(IB),and 85 normal ones,was detected using a nested polymerase chain reaction technique(nPCR),electropherosis and sequence analyse,and serum alanine amniotransferase (ALT)was determined in all neonates.Results In DB neonates,TTV DNA were detected in 7 neo- nates(12.1%,including 3 neonates with hepatitis syndrome);in IB and normal ones,1 neonate had positive TTV DNA(1.1% and 1.25),respectively.Even if there were point mutations in Guangdong's TTV,the homology of Guangdong's TTV(GD1-9)and Japanese TTV(N22)ranged from 87.1%~97.8% at nucleotide level.Conclusion TTV infection may be one of important pathogenesis resulting in neonatal hyperbilirubinemia and liver damage in such patients.Guangdong's and Japanese TTV isolates had the same genotype,some gene mutations maybe increase the pathogenicity in TTV.

Fragility fracture caused by osteoporosis affects the health of the elderly. The International Osteoporosis Foundation (IOF) advocate that the fracture liaison service (FLS) can improve the detection rate, treatment rate and compliance of patients with fragility fractures, and hence to reduce the incidence of secondary fractures. Currently, there are different models of FLS available globally, appropriate management is the key to effective implementation of FLS projects.
9.Butorphanol,midazolam and innovar in relieving traction response during appendectomy

Objective To compare the effects of butorphanol,midazolam and innovar on relieving appendix traction response during appendectomy. Methods Sixty-eight patients who underwent appendectomy were randomly divided into butorphanol group(n=21),midazolam group(n=23) and innovar group(n=24).Butorphanol,midazolam and innovar were intravenously administered to patients during skin incision.The appendix traction response,sedation score,heart rate(HR),mean arterial pressure(MAP) and pulse oxygen saturation(SpO2) were recorded and compared among groups.Results Compared with those before operation,there was no significant changes in HR,MAP and SpO2 during operation.The effective rates of butorphanol,midazolam and innovar in relieving appendix traction response were 95.2%,65.2% and 50.0%,respectively.Butorphanol surpassed midazolam and innovar in sedation and relieving appendix traction response(P
10.Advances in Study on Hedgehog Signaling Pathway in Pathogenesis of Barrett’s Esophagus

Hedgehog( Hh)signaling pathway is evolutionarily conserved in vertebrates,its excessive activation is associated with abnormal cell differentiation, over proliferation, apoptosis resistance and promotion of invasion and metastasis of tumor cells. Hh signaling pathway involves in the formation and maintenance of esophageal columnar epithelium in embryonic stage,however,undetectable or barely expressed in matured esophageal squamous epithelium. Studies have shown that esophageal Hh signaling pathway can be activated by gastric acid and bile salts. Aberrant activation of Hh signaling pathway can cause the gradual transition of squamous epithelium to columnar epithelium and intestinal-type epithelium,ultimately induces the occurrence of Barrett’s esophagus( BE). Therefore,targeted inhibiting the Hh signaling pathway may be a new strategy for the treatment of BE. This article reviewed the advances in study on Hh signaling pathway in the pathogenesis of BE.
