Journal of Chongqing Medical University 1986;0(02):-
Objective:To study the relative bioavailability and pharmacokinetics of domestic acitretin capsules in 22 healthy male volunteers. Methods:A single dose of 50mg of domestic or imported acitretin was administered by randomized crossover way in 22 volunteers and the plasma concentrations of acitrctin were determined by HPLC.The pharmacokinetic parameters were calculated with 3p97 pharmacokinetic program and the bioequivalency was evaluated. Results:The concentration-time curves of two preparations fitted one-compartment model. The peak plasma levels(C max )of domestic and imported acitretin were 490.51?136.50 and 470.91?118.59ng/ml respectively. The peak time(T max ) were 4.13?1.16 and 3.75?1.18h respectively. And AUC (0~24) were 2891.97?809.60 and 2883.01?905.60ng?h?ml -1 respectively. The relative bioavailability of domestic acitretin capsule was 100.23?11.03%.Conclusion:The result of two one-sided tests suggests that the domestic capsule is bioequivalent to the imported capsule.
2.Color Doppler ultrasonographic study of crural arterial diseases
Chinese Journal of Ultrasonography 2003;0(08):-
50%,0-patent lumen. Those legs with dominant crural arterial lesion (gradeⅠ,Ⅱor Ⅲ) on at least one segments and without femoropopliteal arterial obstructions were selected. Results Totally 75 legs were involved. There were 53 legs( 70.7%) with only 1 main artery damaged,14 legs( 18.7%) with 2 main arteries damaged, and 8 legs( 10.6%) with all 3 main arteries damaged.Among the 5 arterial segments, the anterior tibial arterial lesions were found most frequently ( 73.3%,55/75),in which the segmental occlusion (gradeⅡ) was the most common type. The peroneal arterial lesions appeared least ( 20.0%,15/75).In the damaged main arteries,1/3 anterior tibial-dorsal arteries and 1/5 posterior tibial arteries had greatly improved distal vascularity compared with their proximal and middle portion, owing to the collateral arteries around the ankle.Statistical analysis showed that there was close relationship between the blood flow damage of the distal portion of the posterior tibial arterial segments (medial trans-ankle flow) and the peroneal artery with the clinical ischemic manifestations. Conclusions The dominant occlusive crural arterial lesions and the collateral arteries can be well imaged and diagnosed by color Doppler ultrasonography, which is helpful in providing accurate information of the blood supply of the distal part of the lower extremity.
3.Detection of chromosomally abnormal fetuses by nuchal translucency
Chinese Journal of Ultrasonography 1993;0(01):-
Objective To assess the ultrasonographic detection of nuchal translucency or nuchal edema in identifying chromosomally abnormal fetuses. Methods Fetal nuchal translucency thickness was measured between 10~14 weeks of gestation,or nuchal fold thickness after 14 weeks, with serial follow-up ultrasound examinations. Results In a 2-year period,12 aneuploid fetuses had been identified, together with 5 fetuses of normal karyotype but fatal prognosis.Compared with the fetuses of increased nuchal translucency but normal karyotype and normal follow-up,they had a much thicker nuchal translucency in the first trimester (average 6.1 mm vs 3.6 mm),and in most cases the nuchal translucency evolved into nuchal edema in the second trimester, with other abnormalities detected by ultrasound. Conclusions Detection of increased nuchal translucency by ultrasound is effective in detecting chromosomally abnormal fetuses or fetuses with other abnomalities, and the changes of nuchal translucency observed by follow-up ultrasound may play an important role in differential diagnosis and predicting the prognosis.
4.Attenuating effect of chrysin on experimental autoimmune uveitis and its mechanism
Chinese Journal of Experimental Ophthalmology 2015;33(7):588-593
Background Chrysin has many biological activities, and its anti-inflammatory effect has been confirmed. However, whether it can treat experimental autoimmune uveitis (EAU) is still not elucidated. Objective This study was to investigate the therapeutic effect of chrysin on EAU and explore the potential mechanism. Methods EAU animal models were established in 30 SPF C57BL/6J mice by the subcutaneous injection and ball pad injection of interphotoreceptor retinoid binding protein1-20 (IRBP1-20)/complete Freund adjuvant (CFA),and then the models were randomized into the model control group and chrysin-treated group.Chrysin solution dissolved by 10 μl dimethyl sulfoxide (DMSO)+140 μl PBS was administrated by gavaging in the mice with the dlose 25 mg/kg from 3 days before immunization through 21 days everyday in the chrysin group, and equal volume of solvent was used in the same way in the model control group.Retinas were examined by indirect ophthalmoscope once per day,and inflammation and pathological scores of retina were performed based on the criteria of Caspi on the 21st day after injection. The apoptosis of retinal cells was assayed by TUNEL staining, and the relative expressions of proinflammatory cytokines interferon-γ (IFN-γ),interleukin-17 A (IL-17A),IL-6 and tumor necrosis factor-α (TNF-α) and signal transducer and activator of transcription 1 (STAT1),STAT3,p-STAT1,p-STAT3 in mouse retinas were detected by Western blot. Results Compared with the model control group, the inflammation scores and pathological scores of retinal inflammation were significantly reduced in the chrysin group (inflammation scores:0.50± 0.45 vs 1.58±0.92,t=2.600,P=0.026;pathologic scores:0.58±0.38 vs 1.83±0.75,t=3.638,P=0.005).The infiltration of a large number of inflammatory cells, retinal vasculitis and granulomatous lesions were found in mouse retinas in the model control group, however, the morphology of mouse retinas in the chrysin group was normal based on hematoxylin-eosin staining. The number of apoptotic cells was remarkable lessened in the chrysin group compared with the model control group under the fluorescence microscope.Western blot assay resolved that significantly downregulation in the expressions of IFN-γ,IL-17A,IL-6 and TNF-α was seen in the chrysin group in comparison with the model control group (t =7.802,P =0.001;t =14.906,P =0.000;t =10.241,P =0.001;t =3.304,P =0.030),and the relative expression levels of STAT1,STAT3,p-STAT1 and p-STAT3 were considerably lower in the chrysin group than those in the model control group (t=8.965,P=0.001;t=8.358,P=0.001;t=4.864,P=0.031;t=4.730,P=0.009).ConclusionsChrysin or chrysin-like flavonoids ameliorate intraocular inflammatory symptoms in EAU mice by inhibiting the activity of STAT signal pathway.
5.Progress in molecularly targeted anti-tumor drugs derived from natural products or their derivatives
Acta Pharmaceutica Sinica 2020;55(11):2491-2500
Conventional chemotherapy drugs, molecularly targeted drugs, and immune checkpoint inhibitors are the major constituents of anti-tumor drugs in clinical settings at present. Molecularly targeted drugs specifically target the key proteins, genes, or signal transduction pathways in tumor cells which are essential for initiation and development of tumor, resulting in selective activity to induce cell death or growth inhibition. Molecularly targeted drugs have emerged as the mainstream in the research and development of anti-tumor drugs due to its high selectivity and low toxicity. Natural products refer to the chemical constituents or metabolites originated animals, plants, or microorganisms, which have been recognized as one of the important sources of drug discovery with abundant resources and diversified structures. At present, a number of molecularly targeted anti-tumor drugs derived from natural products or their derivatives have been approved for cancer therapy or in clinical trials. This review will summarize the molecularly targeted anti-tumor drugs derived from natural products or their derivatives according to their different cellular targets, and also outline the molecular mechanism, progress, and perspectives of these drugs.
