1.Infection Management in Animal Laboratory of General Hospital

OBJECTIVE To ensure all relative environmental indications achieving stabilization in both animal test region and breeding area in the barrier system by enhancing infection management in animal laboratory.METHODS Standard nosocomial infection management in the animal laboratory was taken strictly and adapted standard management and attendance to the clean-air system;constructed and perfected all the regulations and managements of the barrier facility,taken a series of sterilization measurement in SPF area,and performed standard operating procedure of the airflow,human-being flow,material flow and animal flow strictly.RESULTS Since the barrier system in animal laboratory had been built and used for more than two years,every monitor index including temperature,humidity,ventilation time,airflow velocity,pressure grade,falling bacteria,noises et al,had achieved national standards.CONCLUSIONS Powerful and effective management can guarantee environment keeping cleaning and animal quality standard in the barrier system.
2.Clinical Utilization and Blood Concentration Monitoring of Vancomycin in Our Hospital:Analysis of 41 Cases

OBJECTIVE:To evaluate clinical utilization and blood concentration monitoring of vancomycin in our hospital. METHODS: By retrospective survey method, the clinical information (primary diseases, bacteriological culture, blood concentration of vancomycin, clinical efficacy, renal function) of 41 inpatients who had been treated with vancomycin and their blood concentrations from Nov. 2005 to Dec. 2008 were analyzed statistically. RESULTS: The total response rate of vancomycin was 65.0%. The peak concentration of 62.7% cases were below 25 mg?L-1 while the valley value of 55.2% cases were below 5 mg?L-1. The incidence of renal dysfunction after treatment was 12.2%. CONCLUSION: The consumption and blood concentrations of vancomycin are close to low level in our hospital. It should be paid attention to the blood concentration monitoring of vancomycin to achieve an ideal individual administration according to clinical practice.
3. Construction of eukaryotic expression vectors of two NF2 mutants and their expression in rat schwannoma cell line

Objective: To construct the eukaryotic expression vectors of two NF2 mutants (neurofibromatosis II) and to study their expression in rat schwannoma cell line (RT4). Methods: Site-directed mutagenesis was performed to induce the mutation of the codons for the residue Lys42 in NF2 into Pro in pcDNA3. 1-NF2ΔLys42Pro, and convert the codons for Phe 47 into that of Leu. RT4 cells were transiently transfected with the 3 kinds of plasmids containing the mutations and wide-type NF2 via lipofectin separately, then the expression levels of NF2 mRNA and protein were determined by RT-PCR and Western blotting in 3 groups. The cell proliferation was determined by the MTT after transfection. Results: DNA sequence analysis demonstrated the two-step mutagenesis was successful and the two plasmids of pcDNA3.1ΔLys42Pro and pcDNA3.1-NF2ΔPhe47Leu were obtained, both of the transfectants could produce merlin protein and mRNA. The two mutants had a significantly lower inhibitory rate for RT4 cells compared with wide-type NF2 (P < 0.05). Conclusion: The recombinant plasmids pcDNA3. 1-NF2ΔLys42Pro and pcDNA3. 1-NF2ΔPhe47Leu have been successfully constructed and they can be efficiently expressed in RT4 cells.

Many newly discovered interleukins have been implicated to play an important role in the pathogenesis of the allergic diseases, for example, human interleukin (IL)-17, a T-cell derived cytokine; interleukin (IL)-18, an interferon (IFN)-gamma-inducing cytokine; interleukin (IL)-23, produced by activated dendritic cells, and interleukin (IL)-25, a recently described T helper 2 (Th2) cell-derived cytokine. Understanding their characteristic and roles in diseases may help us to know better the mechanism of the allergic diseases and develop the strategy for treating the disease. [
5.Effect of leupeptin, TLCK and lactoferrin on tryptase release from human colon mast cells

AIM: To investigate the ability of leupeptin, TLCK and lactoferrin on tryptase release from human colon mast cells. METHODS: Human mast cells were dispersed from colon tissue with collagenase and hyaluronidase, and were challenged with stimulus for 15 min at 37 ℃. Tryptase concentrations were measured with a sandwich ELISA procedure. RESULTS: Leupeptin, TLCK and lactoferrin were not able to provoke significant tryptase release from human colon mast cells. But they were able to inhibit anti-IgE and calcium ionophore (CI)-induced tryptase release in a concentration-dependent manner. Preincubation of inhibitors of leupeptin and TLCK with cells for 20 min before challenging with anti-IgE failed to enhance their inhibitory actions. The extent of inhibition by leupeptin was increased when colon mast cells were preincubated for 20 min before CI being added. However, the same treatment failed to improve the action of TLCK. CONCLUSION: We found for the first time that leupeptin, TLCK and lactoferrin were able to inhibit anti-IgE and calcium ionophore-induced tryptase release from human colon mast cells, which may indicate a potential of a novel therapy for the treatment of inflammatory bowel disease or other mast cells-related diseases.
6.Clinical research on application of adjustable sutures in glaucoma filtering operation

AIM:To discuss the application effect of adjustable sutures in glaucoma filtering operation after trabecular resection.
METHODS: Seventy-eight cases ( 101 eyes ) suffered from glaucoma were randomly divided into two groups, observation group and control group. Thirty-nine cases ( 51 eyes ) in the observation group underwent trabeculectomy with adjustable sutures, the control group (39 cases, 50 eyes) only adopted trabeculectomy.RESULTS: Compared Preoperative IOP in two groups, the difference was not statistically significant (P>0. 05). After 6mo, IOP were decreased compared with preoperative in two groups, and that in observation group was lower than control group, the difference was statistically significant ( P < 0. 05 ). Six months after operation, there were 1 eye with shallow anterior chamber Ⅰ and 2 eyes with non- functional bleb in observation group, and the complication rate was 5. 9%. While there were of 6 eyes with shallow anterior chamber, in which 4 eyes at gradeⅠ, each one at gradeII and Ⅲ. Non-functional blebs in 5 eyes and a scleral flap adhesion complications rate in the control group was 24. 0%, significantly higher than that in the observation group, the difference was statistically significant ( P<0.01).
CONCLUSION: The adjustable sutures combined with trabeculectomy for glaucoma can significantly reduce the postoperative complications. The curative effect is exact and clinically applicable.
8.Clinical therapeutic effect of rehabilitative therapy on dysphagia after cerebral stroke

Objective:To explore the clinical therapeutic effect of comprehensive rehabilitative treatment on dyspha-gia after acute cerebral stroke .Methods :A total of 78 patients with dysphagia after acute cerebral stroke ,who visi-ted to our hospital from Jun 2011 to Jun 2012 ,were selected ,randomly and equally divided into Vitalstim electrical stimulus plus swallowing training group (rehabilitation treatment group ) and traditional swallowing training group (routine training group) .Patients received Kubota's water drinking test before and after treatment .Therapeutic effects were observed ,compared and statistically analyzed .Results:Total effective rate of rehabilitation treatment group was significantly higher than that of routine training group (92.3% vs .74.3% ,χ2 = 4.191 , P= 0.041 );mean declining amplitude of swallowing function score in rehabilitation group was significantly higher than that of routine training group [ (2.0 ± 0.5) scores vs .(1.1 ± 0.6) scores , t=5.284 , P=0.032] .Conclusion:Comprehen-sive rehabilitation training can significantly improve dysphagia caused by cerebral stroke .

Epigenetics has become a hot topic in biomedical research in last a few years.Epigenetics refers to as epigenome,and it is interpreted as epigenetic changes (DNA methylation,histone modifications and non-coding RNAs such as microRNAs) in the regulation of genes expression on chromatin-based pathways rather than a single gene.Epigenetic factors such as DNA methylation,histone modification and miRNA serve as an interface for environmental stimuli and interacts each other to regulate gene expression and control phenotypes of cell.All of those epigenetic factors are essential in the keeping homeostasis of human body and contribute to many biologic conditions.In addition,the application of epigenetic drugs has been come into clinical trials especially in the treatment of cancers,therefore the understanding the mechanism of epigenetics in the development of human diseases and the therapeutic potential of epigenetic regulators in the treatment of human disease will bring us into the epigenetics era in biomedical research.
