2.Effects of halofuginone on maturation of dendritic cells and immune tolerance during homograft heart transplantation of rats
Chinese Journal of Biochemical Pharmaceutics 2014;37(7):40-42,45
Objective To explore the role of halofuginone in immune regulation, observe the effects of halofuginone on maturation of dendritic cells and immune tolerance during homograft heart transplantation of rats,in order to provide new ways and means to treat transplantation tolerance and to explore its possible mechanism. Methods Rat bone marrow derived dendritic cells were isolated and the effects of halofuginone in LPS-treated DCs were investigated by flow cytometry;heart transplantation model of rats were established and the effect of halofuginone stimulated dendritic cells in the immune tolerance by pathological analysis and mixed lymphocyte reaction were observed. Results Flow cytometry results show that the expression of cell surface CD80 CD86,OX62 antigens were not high in control group, their expression increased significantly when stimulated by LPS for 24h,and decreased when stimulated by both Changshan ketone and LPS for 24h,and even lower than their expression levels in blank group, which showed that Changshan ketone has the ability to inhibit the maturation of dendritic cells induced by LPS.In rat heart transplantation model, dendritic cells stimulated with Changshan were injected into recipient rats in vivo, and receptor of rat transplantation rejection decreased significantly, which showed that dendritic cells stimulated with Changshan ketone may be used as a new method for cell therapy in treatment of transplant rejection. Conclusion This study systematically observes that halofuginone can inhibit the maturation of rat bone marrow-derived dendritic cells and lymphocyte proliferation for the first time in vivo and in vitro.

Chinese Journal of Bases and Clinics in General Surgery 2003;0(05):-
Objective To investigate the relationship between the development of pancreatic cancer and inflammation, and the therapy strategy. Methods Related articles were reviewed. Results The pathogenesis of inflammation in pancreatic cancer development involves cytokines, NF ?B, COX 2, PPAR ?, DNA damage, gene changes, etc. Based on these mechanisms some medications are under developing. Conclusion Accumulative effects of pancreatic inflammation may lead to DNA changes, and even pancreatic cancer development. Medications aimed at suppressing pancreatic inflammation may help with prevention and treatment of pancreatic cancer.
5.Aplastic anemia and clonal evolution:germline and somatic genetics
Journal of Leukemia & Lymphoma 2017;26(3):138-141
Clonal progression to myelodysplastic syndromes (MDS) or acute myeloid leukemia (AML) remains a dreaded complication for a subset of patients with bone marrow failure (BMF). Recognizing risk factors for the development of MDS or AML would inform individualized treatment decisions and identify patients who may benefit from early or upfront hematopoietic stem cell transplantation. Now that next-generation DNA sequencing is available in the clinical laboratory, research has focused on the implications of germline and somatic mutations for diagnosing and monitoring patients with BMF. Most germline genetic BMF disorders are characterized by a high propensity to develop MDS or AML. Recent studies of somatic variants in marrow failure revealed a high frequency of clonal hematopoiesis with the acquisition of mutations in genes associated with MDS or AML. The evaluation and implications of germline and somatic mutations for the development of clonal disorders in patients with BMF and challenges of clinical genetic testing will be summarized in this paper based on the reports from the 58th American Society of Hematology (ASH) Annual Meeting.

Journal of Xi'an Jiaotong University(Medical Sciences) 1982;0(01):-
Objective To study the significance of serum uric acid in predicting coronary heart disease(CHD) risk. Methods Uric acid and its relationship with coronary heart disease(CHD) was analyzed in 215 cases who underwent coronary angiogaphy(CHD was present in 164 patients and absent in 51 as normal control group). Results Uric acid was significantly higher in patients with CHD than that in the normal control group [(316.67?97.35)?mol/L vs.(286.16?98.33)?mol/L,P
7.Later course accelerated hyperfractionated radiotherapy combine chemotherapy for stage Ⅲ and Ⅳ a nasopharyngeal carcinoma
Cancer Research and Clinic 2006;0(11):-
Objective To evaluated the treatment results of late course accelerated fractionation radiotherapy for Stage Ⅲ and Ⅳa nasopharyngeal carcinoma. Methods 100 patients with stage Ⅲ and Ⅳa nasopharyngeal carcinoma were randomized into two groups. The late course accelerated hyperfractionated radiotherapy combine chemotherapy group(LCAF+CT group) received the induction chemotherapy for two cycles, followed by conventional fractionation radiotherapy to a dose 40 Gy, than changed into accelerated hyperfractionated radiotherapy to a total dose of 70 Gy, and use two cycles adjuvant chemotherapy, The conventional fractionation radiotherapy combine chemotherapy group(CF+CT group) received the chemotherapy that it similar to the LCAF+CT group, and use the conventional fractionation radiotherapy to a total dose of 70 Gy. Results The 3- and 5-year overall survival rate were 70.0% and 62.0% respectively in LCAF+CT group and 58.0 % and 46.0 % respectively in CF+CT group, had the significant difference (P 0.05). There were the cranial nerves complications in two patients of LCAF+CT group. Conclusions The results from this study show that the 3- and 5-year overall survival rate, the 3- and 5-year disease free survival rate and the 3- and 5-year local-regional free survival rate in LCAF+CT group was significant better than in CF+CT group, but the radiation-induced damage to the cranial nerves was improved in LCAF+CT group.
8.The effects of rat brain ischemia on the expression of Acetyl-cholinesterase and Caspase-3 of the immune system
Chinese Journal of Immunology 1985;0(05):-
Objective:To explore the effects of brain ischemia on the immune system and the expression of Acetyl-cholinesterase(AChE) and Caspase-3 after neuro-immunological injury. Methods:The serum IL-4,IL-10,TNF-?,IL-2 and IL-12 concentrations and AChE level in brain, thymus and spleen of the rat brain ischemia at different time spots were detected by ELISA.The active fragments of Caspase-3 P17 and P32 in brain, thymus and spleen were tested by Western blot. The expression of AChE in nerve cells was examined immunohistochemistry. The relationship between amount of AChE and concentrations of the cytokines or NK activity as well as cell apoptosis was analyzed. Results:The concentration of AChE in brain increased gradually after ischemia, the level reached peak after 12 hours, and then decreased afterward. The AChE level in thymus and spleen increased after 1 h in accordance with the ischemic brain. The NK activity, serum TNF-?,IL-2 and IL-12 levels also increased at 0.5 h,1 h and 2 h after ischemia, but the trend reversed after 3 hours.Serum IL-4 and IL-10 levels did not change significantly at 0.5 h,1 h and 2 h after ischemia, but serum IL-10 raised after 3 hours to 48 hours.Serum IL-4 level slightly increased only appearing at ischemia 12 h point. The ratio of P17/(P17+P32) became higher with ischemic time. Conclusion:Sustained rising of AChE level due to brain ischemia induces the apoptosis of cells in the brain, thymus and spleen via activation caspase-3,leading to Th1/Th2 imbalance and immune dysfunctions.

10.Inhibitory Effect of Hydroxycamptothecin on C6 Gliomas Cells
China Pharmacy 2005;0(13):-
OBJECTIVE: To investigate the inhibitory effect of hydroxycamptothecin (HCPT) on C6 gliomas cells in rats. METHODS: The cytotoxicity of HCPT at different concentration(1~0.031 25 mg?mL-1) on C6 in vitro was measured by MTT with conventional therapy for C6 gliomas cells as control. Meanwhile, the antitumor effect of O-HCPT(open ring form) and that of C-HCPT(closed ring form) were compared. RESULTS: At a high concentration,HCPT showed an inhibition ratio of as high as(69.43?4.5)% on C6 gliomas cells in concentration-dependent manner, showing no significant differences as compared with control. The antitumor effect of O-HCPT was lower than that of C-HCPT,and their IC50 were 3.626 mg?mL-1 and 0.135 mg?mL-1,respectively. CONCLUSION: HCPT had remarkable inhibitory effect on C6,and the antitumor effect of C-HCPT is potent than that of O-HCPT.
