1.Mechanism Study of Kang’ai Fangyi Tablets in Inhibiting Angiogenesis of 4T1 Breast Cancer

Objective To study the angiogenesis-inhibitory mechanism of Kang’ai Fangyi Tablets ( KFT) , a Chinese compound recipe with the action of inhibiting cancer metastasis, for 4T1 breast cancer. Methods BALB/c mice were divided into blank control group, model group, Cytoxan ( CTX, 0.04 g·kg-1·d-1) group, and KFT ( 5.2 g·kg-1·d-1) group. Mice model of 4T1 breast cancer was established. Except that the blank control group and model group were given the saline, the mice in the medication groups were given the corresponding medicine. After medication for 4 weeks, the mice were executed, and then we calculated the mass of tumor, the inhibition rate of tumor mass, and the number of lung metastatic nodules. The number of microvessel and expression of vascular endothelial growth factor (VEGF) were measured by immunohistochemical method. Results Compared with the model group, mice tumor mass was decreased ( P<0.05) , the number of pulmonary metastatic nodules was reduced ( P<0.05) , and the number of tumor microvessel and VEGF expression in the isolated tumor mass showed the decreasing trend in KFT group. Conclusion KFT can inhibit 4T1 breast cancer growth and metastasis by reducing the tumor microvessel formation and by affecting the expression of VEGF.
2.Influence of initial periodontal therapy on the presence of oral Helicobacter pylori and the relation of genotype of Helicobacter pylori in oral cavity and with that in stomach

Objective: To investigate whether initial periodontal therapy may affect the presence of oral Helicobacter pylori(Hp), and to compare the genotype of Helicobacter pylori in oral cavity and that in stomach. Methods: 56 patients with gastric Hp and periodontitis were enrolled in this study. PCR was carried out to identify the presence of Hp in the samples before and after initial periodontal therapy(group 1) or medicine therapy(group 2). DNA sequence of PCR products of 5 patients and 1 Hp infected patient's relative was compared and analyzed. 3.Establishment and Noninvasive Echocardiographic Assessment of Mouse Myocardial Infarction Model

Objective To establish a standardized mouse myocardial infarction model,and non-invasively evaluate its cardiac function by using the echocardiography.Methods A total of 20 wild male mice were enrolled into this study.The left anterior descending coronary was ligated through the left fourth intercostal space after intubation.The echocardiographic assessment of myocardial infarction and heart function was performed 3 days preoperatively and 2 weeks postoperatively.The shortening fraction(FS),end systolic dimension(ESD),and end diastolic dimension(EDD) was recorded and analyzed.Results The acute and one-week mortality rates were 20%(4/20) and 40%(8/20),respectively.The echocardiography showed that the myocardial infarction model was successfully established in 10 mice.The heart function of the mice was markedly impaired.The FS significantly decreased from(50.23?8.36)% to(25.41?0.97)%(t=16.18,P
4.The synthesis and preliminary bioactivity of isoflavone derivatives

Objective To synthesize soybean isoflavones, and their 7-alkaline analogues and to evaluate their uterotrophic activities and anti-uterotrophic activities preliminarily. Methods The target molecules were synthesized by multi-step from resorcinol and p-substituted phenylacetic acid as the starting material. Their uterotrophic activities and anti-uterotrophic activities were evaluated by female mouse at the concentration of 1.1?10 -2 ?mol/ml and 1.85?10 -3 ?mol/ml. Results Twenty-two compounds were synthesized, among which 4 intermediates and 12 isoflavones are new compounds. Conclusion All the target molecules except for 6a show weak uterotrophic activities and high anti-uterotrophic activities.

6.Effect of initial periodontal therapy on the eradication of gastric Helicobacter pylori

Objective: To investigate whether periodontitis might adversely affect the outcome of eradication of gastric Helicobacter pylori(Hp) . Methods: Ninety one patients with gastric Helicobacter pylori and periodontitis were enrolled. The patients were treated by medicine therapy, meanwhile 33 of them were treated by initial periodontal therapy. After treatment, the effect of initial periodontal therapy on the eradication of gastric( Hp ) was examined with 13 C urea breath test. The correlation between the gastric eradication rate and periodontitis was analyzed. Objective:To investigate involvement of the Ras/Erk signal pathway in activation of human ??T cells stimulated by Mycobacterium tuberculosis low molecular peptide antigen(Mtb-Ag).Methods:PBMC were isolated from peripheral blood of healthy subjects and simulated in vitro with Mtb-Ag,CD3mAb or PMA and inomysin(IM).CD69 expression of total T cells and ??T cells were measured by flow cytometry at different hours after stimulation, and the number of expanded ??T cells were counted after 10 days of culture. PD98059,a specific inhibitor for Erk pathway was used to treat PBMC for 30 min before stimulation.Results:The proportions of CD69 expressing cells in total T cells and ??T cells were same as 99% at 6 h after stimulation of PBMC with PMA +IM and similar about 55% at 24 h after stimulation with CD3 mAb,whereas those at 24 h stimulation of PBMC with Mtb-Ag were 75.2% and 16.0%,respectively.CD69 expression and proliferation of ??T cells activated by Mtb-Ag were markedly inhibited by pretreatment of PBMC with PD98059.Conclusion:Mtb-Ag is a specific stimulator for activation of ??T cells, and Ras/Erk signal transduction pathway involves in the activation of ??T cells.
8.THREE-DIMENSIONAL MASTICATORY MUSCLE FORCE VECTORS DETERMINED BY CT AND MRI FUSION IMAGE TECHNOLOGY

In order to measure and analyze three dimensional masticatory muscle force vectors in craniofacial region, the multiresolution method based on wavelet pyramid was used for the fusion of cranio jaw facial CT and MRI image, then these images were visualized. The results showed that the area of physiological cross section (PCS) and the maximum muscle force were different during functioning in the masticatory muscles system. The various masticatory muscle forces or various sides in the same muscle were also different in the three dimensional directions. The results suggested that determination of three dimensional masticatory muscle vectors is feasible by CT and MRI fusion image technology in craniofacial region, and can provide significant results for the basic research in the masticatory muscle system.
9.Biological characteristics of bone marrow derived mesenchymal stem cells from patients with coronary artery disease

Objective To establish a method for the isolation and culture of bone marrow derived mesenchymal stem cells (MSCs) from patients with coronary artery disease and investigated its biological characteristics. Methods Bone marrow was extracted from patients' sternum when they were operated for coronary artery bypass graft. MSCs were separated from bone marrow by density gradient centrifugation in Percoll solution and seeded in culture medium supplemented with 10% fetal bovine serum. The morphology and growth characteristic of MSCs were observed in primary and passage generation culture with inverted microscope. The cell phenotype and cell cycle of MSCs were analyzed by flow cytometry. The growth curves were compared between different passages and ages. Results Most adherent MSCs showed spindle morphology whatever passages they were. The MSCs could be expanded for 15 passages. The proliferative potential of MSCs over the fifth passage or from old patients decreased gradually. The MSCs in primary and passage were positive for CD29 and CD44, but negative for CD34 and CD45. Analysis for cell cycle showed most MSCs in G_0/G_1 phase. Conclusion The method of isolation and culture of sternum bone marrow derived MSCs from patients with coronary artery disease is feasible, the MSCs have self-renewal capacity and might be ideal for cellular cardiomyoplasty.
