1.Mechanism Study of Kang’ai Fangyi Tablets in Inhibiting Angiogenesis of 4T1 Breast Cancer

Objective To study the angiogenesis-inhibitory mechanism of Kang’ai Fangyi Tablets ( KFT) , a Chinese compound recipe with the action of inhibiting cancer metastasis, for 4T1 breast cancer. Methods BALB/c mice were divided into blank control group, model group, Cytoxan ( CTX, 0.04 g·kg-1·d-1) group, and KFT ( 5.2 g·kg-1·d-1) group. Mice model of 4T1 breast cancer was established. Except that the blank control group and model group were given the saline, the mice in the medication groups were given the corresponding medicine. After medication for 4 weeks, the mice were executed, and then we calculated the mass of tumor, the inhibition rate of tumor mass, and the number of lung metastatic nodules. The number of microvessel and expression of vascular endothelial growth factor (VEGF) were measured by immunohistochemical method. Results Compared with the model group, mice tumor mass was decreased ( P<0.05) , the number of pulmonary metastatic nodules was reduced ( P<0.05) , and the number of tumor microvessel and VEGF expression in the isolated tumor mass showed the decreasing trend in KFT group. Conclusion KFT can inhibit 4T1 breast cancer growth and metastasis by reducing the tumor microvessel formation and by affecting the expression of VEGF.

4.Establishment and Noninvasive Echocardiographic Assessment of Mouse Myocardial Infarction Model

Objective To establish a standardized mouse myocardial infarction model,and non-invasively evaluate its cardiac function by using the echocardiography.Methods A total of 20 wild male mice were enrolled into this study.The left anterior descending coronary was ligated through the left fourth intercostal space after intubation.The echocardiographic assessment of myocardial infarction and heart function was performed 3 days preoperatively and 2 weeks postoperatively.The shortening fraction(FS),end systolic dimension(ESD),and end diastolic dimension(EDD) was recorded and analyzed.Results The acute and one-week mortality rates were 20%(4/20) and 40%(8/20),respectively.The echocardiography showed that the myocardial infarction model was successfully established in 10 mice.The heart function of the mice was markedly impaired.The FS significantly decreased from(50.23?8.36)% to(25.41?0.97)%(t=16.18,P
5.The synthesis and preliminary bioactivity of isoflavone derivatives

Objective To synthesize soybean isoflavones, and their 7-alkaline analogues and to evaluate their uterotrophic activities and anti-uterotrophic activities preliminarily. Methods The target molecules were synthesized by multi-step from resorcinol and p-substituted phenylacetic acid as the starting material. Their uterotrophic activities and anti-uterotrophic activities were evaluated by female mouse at the concentration of 1.1?10 -2 ?mol/ml and 1.85?10 -3 ?mol/ml. Results Twenty-two compounds were synthesized, among which 4 intermediates and 12 isoflavones are new compounds. Conclusion All the target molecules except for 6a show weak uterotrophic activities and high anti-uterotrophic activities.
6.Biological characteristics of bone marrow derived mesenchymal stem cells from patients with coronary artery disease

Objective To establish a method for the isolation and culture of bone marrow derived mesenchymal stem cells (MSCs) from patients with coronary artery disease and investigated its biological characteristics. Methods Bone marrow was extracted from patients' sternum when they were operated for coronary artery bypass graft. MSCs were separated from bone marrow by density gradient centrifugation in Percoll solution and seeded in culture medium supplemented with 10% fetal bovine serum. The morphology and growth characteristic of MSCs were observed in primary and passage generation culture with inverted microscope. The cell phenotype and cell cycle of MSCs were analyzed by flow cytometry. The growth curves were compared between different passages and ages. Results Most adherent MSCs showed spindle morphology whatever passages they were. The MSCs could be expanded for 15 passages. The proliferative potential of MSCs over the fifth passage or from old patients decreased gradually. The MSCs in primary and passage were positive for CD29 and CD44, but negative for CD34 and CD45. Analysis for cell cycle showed most MSCs in G_0/G_1 phase. Conclusion The method of isolation and culture of sternum bone marrow derived MSCs from patients with coronary artery disease is feasible, the MSCs have self-renewal capacity and might be ideal for cellular cardiomyoplasty.
7.Clinical application and comparison of rapid and accurate identification of mycobacterium by gene chip microarray and smear acid-fast staining

Objective To compare the clinical effect of application of gene chip microscopy technique for rapid identification of Mycobacterium and classic smear acid-fast staining,and to assess the advantages and disadvantages of the wo methods.Methods From 201 1 to 2014,gene chip microarray and smear acid fast staining were used to identify the mycobacterium tuberculosis in speci-mens suspicious of the infection from all the general hospitals of Shenzhen city.Chi-square test was used to compare the positive rates of the two methods.Results A total of 2 481 specimens were collected from clinic.With smear acid-fast staining technique, the positive specimens of 1 93 cases werefound and the positive rate was 7.8%.Meanwhile,31 7 positive samples were detected by the technology of gene chip microarray,and the positive rate was 12.8%.The positive rate of Gene chip microarray technology was higher than that of the smear acid fast staining,and there was significant difference between them (P < 0.05 ).The 31 7 positive samples identified by Gene chip microarray,included 263 cases of Mycobacterium tuberculosis,27 cases of Mycobacterium absces-sus,18 cases of Mycobacterium intracellulare,3 cases of Mycobacterium gastric uLcer,3 cases of Mycobacterium avium,1 case of Mycobacterium Gordonae,1 case of Mycobacterium marinum and 1 case of Mycobacterium Kansas.Conclusion The gene chip mi-croarray technology is fast,accurate,and its positive rate is higher than that of smear acid-fast staining technique.Classification and identification of Mycobacterium is very helpful for clinical individualized treatment of anti mycobacterium infection.

10.Advances in Study on Hedgehog Signaling Pathway in Pathogenesis of Barrett’s Esophagus

Hedgehog( Hh)signaling pathway is evolutionarily conserved in vertebrates,its excessive activation is associated with abnormal cell differentiation, over proliferation, apoptosis resistance and promotion of invasion and metastasis of tumor cells. Hh signaling pathway involves in the formation and maintenance of esophageal columnar epithelium in embryonic stage,however,undetectable or barely expressed in matured esophageal squamous epithelium. Studies have shown that esophageal Hh signaling pathway can be activated by gastric acid and bile salts. Aberrant activation of Hh signaling pathway can cause the gradual transition of squamous epithelium to columnar epithelium and intestinal-type epithelium,ultimately induces the occurrence of Barrett’s esophagus( BE). Therefore,targeted inhibiting the Hh signaling pathway may be a new strategy for the treatment of BE. This article reviewed the advances in study on Hh signaling pathway in the pathogenesis of BE.
