Objective:To construct the plasmid containing short hairpin RNA(shRNA)of survivin and modify human bladder transitional cell carcinoma(BTCC)T24 cell line with it to evaluate its inhibition on tumor growth and proliferation and further discuss its functions in T24 cells.Methods:Survivin specific oligonucleotides were designed and synthesized.These oligonucleotides were annealed to form double strand DNA fragment and this fragment was inserted into plasimid PTZU6+1 to generate the plasimid pshRNA-survivin2.Then T24 cells were inoculated into the right-back legs of BALB/c nude mice to establish bladder cancer model.Through plasimid PTZU6+1,T24/survivin-siRNA expressing survivin was successfully constructed to study the effects of survivin-siRNA on BTCC growth and proliferation.Microscope was used to observe the morphological changes.Histological examination was under HE staining.Survivin was detected by SABC to evaluate the expression of survivin after the treatment of survivin-siRNA through PTZU6+1.Results:The expression of survivin in transfected T24 cells in vivo was markedly depressed as compared with control groups(P

4.The differentiation and treatment of acute leukemia after chemotherapy from Jueyin

After chemotherapy, the healthy qi of acute leukemia (AL) patients was severly demaged, remnant toxin with internal amassment, qi and blood deficiency, blood stasis, vertigo with syndrome of stagnation and blockade of phlegm-damp. The clinical symptoms with cold-heat intermingled together which are complicated and changable were quite similar to those of Jueyin disease. Therefore, this paper tried to explore the differentiation and treatment of AL after chemotherapy from Jueyin. It was concluded that the treatment of AL after chemotherapy should be treated with cold and heat method, attacking and supplementing together, with nourishing the liver as the main part. At the same time, it was importance to protect healthy qi. Combined with the basic pathogenesis of AL, it was proposed that Danggui-Sini Decoction was suitable for the treatment of AL after chemotherapy. All in all, the paper explored the differentiation and treatment of AL after chemotherapy from Jueyin in order to use more Chinese medicine therapy in the field of AL treatment.

Objective To investigate the effects of regional infusion of hypothermic normal saline contained captopril on free radicals level and myeloperoxidase(MPO) activity in rabbits during spinal cord ischemia-reperfusion(I/R).Methods The 80 rabbits were randomly divided into four equal groups,group A(sham operated,no I-R injury,n=8),group B(control,only I/R,n=24),group C(I/R + intraaortic hypothermic normal saline,n=24),and group D(I/R + N.S plus 4mg/kg captopril,n=24).The model of spinal cord ischemia in the rabbit was induced with the aorta clamped.Hypothermic solution was infused into the isolated aortic segments.The content of spinal cord MDA and the activities of superoxide dismutase(SOD),MPO were measured at 8h,24h and 72 h after the reperfusion respectively.Results The level of spinal cord MDA increased,the activity of SOD decreased and the activity of MPO increased markedly after the reperfusion.Compared with group B,the MDA content decreased significantly at the three observation time points and the MPO activity decreased significantly in groups C and D(all P
7.Diagnoses of Subacute Thyroiditis by Fine Needle Aspiration Cytolgy

Objective To explore the diagnostic value of subacute thyroiditis by fine needle aspiration cytology(FNAC).Method 3665 patients with thyroid disease were detected by FNAC method in 8 years(from Feb 1990 to Dec 1998).Results 353 cases(9 63%) of subacute thyroidits were definitely diagnosed by FNAC with vital staining.Conclusions As compared with other clinical examination including CT,ultrasonic B and radioimmunological assay.FNAC test is all the more simple,safe and valuable diagnostic method for subacute thyroiditis.
8.Current research of role of Hox genes in pathogenesis of equinus deformity

Congenital clubfoot(CCF), which is also known as equines deformity, is a common congenital malformation that affect children′s life quality. However, its cause is still to be elucidated. Currently, polygenetic and environmental fac?tors are both believed to play important roles in CCF pathogenesis. Several genes including HOX, PITX1, NAT2, P63, DTDS and COL9A were shown to contribute to congenital clubfoot, but which is the most critical gene remains unclear. Several re?ports have revealed that Hox genes are closely related to the cause of CCF. Hox genes are regulators of body morphogenesis, and its mutation result in limbs and trunk deformity in human. Here, we systematically reviewed the latest literature that stud?ied the role of Hox genes in pathogenesis of Congenital clubfoot, with the prospect of laying a foundation for its future clinic treatment.
9.Analysis and prevention of pulmonary embolism after urologic surgery

Objective To analyze the clinical features of pulmonary embolism(PE) after urologic surgery in order to provide the theory base for preventing pulmonary embolism Methods CNKI database was used to look for our subjects who were with PE after urologic surgery from January 2005 to December 2011.Ten cases were selected.Three related patients simultaneously in Xuancheng Central Hospital were also recruited as our subjects.The information of diagnosis and treatment information and clinical features were collected.Results Among the 13 patients,3 were women and 10 were men with confirmed PE.Their age ranged from 48 to 79 years old and the average was 61.Among the 13 patients,2 conducted routine surgery,and the other 11 conducted mini-invasion operation.The 13 patients underwent PE from 2 to 18 days after urologic surgery and 7 patients (53.85%) died of PE.Conclusion No specificity of clinical feature for PE was found to contribute to cure rate.Early diagnosis and prevention of PE are important,and anticoagulant treatment and immediate thrombolytic are critical.
10.Fabrication of Flower-Shaped Pt-Au-graphene Nanostructures and Their Application in Electrochemical Detection of Glucose

The flower-shaped Pt-Au-graphene oxide ( GO ) nanostructures were synthesized by one-step electrochemical reduction method. The electrocatalytic activities of the nanostructures toward glucose oxidation were investigated by cyclic voltammetry. The results indicated that the nanocomposites exhibited a higher electrocatalytic activity than the Pt-Au nanoparticles. A glucose nonenzyme sensor could be easily developed with the use of the flower-shaped Pt-Au-GO nanostructure. The sensor exhibited low detection limit (4. 0 μmol/L), a wide and useful linear range (1. 0-25. 0 mmol/L), and high sensitivity (26. 3 μA cm-2 ( mmol)-1 ) . In addition, the sensor exhibited excellent reproducibility, stability and selectivity for the determination of glucose, and it could be used to analyze real samples.
