1.Effect of Fosinopril on oxidative stress of pancreatic tissue in diabetic rats

Objective To study the oxidative stress in diabetic rat pancreas,and the changes of islet function after intervention of Fosinopril.Methods Wistar rats were randomly divided into control group,diabetes group and Fosinopril treated group.The diabetes was induced by feeding the rats from diabetes group and Fosinopril group with high fat diet for 3 weeks and then once intraperitoneal injection of STZ(35 mg/kg),while additionally Fosinopril(5 mg/kg) was given to the diabetic rats in Fosinopril treated group.Blood glucose,body weight,nitrotyrosine levels in pancreatic tissues were detected and the histopathologic changes of pancreatic tissues were observed.Results Nitrotyrosine content was significantly higher in diabetes group than that in control group(P
2.Clinical effect of percutaneous transluminal angioplasty for the treatment of diabetic lower extremity arterial disease

Objective To observe the clinical effect of percutaneous endovascular angioplasty ( PTA) for the treatment of diabetic lower extremity arterial disease( LEAD) , so as to provide a reference for the selection of the best treatment for diabetic lower extremity arterial dis-ease(LEAD). Methods In this parallel study, totally 60 patients were equally divided into the control group and the intervention group. The control group were treated by the conventional method, and the intervention group were treated by percutaneous endovascular angioplasty (PTA). The changes of ankle brachial index (ABI),average blood flow velocity of dorsalis pedis,pulsation index(PI) and the maximum walking distance were observated in 12 weeks and 24 weeks after the treatment. Results Compared with the control group, the ankle brachi-al index ( ABI) , average blood flow velocity of dorsalis pedis, pulsation index ( PI) and the maximum walking distance in the intervention group were significantly improved 12 weeks and 24 weeks after treatment, and the difference between the two groups was statistically signifi-cant (P<0. 05). And compared with thses data before treatment in the intra-group comparison, there were statistically significant difference (P<0. 05). However, the changes in 24 weeks and 12 weeks after treatment in the same group showed no significant difference (P>0. 05). Conclusion Percutaneous endovascular angioplasty ( PTA) is better than traditional drug therapy in the treatment of diabetic lower extremity arterial disease( LEAD) . It is a feasible clinical treatment of lower extremity arterial disease( LEAD) .
3.A study on trends of the incidence of gestational diabetes mellitus and its relevant factors in Chongqing city from 2005 to 2009

By using the Medical Case Inquiry System and registries of infant's birth,the number of women with gestational diabetes mellitus(GDM)and the total number of women delivering in the First and Second Affiliated Hospital of Chongqing Medical University,Chongqing Xinqiao Hospital,and Chongqing Health Center For Women and Children from 2005 to 2009 were obtained.The data of 540 pregnant women with GDM were further analyzed.From 2005 to 2009,the incidence of GDM increased from 2.29% to 3.81 %(P<0.05).The diagnosis of GDM was made earlier by 28.0 weeks in 2009 compared with 31.3 weeks in 2005(P<0.05).Body mass index during GDM diagnosis manifested a growing trend(25.6-28.1 kg/m2),but no significance(P>0.05).From 2005 to 2009,the other related factors,including the average maternal age,the constituent ratio of women with advanced maternal age,pregnancy history,delivery history,and family history of diabetes showed insignificant changes(P>0.05).
4.Research on the value of data middle platform in the management of data assets for medical consumable

Objective:To establish a data middle platform for data assets of medical consumables,to provide open data sharing services for the management and application development of medical consumables,and to reduce the risk of data silos.Methods:Based on the data resource management model of data middle platform,data aggregation and big data development technology were adopted to realize data loading,integration and quality control between multi-business systems.A hierarchical data management system was established through data warehousing and object modeling methods.A data assets management system was built based on the data governance process to provide functions of data standards and data lineage maintenance,metadata management,data permission control,and data lifecycle management for medical consumables data resources.A microservice architecture was adopted to provide functions such as data service generation,registration,discovery and subscription for medical consumables management.The front-end data application development workload of business data and application operation time before and after the application of data middle platform service of medical consumables were compared in the South Campus of the Fifth Medical Center of PLA General Hospital from March to May 2020.Results:The medical consumables data middle platform realized the connection between the front-end application interface and the back-end data service,and provided synchronized data services for the front-end pages.The workload of front-end data application development for data middle platform data services decreased from 64 person-months of traditional data warehouses to 6 person-months,and the operation time for testing applications reduced from 430 minutes to 16 minutes.Conclusion:The data middle platform management mode is helpful to improve the information management capabilities and efficiency of medical consumables management,and provides reference for the digital transformation of medical consumable management.
5.Preliminary investigation of the mechanism of action of metformin in the treatment of Hashimoto′s thyroiditis

Objective:
To preliminarily investigate the mechanism of action of metformin in the treatment of Hashimoto′s thyroiditis.
Methods:
Forty-five healthy SD female rats were randomly divided into control group, model group and metformin-treated group, and the rats were simulated with a model of Hashimoto′s thyroiditis by immuno-injection of high iodine water combined with thyroglobulin and Freund′s adjuvant. After successful modeling, the control group and the model group were perfused with saline, and the metformin group was administered with 300 mg/kg metformin by gavage for 4 weeks, and the rats in each group were executed and sampled. Serum thyroid function indexes(TSH, T3, T4, TGAb, TPOAb) and thyroid tissue inflammatory factors(TNF-α, IL-10, IL-6,IL-12) levels were measured by ELISA; thyroid tissue pathological changes were observed by HE staining; thyroid tissue apoptosis was detected by TUNEL fluorescence staining; thyroid tissue expression of Fas, Fas-L was detected by immunohistochemistry; the expression of Fas, Fas-L, FADD and cleaved caspase-3 in thyroid tissue was determined by Western blot; Fas, Fas-L and FADD mRNA expression in thyroid tissue was detected by qRT-PCR.
Results:
Compared with the control group, serum levels of TSH, TGAb, TPOAb, positive cell ratio, levels of TNF-α, IL-6 and expression of Fas, Fas-L, FADD and cleaved caspase-3 in thyroid tissue were significantly higher in the model group, and serum FT3 and FT4 levels and IL-10 and IL-12 levels in thyroid tissue significantly decreased(P<0.01). Compared with the model group, serum levels of TSH, TGAb, TPOAb, positive cell ratio, TNF-αand IL-6 contents and expression of Fas, Fas-L, FADD and cleaved caspase-3 in thyroid tissue were significantly lower in the metformin group, and serum FT3 and FT4 levels and IL-10 and IL-12 contents in thyroid tissue were significantly higher(P<0.05).
Conclusion
Metformin inhibits the progression of Hashimoto′s thyroiditis, and its mechanism of action may be related to the inhibition of the Fas/Fas-L-mediated apoptotic pathway.
