The effect of selenium on the synthesis of nucleic acids in human am-nion cells and monkey kidey cells was studied by the techniques of cell culture and radioisotope assay. The cells were cultured in a medium containing varied dosages of sodium selenite for 24 hours at 37℃ in a CO2 incubator (5% CO2), then 3H-labeled precursors of DNA and RNA were added. The synthetic activity of DNA and RNA was determined by assaying the incorporation amount of radioisotope. The result suggested that selenium can promote intracellular DNA and RNA synthesis and may be essential to cell growth.
3.Human erythropoietin-modified human amniotic mesenchymal stem cellsvia subarachnoid transplantation promote neurologic recovery from brain injury

BACKGROUND:Studies have shown that as a regulator of bone marrow functionerythropoietinis a glycoprotein that controls the development of the central nervous system and has neurotrophic and neuroprotective potential. Therefore, transplantation of human amniotic mesenchymal stem cels geneticaly modified by human erythropoietin is a new choice for brain injury treatment.
OBJECTIVE:To observe the effect of transplantation of human amniotic mesenchymal stem cels geneticaly modified byhuman erythropoietin on the functional recovery from brain injury in rats.
METHODS:Eukaryotic expression plasmid pcDNA3.1 carrying erythropoietin was successfuly constructed and transferred into amniotic mesenchymal stem cels culturedin vitro. Expression of erythropoietin was detected using western blot assay before and after transfection. Rat models of middle cerebral arterial occlusion was made and given transplantation of transfected amniotic mesenchymal stem celsviathe tail vein (transfection group). Additionaly, model and simple cel transplantation groups were set in a comparative study.
RESULTS AND CONCLUSION:Findings from western blot detection showed that transfected cels could express human erythropoietin. Compared with the other groups, modified neurologic severity scores, growth-associated protein 43 and aquaporin 9 at mRNA and protein levels were al decreased significantly in the transfection group. Furthermore, the number of cels positive for CM-Dil was highest in the transfectiongroup, folowed by simple cel transplantation group, and lowest in the model group (alP<0.05). Overal findings from this study show that human erythropoietin-modified human amniotic mesenchymal stem cel transplantation promotes neurologic recoveryfrom brain injury through eliciting a reduction in growth-associated protein 43 and aquaporin 9 at mRNA and protein levels as wel as inhibiting cel apoptosis.

5.Expression of miR-483-5p in epithelial ovarian cancer and its effects on cisplatin resistance in epithelial ovarian cancer cells

Background and purpose:Although cisplatin-based chemotherapies are used as the first-line treatment for ovarian cancers, the majority of patients eventually progress with platinum-resistant disease. miR-483-5p is overexpressed in lung cancer. However, the research on miR-483-5p in epithelial ovarian cancer (EOC) is still unclear. This study aimed to investigate the expression of miR-483-5p in EOC and its effects on cisplatin resistance in EOC cells.Methods:This study analyzed the expression of the miR-483-5p by real-time lfuorescent quantitative polymerase chain reaction (RTFQ-PCR) in EOC tissues, normal ovarian tissues, and EOC cells. The role of miR-483-5p in EOC was evaluatedin vitro by lentivirus-mediated knockdown of miR-483-5p or overexpression of miR-483-5p in EOC cell lines. Drug sensitivity assay was carried out by CCK-8 kit.Results:miR-483-5p was upregulated in EOC tissues as compared with normal tissues (P<0.01). Furthermore, miR-483-5p expression in advanced stage (Ⅲ–Ⅳ) EOC was significantly higher than that in early stage (Ⅰ–Ⅱ) EOC (P<0.05). Interestingly, miR-483-5p expression was higher in cisplatin-resistant A2780/CP cells than other cells. Increased miR-483-5p expression caused EOC cell resistance to cisplatin and downregulated the expression of p21 and Bcl-2, whereas reduced miR-483-5p expression induced its sensitivity and upregulated the expression of p21 and Bcl-2.Conclusion:The results suggest that miR-483-5p is highly expressed in EOC and contributes to cisplatin resistance. Thus, miR-483-5p is a potential therapeutic target for ovarian cancer.
6.Role of whey protein hydrolyzed formula as an adjunctive therapy for treating patients with infantile eczema

Objective To study the clinical value of extensively whey protein hydrolyzed formula and partially whey protein hydrolyzed formula as an adjunctive therapy in treating infantile eczema.Methods Totally 59 bottle-feeding babies with infantile eczema were divided into three groups depending on different formula feeding:extensively hydrolyzed formula feeding group (eHF group,n =18),partially hydrolyzed formula feeding group (pHF group,n =22),and conventional cow's milk formula feeding group (CMF group,n =19).Meanwhile,all patients received the same drug treatment.The effective rate and remission rate were evaluated using the SCORing Atopic Dermatitis (SCORAD) index.Results After 2 weeks of treatment,the effective rate was 38.9% and the remission rate was 50.0% in the eHF group,which were significantly different from those in the CMF group (5.3% and 31.6%) (x2 =12.225,P =0.002).The effective rate was 9.1% and the remission rate was 40.9% in the pHF group,showing no significant difference when compared with the CMF group (x2 =0.761,P =0.683).After 8 weeks of treatment,the effective rate was 55.6% and the remission rate was 38.9% in the eHF group,comparing to 15.8% and 47.4% in the CMF group,the difference was statistically significant (x2 =8.498,P =0.014).The effective rate was 31.8 % and the remission rate was 59.1% in the pHF group,a trend towards higher than that of the CMF group,but the difference was not statistically significant (x2 =4.912,P =0.086).Before (F =0.773,P =0.466) and after (F =1.313,P =0.277) the treatment,the growth and development data in different groups showed no significance differences.Conclusion Both the extensively and partially whey protein hydrolyzed formula play an adjunctive role in treating infantile eczema,although the treatment effectiveness of the extensively hydrolyzed formula appears earlier than that of the partially hydrolyzed formula.

8.Association between Airborne Pollen Concentrations and Human Respiratory Diseases

Objective To investigate the relationship between airborne pollen concentrations and the number of medical consultations in out-patient and emergency for respiratory diseases in Haidian district,Beijing. Methods To collect the data of airborne pollen concentrations,atmospheric concentrations of total suspended particles(TSP),the related meteorological factors (average temperature,maximum temperature,minimum temperature,average air pressure,precipitation and relative humidity) and the data of patients who were diagnosed with respiratory diseases including acute pharyngitis,acute laryngitis,acute bronchitis,anaphylactic rhinitis and bronchial asthma from database on medical insurance,from April to September in 2000 to 2002 in Haidian district,Beijing. The partial correlation coefficients between airborne pollen concentrations and the number of medical consultations for respiratory diseases in spring (from April 1st to June 16th),and in summer-autumn (from July 15th to September 30th) were calculated after controlling TSP concentrations and the six related meteorological factors. Results The airborne pollen concentrations in these three years showed two obvious peaks in spring and summer-autumn,and each of the five respiratory diseases has its specific change character. The number of medical consultations for anaphylactic rhinitis was significantly associated with the airborne pollen concentrations in summer-autumn (the partial correlation coefficient was 0.802,P
9.Association between Ambient PM10 and Daily Hospital Visits:a Time-Series Analysis

Objective To estimate quantitatively the impact of the ambient PM10 on the hospital outpatients for cardiovascular diseases of local residents. Methods Time serial analysis using generalized addictive model (GAM) was applied. After controlling for those confounding factors such as long-term trend, weekly pattern and meteorological factors, considering lag effect and the influence of other air pollutants, excess relative risks (ER) of daily hospital visits associated with increasing PM10 level were estimated by fitting a Poisson regression model. Results A 10 ?g/m3 increase in PM10 levels was associated with an ER of 0.380% (95%CI: 0.326% ~0.433%) for hospital visits for cardiovascular diseases. Lag effect of 4 days with an ER of 1.166% (95%CI:1.121%~1.212%) were observed. The ER value increased when CO, NO2, SO2 concentrations were introduced. Conclusion The ambient PM10 concentration is positively associated with daily hospital visits for cardiovascular diseases in Beijing.
10.Shenmai Zhusheye vs vitamine C injection in adjuvant treatment of dilated cardiomyopathy

AIM: To observe the therupeatic effects of Shemai Zhusheye vs vitamine C injection in treating dilated cardiomyopathy (DCM). METHODS: Fifty-two patients with DCM were divided into 2 groups, 32 patients in treatment group (M 20, F 12; age 46 a± s 13 a), 20 patients in control group (M 11, F 9; age 47 a±14 a). Besides the patients in two groups were treated with conventional therapy, the patients in treatment group were given Shenmai Zhusheye 30 mL and 5% glucose injection 100 mL, iv, gtt, qd for 2 wk. The patients in control group were given vitamine C 5.0 and 5% glucose injection 100 mL, iv, gtt, qd for 2 wk. RESULTS: Treatment group total effective rate was 84%. Control group total effective rate was 65% (P>0.05). CONCLUSION: Shenmai zhusheye has better effect in treating DCM. Symptom improving rate of Shenmai Zhusheye has better than vitamin C.
