1.The expression of cIAP and Caspase-7 in endometrial adenocarcinoma and its relationship with the prognosis

Objective To study the expression of cIAP and Caspase‐7 in endometrial adenocarcinoma tissue and its relation with prognosis .Methods Fifty‐five cases tissues of endometrial adenocarcinoma patients were detected by immunohistochemistry assay from the First Affiliated Hospital of Henan University of Science and Technology from 2009 to 2010 .Histological classifica‐tion ,pathological stage ,lymph node metastasis ,age and other parameters were collected ,the survival and overall survival of the tumor were followed up ,immunohistochemistry was used to observe the expression of cIAP and Caspase‐7 in paraffin sections of pathological specimens ,and the relationship between them and the parameters were analyzed .Results The expression level of cIAP and Caspase‐7 was related to the histological classification ,pathological stage and lymph node metastasis of endometrial carcinoma (P<0 .05) ,but it was not related with the age ,muscular layer infiltration(P>0 .05) .There was significant difference between the positive expression and the negative expression of cIAP and Caspase‐7(P<0 .05) .Conclusion The expression level of cIAP and Caspase‐7 in patients with endometrial carcinoma is correlated with tumor grade and overall survival ,or can be used as a prognostic indicator for endometrial cancer .

OBJECTIVE To investigate the infectious condition of urogential mycoplasma and chlamydia among STD high risk population.METHODS Urethral specimens from 932 cases of patients with NGU were collected and detected for Chlamydia trachomatis(Ct) and mycoplasma.RESULTS The infectious rates of mycoplasma and chlamydia among 932 STD high-risk population were 34.3%.from then rates were in men 31.9%,and were in women were 38.1%.Among these patients,positive CT,positive UU and positive Mh were found in 77 patients(8.3%),122 patients(13.1%) and 70 patients(7.5%),respectively,And the mixed infection by Mycoplasma hominis(Mh) and Ureaplama urealyticum(Uu),Uu and Ct,Ct and Mh,Ct and Uu with Mh were 7 cases(0.8%),14 cases(1.5%),3 cases(0.3%) and 1 case(0.1%),respectively.Compared with the infectious rates(8.7%) in healthy people,there was statistically significant difference(P
4.Genitourinary Tract Infections and Antibiotics Resistance of Mycoplasma

OBJECTIVE To investigate the infection status and drug resistance of Ureaplasma urealyticum(Uu) and Mycoplasma hominis(Mh) in local region.METHODS A total of 2042 genitourinary samples of NGU patients were detected for the presense of mycoplasma with culture and drug sensibility.RESULTS From them 881(43.1%) were infected with mycoplasma,including Uu infection(650cases,31.8%),Mh infection(74 cases,3.6%) and mixed infection of Uu plus Mh(157 cases,7.6%);significant differences of positive cultivation rate were found between the male and female and in different age groups.Among 18 antibiotics,the lowest resistance drugs were josamycin,doxycycline and clarithromycin(3.6%,4.1%,and 4.2%).The highest resistance drugs were ofloxacin,fleroxacin and thiamphenicol(38.2%,18.3% and 14.6%).CONCLUSIONS The infection of mycoplasma in local region is increasing.Mycoplasma has increasing resistance to the common antibiotic.The detection of drugs resistance of mycoplasma has an improtant significance in guiding clinical use of the drugs.
5.Drug Susceptibility and Resistant Mechanisms of 3170 Strains of Clinical Isolates

OBJECTIVE To investigate the drug susceptibility of clinical isolates in local region for using antibiotic reasonably. METHODS Totally 3 170 strains of clinical isolates were identified by API and Microscan and tested for drug resistance against antimicrobial agents by K-B method. WHONET5.4 was applied for analysis. RESULTS The commonly encountered bacteria were Pseudomonas aeruginosa (24.3%), Acinetobacter baumannii (10.9%), Klebsiella pneumoniae (10.4%), Escherichia coli (8.9%), and Staphylococcus aureus (SA,8.0%). In Gram-negative isolates, the resistance rate to meropenem was 19.7%, and to piperacillin-tazobactam was 26.5%. The incidences of E.coli and K. pneumoniae isolates producing extended spectrum beta-lactamases (ESBLs) were 49.1% and 33.5%, respectively. In Gram-positive isolates, the susceptibility rate to vancomycin and teicoplanin both was 100.0%. The oxacillin resistant rates of SA and coagulase negative Staphylococcus (CNS) were 54.2.0% and 82.3%. CONCLUSIONS The production ratio of ESBLs and oxacillin resistance of bacteria in local region are high. It is important to promote the rational use of antimicrobial agents and take effective contaminant methods to reduce resistant rates of bacteria and dissemination of multi-resistant bacteria.
6.The extraction of sperm DNA from mixed stain using DNase-Ⅰ purification combined with alkaline lysis method

Objective To establish a method of sperm DNA extraction in mixed stain by using DNase-Ⅰ purificationcombined with alkaline lysis method in forensic science.Methods 79 mixed stain samples of criminal cases were collected.Sperm DNA was extracted using the purification of DNase-Ⅰ binding alkaline lysis method.16 STR loci were genotyped with fluorescent multiplex amplification system.The typing results were compared with that of extracted using two-step differential extraction procedure.Results Of all 79 mixed stain samples,64 samples were genotyped successfully by using DNase-Ⅰ purification combined with alkaline lysis method while 57 samples were genotyped successfully with two-step differential extraction procedure.There was significant difference between two methods(P=0.039).The purification of DNase-Ⅰ binding alkaline lysis method had a higher success rate and lower cost than that of two-step differential extraction procedure.Conclusion Purification of DNase-Ⅰ binding alkaline lysis method can increase the typing success rate of the mixed stain samples.The method is simple,rapid and easy to be automated,and suitable for forensic identification test.

8.Effect of angiotensin Ⅱ and aldosterone in patients with congenital heart disease by preoperative nursing interventions at high altitude area

Objective To study the effect of angiotensin Ⅱ and aldosterone (ALD) in patients with congenital heart disease by preoperative nursing interventions at high altitude area. Methods 86 patients with congenital heart disease were divided into the experimental group and the control group on average by random. The experimental group was treated with preoperative nursing interventions and routine nursing before operation and the control group was treated with routine nursing only. Angiotensin Ⅱ and aldosterone were detected before anesthesia, 30min after operation and after pulling out of endotracheal tube respectively. Results The concentration of Angiotensin Ⅱ and aldosterone was significantly lower in the experimental group than the control group. Conclusions The risk of open surgical operation for congenital heart disease at high altitude can be reduced by preoperative nursing interventions.
9.Noninvasive prenatal diagnosis ofβ-thalassemia by enriching cell-free fetal DNA in materal plasma

AIM:To establish a kind of simple and efficient method for cell-free fetal DNA ( cff-DNA) enrich-ment and to investigate its range of applications and the advantages and disadvantages.METHODS:(1) The single nucleo-tide polymorphisms( SNPs) , which linked to paternalβ-thalassemia mutations, were screened.We analyzed the contact be-tween the SNPs inβ-thalassemia gene ( HBB gene) and haploid type by the Haploview software, and then selected these close SNPs which have higher heterozygosity with the HBB gene.(2) We selected 4 cases of different β-thalassemia muta-tions with their husband, and then we used TT-FAST-COLD-PCR to enrich the IVS-II-654 mutations in maternal plasma.If the IVS-II-654 mutation was not detected, we detected the SNP which linked to the IVS-II-654 mutation.Similarly, we used TT-FULL-COLD-PCR to enrich the CD41-42 mutations in the maternal plasma.At the same time, we used the conventional PCR to enrich CD41-42 mutation and IVS-II-654 mutation in the maternal plasma.RESULTS:(1) Nine cases of the SNP ( rs7480526) linked to the mutation at IVS-II-654 in HBB gene, and 11 cases of the SNP ( rs10768683) linked to the muta-tion at CD41-42 in HBB gene were detected.( 2 ) We detected 1 case who inherited the paternal β-thalassemia mutation (IVS-II-654).We did not directly detect patermal IVS-II-654 mutation in maternal plasma, but detected the SNP linked to the IVS-II-654 mutation in the other case and had 100%detection, and 2 cases inherited the paternal β-thalassemia muta-tions (CD41-42) in the maternal plasma by TT-FULL-COLD-PCR and had 100%detection.However, we detected nothing by conventional PCR.CONCLUSION:TT-COLD-PCR is applicable to enrich cell-free fetal DNA in maternal plasma and is a method in the field of noninvasive prenatal diagnosis.
10.Interventional effects of Tongxinluo combined with Atorvastatin and Aspirin(ATS) on the angiogenesis of vasa vasorum in the early stage of atherosclerosis

Aim To observe the effect of a treatment proposal named “Golden Triangle” ( Atorvastatin, Tongxinluo,and Aspirin) on the vasa vasorum angio-genesis of early atherosclerosis lesions in rabbits carotid artery. Method Seventy-two healthy New Zealand rabbits with half males and half females were divided into 6 groups randomly ( n =12 ):control group, model group, Tongxinluo group ( TXL ) , atorvastatin group ( ATO ) , aspirin group ( ASP ) , golden triangle group ( ATS) . The control group was fed with common feed-stuff, and all the other groups′ right carotid arteries were equipped with the silicone tube,and were then fed with fatty feedstuff. The Tongxinluo group, the Atorvas-tatin group and the Aspirin group were given suspen-sion of Tongxinluo supermicro powder(0. 3 g·kg-1 · d-1 ) , Atorvastatin ( 2. 5 mg · kg-1 · d-1 ) and Aspirin (12 mg·kg-1·d-1),the golden triangle group were given suspension of Tongxinluo supermicropowder (0. 3g·kg-1 ·d-1),atorvastatin(2. 5 mg·kg-1 · d-1 ) and Aspirin ( 12 mg · kg-1 · d-1 ) . All the groups were fed with medicine for 4 weeks. Tissue slice of carotid artery was stained with HE and observed un-der light microscope. The change of blood liquid was detected by biochemical assay. Immunohistochemical staining was used to detect the protein expression of CD34 around the carotid artery adventitia. Color micro-sphere method was used to detect the blood flow vol-ume change of the cartoid artery microvascular. VEFG, VEGFR-2 gene and protein expression in the cartoid artery were detected by Real-time PCR and Western Blot. Result Compared with the control group,the content of VEGF, VEGFR-2 gene and pro-tein expression and the microvascular blood flow vol-ume of cartoid artery microvascular in the model were significantly increased ( P <0. 01 ) . But those in each drug group were lighter than those in the model group (P<0. 01,P <0. 05). In the ATS group,the content of VEGF, VEGFR-2 gene and protein expression and the microvascular blood flow volume of cartoid artery microvascular were lower than those in the TXL, ATO and ASP group ( P <0. 01 , P <0. 05 ) . Compared with the ASP group,the content of VEGFR-2 protein expres-sion was significantly decreased(P<0. 01)in the TXL and ATO group. VEGF,VEGFR-2 gene and protein ex-pression in different subgroups showed no significant difference( P >0. 05 ) . The content of CD34 was de-creased in TongxinLuo group,atorvastatin group,aspirin group and ATS group. Conclusion The ATS project can reduce the expression of VEGF,VEGFR-2, inhibit the vasa vasorum angiogenesis and decrease proinflam-matory substances in the tunica media and intima of vascular wall. It plays an important role in intervening in the process of AS.
