1.Significance of pancreatic stone protein in pancreatic injury and sepsis

Pancreatic stone protein (PSP) is the product of exocrine pancreas mainly synthesized and secreted by the pancreas acinar cells, a small amount secreted by gastrointestinal cells. PSP is one of stress protein family members secreted in the pancreas. PSP increased in acute and chronic pancreatitis, diabetes, inlfammatory bowel disease and gastrointestinal tumors. In recent years, some studies showed that PSP has potential clinical value in pancreatic injury and sepsis. In this article, we reviewed the progress of pancreatic stone protein in pancreatic injury and sepsis.
2.Influence of rosuvastatin on inflammatory factors and immune function in chronic obstructive pulmonary disease with pulmonary heart disease

Objective To investigate the influence of rosuvastatin on inflammatory factors and immune func-tion in chronic obstructive pulmonary disease(COPD)with pulmonary heart disease.Methods 80 COPD patients with pulmonary heart disease were randomly divided into two groups,40 cases in each group.The control group was treated with conventional medicine comprehensive treatment,the observation group was given rosuvastatin based on the control group.The inflammatory cytokines (TNF -α,IL -1 and hs -CRP)changes,immunoglobulin IgM,IgG and IgA changes situation,and heart,lung function changes related indicators were compared between the two groups before and after the treatment.Results After treatment,the TNF -α,IL -1 and hs CRP levels of the observation group were significantly lower than those before treatment(t =145.5,35.35,57.5,all P <0.05)and those of the control group(t =160.75,11.45,32.99,all P <0.05).After treatment,the IgM,IgG and IgA levels of the observa-tion group were significantly lower than those before treatment (t =4.89 of 18.29,3.42,all P <0.05)and those of the control group (t =2.55 9.67,2.342,all P <0.05 ).After treatment,the FEV1 ,FEV1 /FVC and LVEF of the observation group were significantly higher than those before treatment(t =40.44 33.64,33.79,all P <0.05),while the PAP level was significantly lower than that before treatment (t =24.12,P <0.05),and the FEV1 ,FEV1 /FVC and LVEF of the observation group were significantly higher than those of the control group after treatment(t =16.30, 15.82,18.72,all P <0.05),while the PAP level was significantly lower than that of the control group after treatment (t =13.43,P <0.05).Conclusion Combined rosuvastatin therapy for COPD patients with pulmonary heart disease can effectively reduce body inflammatory response,improve immune function and myocardial contractility,so it can promote functional recovery of heart and lung in patients.
3.Effects of pumpkin polysaccharide on Fas、Fas-L、Bcl-2 and Bax expression in the pancreas of diabetic rats

Aim To investigate the effects of pumpkin polysaccharide(PP) on Fas、Fas-L、Bcl-2 and Bax expression in the pancreas of diabetic rats.Methods Rats were injected intraperitoneally(ip) with streptozocin(STZ) 60 mg?kg-1 to induce diabetic model.The diabetic rats were given PP 150 mg?kg-1,300 mg?kg-1,600 mg?kg-1.The blood sample were taken after three weeks.Blood glucose was measured by glucose oxidase method;The expression of the Fas and Fas-L have been determined by using immunohistochemistry(S-P method);The expression of Bax mRNA and Bcl-2 mRNA were checked up by Reverse transcription PCR(RT-PCR).Results PP could decrease the level of blood glucose in streptozotocin-induced diabetic rats;After three weeks,The expressions of the Fas and FasL in ? cell of streptozotocin-induced diabetic rats were obviously lower;The expression of Bax mRNA was obviously down-regulated,the expression of Bcl-2 mRNA was obviously up-regulated,and the ratio of Bcl-2/Bax was advanced after the diabetic rats were given PP.Conclusions PP could decrease the level of blood glucose of the diabetic rats.The mechanism was related to decreasing the expressions of the Fas and FasL and regulating the expressions of Bcl-2 and Bax in the pancreas of diabetic rats.
4.Correlation of ER with Bcl-2 in cervical cancer and pre-cancerous lesion

Objective:This study aimed to investigate the relationship between estrogen receptor (ER) and Bcl-2 in cervical can-cer and precancerous lesions. Methods:ER and Bcl-2 were evaluated by immunohistochemistry in 20 non-tumor tissues, 30 low-grade cervical intraepithelial neoplasias, 70 high-grade cervical intraepithelial neoplasias, and 80 cervical cancers. Results:The positive rate of ER was 75%in non-tumor tissues;this rate decreased with disease severity (χ2=24.266, P<0.01). The overexpression of ER protein was remarkably lower in cervical intraepithelial neoplasias (CIN) Ⅱ-Ⅲ and carcinomas than in non-tumorous cervices. Differences were not significant between CINⅠand non-tumorous cervices (χ2=0.751, P=0.386). Furthermore, ER expression was not correlated with histological type and tumor grade. The positive rate of Bcl-2 was 75.38%in squamous cell carcinoma. This rate increased with dis-ease severity (χ2=27.715, P<0.01). The Bcl-2 protein overexpression was remarkably higher in CINⅠ, CINⅡ-Ⅲ, and carcinomas than in non-tumorous cervices. Bcl-2 expression was also not correlated with histological type (P=0.500);by comparison, Bcl-2 expression was correlated with tumor pathological grade (χ2=4.862, P=0.027). The co-expression of ER and Bcl-2 was 38.57%and 15.38%in CINⅡ-Ⅲand squamous cell carcinoma, respectively;the differences between these two values were significant (χ2=9.108, P=0.003). A cor-relation between ER and Bcl-2 was observed in CINⅡ-Ⅲ(r=0.506, P<0.01);no correlation was observed between other groups. Con-clusion: ER expression was inversely correlated with Bcl-2 in cervical cancer progression. The co-expression of these markers may have an important function in the course of high-grade cervical intraepithelial neoplasia.
5.Construction of a new teaching quality control model though collective teaching demonstration in English

In the research and prac-tice of teaching quality monitoring mode,we explored and constructed a new monitoring mode-collec-tive teaching demonstration in English,which laid a solid foundation for improving bilingual teaching quality and realizing the sustainable development.
6.Problem-based learning (PBL)in clinical teaching of pediatric interns

Objective To investigate problem-based learning(PBL)in clinical teaching of pediatric interns.Methods To compare and analyze the grades of pediatric interns’case writing before and after PBL.Results There was significant difference between the two groups and the grades of interns by PBL were notably higher.Conclusion PBL can improve the interns’ability of case writing,clinical thoughts,operation ability,and enhance their enthusiasm in learning.
7.Correlation Analysis on Life Quality and TCM Constitutions of Elderly Patients with Hypertension in a Community in Shanghai

Objective To explore the relationship between quality life and TCM constitutions of elderly patients with hypertension in a community in Shanghai; To provide the basis for the suitable and feasible community elderly hypertension management method. Methods Through multistage stratified sampling method, 808 elderly patients with hypertension in the community were investigated for their life quality and TCM constitutions on the basis of inform consent and voluntary participation. The life quality and TCM constitution characteristics as well as the relationship were analyzed by applying the statistic analysis software. Results Among the investigated elderly patients with hypertension in the community, 97.28% cases’ overall life quality was above the middle level and 91.46% cases’ self-evaluated health and quality of life scores were above the middle level. 54.70% cases were gentleness type, and the biased constitution was dominated by qi deficiency, yang asthenia, yin asthenia and phlegmatic hygrosis. The relationship between life quality and TCM constitutions may be existent. If TCM constitutions tended to be gentleness type, the physical score and the psychological score would be higher, which leaded to better quality of life. Conclusion TCM constitution characteristics are associated with quality life of elderly hypertension patients in the community. Intervention targeted on the results of TCM constitution identification would be better to prevent and control hypertension and improve health level and quality of life.
8.Changes in pancreatic stone protein/regenerating protein of children with sepsis and pancreatic damage and its significance

Objective To determine the levels of blood pancreatic stone protein/ regenerating protein(PSP/reg)and to explore the value of PSP/ reg in assessing severity and predicting prognosis of sepsis with or without panc-reatic damage. Methods In this prospective study,the clinical data and blood samples to measure PSP/ reg levels in 1,3 and 7 days after admission in the Pediatric Intensive Care Unit,Hunan Children's Hospital,from the children with sepsis from September 2013 to September 2014 were collected. According to levels of pancreatic trypsin(amylase or li-pase),the children were divided into normal group,mildly elevated group(1 - 3 times),and severely elevated group (more than 3 times);based on the condition of sepsis,the children were divided into sepsis group and severe sepsis group. Chi - square test,nonparametric rank sum test,Spearman correlation and area under the receiver operating cha-racteristic curve(AUC),Kaplan - Meier survival curves were used for statistical analysis. Results (1)A total of 226 children with sepsis were included in this study,among them pancreatic trypsin was normal in 159 cases(70. 4% ), pancreatic trypsin mildly elevated in 44 cases( 19. 4% ),and pancreatic trypsin severely elevated in 23 cases (10. 2% );125 cases(55. 3% )were sepsis,and 101 cases(44. 7% ,of which 29 cases were sepsis shock)were severe sepsis.(2)The correlation between PSP/ reg and serum amylase was maximum,while PSP/ reg had some correlation with serum lipase,but there was no correlation with C - peptide,insulin,glucose.(3)When pancreatic trypsin was normal or mildly elevated,severe sepsis group had higher levels of PSP/ reg than that of sepsis group(Z = - 7. 627,- 2. 965,P =0. 000,0. 003);while pancreatic trypsin was severe elevated,there was no significant difference in PSP/ reg levels be-tween sepsis group and severe sepsis group(Z = - 0. 134,P = 0. 894).(4)The AUC of PSP/ reg and procalcitonin to assess severity of sepsis were greater than 0. 7 in the children whose pancreatic trypsin was normal or mildly elevated, and there was no significant difference between them(P ﹥ 0. 05);while the AUC of C - reactive protein and WBC were less than 0. 7.(5)PSP/ reg levels gradually increased in non - survival children in 3 and 7 days,while survival children gradually declined. The AUC of PSP/ reg to predict prognosis was 0. 775,and 60 μg/ L was viewed as the critical point, sensitivity 89% ,specificity 67% . Children with PSP/ reg levels ≤60 μg/ L had higher rate of survival than children with ﹥ 60 μg/ L(χ2 = 36. 874,P = 0. 000). Conclusions PSP/ reg is closely associated with pancreatic exocrine func-tion,but PSP/ reg still can assess severity and predict prognosis in children with sepsis and pancreatic damage.
9.Effects of AMP-activated protein kinase on HMGB1 release from PC12 cells after oxygen-glucose deprivation and reoxygenation and its mediated inflammatory response in BV2 cells

Objective To investigate the effects of adenosine monophosphate-activated protein kinase (AMPK) on high-mobility group box 1 (HMGB1) release from PC12 cells after oxygen-glucose deprivation and reoxygenation (OGD/R) and its mediated inflammatory response in BV2 cells.Methods PC12 and BV2 cells were cultured,respectively.The PC12 cells were used to induce a model of oxygen glucose deprivation for 12 h and reoxygenation for 24 h.After giving 5-aminoimidazole-4-carboxamide (AICAR) 5,50 and 100 μmol/L as well as Compound C 0.1,1 and 10 μmol/L activation or inhibition of AMPK phosphorylation,respectively,methyl thiazolyl tetrazolium (MTT) was used to detect the PC12 cell activity.Enzyme-linked immunosorbent assay was used to detect the HMGB1 release level in the PC12 cell culture media.After OGD/R in each group,the PC12 culture media were acted on normal cultured BV2 cells for 24 h respectively.Westem blotting and Enzyme-linked immunosorbent assay were used to detect the NFκB inhibitory protein (inhibitor of NFκB,IκB) phosphorylation level and TNF-α release level in BV2 cells,respectively.Results After OGD/R,the PC12 cell activity was decreased significantly (68.84%±6.60% vs.100.04% ± 8.82%;P < 0.01);the AMPK phosphorylation level was increased significantly (1.95 ±0.39 vs.1.00 ±0.20;P<0.05),and the extracellular HMGB1 release was increased significantly (287.66 ± 26.42 pg/μl vs.53.05 ± 9.11 pg/μl;P < 0.01).Compared with the OGD/R group,AICAR 100 μmol/L significantly increased the survival rate of PC12 cell after OGD/R (78.6% ± 3.75% vs.68.84% ± 6.60%;P < 0.05),promoted AMPK phosphorylation (3.32 ± 0.66 vs.1.95 ± 0.39;P < 0.01),and reduce the release of extracellular HMGB1 (164.06 ± 12.77 pg/μl vs.287.66 ± 26.42 pg/μl;P <0.01).In contrast,Compound C 10 μmol/L significantly reduced the cell survival rate of PC12 (40.44% ±3.79% vs.68.84% ±6.60%;P <0.01),inhibited AMPK phosphorylation (1.07 ± 0.21 vs.1.95 ± 0.39;P<0.05),and increased the release of HMGB1 (337.97 ± 18.9 pg/μlvs.287.66 ± 26.42 pg/μl;P<0.01).The conditioned medium from the AICAR 100 μmol/L group significantly inhibited IκB phosphorylation (1.68 ±0.51 vs.3.09 ± 0.10;P < 0.05) and reduced the release of TNF-α (669.53 ±38.58 pg/μlvs.841.76 ± 45.82 pg/μl;P< 0.05) in BV2 cells.The conditioned medium from the compound C 10 μmol/L group significantly promoted IκB phosphorylation (4.98 ± 1.24 vs.3.09 ± 0.10;P <0.01) and increased the release of TNF-α (1 035.32 ± 128.06 pg/μl vs.841.76 ± 45.82 pg/μl;P <0.05) in BV2 cells.Conclusions Promoting AMPK phosphorylation activation may reduce the release of HMGB1 from PC12 cells after OGD/R,and inhibit its mediated NF-κB inflammatory pathway and reduce the release of TNF-αin BV2 cells,and thus reducing neuroinflammatory injury.On the contrary,inhibiting AMPK phosphorylation may promote the release of HMGB1 from PC12 cells after OGD/R and aggravate its mediated inflammatory reaction in BV2 cells.
10.Regulation of saw palmetto extract on immune function in GL261 glioma

Objective:To investigate the effect of saw palmetto extract ( SR ) on GL261 glioma in rats and immune system.Methods:The 40 rats were divided into 4 groups randomly,one was the control group without tumor( the blank group) ( n=10) ,the other 30 rats were given subcutaneous inoculation of tumor cells and then divided into 3 groups:the tumor-bearing group( n=10) ,low dose SR group( n=10) ,and high dose SR group( n=10).After 1 week fed,the rats of SR groups were given the saw palmetto extract,low dose group 50 mg/kg once a day every other day and 300 mg/kg of high dose group every other day.The tumor-bearing groups received the same dose of distilled water.After 4 weeks fed, we measured the tumor weight and the inhibition rate was calculated.The tumor cell apoptosis was detected by TUNEL staining.The activity of splenic macrophages was detected by macrophage phagocytosis of chicken erythrocytes and CD4+lymphocytes by flow cytometry.Results:①The SR groups compared with tumor group, the tumor weight was significantly reduced(F=62.678,P=0.000).The tumor inhibition rate was significantly higher in high dose group.②The apoptosis of tumor cells in tumor-bearing group was significantly less than SR groups and the apoptosis was significantly increased after treatment with SR ,especially in high dose SR group(F=1.287,P=0.000).③Compared with the blank group,the macrophage activity decreased remarkablely.SR enhanced the macrophage activity obviously, positively related to doses of SR ( F=141.205,P=0.000;F=126.903,P=0.000).④In the tumor-bearing group,the CD4+lymphocyte count decreased significantly.After SR treatment,the CD4+lymphocyte increased and were positively related to the dose of SR.Conclusion: Saw palmetto extract can strengthen the immune system.
