1.The effect of celecoxib on Wnt/β-catenin signaling pathway and matrix metabolism of chondrocytes in osteoarthritis

Objective To study effect of celecoxib on Wnt/β-catenin signaling pathway and matrix metabolism of chondrocytes in osteoarthritis (OA).Methods Use a modified way of mechanical fixation to make rabbit OA model.Knee OA separated and extractd rabbit chondrocytes were and then culture in medium.The cultured chondroctyes were divided into the Dimethyl sulfoxide (DMSO) control group and the celecoxib group, which were given 1% DMSO and 10 μmol/L celecoxib respectively.After 48 hours' intervention, the protein expression and gene expression of Wnt/β-catenin signaling pathway related factors, including β-catenin,matrix metalloproteinase (MMP)-3, MMP-13, collagen Ⅱ were detected.T-test was used for the comparison between the two groups.Results Compared with the control group, the levels of type Ⅱ collagen [(275±19) ng/ml vs (202±12) ng/ml, t=-6.21, P<0.05] and proteoglycan [(58±12) ng/ml vs (30±3) ng/ml, t=-9.23, P<0.05] significantly increased in celecoxib group (P<0.05).Meanwhile, compared with the control group, the levels of β-catenin, MMP-3, MMP-13 mRNA and protein in celecoxib group were significantly reduced (P<0.05).Conclusion In rabbit OA chondrocytes, celecoxib can inhibit Wnt/β-catenin signaling pathway, reduce downstream factors expression including MMPs, thereby increase the cartilage matrix type Ⅱ collagen and proteoglycan levels.

3.The clinical significance of serum and joint fluid osteopontin, and thrombin-cleaved osteopontin levels in osteoarthritis

Objective To explore the relationship between osteopontin (OPN),its downstream product thrombin-cleaved osteopontin (NTOPN) and radiographic changes in osteoarthritis (OA).Methods The levels of OPN and NTOPN in serum and synovial fluid were tested by enzyme-linked immuno sorbent assay (ELISA) kit in 22 OA patients and 21 health controls.The correlation between the above parameters and clinical data such as radiology changes of OA patients was analyzed.Results The serum level of OPN in the OA group was positively correlated with C reactive protein (CRP) (r =0.574,P =0.005).The serum level of OPN in the OA group was slightly lower than that in the healthy control group [(1.8 ± 0.5) μg/L vs (1.7 ± 1.1) μg/L,P > 0.05].Similarly,the serum level of NTOPN in the OA group was significantly higher than that in control group [(21.1 ± 11.7) μg/L vs (14.7 ± 3.6) μg/L,P < 0.05].Meanwhile,serum NTOPN level in female patients was significantly higher than that in male patients [(23.2 ± 11.80) μg/L vs (11.1 ± 4.9) μg/L,P < 0.05].In synovial fluid of OA group,the level of OPN was (7.75 ± 2.33) μg/L,NTOPN was (0.91 ± 0.49) μg/L and the ratio of NTOPN/OPN was 0.12 ± 0.08.The level of OPN of synovial fluid in the OA group was positively correlated with CRP (r =0.574,P =0.005).NTOPN-to-OPN ratio of synovial fluid in the OA group was positively correlated with Kellgren-Lawrence grade (r =0.535,P =0.022).Conclusions The serum level of OPN is correlated with severity of inflammation in the OA patients.Meanwhile,NTOPN-to-OPN ratio of synovial fluid is correlated with OA severity.As a downstream bioactive product of OPN,NTOPN may play an important role in OA pathogenesis.Whether these parameters can be used as predictors of joint lesion severity and inflammatory status is worth further study.
4.The Effect of Astragalus Membranaceus and Angelica Sinensis on Grafted Cochlea Stem Cells Apoptosis in Hearing Impaired Rats

Objective To investigate the effects of astragalus membranaceus and angelica sinensis on grafted cochlea stem cells apoptosis in hearing imparied rats.Methods The hearing impaired rats were divided into 3 groups:cochlea stem cells group(45 rats),astragalus membranaceus and angelica sinensis group(45 rats),and the control group(35 rats).Cochlea stem cells only were grafted into the first group cochlea regions,mixture of stem cells and astragalus membranaceus and angelica sinensis were grafted into that of the second group.Saline was grafted into that of control rats.Observed was apoptosis at 1 day,3 days,5 days,10 days,15 days after the transplantation by CASPASE-3 and Bcl-2 immunofluorescence.Results After transplantationn 1 day,3 days,5 days,10 days,and 15 days of five time,for the cochlea stem cell group,the rates of apoptotic cells were 27.84%,49.37%,44.78%,39.37%,and 34.82%,respectively.The rates of astragalus membranaceus and angelica sinensis group were 18.35%,26.92%,23.18%,21.38%,and 18.19%,respectively.The percent of grafted cochlea stem cells apoptosis in astragalus membranaceus and angelica sinensis group was much lower than that in cochlea stem cells group and control group.The majority of apoptotic cells occurred within three to five days after transplantation.Conclusion The majority of apoptotic cells occur within three to five days after transplantation in the hearing impaired rats.The astragalus membranaceus and angelica sinensis could reduce the apoptotic rates of transplanted stem cells.
5.Clinical analysis of complete left bundle branch block after transcatheter closure of ventricular septal defect

Objective To evaluate the clinical features and prognosis of patients with complete left bundle branch block(CLBBB)following transcatheter device closure of ventricular septal defect(VSD)closure. Methods Clinical feathers of 11 patients with postoperative CLBBB in Department of Pediatric Cardiology,Guangdong General Hospital from January 2011 to December 2013 were collected and reviewed retrospectively. They were treated with dif-ferent protocol based on the appeared time of CLBBB occurrence and clinical symptoms. The patients were followed up, and the prognosis was recorded. Results The median age of 11 patients was 3. 9 years(3. 4 to 17. 5 years old). The median interval of intervention therapy to first attack of CLBBB was 2. 8 months(1 day to 25. 4 months). CLBBB oc-curred within 1 week to 1 month postoperatively in 4 patients,another 1 case suffered from CLBBB between 1 week to 1 month postoperatively,meanwhile 6 cases underwent CLBBB after 6 months postoperatively. The longest term of CLBBB attack postoperatively was 25. 4 months in 1 patient. The electrocardiograms on 5 patients returned to normal by only drug treatment. However,3 patients failed to recover with drug therapy,2 of them undertaken surgical procedure to re-move the occluder associated with VSD repair,1 patient recovered to normal and another converted to incomplete right bundle branch block. One of them refused to undertake surgical procedure and still bothered with persistent CLBBB. Another 3 cases did not receive special treatment due to the later attack of CLBBB(≥6 months)without clinical symp-toms. By the end of observation,the electrocardiogram(ECG)in 4 patients returned to normal,4 patients presented with persistent CLBBB. One patient's ECGs were presented with right bundle branch block. After ECG successfully returning to normal ECG by drug therapy,2 patients relapsed during follow - up,and 1 of them developed to an enlarging left ven-tricle and heart failure which led to death. Conclusions CLBBB may occur in short or long - term after VSD closure. ECG may become normal after early and appropriate treatment postoperatively. Systolic dyssynchrony and cardiac dys-function may be caused by persistent CLBBB. Therefore,patients with CLBBB after VSD closure should be treated ap-propriately without delay,and more frequent and longer follow - ups are required.
6.The effect of hBSP over-expression on the biological behaviors of breast cancer cells

Objective Recent researches show that bone sialoprotein is closely related with metastasis of breast cancer to bone. In order to investigate the effect of bone sialoprotein in the course of metastasis of breast cancer to bone, the following study was undertaken. Methods The growth curve, clone formation and adherence of breast cancer cells experiments were carried out to study the effect of hBSP over-expression on biological behaviors of breast cancer cells. Results It was found that breast cancer cells MDA-MB-231BO(pI) and MDA-MB-231BO(pID) stably transfected with hBSP cDNA were not inhibited through proliferation assay of breast cancer cells. The activity and tumorigenicity of breast cancer cells with hBSP over-expression were enhanced. Comprising with MDA-MB-231BO non-transfected the colony formation percentage of breast cancer cells MDA-MB-231BO(pI) and MDA-MB-231BO(pID) increased from 8.90%?0.53% to 15.6%?0.94% and 12.8%?0.77%. The OD_ 620 value of adherence of breast cancer cells MDA-MB-231BO(pI) and MDA-MB-231BO(pID)increased from 0.29?0.017 to 0.52?0.031 and 0.43?0.026 respectively in cells attachment assay, which suggested that over-expression of hBSP enhances breast cancer cell MDA-MB-231BO adherence and attachment property. Conclusion The study results suggested that hBSP over-expression had autocrine effect on breast cancer cells and could enhance the ability of breast cancer cells metastasizing to bone.
7.Dermoscopic features of 82 cases of molluscum contagiosum

Objective:To investigate dermoscopic features of molluscum contagiosum.Methods:A total of 82 outpatients with molluscum contagiosum were collected from Dalian Dermatosis Hospital between August 2019 and January 2020, and the dermoscopic features of 227 skin lesions were analyzed retrospectively.Results:Of the 227 skin lesions, hilar depression and orifices were observed in 120 (52.86%) by naked eyes and 198 (87.22%) by dermoscopy, and blood vessels were observed in 20 (8.81%) by naked eyes and 214 (94.27%) by dermoscopy. Different vascular patterns were identified by dermoscopy, including crown-shaped vessels (132 lesions, 58.15%) , punctate vessels (34 lesions, 15.00%) , and a mixed pattern (48 lesions, 21.15%) . The proportion of punctate vessels was significantly higher in lesions with inflammation or exfoliation (86.00%) than in those without (22.03%, χ2=81.685, P < 0.001) , and significantly higher in lesions with eczematous changes (50.00%) than in those without (33.85%, χ2=17.784, P < 0.001) . The most frequently observed structures were round, white-yellow, amorphous structures (40.53%) , followed by four-leaf clover-like pattern (33.48%) , polylobular pattern (20.26%) and nonspecific pattern (5.73%) . Conclusion:Orifices, vascular structures and white-yellow amorphous structures can be observed in molluscum contagiosum by dermoscopy, and the most common vascular pattern is the crown-shaped vessel.
8.Measurement of caspase-1 and its downstream factor interleukin-18, interleukin-33 in patients with rheumatoid arthritis

Objective To measure the levels of caspase-1 and its downstream factor interleukin (IL)-18 and IL-33 in rheumatoid arthritis (RA) and explore their possible mechanisms.Methods Blood samples were drown from 56 patients with RA and 22 healthy subjects.Serum levels of caspase-1,IL-18 and IL-33were tested by the method of enzyme linked immunosorbent assay (ELISA).Kruskal-Walls and Mann-Whitney test were used to compare the levels of caspase-1,[L-18 and IL-33 and Spearman's correlation test was used for correlation analysis.Results The level of caspase-1 was significantly increased in RA group compared to healthy group [(32±26) ng/ml vs (15±6) ng/ml,P<0.01].Meanwhile,the active disease groups showed a higher level than the remission group,and level in the untreated group was higher than the treated group [(47±27) ng/ml vs (25±22) ng/ml,P<0.01].The levels of IL-18 and IL-33 were significantly increased in RA group compared to healthy group [(121±121) ng/L vs (58±33) ng/L,(1032±1011) ng/L vs (510±231)ng/L,respectively,P<0.05].Meanwhile,the active disease groups had a higher level than the remission group and the untreated group had higher levels than the treated group [IL-18 and IL-33 were (172±139) ng/L vs (97±106) ng/L,(1469±1039) ng/L vs (825±941) ng/L,respectively,P<0.05].Caspase-1 was correlated withIL-18 and IL-33 (r=0.824,0.854,P<0.01) and IL-18 was correlated with IL-33 (r=0.800,P<0.01).But neither of the three factors was related with clinical indexes including disease duration,RF,anti-CCP antibody,tender joints count and swollen joints count.Conclusion Caspase-1 and its downstream factor IL-18,IL-33 increase in RA,and they may play important roles in RA.
9.Bioinformatics analysis of lung cancer metastasis-related miR-29b

Objective:This paper aims to bioinformatically analyze the target genes of miR-29b and to provide clues for cancer research targeting miR-29b. Methods:The differential expression levels of miRNAs in CD133+and CD133-A549 cells were detected using the miRNA PCR chip. Real-time polymerase chain reaction was performed to verify the partially differential expression of miR-NAs. Target genes of miR-29b were predicted by miRecords and analyzed by gene ontology and signal transduction pathway enrich-ment analysis. Results: The miR-29b expression was significantly decreased in the CD133+A549 cells compared with that in the CD133-cells. The number of miR-29b target genes was 106. The functions of these target genes were enriched in binding and extracel-lular matrix structural constituent (P<0.01). The JAK-STAT and TGF-βsignal transduction pathways were significantly enriched (P<0.05). Conclusion:The abnormal expression of miR-29b may be related to metastasis. Some of the predicted target genes of miR-29b were significantly enriched in the signaling pathways in relation to the tumors.

A new organic-soluble co-reactant of N,N,N’,N’-tetrakis(propyl)-pentanediamine (TPPD) was synthesized and co-immobilized with tris ( 2, 2’-bipyridine ) ruthenium ditetrakis ( 4-chlorophenyl ) borate ([Ru(bpy)3][4-(Clph)4B]2) on an indium tin oxide (ITO) electrode for electrochemiluminescent (ECL) study. The hydrophobicity of TPPD prevented its leakage from the electrode surface, thus the long-term stability of the ECL electrode was enhanced. The TPPD/[Ru(bpy)3][4-(Clph)4B]2 modified-ITO electrode exhibited strong, stable and reproducible ECL signal. The ECL signal could be quenched efficiently by phenol with a quenching efficiency of 95. 4% in the presence of 0. 1 mmol/L phenol, demonstrating the potential of the modified electrode in determination of phenolic compounds. The transient-state of the electrogenerated chemiluminescence reaction was also investigated, which revealed that the TPPD/[Ru(bpy)3][4-(Clph)4B]2 modified-ITO electrode had a longer lifetime than conventional TPA-Ru(bpy)2+3 co-reactant system.
