Objective To study the effect of Yiqimingmu pill on ATPase and LD of retinal ischemia rabbit, and probe into its mechanism. Methods Thirty-two Japanese large-ear white rabbits were divided into four groups, normal group (A group), model group (B group), comparison group (C group) and treated group (D group). Retinal ischemia was made in B, C and D group. After the treatment of one month, testing the correspondent indexes and studying with statistics. Results After the treatment of one month, in B and C group, the content of ATPse was lower, while the content of LD was higher compared with A group. Conclusion Yiqimingmu pill is reliable in the treatment of retinal ischemia, and superior to Buzhongyiqi wan.
3.Ultrasonography and MRI in the Diagnosis of Cesarean Scar Pregnancy

Purpose The incidence of cesarean scar pregnancy (CSP) shows a rising trend in recent years, and misdiagnosis or improper treatment of it can cause serious consequences. In this paper, we analyzed the characteristics of MRI and ultrasonography of CSP to provide a basis for choice of clinical imaging techniques. Materials and Methods Fifty-eight patients with cesarean scar pregnancy were studied retrospectively, all the cases underwent preoperative color Doppler ultrasound (trans-vaginal or together with trans-abdominal), and 31 cases were examined with MRI, the accurate imaging features and diagnosis rate of each was analyzed.Results 48 patients (82.8%) were diagnosed accurately by color Doppler ultrasound, including 20 cases of out-growth type (95.2%, 20/21), 18 cases of inner-growth type (90.0%, 18/20) and 10 cases of mass type (58.8%, 10/17); 27 cases (87.1%) were diagnosed accurately by MRI, including 8 cases of out-growth type (100.0%, 8/8), 4 cases of inner-growth type (66.7%, 4/6), and 15 cases of mass type (88.5%, 15/17).Conclusion The diagnostic accuracy of sonography for CSP is close to that of MRI, thus can act as the first choice; MRI can be chosen for combine examination to improve the accurate diagnostic rate when the CSP case is mass type detected by ultrasonic findings which is difficult to be diagnosed.
5.Determination of hypericin in Hypericum sampsonii

Objective: To explore the contents of hypericin from different parts of Hypericum sampsonii. Methods : The hypericin in Hypericum sampsonii and H. Officinalis were measured by HPLC with Novapak C 18 column, mobile phase(methanol∶ethanol∶0.1mol?L -1 NaH 2SO 4=200∶300∶100), flow rate 1.0mol?L -1 , column temperature at 30 ?C , wavelength at 588nm. Results:Contents of H. Sampsonii in flower and flower itself were equal content to H. officinalis. Conclusion:It is possible to sift out the plant of H. Sampsonii with more Hypericin.
6.Adsorptive Voltammetric Behavior of Riboflavine Tetrabut yrate and Its Measurement of Rate Constant at the Mercury Electrode

Cyclic voltammetry shows that, in a supporting ele ctrolyte of NaOH,riboflavine tetrabutyrate(RT)has a pair of cathodic and anodi c peaks at Hg electrode. The peak potentials Epc=Epa=-0.64 V(vs.Ag/Agcl). The adsorptive characteristics of RT are explored in detai ls with various methods. The adsorbed species is most probably neutral molecule of RT.The saturated adsorption amount of RT at Hg electrode is 5.42×10-11 mol/cm2 and every RT molecule occupied 3.06 nm2. On the surface of HMDE ,the adsorption of RT obeys Frumkin dasorption isotherm. The adsorptive coeffic ient β,the attractive factor γ are 4.60×105,1.10 and the adsorption free energy ΔG, is -32.30 kJ/mol, respectively. The electrode proc ess kinetic behavior has also been studied by using cyclic voltammetry. Under th e experimental conditions the rate constants for electron transfer are determine d. The values of ks are in good agreement with literature′s results.
7.Determination of artesunate in artesunate and amodiaquine hydrochloride tablets by HPLC

Objective To establish an HPLC method for the determination of artesunate in artesunate and amodiaquine hydrochloride tablets. Methods WondaSil C18-WR column was used with mobile phase consisted of acetonitrile:phosphoric acid aqueous solution(adjust pH to 3,gradient elution);wavelength was 210 nm; flow rate was 1 mL/min and the column temperature was 30℃.Results The standard curve was linear in the range of 0.2~3.2 mg/mL(r=0.9997), average recoveries were 99.0%(RSD=1.35%, n=6).Conclusion The method is accurate and sensitive, and it can be used to control the quality of artesunate and amodiaquine hydrochloride tablets.
8.The Effect of Astragalus Membranaceus and Angelica Sinensis on Grafted Cochlea Stem Cells Apoptosis in Hearing Impaired Rats

Objective To investigate the effects of astragalus membranaceus and angelica sinensis on grafted cochlea stem cells apoptosis in hearing imparied rats.Methods The hearing impaired rats were divided into 3 groups:cochlea stem cells group(45 rats),astragalus membranaceus and angelica sinensis group(45 rats),and the control group(35 rats).Cochlea stem cells only were grafted into the first group cochlea regions,mixture of stem cells and astragalus membranaceus and angelica sinensis were grafted into that of the second group.Saline was grafted into that of control rats.Observed was apoptosis at 1 day,3 days,5 days,10 days,15 days after the transplantation by CASPASE-3 and Bcl-2 immunofluorescence.Results After transplantationn 1 day,3 days,5 days,10 days,and 15 days of five time,for the cochlea stem cell group,the rates of apoptotic cells were 27.84%,49.37%,44.78%,39.37%,and 34.82%,respectively.The rates of astragalus membranaceus and angelica sinensis group were 18.35%,26.92%,23.18%,21.38%,and 18.19%,respectively.The percent of grafted cochlea stem cells apoptosis in astragalus membranaceus and angelica sinensis group was much lower than that in cochlea stem cells group and control group.The majority of apoptotic cells occurred within three to five days after transplantation.Conclusion The majority of apoptotic cells occur within three to five days after transplantation in the hearing impaired rats.The astragalus membranaceus and angelica sinensis could reduce the apoptotic rates of transplanted stem cells.
9.Effect of Treg on intestinal T-cell immunity and endotoxin translocation in rats after severe burns

Objective To observe the change of regulatory T cells expression in severely burned rats gut ,To investigate the effects of regulatory T cells on CD3+CD4+ /CD3+CD8+lymphocytes and its relation with gut-origin endotoxin translocation .Meth-ods Fifty SD male rats were randomly divided into normal control group(n=10)and burn model groups(n=40) .Rats were burned to achieve Ⅲ degree scalding ,and a 30% total body surface area(TBSA) burn model was made .Rats were sacrificed before(normal control group) and after 0 .5 ,1 ,2 ,4 burn hour(PBH groups) .Flow cytometry techniques were used for the detection of the expres-sions of regulatory T cells and CD3+CD4+ /CD3+CD8+ lymphocyte in intestinal lymph nodes which were separated .The dynamic turbidity method was used for detection of endotoxin levels in portal vein blood .Results The expression of regulatory T cells was negativelycorrelatedwithCD3+CD4+ /CD3+CD8+ lymphocyteratio(r= -0.827,P<0.01)inintestinallymphnodeofrats,while the regulatory T cells was positively correlated with ET levels in portal vein blood plasma .(r=0 .782 ,P<0 .01) .Conclusion The expression of the regulatory T cells in intestinal lymph node in severely burned rats was increased compared to that in normal con-trol group .Regulatory T cells suppressed the expression of intestinal T lymphocytes ,leading to gut immune inhibition .The translo-cation of intestinal endotoxin has a close relationship with regulatory T cells in severely burned rats .Regulatory T cells could have portal effects on intestinal immunity barrier .
10.Role of Toll-like receptor 4 in spinal cord in a rat model of persistent postoperative pain evoked by skin/ muscle incision and retraction

Objective To investigate the role of Toll-like receptor4 (TLR4) activation in spinal cord in a rat model of persistent postoperative pain evoked by skin/muscle incision and retraction(SMIR).Methods Ninetysix male SD rats weighing 200-250 g were randomly divided into 4 groups(n =24 each):group sham operation; group SMIR; group SMIR + IT scramble siRNA and group SMIR + IT TLR4siRNA.The rat model of persistent postoperative pain evoked by SMIR was established according to the method described by Flatters.The TLR4 siRNA were administered intrathecally for 7 days starting from 1 day beforc surgcry.Pain behavior was assessed by paw mechanical withdraw threshold (MWT) to Electronic von Frey Anesthesiometer stimulation at 1 day before and 1,3,7,12,and 22 days after operation.Four animals were sacrificed at each time point in each group for detection of the expression of TLR4 protein in the spinal cord by Western blot analysis.Results Compared to group sham group,MWT was significantly descreased at 3,7,12,and 22 days after operation,while the expression of TLR4 protein in the spinal cord were significantly increased at 3,7,12 days after operation in group SMIR and group SMIR + IT scramble siRNA ; IT TLR4siRNA significantly attenuated the hyperalgesia induced by SMIR and descreased the expression of TLR4 protein at 3,7,12 days after operation in group SMIR + IT TLR4siRNA.Conclusion TLR4 activation in spinal cord plays an important role in the development of SMIR-evoked persistent postoperative pain in rats.
