1.Intervention therapy of chemoradiotherapy-induced oral mucositis

Oral mucositis is a common and serious complication secondary to chemo-radiotherapy for head and neck cancer that has a high morbidity and affects the treatment outcomes. Currently available interven-tions such as three-dimensional conformal radiotherapy and intensity-modulated radiation therapy are directed at minimizing local radiation exposure and providing symptomatic relief. Recently, a number of new treatment mo-dalities have been expected to be introduced for reduction of severity and duration of mucositis, such as kerati-nocyto growth factor, amifostine, zinc sulfate,anti-inflammation agents,glutamine,etc. These efforts are likely to improve the life quality of cancer patients significantly.
2.A Systematic Review of Randomized Controlled Trials for Clinical Efficacy and OS on Xi-Pa-Yi Mouth Rinse for Simple Gingivitis

This study was aimed to evaluate the clinical efficacy of Xi-Pa-Y i (XPY) Mouth Rinse in the treatment of simple gingivitis and improved value for halitosis. Related literatures were systematically searched in the China Na-tional Knowledge Infrastructure (CNKI, 1979-2012), Weipu Database for Chinese Technical Periodicals (1989-2013), PubMed and Cochrane Library. The methodological quality of included trials was assessed based on the Jadad scale. And the available data were analyzed with RevMan software (version 5.2.0). The results showed that a total of 4 stud-ies involving 360 participants satisfied the minimum criteria for meta-analysis. The evidences showed that the XPY Mouth Rinse was compared with saline or western medicine for treatment of the OS of simple gingivitis WMD (95%confidence interval) were -0.48 (-0.81, -0.16), 0.04 (-0.15, 0.22), respectively; the XPY Mouth Rinse was com-pared with saline or western medicine for treatment of simple gingivitis clinical efficacy's RR (95% confidence inter-val) were 1.67 (1.29, 2.15), 1.22 (0.35, 4.20), respectively. It was concluded that the current clinical evidence showed that the effectiveness of XPY Mouth Rinse in the treatment of simple gingivitis was higher than the saline. It had similar effects with western medicine, which can reduce the OS, and with no significant side effects and easy to take. However, due to the low quality studies and small sample amount, it is still need to be confirmed by the high-quality, multi-center, large sample, randomized, double-blind clinical controlled trials.
3.Influence of White Prepared Lateral Root of Aconite on Blood Biochemical Indicators in Normal and Kidney Yang Deficient Rats

Objective To explore the differences of blood biochemical indicators between normal and kidney Yang deficient rats administered orally different doses of white prepared lateral root of aconite.Method Eighty Sprague-Dawley(SD) male rats were divided randomly into normal group and kidney Yang deficient model group,forty rats in each group.After producing kidney Yang deficient model by injecting hydrocortisoni natrii succinas in intramuscular,normal and model rats were respectively and randomly divided into little,low,medium,great dose group and blank group,eight rats in each group.Those rats were administered orally with different concentrations of white prepared lateral root of aconite every day for two weeks.Twenty-four hours after the last oral administration,the blood were obtained and used for testing adrenocorticotrophic hormone(ACTH),cyclic adenosine monophosphate(cAMP),cyclic guanosine monophosphate(cGMP),glucose(GLU),cholesterol(CHO),alanine aminotransferase(ALT),urea nitrogen(BUN).Results Before taking medicine,model rats' cAMP/cGMP and ACTH were lower than normal rats',and they showed a trend of rise with adding dose.Before taking medicine,model rats' GLU was lower than normal rats'.With adding dose,model rats' GLU increased,however,normal rats' GLU reduced.When taken medium dose white prepared lateral root of aconite,model rats' CHO was higher than normal rats'.Taking medicine made both groups ALT rise,but normal rats' went up more quickly than model rats'.Model rats' BUN was higher than normal rats',and taking low dose or little dose medicine could lead to incline of reduction.Conclusion White prepared lateral root of aconite caused different blood biochemical effects on normal and kidney Yang deficient rats,and more remarkable toxicity effect on normal rats.
4.Setting up of Intraluminal Thread Approach in Making Focal Cerebral Ischemia/Reperfusion Model in Rabbit and the Evaluation of Image Features

Objective To establish a stable model of intraluminal thread approach in making focal cerebral ischemia/reperfusion modelin rabbit.Methods Forty New Zealand white rabbits were randomly divided into three groups:A,B,and C.Angiography of ICA wasperformed in group A(n=5). Control group B(n=5) was sham-operated.In group C(n=30),the focal cerebral ischemia/reperfusionmodel was made by inserting a wire ( d=0.45 mm ) into MCA through ICA and CCA directly and pulling it out 3 hours later . MRI andneurological deficit score were used to evaluate the effectiveness of models.Results Angiography could show the course and bifurcation of ICA clearly.22 models were successful.The pathological changes were shown on MRI.The neurological deficit score (reperfusion 1 h) were 2.8?0.45.Conclusion The focal cerebral ischemia/reperfusion model made by inserting a wire(d=0.45 mm) into MCA through ICA and CCA directly and pulling it out 3 hours later was stable and fit for imaging research.

7.Effects of continuous venovenous hemodiafiltration on plasma LPS, IL-1β, IL-4, HMGB-1 in multiple organ dysfunction syndrome dogs

Objective To investigate the effects of continuous venovenous hemodiafiltration (CVVHDF) on plasma lipopolysaccharide (LPS),interleukin(IL) 1β,IL-4,high mobility group box 1 (HMGB-1) in multiple organ dysfunction syndrome (MODS) dogs,to explore the therapeutic mechnism of CVVHDF.Methods Dogs were subjected to hemorrhagic shock plus resuscitation and endotoxemia to establish MODS model,then they were randomly divided into 2 groups:CVVHDF group (n=6) and MODS group (n=6).After endotoxin injection completed,the CVVHDF group received CVVHDF treatment for 24 h; MODS group did not receive CVVHDF treatment.The LPS,IL-1β,IL-4,HMGB-1 levels in plasma and ultrafiltrate were measured at different time points(before operation,before intravenous infusion of endotoxin,0 h,3 h,6 h,9 h,12 h,18 h,24 h after intravenous infusion completion of endotoxin),then the sieving coefficient (SC) was calculated.Results Compared with MODS group,the LPS,IL-1β,IL-4,HMGB-1 levels in plasma were significantly decreased in the CVVHDF group at 9 h,12 h,18 h,24 h (P < 0.05).Some concentration of IL-1β,IL-4 and HMGB-1 was detected in the ultrafiltrate,the SC for IL-1β,IL-4 and HMGB-1 was 0.60±0.12,0.83±0.11,0.70±0.09 respectively,and LPS could not be detected in the ultrafiltrate,its SC was 0.After treatment,the damage level of lung and renal had been significantly reduced in CVVHDF group.Conclusion CVVHDF can effectively reduce the levels of pro-inflammatory mediator and anti-inflammatory cytokine related to MODS,cut off the vicious circle of cytokine network and reduce the damage level of organism in the treatment of MODS.
8.Capillary electrophoresis and laser-induced fluorescence (CE-LIF) for screening of wild type genes based on molecular beacons hybridization

Objectives A new method for the detection of PCR ampli fied DNA in capillary electrophoresis and laser induced fluorescence (CE LIF) system based on molecular beacons hybridization is described.Methods The method is based on the hybridization of molecular beacons into the amplification product, then separating the hybridized product with CE LIF system. This method was applied to the detection of p73exon3, p53exon5 and p16exon2 for gastric carcinoma. Results Detected with CE LIF molecular beacons method, the wild type percent of p73exon3 is 100%, same as SSCP; p53exon5 is 60 0%, while SSCP is 63 3%, P=0 99; p16exon2 is 20 0%, same as SSCP(20 0%).Conclusion The results demonstrate that the specific peaks appeared in wild type samples while no peaks or unspecific peaks in mutation ones. The results showed correlation with the results of PCR SSCP. The method is able to develope as a clinical screening method for tumor suppressor genes to diagnose carcinoma.
9.Effects of different load exercises on sex hormone and sperm quality in male obese mice

BACKGROUND:Overweight and obesity can lead to a disorder of sex hormone in men. The increase in female hormone levels may inhibit the synthesis and secretion of male hormone, increase fat accumulation and form a vicious circle. Exercise can effectively reduce body fat. OBJECTIVE: To investigate the effects of different exercise loads on sex hormone and the quality of sperm in obese male mice. METHODS: Weanling male C57BL/6J mice were divided into normal control group and obesity group. Mice in the obesity group were given high fat diet for 10 weeks to establish mouse model of obesity. The amount of food and water was recorded daily. Body weight was weighed once every week. After model induction, models were assigned to obesity moderate load exercise group and obesity high load exercise group. These models did exercises for 8 weeks. Body length was measured. Body weight, abdominal fat, testis, epididymis and seminal vesicle were weighed. Sperm activity and motility were observed by the sperm counting method in the epididymis tail. Enzyme linked immunosorbent assay was used to measure serum progesterone, folicle stimulating hormone, testosterone and estradiol. RESULTS AND CONCLUSION:Compared with the normal control group, body weight, abdominal fat weight, and lee’s index were increased (P < 0.01); the coefficient of testis and seminal vesicle were significantly decreased (P < 0.01); serum levels of luteinizing hormone, folicle stimulating hormone and testosterone were significantly decreased and estradiol level was significantly increased (P< 0.05); sperm count and activity were significantly decreased in the obesity group (P < 0.01). Compared with the obesity group, body weight, abdominal fat weight and lee’s index were significantly decreased (P < 0.05 orP < 0.01); the coefficient of testis and seminal vesicle were significantly increased in the obesity moderate load exercise group and obesity high load exercise group (P < 0.05 orP < 0.01). Serum luteinizing hormone, folicle stimulating hormone and testosterone levels were significantly increased (P < 0.05 orP < 0.01); estradiol levels were significantly decreased (P < 0.05); sperm count and activity were significantly increased (P < 0.01,P < 0.05) in the obesity moderate load exercise group. Compared with the obesity moderate load exercise group, abdominal fat weight and lee’s index were significantly reduced (P < 0.05); serum luteinizing hormone, folicle stimulating hormone, testosterone, sperm count and activity were decreased in the obesity high load exercise group (P < 0.01). These results indicate that long-term high fat diet leads to early obesity in males, inhibits the development of the reproductive gland and reproductive organs, and causes the decrease of the level of male hormone and sperm quality. Long-term moderate load exercise effectively reduces body fat, improves the inhibitory effect on male reproductive organs and glands, and relieves the negative effect of obesity on reproductive function. The effect of long-term large load exercise on reducing body fat is better than medium load exercise, but it has little effect on improving the level of male hormone in obese mice or on relieving the negative effect of obesity on reproductive function, even has a tendency to aggravate.
