Objective To investigate the effects of different hemostatic methods in laparoscopic ovarian cystectomy on ovary functions . Methods A total of 90 cases of laparoscopic ovarian cyst resection from June 2010 to June 2013 were selected .They were divided into bipolar electrocoagulation group , monopolar electrocoagulation group , and suture group according to the doctor ’ s favor. The FSH, LH, and E2 before and after surgery were compared among the three groups . Results All patients were followed up for more than 1 year. The rate of patients with menstrual disorder in monopolar electrocoagulation group [33.3%(10/30)] was significantly higher than that in bipolar coagulation group [10.0%(3/30),χ2 =4.812,P=0.028] and suture group [6.7%(2/30),χ2 =6.667,P=0.010].There was no significant difference between bipolar premature coagulation group and suture group (χ2 =0.000, P=1.000).There was no significant difference in FSH ,LH,and E2 among the three groups before and 1 month after surgery (P>0.05).There was no significant difference in FSH ,LH, E2 between bipolar premature coagulation group and suture group 6 months after surgery (P>0.05).In the monopolar electrocoagulation group , the FSH [(10.43 ±2.04) U/L] and LH [(18.58 ± 3.82) U/L] levels were significantly higher than those in the bipolar electrocoagulation group [(8.12 ±1.82) U/L and (13.31 ± 2.53) U/L, P<0.05], and in the suture group [(7.08 ±1.68) U/L and (12.61 ±2.68) U/L, P<0.05].The E2 level in the monopolar electrocoagulation group [(252.5 ±26.9) pmol/L] was significantly lower than that in the bipolar electrocoagulation group [(321.3 ±28.2) pmol/L, P<0.05] and in the suture group [(313.7 ±31.4) pmol/L, P<0.05]. Conclusion In laparoscopic ovarian cystectomy , bipolar coagulation hemostasis is safer than monopolar electrocoagulation hemostasis , being worthy of clinical application .

Methods The human liver tumor cells were treated with alkylating agent(nitrogen mustard) and total RNA was extracted, the expression levels of Gadd genes were determined by RT-PCR. Results Treated with 1 ng/?l nitrogen mustard for 3 h, 24 h, 48 h respectively and with 1 ng/?l, 5 ng/?l nitrogen mustard for 24 h respectively, the expression levels of Gadd genes increased at first (P0.05). Conclusion Gadd genes expression can be used as the indexes in the surveillance of DNA damage induced by nitrogen mustard.

Objective To investigate the effects of ketogenic amino acid ( KAA) replacement diet on insulin resistance in mice fed with high fat diet(HFD) and to analyze the possible mechanism. Methods C57BL mice were fed with a control diet, HFD, and KAA-fortified HFD(HFDKAAR)from the age of 8 weeks, and 8 weeks after HFD initiation, the HFD-fed mice were divided into two groups:one group of mice were fed the same HFD, the other group were fed HFDKAAR ( HFD→HFDKAAR ) . The metabolic evaluations were performed at the end of 16 weeks. Blood glucose levels were measured at 0, 15, 30, 60, 90, and 120 min after the injection of glucose ( 1 g/kg BW intraperitoneal glucose tolerance test, ipGTT) . The insulin,β-hydroxybutyrate, and acetoacetate levels in the plasma were measured via ELISA. The insulin resistance index ( IRI) and area under curve ( AUC) were calculated. The expression of hepatic LKB1 ( liver kinase B1 ) , AMP-activated protein kinase ( AMPK ) , and mTOR ( Mammalian target of rapamycin ) protein, and mcp-1 mRNA were measured by western blot and real-time PCR respectively. Results HFD-fed group of mice displayed significantly heavier body weight, heavier intra-abdominal fat weight, and significant deterioration of glucose tolerance at the end of 16 weeks in addition to higher insulin levels( all P<0. 05), HFDKAAR-fed mice exhibited significantly ameliorated high fat diet-induced obesity and glucose intolerance compared to the HFD-fed mice, which was associated with decreased insulin levels, IRI, AUC, and mcp-1 mRNA expression (all P<0. 05). HFD suppressed hepatic LKB1 and AMPK phosphorylation expression, and increased mTOR phosphorylation levels compared to the control diet-fed mice(all P<0. 05). In contrast, treatment with the HFDKAAR diet increased LKB1and p-AMPK expression, which was associated with suppressed p-mTOR levels compared to the HFD-fed mice(all P<0. 05). Conclusion KAA may ameliorate high fat diet-induced obesity, glucose intolerance, via normalizing the hepatic LKB1-AMPK-mTOR nutritional signal passageway. KAA replacement diet seems to be a potential nutritional intervention for the treatment for patients with metabolic defects, such as obesity, glucose intolerance, as well as metabolic syndrome.

AIM:To observe the expression of Smad7 and Smad ubiquition regulatory factor-Smurf2 in rat glomerular mesangial cells (GMC) stimulated by the high concentration of glucose, and to investigate the effect of the ubiquition on Smad signaling by adding MG132 as a proteasome differential inhibitor. METHODS:Cultured rat GMC were divided into normal group (the concentration of glucose:5.6 mmol/L),high glucose group (20 mmol/L,30 mmol/L,respectively),therapy group (30 mmol/L glucose with MG132). The expressions of Smurf2 and Smad7 in each group were measured by indirect immunofluorescence and laser scanning confocal microscope. RESULTS:(1) The expression of Smurf2 in GMC in normal group was weak (25.93?3.35) whereas the expression of Smad7 was strong (64.09?7.43). (2) The expression of Smurf2 in high glucose group was stronger than that in normal group (P

AIM: To study the regulatory effect of curcumin on expression of heme oxygenase-1 (HO-1) in the lung of rat treated with LPS. METHODS: Eighteen rats were divided into three groups injected with different agents via lingua vein: control group (animals received equivalent saline) , LPS group (animals received a bolus dose of LPS 5 mg?0.5 mL-1?kg-1) and LPS+ curcumin group (animals received AP- 1 inhibitor curcumin 20 mg?0.5 mL-1?kg-120 min before the injection of LPS 5 mg ?0.5 mL-1?kg-1) . The expression of HO-1 mRNA and protein in the lung were examined 7 h after LPS administration by reverse transcribed polymerase chain reaction (RT- PCR) and Western blotting, respectively. Carboxyhemoglobin (HbCO) formation within pulmonary tissue was measured to represent CO content. RESULTS: The results showed that HO- 1 mRNA and protein expression as well as CO content in the lung of rats in LPS group were significantly higher than those in control group (P
