1.Therapeutic effects of modified Bushen Huoxue granules on ovariecto-mized osteoporosis rats

AIM: To observe the therapeutic effect of modified Bushen Huoxue granules (MBHG) on ovariec-tomized osteoporosis rats.METHODS: Female SD rats were randomly given sham operation (n =10) and ovariectomy, and then the model rats were further randomly divided into model group, MBHG treatment groups at doses of 200 mg/kg, 100 mg/kg and 50 mg/kg respectively, and positive control (estradiol valerate) group, with 10 rats in each group by intra-gastric administration for 12 weeks.The morphology, area, thickness, spacing and area percentage of trabecular bone in the rats were observed.The serum levels of calcium (Ca), phosphorus (P) and alkaline phosphatase (ALP) were mea-sured by automatic analyzer.Bone mineral density (BMD) was analyzed.Serum estradiol (E2 ), osteocalcin (BGP), os-teoprotegerin (OPG), receptor activator of nuclear factor-κB (RANK) and receptor activator of nuclear factor κB ligand (RANKL) levels were detected by ELISA.RESULTS: Compared with model group, trabecular bone significantly widened in all treatment groups with large number, and net-like structure restored partially.The thickness, area and area percenta-ges of trabecular bone in treatments groups were higher than those in model group,and trabecular spacing was less than that in model group (P <0.05).The serum Ca, P, E2 and OPG, and femoral BMD were significantly higher in treatment groups than those in model group, and the levels of ALP, BGP, RANK and RANKL were significantly lower than those in model group (P <0.01).CONCLUSION: MBHG has a significant therapeutic effect on ovariectomized osteoporosis rats. The mechanism may be related to the regulation of OPG and inhibition of RANKL secretion.
2.Effect of Xingnaojing Injection Combined with Conventional Therapy for Patients with Severe Traumatic Brain Injury

Objective:To observe the effect of Xingnaojing injection in the treatment of severe traumatic brain injury .Methods:From August 2013 to December 2014 , 110 patients with severe traumatic brain injury were randomly divided into the control group and the research group with 55 ones in each .The control group was treated with the conventional treatment , including removal of intracrani-al hematoma, bone disc decompression according to surgical indications , anti-infection, dehydration intracranial pressure reduction and nutritional support treatment .The research group was treated with Xingnaojing injection additionally .After the one-week, two-week and 4-week treatment , the GCS score of the two groups were compared .The rate of lung infection and stress ulcer in 4 weeks were com-pared.The clinical outcome of the two groups after the 4-week treatment was also evaluated .Results:The GCS score of the research group was higher than that of the control group after the 2-and 4-week treatment (P<0.05).There was no significant difference in the incidence of stress ulcer between the two groups , while the incidence of pulmonary infection in the research group was lower than that in the control group (P<0.05).The total effective rate of clinical prognosis in the research group (90.9%) was significantly higher than that in the control group (72.7%, P<0.05).Conclusion:Xingnaojing injection combined with the conventional treatment for the patients with severe traumatic brain injury can decrease the clinical complications and improve the clinical symptoms of patients , which is worthy of clinical promotion .
3.Determination of Six Phendic Acids in Mailuoning Injection by RP-HPLC

Objective To establish a method for determination the contents of six phendic acids in Mailuoning injection by HPLC at same time. Methods Separation was performed on a Kromasil-C18 (4.6 mm? 250 mm, 5 ?m) column with mobile phase consisting of 0.1% phosphoric acid-water and Acetonitrile with gradient elute at the flow rate of 1 mL/min. The UV detection wavelength was set at 327 nm and the column temperature was set at 25 ℃. The sample size was 20 ?L. Results The standard cures of neochlorogenic acid, chlorogenic acid, cryptochlorogenic acid, caffeic acid, 3,5-Dicaffeoylquinic acid, 3,4- Dicaffeoylquinic acid were linear within the contentration range of 0.010 90~0.544 8 ?g (r=0.999 9), 0.024 06~1.203 2 ?g (r =1), 0.011 06~0.552 8 ?g (r =1), 0.015 94~0.796 8 ?g (r =1), 0.009 104~0.455 2 ?g (r =1), 0.010 37~0.518 4 ?g (r=1) respectively. And the recovery rates of them were 101.06%, 100.22%, 101.54%, 99.42%, 100.21%, 101.65% respectively. Conclusion The method appeared to be simple, reliable, accurate and can be used to determine the contents of six phendic acids in Mailuoning injection.
4.Three-dimensional finite element analysis of stress in the bending root of maxillary first molar

METHODS:The maxil ary first molars with normal roots and bending roots were selected, and then the finite element models of maxil ary first molars and periodontal ligaments were established through CT scan and
Workbench 11.0 finite element analysis software. The models were loaded with six different methods to analyze the stress distribution.
RESULTS AND CONCLUSION:The stress distribution of bending root was mainly concentrated in the tooth
neck, fol owed by the apex. During the overal tooth movement, the stress was smal est on the root, periodontal ligament and alveolar bone. Correction of bending roots needs more accurate positioning and traction.
5.Analysis of dermoscopy and reflectance confocal microscopy features of verruca plana

Objective To evaluate imaging features of verruca plana by dermoscopy and reflectance confocal microscopy (RCM).Methods Totally,88 untreated patients with verruca plana were enrolled into this study.One typical skin lesion was chosen from each patient and subjected to dermoscopy and RCM separately,and imaging features were recorded and analyzed.Results Dermoscopy revealed a light red background in 48 cases (54.55%),a light yellow background in 40 cases (45.45%),and a various number of punctate hemorrhages in 84 cases (95.45%).RCM showed concentric onion-skin-like structures in 48 patients (54.55%).Among 57 patients with disease duration > 1 year,44 (77.19%) had the concentric onion-skin-like pattern,while only 4 (12.90%) patients had similar structures among 31 patients with disease duration < 1 year.A significant difference in the prevalence of concentric onion-skin-like structures was observed between patients with disease duration > 1 year and those with disease duration < 1 year (x2 =33.47,P < 0.05).Of 88 patients,86 (97.73%) had hyperpigmentation in the basal layer at the dermo-epidermal junction,intact dermal papillary rings,vasodilatation,hyperemia and 1-2 dilated capillaries in the dermal papillae.Eight patients were treated for 2 weeks,and the above features disappeared after the treatment under dermoscopy and RCM.Conclusion Based on clinical manifestations,dermoscopy and RCM features may be helpful for the diagnosis of verruca plana.
6.Analysis of Individualized Drug Use of Clopidogrel through Drug Genetic Testing by Clinical Pharmacists

Objective:To study how to guide the individual dose of clopidogrel in line with the genetic testing results. Methods:Clinical pharmacists decided how to optimize the prescription of clopidogrel according to the genotype combined with the drug metabolism and drug interactions for three patients respectively with slow clopidogrel metabolism,intermediary metabolism and super fast metabolism. Results:The slow clopidogrel metabolism patient with subacute stent thrombosis after half a month of coronary stenting was switched to orally administrate with ticagrelor. The super fast metabolism patient suffered from repeatedly subcutaneous hemorrhage with antiplatelet therapy was suggested to lower the dose of clopidogrel,temporarily withdraw Maixuekang capsules and conventionally administrate with vitamin C tablets orally. The intermediary metabolism patient with late stent thrombosis co-treated with lansoprazole was suggested to increase the dose of clopidogrel or use ticagrelor instead,and when it was necessary,panxitorazole,ray Bella or the other ranitidine acid suppression drugs such as ranitidine could be considered. Conclusion:Through genetic testing and drug interactions,clinical pharmacists guide the clinical use of clopidogrel and the optimization of antiplatelet therapy.
7.Effects of rosiglitazone on activation of human umbilical vein endothelial cells in high glucose environment

Objective To investigate the effects of rosiglitazone on proangiogenesis function of human umbilical vein endothelial cells ( HUVEC) in high glucose environment. Methods HUVECs were cultured in high glucose environment and stimulated by rosiglitazone. MTT, cell scratch test and Transwell assay were used to detect HUVEC proliferation and migration. The concentration of VEGF, SDF-1 was also detected in the supernatant. Results Rosiglitazone could effectively promote HUVEC proliferation and migration. The concentration of VEGF and SDF-1 in rosiglitazone stimulated supernatant was higher than that in high glucose group. The inhibition of AKT signal could block the promotion of rosiglitazone on the HUVEC proliferation, migration and secretion. Conclusion Rosiglitazone could significantly promote HUVEC secretion, proliferation and migration in high glucose environment. AKT signal played an important role in this process.
8.Inhibitory effects of lobaplatin on human ovarian cancer and its regulation of apoptosis-related genes in nude mice

Objective To explore the inhibitory effects of lobaplatin and cisplatin and their regulation of apoptosis-related genes in ovarian cancer cells in nude mice.Methods SKOV3 cells were implanted into nude mice.In monotherapy treatment study,the nude mice bearing human SKOV3 cells were randomly divided into control,lobaplatin,and cisplatin groups,with 7 mice in each group.The mice in each group were received corresponding treatment.The volume of tumor and the weight of nude mice were measured three times per week,respectively.Tumor inhibitory rate was calculated.The protein expressions of bax and bcl-2 were detected by flow cytometry.Results The growth inhibitory rate was 47.2% in lobaplatin group and 42.8% in cisplatin group,without significant difference between two groups (P > 0.05).The expression of bcl-2 was decreased but the bax was increased in lobaplatin and ciaplatin groups compared to the control group.Conclusions Lobaplatin can significantly inhibit the growth of ovarian cancer cells,induce apoptosis by down-regulation of bcl-2 and up-regulation of bax.
9.Effects of AMP-activated protein kinase on HMGB1 release from PC12 cells after oxygen-glucose deprivation and reoxygenation and its mediated inflammatory response in BV2 cells

Objective To investigate the effects of adenosine monophosphate-activated protein kinase (AMPK) on high-mobility group box 1 (HMGB1) release from PC12 cells after oxygen-glucose deprivation and reoxygenation (OGD/R) and its mediated inflammatory response in BV2 cells.Methods PC12 and BV2 cells were cultured,respectively.The PC12 cells were used to induce a model of oxygen glucose deprivation for 12 h and reoxygenation for 24 h.After giving 5-aminoimidazole-4-carboxamide (AICAR) 5,50 and 100 μmol/L as well as Compound C 0.1,1 and 10 μmol/L activation or inhibition of AMPK phosphorylation,respectively,methyl thiazolyl tetrazolium (MTT) was used to detect the PC12 cell activity.Enzyme-linked immunosorbent assay was used to detect the HMGB1 release level in the PC12 cell culture media.After OGD/R in each group,the PC12 culture media were acted on normal cultured BV2 cells for 24 h respectively.Westem blotting and Enzyme-linked immunosorbent assay were used to detect the NFκB inhibitory protein (inhibitor of NFκB,IκB) phosphorylation level and TNF-α release level in BV2 cells,respectively.Results After OGD/R,the PC12 cell activity was decreased significantly (68.84%±6.60% vs.100.04% ± 8.82%;P < 0.01);the AMPK phosphorylation level was increased significantly (1.95 ±0.39 vs.1.00 ±0.20;P<0.05),and the extracellular HMGB1 release was increased significantly (287.66 ± 26.42 pg/μl vs.53.05 ± 9.11 pg/μl;P < 0.01).Compared with the OGD/R group,AICAR 100 μmol/L significantly increased the survival rate of PC12 cell after OGD/R (78.6% ± 3.75% vs.68.84% ± 6.60%;P < 0.05),promoted AMPK phosphorylation (3.32 ± 0.66 vs.1.95 ± 0.39;P < 0.01),and reduce the release of extracellular HMGB1 (164.06 ± 12.77 pg/μl vs.287.66 ± 26.42 pg/μl;P <0.01).In contrast,Compound C 10 μmol/L significantly reduced the cell survival rate of PC12 (40.44% ±3.79% vs.68.84% ±6.60%;P <0.01),inhibited AMPK phosphorylation (1.07 ± 0.21 vs.1.95 ± 0.39;P<0.05),and increased the release of HMGB1 (337.97 ± 18.9 pg/μlvs.287.66 ± 26.42 pg/μl;P<0.01).The conditioned medium from the AICAR 100 μmol/L group significantly inhibited IκB phosphorylation (1.68 ±0.51 vs.3.09 ± 0.10;P < 0.05) and reduced the release of TNF-α (669.53 ±38.58 pg/μlvs.841.76 ± 45.82 pg/μl;P< 0.05) in BV2 cells.The conditioned medium from the compound C 10 μmol/L group significantly promoted IκB phosphorylation (4.98 ± 1.24 vs.3.09 ± 0.10;P <0.01) and increased the release of TNF-α (1 035.32 ± 128.06 pg/μl vs.841.76 ± 45.82 pg/μl;P <0.05) in BV2 cells.Conclusions Promoting AMPK phosphorylation activation may reduce the release of HMGB1 from PC12 cells after OGD/R,and inhibit its mediated NF-κB inflammatory pathway and reduce the release of TNF-αin BV2 cells,and thus reducing neuroinflammatory injury.On the contrary,inhibiting AMPK phosphorylation may promote the release of HMGB1 from PC12 cells after OGD/R and aggravate its mediated inflammatory reaction in BV2 cells.
10.The Onphonation Threshold Pressure and Nasalance in Normal Subjects

Objective To investigate the difference and the correlation of the vowel's phonation threshold pressure and nasalance in normal subjects .Methods The Aeroview (GLOTTAL Enterprises)with the materials /pa/,/pi/,/pu/ was used to obtain phonation threshold pressure values and The NasalviewTM (Tiger Electronics Inc .,Seattle ,Shanghai) with the materials /a/,/i/,/u/was used to obtain nasalance values .A total of 30 subjects (15 males ,15 females ,and aged 19~30) years old received the tests .The results of the vowel's phonation threshold pressure and nasalance gender score difference were compared .Results The PTP gender scores of /a/,/i/,/u/in maleswere4.53±0.70cm,5.21±0.80cm,5.87±1.45cm,andinfemaleswere4.09±0.30cm,4.47±0.30cm, and 4 .77 ± 0 .32 cm ,respectively .There were significant differences between males and females in PTP values (P<0 .05) .The PTP gender score differences showed that males were greater than that of females .There were signifi‐cant differences in different vowels (P<0 .05) .The nasalance gender scores of /a/,/i/,/u/in males were 30 .2 ± 5 . 75% ,38 .8 ± 8 .54% ,26 .5 ± 6 .03% ,and in females w ere32 .4 ± 7 .14% ,40 .5 ± 11 .8% ,and 30 .1 ± 4 .83% ,respec‐tively .There were no significant differences between males and females in nasalance values (P> 0 .05) .The na‐salance gender score differences showed that females were greater than males .There were significant differences in different vowels(P<0 .05) .This study found a weak correlation between PTP and nasalance .Conclusion The relationship between oral pressure and nasal flow cannot be accurately measured by measuring PTP ,and /pa/is a relatively suit‐able material for testing oral pressure .
