1.Research Progress on Multiple Origins Theory of Epithelial Ovarian Carcinoma

[Summary] Epithelial ovarian carcinoma has the highest fatality rate among gynecologic malignant tumors.Lacking of effective early diagnosis and screening method are the main reasons for its poor prognosis.There is a close relationship between the heterogeneity of epithelial ovarian carcinoma and the diversity of cancerous origins.According to the traditional theory, epithelial ovarian carcinoma derives from germinal epithelium at ovarian surface.The “secondary Mullerian system” theory has challenged above view.This“dualism” theory completely subverts the traditional one.However, both of the theories have their limitations.In recent years, the multiple origins theory of epithelial ovarian carcinoma has gradually become a hot point of discussion.
2.Rho-Associated Kinase and Cerebrovascular Diseases

The disability and mortality of cerebrovascular diseases are high,and they cause significant harm to human health.A number of studies have found that a great many factors participated in the pathophysiological processes of cerebrovascular diseases including Rho-associated kinse.This article reviews the mechanisms of action on Rho-associated kinase in the pathophysiological processes of cerebrovascular diseases.
4.PRODUCTION OF ACTIVE OXYGEN SPECIES AND CHANGES OF ENZYMES IN GRIFOLA UMBELLATA INDUCED BYARMILLARIA MELLEA ELICITOR

The changes of active oxygen species and some enzymes in Grifola umbellata induced by Armillaria mellea elicitor were studied.The results showed that active oxygen species appeared in both mycelia and sclerotia of G.umbellata after treated with A.mellea.There were two phases of active oxygen production upon addition of A.mellea elicitor.Phase I occured at 10 minute after addition of A.mellea elicitor.Phase Ⅱ occurred about 90 minute.The changes of some enzyme activity were also studied in this paper.Compared with control,the A.mellea elicitor could reduce the activity of superoxide dismutase and peroxidase.The catalase activity changed only little.The phenylanine ammonia lyase activity declined in the early stages and then increased in the late stages.
5.Expression of DSP,DMP1,CBFA1,BMP2 in rat dental pulp stem cells

Objective:To examine the expression pattern of DSP,DMP1,CBFA1,BMP2 in rat dental pulp stem cells(RDPSCs).Methods:Immunohistochemical staining was carried out using antibodies against DSP,DMP1, CBFA1 ,BMP2 in rat dental pulp stem cells. Mineralization was induced in the RDPSCs and expression of DSP and DMP1 was measured after induction.Results:CBFA1 and BMP2 were positive in RDPSCs. Only a few RDPSCs were stained positive for DMP1. DSP expression was observed in the minority of these cells. However, the majority of the RDPSCs were found strongly positive for DSP and DMP1 after mineralization induction.Conclusion:Positive expression of CBFA1 and BMP2 indicates the premature nature of RDPSCs. The dentin-specific expression of DSP demonstrates that the RDPSCs can differentiate along odontoblastic lineage.
6.Sleeping quality and the associated factors analyzing among clinical nurses

Objective To know the sleeping quality of clinical nurses in Urumqi, and analyze the influencing factors, put forward the corresponding countermeasures for different factors to improves the sleeping quality. Methods Randomly selected 407 nurses in clinical departments as the research objects in Urumqi top three hospitals, using the Pittsburgh sleep quality index scale (PSQI) and general situation questionnaire to investigate, and then analyzed the results of the survey. Results The incidence rate of sleeping disorders in emergency nurses was the highest (80.00%), followed by pediatric, intensive department, internal medicine, surgical department, operating room, outpatient and others was the lowest (39.02%). Gender (t=-2.015, P =0.045), age (F=8.509, P =0.000), establishment(F=3.609, P =0.013) were the influence factors of PSQI scores. Only the difference of the scores of hypnotic drugs between different ethnic groups was significant, P<0.05, while the others had no significant difference, P>0.05. Conclusions Poor sleeping quality of nurses existed in every department, its sleeping quality status, department, gender, age and establishment have certain correlation, interventions should be strengthened, to improve the overall level of sleeping.
7.Research of the Rho kinase in perihematomal cerebral tissue after experimental intracerebral hemorrhage

Objective To observe the expression of the Rho kinase in perihematomal cerebral tissue after experimental intracerebral hemorrhage(ICH) and the relationship between which and the neurological function deficit.Methods The ICH models rats were randomly divided into groups 1 d,3 d,5 d and 7 d after ICH.At the certain time point,the score of neurological function in each model was measured before killed.The expression of Rho kinase protein and mRNA were detected by immunohistochemistry and in situ hybridization.Results After ICH 1 d,3 d,5 d and 7 d groups,the scores of neurological function were 1.63,2.25,3.13 and 2.88,respectively;the number of Rho kinase protein positive cells in perihematomal cerebral tissue were 26.63,30.75,38.88 and 35.00,and the number of Rho kinase mRNA positive cells were 27.25,32.25,40.88 and 38.00,respectively.There were significantly differences among the groups 1 d,3 d and 5 d in the scores of neurological function and the expression of the Rho kinase protein and mRNA(P
8.Accuracy of Endometrial Sampling Device in Diagnosing Endometrial Lesions by Cytology

Objective To probe into the diagnostic accuracy of endometrial sampling device in the detection of endometrial lesions by cytology. Methods Endometrial cytological examination was performed on totally 193 out-patients from January to May 2008,the results were compared with the histological results by diagnostic curettage (D&C). Results The sampling qualified rate of endometrial sampling device was 93.8% (181/193) while that of D&C was 99.0% (191/193,?2=7.412,P=0.006). Thirteen patients were diagnosed with endometrial malignant lesions by D&C. In these patients,the sensitivity and specificity of cytology was 69.2% (9/13) and 100.0% (166/166),respectively;and its accuracy rate was 97.8% (175/179). By D&C 18 patients were diagnosed with endometrial hyperplasia,in whom the sensitivity and specificity of cytology was 27.8% (5/18) and 98.8% (159/161),respectively,and the accuracy rate was 91.6% (164/179). Ten patients were confirmed as having endometrial benign lesions without hyperplasia by D&C,for whom the sensitivity and specificity of cytology was 60.0% (6/10) and 98.8% (167/169) respectively,and the accuracy rate was 96.6% (173/179). The accuracy rate of endometrial cytological examination for 80 infertility patients with satisfactory sample was 100%,when the main objective was to determine whether the endometrial gland secreted or not. Conclusion Cytological examination by endometrial sampling device is feasible in the detection of endometrial malignant lesions and inspection of endometrium for infertility patients,but further discussion is necessary in the detection of endometrial benign lesions.
9.Mechanism of Anticancer Activities of the Extracts of Prescription 921

Objective To investigate the mechanism of anticancer activities of the extracts of prescription 921. Methods The inhibitory effects of the extracts of prescription 921 on DNA and RNA biosyntheses in cultured prostate PC-3M were investigated with 3H-TdR or 3H-UR incorporation. Flowcytometry was applied to analyze the regulating cell cycle of the extracts of prescription 921. The expression of cell cycle related factors CyclinA, CyclinB, p21 and p27 were detected by Western-blot. Results The extracts of prescription 921 showed inhibitory effect on DNA biosyntheses. The cells of PC-3M were arrested in S-phage when treated with the extracts for 72 h. The expression level of CyclinA was down-regulated while expression of CyclinB, p21 and p27 was not altered. Conclusion The extracts of prescription of 921 play its anticancer activities by inhibiting DNA biosyntheses and arresting the cells in S-phage by down-regulating the expression of Cyclin A.
10.Construction of pIRES2-hBSP-EGFP vector and stably expression in MDA-MB-231BO breast cancer cells

Objective To constitute a higher expression system and to obtain the breast cancer cell strains in which BSP is stably expressed. Methods hBSP gene was subcloned from pB-hBSP vector by PCR. The PCR fragment was inserted into the eukaryon expression vector, pIRES2-EGFP, which allow exogenous protein and EGFP to express respectively. The recombinant vector, pIRES2-hBSP-EGFP, was transfected into human breast cancer cells with Lipofectamine TM 2000. The expression of symbol protein EGFP, could be conveniently observed with fluorescent microscope. Results The recombinant pIRES2-hBSP-EGFP plasmid was constituted and successfully transfected into breast cancer cells. In the breast cancer cell strain hBSP and EGFP were expressed. Conclusion The successful constitution and transfection of hBSP and EGFP nonfusion expression vector laid a foundation for the further study on the effect of BSP on breast cancer metastasizing to bone in vivo or in vitro.
