1.Isolation,purification and culture in vitro of stromal cells of human endometrium

Objective To isolate,purify human endometrial stromal cells,and culture these cells in vitro,for the further research of pathogenesis of intrauterine adhesions.Methods Human endometrial stromal cells were separated and purificated by enzymo ly-sis,mesh filtration and pasted wall,then the stromal cells of purification were cultured.Phenotype of stromal cells was identified by immunohistochemistry.Results The stromal cells became adherent 30 min after culture.Two types of stromal cells were found. One was spindle and the other was polygonal.Immunocytochemical staining method revealed that vimentin was positive in both types of stromal cells,and the positive rate was above 9 5%;meanwhile,cytokeratin was negative in both types of stromal cells, which indicated that these cells were endometrial stromal cells.The isolated endometrial stromal cells can be cultured in vitro.Con-clusion Highly purified stromal cells of human endometrium could be obtained by enzymolysis,mesh filtration and pasted wall pur-ification,and the cultured stromal cells could passage to limited generations.
2.The combination ofKangfuxing liquid, ethacridine lactate and local oxygen therapy in the treatment of radiation-induced skin damage

Objective To investigate the efficiency ofKangfuxing liquid, ethacridine lactate in combination with local oxygen therapy for radiation-induced skin damage.Methods A total of 96 patients affected by radiation-induced skin damage were enrolled and randomly allocated to a treatment group and a control group by random number table method, with 48 patients in each group. All patients in the control group were treated with trolamine cream, and those in the treatment group were treated withKangfuxing liquid, ethacridine lactate in combination with local oxygen therapy. Skin dermatitis was scored utilizing Radiation Therapy Oncology Group (RTOG) toxicity scale.Results The total effective rate in the treatment group was significantly higher than that in the control group (97.9%vs.79.1%;χ2=10.119,P=0.002). The distribution of patients with RTOG grades 0 to 4 showed different degrees of improvement, and the number of patients with RTOG 0-4 grades changed from 0, 3, 9, 12, 24 before the treatment to 8, 12, 22, 6, 0 after the treatment in the treatment group, 0, 2, 10, 25, 11 to 2, 5, 18, 23, 0 in the control group. The percent of patients with RTOG grade 0 in the treatment group was significantly increased than that in the control group (16.7%vs. 4.2%;χ2=4.019, P=0.045) and the percent of patients with RTOG grade 3 significantly decreased (12.5%vs.47.9%;χ2=12.649, P<0.001). The percents of patients with dispirited spirit (31.3%vs. 58.3%;χ2=6.066,P=0.014), dry and chapped skin (20.8%vs. 58.3%;χ2=12.588,P=0.000) in the treatment group were all significantly lower than those in the control group.ConclusionKangfuxing Liquid, ethacridine lactate in combination with local oxygen therapy can improve radiation-induced skin damage.
3.Application of sinusitis mixture on functional endoscopic sinus after surgery

Objective To investigate effects of sinusitis mixture on functional endoscopic sinus after surgery .Methods According random number table,36 cases of chronic nasosinusitis patients were divided into the treatment group and control group,the two groups were routinely 2%NaCl saline nasal irrigation,use of nasal corticosteroids and mucous discharging agent mixture used in postoperative patients with sinusitis in the treatment group oral therapy. Results The treatment group 100%,total efficiency in the control group (89%) (χ2 =4.36,P<0.05),there was a statistically significant difference.Conclusion Sinusitis mixture has significant efficacy on the recovery of patients afterfunctional endoscopic sinus surgery.

This study was conducted under the background of the internationalization of traditional Chinese medicine (TCM) terminology. Current TCM symptom terminology related to sweat was taken as basis in the extended study. The ICD classification of multiple axis and hierarchical level was used as guidance thought. This paper proposed the principle of detailed classification of TCM sweat symptoms and finally made classification rules for TCM sweatsymptoms. This study has promoted the development of TCM terminology standardization.
5.Clinical Observation of Lung Protective Ventilation Strategy in Gynecologic Laparoscopic Surgery

Objective:By monitoring the lung compliance,oxygenation index(OI),CC16、IL-8 in plasma,observe whether the protective lung ventilation in patients undergoing gynecological laparoscopic surgery can improve the respiratoryw function and reduce lung injury.Methods:40 cases of ASA grade Ⅰ-Ⅱ were selected,and 40 patients were treated by elective laparoscopic radical resection of cervical cancer.The patients were divided into two groups according to the random number table method in 40 cases.Mechanical ventilation in general anesthesia,A group used traditional intermittent positive pressure ventilation mode,tidal volume is set to 10ml/KG (PBW);B group used lung protective ventilation strategy,tidal volume is set to 6ml/KG (PBW) + 5cmH2O PEEP + recruitment manoeuvres.For patients before pneumoperitoneum(T0),2 hours after pneumoperitoneum (T1),4 hours after pneumoperitoneum (T2)MAP,HR,CVP,RR and C were observed.In pneumoperitoneum (T0),2 hours after pneumoperitoneum(T1),4 hours after pneumoperitoneum (T2),2 hours after operation(T3),24 hours after surgery(T4) venous,IL-8,CC16 content detection.Results:Before pneumoperitoneum,two groups of patients with pulmonary compliancethe difference was statistically significant(P <0.05).Compared T1 、T2 with T0 in A group and B group,the pulmonary compliance all decreased and there existed significant deference(P<0.05).Notably,A group decreased evidently(P<0.05).T0,T1,T3,T4 time points in two groups of patients with oxygenation index,the difference was statistically significant (P < 0.05).Two groups of patients with CC16,IL-8content in T1,T2,T3,T4 time point,the difference was statistically significant (P < 0.05).Conclusions:Lung protective ventilation strategy can improve respiratory function in patients with gynecological laparoscopic surgery,and reduce the incidence of lung injury.
6.Effect of Hushengujingfang on the Proliferation of Rat Mesangial Cells

Objective To study the effect of the Hushengujingfang (HSGJF), a traditional Chinese prescription, on the biological behaviour of rat mesangial cell (MC). Methods MC was cultured in vitro, and the serologic pharmacology method was employed. The cells were divided into serum groups of HSGJF, LPS group and control group. Added the different concentration actors to MC and cocultured then made into unicell suspension. After examining it by flow cytometry, the percentage of G 0 /G 1 period, S period, G 2 /M period in cell cycle and the inhibition percentage of S period and cell proliferation index were calculated. Result The actors of different concentration had different effects to the multiplication cycle of MC and delayed the entry of cell cycle at G 0 /G 1 period. HSGJF induced apoptosis of MC. Conclusion One of action mechanism of HSGJF treating mesangial proliferative nephritis and other renal glomerular diseases is to inhibit MC proliferation and promote apoptosis.
7.Adsorptive Voltammetric Behavior of Riboflavine Tetrabut yrate and Its Measurement of Rate Constant at the Mercury Electrode

Cyclic voltammetry shows that, in a supporting ele ctrolyte of NaOH,riboflavine tetrabutyrate(RT)has a pair of cathodic and anodi c peaks at Hg electrode. The peak potentials Epc=Epa=-0.64 V(vs.Ag/Agcl). The adsorptive characteristics of RT are explored in detai ls with various methods. The adsorbed species is most probably neutral molecule of RT.The saturated adsorption amount of RT at Hg electrode is 5.42×10-11 mol/cm2 and every RT molecule occupied 3.06 nm2. On the surface of HMDE ,the adsorption of RT obeys Frumkin dasorption isotherm. The adsorptive coeffic ient β,the attractive factor γ are 4.60×105,1.10 and the adsorption free energy ΔG, is -32.30 kJ/mol, respectively. The electrode proc ess kinetic behavior has also been studied by using cyclic voltammetry. Under th e experimental conditions the rate constants for electron transfer are determine d. The values of ks are in good agreement with literature′s results.
8.Effect of photon on blood biologic components in photochemical treatment

Objective To explore the effect of photon on blood biologic components in blood photochemical treatment. Methods After the blood sample was adjusted to an appropriate density, it was treated with 0.1 nmol/ml 8-MOP (8-methoxypsoralen), 0.1 nmol/ml TFO (triple helix-forming oligonuletide) and UVA (ultraviolet A radiation) at the intensity of 1 800 μW/cm2 for 3~20 min. The changes of biologic activities of major components in blood were measured with automatic blood gas analyzer, platelet aggregation analyzer, blood coagulation analyzer, micropipette aspiration system and assay of poly-lysine adsorption. Results The oxygen content in blood was increased gradually. The resilience of erythrocyte was enhanced ,but its adhesiveness was decreased. The parameters related to blood coagulation had some changes but all remained within the normal ranges. Conclusion Under the definite condition of blood virus being inactivated effectively, the nonspecific effect of photosensitive response may improve blood oxygen content, enhance the transfiguring ability of erythrocyte and decrease the blood viscosity, but having no obvious change on blood coagulation.
9.Cloning and prokaryotic expression of rat surfactant protein C in E.coli

Objectives To construct prokaryotic expression plasmid of rat surfactant protein C (sp-c) gene under hyperoxia and expression in E.coli. Methods Twenty-one-day-old SD premature rats were exposed to 85%hyperoxia 12 hours after birth. The rats were executed after 7 days and their RNA were extracted from lung and cDNA was synthesized and amplified. And then the cDNA was cloned into pMD18-T vector and confirmed by enzyme digestion and sequencing. After the prokaryotic expression vector pET-28a(+)-sp-c was constructed, the recombinant plasmid was induced by IPTG and expressed in E.coli BL21 strain. The fusion protein was analyzed by SDS-PAGE and Western blotting. Results The pET-28a(+)-sp-c plasmid was constructed and the fusion protein with relative molecule mass of 21000 was highly expressed. Conclusions SP-C is successfully expressed in E. coli, which can be used to study the mechanism of action between SP-C and endoplasmic reticulum, the expression of SP-C in lung epi-thelial cell A549 and the impact of SP-C on proliferation, differentiation and apoptosis of AECIIs in future.
10.Expression of GRP78 and CHOP in lungs of rats with bronchopulmonary dysplasia and their relationship with pneumonocyte apoptosis

Objectives To investigate the expressions of endoplasmic reticulum stress (ERS)-related factors, glucose regulated protein 78 (GRP78) and C/EBP homologous protein (CHOP), in lungs of rats with bronchopulmonary dysplasia (BPD) and their relationship with pneumonocyte apoptosis, and further to explore the role of ERS-induced apoptosis in the pathogen-esis of BPD. Methods A total of 24 neonatal SD rats were randomly divided into control group and BPD group with 12 rats each. Rats in BPD group were exposed to 85%O2, while rats in the control group were exposed to air. Four rats in each group were sacriifced at 7, 14 and 21 days respectively after exposure. The expression of GRP78 in the lung tissues was examined by immunohistochemistry, the mRNA and protein levels of GRP78 and CHOP were detected respectively by real-time PCR and Western blot, and the apoptosis in lung cells were evaluated by TdT-mediated dUTP nick end labeling (TUNEL) assay. Results The mRNA and protein levels of GRP78 and CHOP, and the cell apoptosis in BPD group were signiifcantly different from those in control group (P<0.01) at different time points and increased over the time of hyperoxic exposure (P<0.01). Conclusions En-doplasmic reticulum stress may be initiated by hyperoxic exposure and apoptosis is induced via CHOP signal pathway, which is involved in the pathogenesis of BPD.
