1.Inhibitory effects of antisense Snail on invasion and migration of ovarian carcinoma cells

Objective To investigate the inhibitory effects of Snail—— a zinc finger transcription factor, anti-sense plasmid on the invasion of ovary carcinoma cell lines in vitro. Methods Four human ovarian carcinoma cell lines ES-2,OVCAR3,HO8910,HO8910PM were analyzed for the expression of Snail and E-cadherin mRNA by RT-PCR.Then anti-Snail plasmid transfection was introduced into HO8910 cells using lipofectin 2000 reagent to investigate the inverse correlation between E-cadherin and Snail expression, and the inhibitory effects of anti-sense Snail were also detected by Transwell motility assay and Matrigel invasion assay. Results The results demonstrated the presence of Snail mRNA in ES-2,HO8910,HO8910PM detected by RT-PCR. By contrast, E-cadherin mRNA was only detected in OVCAR3 by RT-PCR.The inverse relationships were further observed by transient transfection of anti-sense Snail into HO8910 cells, which showed the down-regulated expression of Snail and the re-expression of E-cadherin. Objective To study the effect of Gadd genes expression in surveillance of DNA damage. Methods The human liver tumor cells were treated with alkylating agent(nitrogen mustard) and total RNA was extracted, the expression levels of Gadd genes were determined by RT-PCR. Results Treated with 1 ng/?l nitrogen mustard for 3 h, 24 h, 48 h respectively and with 1 ng/?l, 5 ng/?l nitrogen mustard for 24 h respectively, the expression levels of Gadd genes increased at first (P0.05). Conclusion Gadd genes expression can be used as the indexes in the surveillance of DNA damage induced by nitrogen mustard.

ObjectiveTo investigate the application effect of Delphi method to establish the preoperative visit mode. MethodsTaking the Delphi method to establish the standardized procedures of preoperative visit.The average length of stay,hospital costs and satisfaction degree of patients before and after application of standardized procedures of preoperative visit were compared. ResultsThe study showed that after taking the Delphi method to establish the preoperative visit mode,the patients' hospital stay for the same type of surgery,treatment costs and satisfaction degree of patients improved significantly than those before.The differences before and after treatment were significant. ConclusionsUsing Delphi method to establish a standardized procedures of preoperative visit has the advantage of strong target,strong representation,high degree of authority.It takes advantage of the excellent medical care experience and provides uniform and stable preoperative preparation work.It lays a foundation for patients to get better medical care and is worthy of clinical application.
5.Activity characteristics of diverse autonomic nervous system of armymen with different mental quality levels during dread emotion processes

ObjectiveTo explore activity characteristics of diverse autonomic nervous system of armymen with different mental quality levels during dread emotion processes.MethodsMental Quality Questionnaire of Armymen was used to select participants.The activities of the diverse autonomic nervous system of armymen withdifferent mental quality levels during dread emotion processes were discussed by physiological - psychological experiments.Results ( 1 ) Under neutral condition blood volume pulse (BPV) is greater in low mental quality group( ( -0.87 ±0.64)mm Hg) than in high( -0.28 ±0.24)mm Hg).(2) During dread emotion processes the normalized low frequency spectral component(LF) and skin conductance (SC) are greater in low mental quality group ( (6.54 ± 2.96) nU),(0.78 ± 0.11 ) umho) than in moderate ( ( 1.32 ± 1.25 ) nU,( (0.22 ± 0.15 ) umho) and high ((3.51 ±2.28 )nU,((0.30 ±0.12) umho).(3) During dread emotion processes the recovery time of HR and the average recovery time of skin conductance( SC ),skin temperature( ST),blood volume pulse(BVP) and HR are greater in low mental quality ( 61.58 ± 35.66 ) s,( 36.78 ± 18.58 ) s ) than in high ( ( 22.36 ± 6.87 ) s,(56.87 ± 23.98 ) s).(4) In emotional arousal,and low mental quality (0.88 ± 2.54 ) in neutral conditions change their arousal'was significantly greater than the middle (0.57 ± 2.88) and high psychological quality group (0.15 ± 1.87) ; Low mental quality group (2.17 ±2.39) under the conditions of pleasure in the dread was significantlygreater than the middle ( 1.87 ± 1.45 ) and high psychological quality group ( 1.24 ± 1.66).ConclusionThe findings indicate that armymen with low mental quality is more likely to show corresponding physiological arousal and alert status in face of the neutral or weak emotional stimuli.When facing dread emotional stimuli,they would show stronger physiological arousal mainly basing on the excitement of sympathetic nerve system,and such statuswould last much longer to bring about more fierce pain experience.

Early diagnosis and accurate staging are important for planning therapeutic intervention to improve outcomes in patients with malignant melanoma (MM).Noninvasive molecular imaging has been extensively studied in cancer diagnosis,staging,therapeutic evaluation,and prognosis.Several radiopharmaceuticals have been developed to diagnose and treat MM,such as monoclonal antibodies,melanocortin receptors,benzamide (BZA) and BZA analogs.In this review,the characteristics and limitations of these radionuclide molecular probes are summarized and discussed.
7.Clinical Observation of Recombinant Human Brain Natriuretic Peptide Combined with Levosimendan in the Treatment of Acute Decompensated Heart Failure

OBJECTIVE:To observe the clinical efficacy of recombinant human brain natriuretic peptide (rhBNP) combined with levosimendan in acute decompensated heart failure(ADHF). METHODS:In retrospective study,120 cases diagnosed as AD-HF were divided into observation group and control group according to treatment plan,with 60 cases in each group. The patients of control group received rhBNP on the basis of conventional treatment,intravenously pulsed with dose of 0.15 μg/kg firstly,then maintained dose of 0.007 5 μg/kg viacontinuous intravenous pumping for 24-72 h;On the basis of control group,treatment group received levosimendan with loading-dose of 12 μg/(kg·min)for 10 min,maintenance dose of 0.1 μg/(kg·min)via intravenous pump,for 24 h,adjusted according to clinical manifestations of patients. The vital signs,hemodynamic and UCG indexes,the rate of dyspnea recovery,plasma level of BNP,urine and the occurrence of ADR were recorded in 2 groups. RESULTS:The vital sign and hemodynamic indexes of control group had no significant change 1 h after treatment;the levels of HR,RR,SBP,MPAP and MPCWP in 2 groups after treatment were significantly lower than before,while the levels of SaO2 and CO were significantly higher than before,with statistical significance (P<0.05). The levels of HR,RR,SBP,MPAP and MPCWP in observation group 1,2, 4 h after treatment were significantly lower than in control group,while the levels of SaO2 and CO were significantly higher than control group,with statistical significance (P<0.05);there was no statistical significance in vital sign and hemodynamic indexes between 2 groups at other time points (P>0.05). 48 h after treatment,LVEF of 2 groups were increased while plasma levels of BNP were decreased,compared to before treatment;the improvement of observation group was more significant than control group,with statistical significance(P<0.05). And the urine volume of observation group were significantly higher than that of con-trol group 24,48 h after treatment,with statistical significance(P<0.05). There was no statistical significance in the rate of dys-pnea recovery at different time points and the incidence of ADR after treatment between 2 groups (P>0.05). CONCLU-SIONS:rhBNP combined with levosimendan could effectively improve vital sign,hemodynamic indexes,UCG indexes and dys-pnea,and decrease the plasma level of BNP with good safety.

Objective:To develop a personality disorder inventory for undergraduates. Method:Based on three diagnostic criteria and clinic experience,the questions were collected after item revision and item analysis of three samples. A total of 425 undergraduates responded to the formal questionnaire. The data of formal questionnaire were used to evaluate the reliability and quality.Result:The whole questionnaire was compose of 160 items including five repeated items for reliability of the respondents. The whole inventory was consisted of ten factors, they were the borderline, the antisocial, the schizoid, the schizotypal, the avoidant, the paranoid, the obsessive-compulsive, the histrionic, the dependent, and the narcissistic. 9.Clinical and image features of breast cancer in early stage

Objective To study the features of mammography,breast ultrasonography(US)and physical examination(PE)on 0-Ⅰstage(early stage)breast cancer,and to approach a diagnostic measure for early stage breast cancer(EBC).Methods Physical examination,ultrasonography and mammography were performed in 28 patients with pathologically proved breast cancer in 0-Ⅰ stage,and the data were analyzed.Results The accuracy of PE for 0-Ⅰstage of EBC was 57.1%(16/28),and the accuracy by US or mammography was 85.7%(24/28)and 89.3%(25/28),respectively.As the main clinical symptom of 0-Ⅰ stage EBC,breast lumps were found in 14 of 16 PE positive cases(87.5%).The main feature of EBC by mammography was the finding of a lump(14/25,56%).The low echo of a solid space occupying lesion was the main US feature of EBC(18/24,75%).The diagnostic accuracy of mammography for 0 stage EBC was 100.0%(8/8),while only 50%(4/8)by both US and PE.Malignant calcification was the main feature of o stage breast cancer in mammography.The diagnostic accuracy of US,mammography and PE for I stage EBC was 100.0%(20/20),and was 85.0%(17/20)and 40%(8/20),respectively.US was advantagious in detecting cancer in young patient with compact breast.Conclusion Early stage breast cancer often displayed atypical features in PE,US and mammography,which are important for the early-diagnosis of breast cancer,and false negative findings are inevitable in examination.Therefore,the diagnosis of early stage breast cancer should be made with.The findings of PE,US and mammography should be analyzed comprehensively,however,when one of examinations gives a positive finding,the diagnosis of breast cancer should be suspected,and further examination should be carried out to make an early diagnosis.
10.Evaluation of clinical physical examination, ultrasonography and mammography in the diagnosis of breast cancer in Chinese

Objective To evaluate the significance of clinical physical examination, breast ultrasonography, and mammography in the diagnosis of breast cancer in Chinese, to compare their sensitivity, specificity and accuracy, and to elucidate the significance of combined application of the 3 methods. Methods A total of 112 patients suspected to have breast cancer as a result of mass screening (38 cases) and OPD examination (74 cases), were subjected to clinical physical examination, breast ultrasonography and mammography, and biopsy specimens were obtained from all the cases by puncture or surgery, then the final diagnosis was confirmed by pathological examination. The diagnosis made from clinical physical examination, breast ultrasonography or mammography was compared with pathological findings, and the sensitivity, specificity, accuracy, positive predictive value, and Kappa index of the 3 methods were then calculated. Results 61 cases of breast cancer and 51 cases of benign breast disease were identified. The diagnostic sensitivity of clinical physical examination, ultrasonography and mammography was respectively 68.85%, 88.52% and 72.13%, the specificity was respectively 88.23%, 21.57% and 56.86%, while the accuracy was respectively 77.68%, 58.04% and 65.18%.
