1.Effect of hesperidin on locomotor activity of mice with infective brain injury induced by LPS

Objective To study the effect of hesperidin on the locomotor activity of mice with infective brain injury induced by li-popolysaccharide (LPS)in the dark field environment.Methods 45 ICR mice were randomly divided into 3 groups:Saline group, LPS group and LPS+Hesperidin group,15 mice in each group.The moving distance and the number of crossing line in the dark field were recorded using the ZH-ZFT locomotor activity video analysis system.ResultsThe LPS group had significantly shorter moving distance as well as lower frequency of the number of crossing line than those of the Saline group(P<0.05);compared with the LPS group,the Hesp group moved longer and crossed line more frequently,the differences between the two groups had statisti-cal significance (P<0.05).Conclusion Hesperidin could significantly improve the capacity for action in the dark field of mice with infective brain inj ury induced by LPS.
2.Therapeutic effects of modified Bushen Huoxue granules on ovariecto-mized osteoporosis rats

AIM: To observe the therapeutic effect of modified Bushen Huoxue granules (MBHG) on ovariec-tomized osteoporosis rats.METHODS: Female SD rats were randomly given sham operation (n =10) and ovariectomy, and then the model rats were further randomly divided into model group, MBHG treatment groups at doses of 200 mg/kg, 100 mg/kg and 50 mg/kg respectively, and positive control (estradiol valerate) group, with 10 rats in each group by intra-gastric administration for 12 weeks.The morphology, area, thickness, spacing and area percentage of trabecular bone in the rats were observed.The serum levels of calcium (Ca), phosphorus (P) and alkaline phosphatase (ALP) were mea-sured by automatic analyzer.Bone mineral density (BMD) was analyzed.Serum estradiol (E2 ), osteocalcin (BGP), os-teoprotegerin (OPG), receptor activator of nuclear factor-κB (RANK) and receptor activator of nuclear factor κB ligand (RANKL) levels were detected by ELISA.RESULTS: Compared with model group, trabecular bone significantly widened in all treatment groups with large number, and net-like structure restored partially.The thickness, area and area percenta-ges of trabecular bone in treatments groups were higher than those in model group,and trabecular spacing was less than that in model group (P <0.05).The serum Ca, P, E2 and OPG, and femoral BMD were significantly higher in treatment groups than those in model group, and the levels of ALP, BGP, RANK and RANKL were significantly lower than those in model group (P <0.01).CONCLUSION: MBHG has a significant therapeutic effect on ovariectomized osteoporosis rats. The mechanism may be related to the regulation of OPG and inhibition of RANKL secretion.

Methods:Fifty C57BL/Ksj-db/+m mice were randomly divided into 2 groups, db/db diabetic group and control group. The morphological changes of submandibular gland and the expression of PCNA were observed by using light microscope (LM) and immunohistochemistry. AIM: To investigate biological effect of endothelin-1 (ET-1) on differentiation of cardiomyogenic cells (CGCs) induced from rabbit bone mesenchymal stem cell (BMSCs) in vitro. METHODS: The BMSCs from shaft of femur of Japan rabbits were isolated and propagated for cell culture. Cultured BMSCs were randomly divided into six groups: Group Ⅰ: the control group (uninduced group); Group Ⅱ: the ET-1 group, added ET-1 (30 nmol/L) into culture medium; Group Ⅲ: 5-aza group, added 5-aza (10 ?mol/L) into culture medium; Group Ⅳ: 5-aza+ET-1 group, after inducing with 5-aza for 3 weeks, ET-1 was added into culture medium. This group was divided into three parts-Ⅳ 1、Ⅳ 2、Ⅳ 3, and separated to add 10、 30、 50 nmol/L ET-1. During the cell culture, growing and differentiation of BMSCs were observed. After inducing for 4 weeks, the differentiation rate and the diameter of cardiomyogenic cells were calculated; Western-blot was employed to analyze the expressions of GATA-4 protein and phosphorylation level. The expression of ?-MHC mRNA was assessed by RT-PCR. Immunohistochemistry staining of Troponin-I and ultrastructure observation of induced cardiomyogenic cells were also completed simultaneously. Objective To study the dynamic changes of regional cerebral blood flow(rCBF) in hematoma, perihematoma and their enantiomorph in the contralateral hemisphere in the patients with acute intracerebral hemorrhage(ICH). Methods 15 patients of acute basal ganglia hemorrhage were divided into different groups according to the Chinese Stroke Scales, 10 patients were minor, the other 5 patients were moderate. 99mtechnetium-ethyl -cysteinate-dimer single-photon emission CT ( 99mTc-ECD SPECT) were performed within 3 days after onset of the disease and re-examined at 16 days. The method of region of interesting (ROI) was used to measure the uptake counts in the core of hematoma, frontal or parietal region around the lesion, the center of cerebellum and their enantiomorphs in the contralateral hemisphere, respectively,and to calculate the ratio of uptake counts.Results The uptake counts showed difference in the second time exam between perilesion and its enantiomorph in minor patients ( P0.05). 6.Molecular mechanism of Wendan Decoction in treating hypertension based on network pharmacology

Objective:To explore the molecular mechanism of Wendan Decoction in the treatment of Hypertension by the network pharmacolog. Methods:The potential active components and targets of Wendan Decoction were screened in TCMSP and TCMIP database. The TTD, genecards and OMIM databases were used to collect hypertension targets. Then the protein interaction was analyzed by using String database. The software of Cytoscape v3.7.1 was used to construct the network diagram of Wendan Decoction in the treatment of Hypertension and using the software of ClueGO to analyze GeneOntology and KEGG pathway of the potential targets. Finally, we used molecular docking technology for the appropriate of molecules and targets. Results:A total of 141 potential active components and 108 potential therapeutic targets were selected from Wendan Decoction. And 20 of core targets selected by MCC in CytoHubba. The GeneOntology analysis contained 164 GO terms, 133 of biological processes, 24 of molecular functions, and 7 of cell components. Seven key pathways were selected by enrichment analysis of KEGG pathway. Molecular docking results showed good docking between key components and targets.Conclusion:Wendan Decoction can treat hypertension through multiple ways and targets. This study predicts the possible mechanism of Wendan Decoction and provides data for its clinical application.
7.The clinical significance of serum and joint fluid osteopontin, and thrombin-cleaved osteopontin levels in osteoarthritis

Objective To explore the relationship between osteopontin (OPN),its downstream product thrombin-cleaved osteopontin (NTOPN) and radiographic changes in osteoarthritis (OA).Methods The levels of OPN and NTOPN in serum and synovial fluid were tested by enzyme-linked immuno sorbent assay (ELISA) kit in 22 OA patients and 21 health controls.The correlation between the above parameters and clinical data such as radiology changes of OA patients was analyzed.Results The serum level of OPN in the OA group was positively correlated with C reactive protein (CRP) (r =0.574,P =0.005).The serum level of OPN in the OA group was slightly lower than that in the healthy control group [(1.8 ± 0.5) μg/L vs (1.7 ± 1.1) μg/L,P > 0.05].Similarly,the serum level of NTOPN in the OA group was significantly higher than that in control group [(21.1 ± 11.7) μg/L vs (14.7 ± 3.6) μg/L,P < 0.05].Meanwhile,serum NTOPN level in female patients was significantly higher than that in male patients [(23.2 ± 11.80) μg/L vs (11.1 ± 4.9) μg/L,P < 0.05].In synovial fluid of OA group,the level of OPN was (7.75 ± 2.33) μg/L,NTOPN was (0.91 ± 0.49) μg/L and the ratio of NTOPN/OPN was 0.12 ± 0.08.The level of OPN of synovial fluid in the OA group was positively correlated with CRP (r =0.574,P =0.005).NTOPN-to-OPN ratio of synovial fluid in the OA group was positively correlated with Kellgren-Lawrence grade (r =0.535,P =0.022).Conclusions The serum level of OPN is correlated with severity of inflammation in the OA patients.Meanwhile,NTOPN-to-OPN ratio of synovial fluid is correlated with OA severity.As a downstream bioactive product of OPN,NTOPN may play an important role in OA pathogenesis.Whether these parameters can be used as predictors of joint lesion severity and inflammatory status is worth further study.

Objective The medical code of biology-psychology-societyplays a vital role in modern treatment .The arti-cle aimed to investigate the clinical value of He-Ne laser combined with psychological intervention in the treatment of acute herpes and zoster. Methods Retrospective analysis was made on the clinical data of 81 patients with acute herpes and zoster in our hospital from Jan 2015 to Jan 2016 .The patients were divided into 3 groups randomly .Patients in group A were treated with antiviral and neu-rotrophic medication , psychological intervention and analgesic drugs . Patients in group B were treated with antiviral and neurotrophic medi-cation, psychological intervention and He-Ne laser.Patients in group C were treated with antiviral and neurotrophic medication , psychologi-cal intervention, analgesic drugs and He-Ne laser.Visual analogue scale ( VAS) was evaluated before treatment in hospital on the 3rd day, the 5th day of treatment, and telephone follow -up after 4 weeks of treatment.VAS and average length of stay were analyzed . Results No difference was found as to VAS before treatment in hospital among three groups (8.1 ±1.1, 8.1 ±0.9 vs 7.9 ±1.0, P<0.05).VAS was decreased in all groups after nursing propaganda , psychological intervention and placebo (P<0.05).Compared with the VAS before treatment, VAS in three groups decreased significantly on the 3rd day and the 5th day of treatment.From the re-sults on the 5th day and telephone follow -up after 4 weeks of treatment , VAS was still at a low level in group A and group B , on the contrary, VAS was decreased continuously in group C . Conclusion Combined treatment of antiviral , neurotrophic, analgesic drugs and He-Ne laser can relieve pain , decrease VAS and shorten the average length of stay for patients with acute herpes and zoster .
9.CT Diagnosis and Types of Malignant Ovarian Tumor

Objective To improve the CT diagnostic rate of malignant ovarian tumor(MOT).Methods The CT appearences of 42 cases with MOT confirmed by operation and pathology were analyzed retropectively.The CT findings and types,differential diagnosis were emphatically discussed.Results (1)Ovanian tumors were most frequently located in the adnexal and the pouch of Douglas(78.6%);(2)The size of tumors was mostly beyond 5 cm in diameter(95.2%);(3)The most of tumors apperared flower-shaped(71.4%);(4)The tumors were classified as cystic,cyst-solid and solid types,but cyst-solid type was common(61.9%);(5)The solid and solid portion of the mass were enhanced obviously(90.8%);(6)Ascites and invasion of the adjacent organs were common appearences of the metastasis;(7)Arched,small nodelar and nodular calcification in a solid mass or solid portion of mass could be found.Conclusion CT examination is of great value in diagnosis of MOT,which may predict the pathologic characteristics of MOT.
10.Quantitative detection of Porphyromonas gingivalis by real-time PCR of orthodontic patients after removal of orthodontic appliances

Objective To detect Porphyromonas gingivalis and clinical index in subgingival specimens after removal of orthodontic appliances and to evaluate clinical index and P.gingivalis factors associated with orthodontic appliances during an episode of gingival inflammation and the impact of appliance removal on periodontal health.Methods Choose 20 patients prepare to removal of orthodontic appliances.Plaque index(PLI),sulcus bleeding index(SBI) and probing depth(PD) of observed teeth were examined and subgingival plaques were collected at prior to appliance removal(baseline) and 1,3 and 6 months after appliance removal.The numbers of P.gingivalis,total bacteria,the proportion of P.gingivalis and the prevalence of P.gingivalis in the samples were detected with the TaqMan real-time PCR.Results The prevalence of P.gingivalis decreased from baseline to the 6th month after fixed appliance removal in two groups,and significant change was found in 6th month(P < 0.05); The numbers of P.gingivalis significantly reduced from baseline to the 6th month(P < 0.05); The proportion of P.gingivalis in subgingival plaque decreased from baseline to the 6th month(P <0.05).PLI,SBI and PD significantly decreased from baseline to the 6th month after fixed appliance removal.Conclusion After fixed appliance removal,most of patients can recover periodontal tissue health in good oral hygiene.The percentage of P.gingivalis are closely associated with periodontal status.TaqMan real-time polymerase chain reaction has a high sensitivity and specificity.It was demonstrated the broad potential of application on detection of oral microbes.
