1.Investigation of the effect of verapamil and chemotherapeutic agents on the P-glucoprotein of the KBV200 cell line

Objective: To observe the effect of verapamil and 6 chemotherapeutic agents vincristine,adriamycin,bleomycin,cyclophosphamide,5-Fu,cisplatin on the P-glucoprotein(P-gp) of the KBV200 cell line.Methods: Flow cytometer(FCM) assay was used to investigate the expression of P-gp on the KBV200 cell line in the different conditions.Results: Verapamil act with vincristine,adriamycin,pingyangmycin and cyclophosphamide separately decreased expression of P-gp on KBV200 cell line by 80.17%,89.69%,19.19% and 53.48% Among all the combinations,the synergistic effects of varapamil act with vincristine and adriamycine show a better effect.Conclusion: Verapamil act with vincristine,adriamycin,pingyangmycin and cyclophosphamide separately can decrease expression of P-gp on KBV200 cell line significantly,and can reverse the drug resistance of KBV200 cell line.

Objective: To make a study of KBV200 cell line on its multidrug resistance and provide experimental data for clinical individual chemotherapy.Methods:MTT reduction assay was used to investgate the inhibition of 8 common chemotherapetic agents on KBV200 cell line.Result:Vincristine、Adriamycin、Bleomycin、Cyclophosphamide and Arabinosyl Cytosine have poorer inhibition than Harrihgtonine、Cicplatin and 5-Fu on KBV200 cell line.Conclusion:KBV200 cell line is multidrug resistance to Vincristine、Adriamycin、Bleomycin、Cyclophosphamide and Arabinosyl Cytosine.
3.Bioequivalence of Ambroxol Hydrochloride Orally Disintegrating Tablets

OBJECTIVE:To study the pharmacokinetic profiles and bioequivalence of test ambroxol hydrochloride vs.refe-rence preparation.METHODS:18 healthy volunteers were administered with a single oral dose of test(ambroxol hydrochloride orally disintegrating tablets)and reference ambroxol hydrochloride(60 mg)by randomized crossover way.The plasma concentrations of ambroxol hydrochloride were determined by LC-MS.The pharmacokinetic parameters and the bioavailability were computed using DAS 1.0 program.RESULTS:The main pharmacokinetic parameters of test and reference ambroxol hydrochloride were as follows:Cmax:(84.7?29.3)and(81.2?28.2)ng?mL-1;tmax:(1.7?0.5)and(1.8?0.4)h;t1/2:(7.9?0.9)and(8.3?1.0)h;AUC0~24:(638.7?180.8)and(591.4?148.3)ng?h?mL-1;AUC0~∞:(717.5?211.1)and(671.1?173.2)ng?h?mL-1,respectively.The relative bioavailability of test ambroxol hydrochloride orally disintegratiog tablets was(108.2?16.5)%.CONCLUSION:The test and reference preparations of ambroxol hydrochloride were bioequivalent.
4.Inhibitory effect of 5 chemotherapeutic agents on KBV200 cell line and reverse effect on multidrug resistance of Matrine

Objective To observe the inhibitory effect of 5 chemotherapeutic agents including vincristine,adriamycin,pingyangmycin,cisplatin,cyclophosphamide on the KBV200 cell line and to study the reverse effect on the multidrug resistance of Matrine.Methods MTT assay was used to investigate the inhibition ability of 5 drugs on the cell line and the reverse effect on the multidrug resistance of Matrine.Results Vincristine,pingyangmycin,cyclophosphamide had less inhibitory effects than adriamycin,cisplatin on the KBV200 cell line.Matrine had the ability to reverse the multidrug resistance of the cell line to vincristine,adriamycin.Conclusion KBV200 cell line is multidrug-resistant to vincristine,pingyangmycin,cyclophosphamide.Matrine can enhance the sensitivity of KBV200 to the vincristine,adriamycin.
5.Clinical observation of injection of triamcinolone acetonide into the anterior chamber for anterior uveitis after cataract combined with anti-glaucoma surgery

Background Anterior uveitis is a common complication following the combination procedure of cataract extraction and anti-glaucoma surgery.More and more researches demonstrated the anti-inflammatory effectiveness of intraocular injection of triamcinolone acetonide (TA).Objective This study aims to observe the anti-inflammatory effectiveness of anterior chamber injection of TA for anterior uveitis following cataract extraction combined with anti-glaucoma surgery.Methods This is a case-control study.The clinical data from 42 eyes of 42 cases underwent the combination surgery of catarat extraction and anti-glaucoma was retrospectively reviewed and analyzed.The operation was performed by the same ophthalmologist on all patients.Twenty-one eyes (21 case) receiving conjunctiva injection of dexamethasone (Dex) in intraoperation served as the control group and the matched cases who received anterior chamber injections of TA in intraoperation were regarded as the TA group.The anterior uveitis was graded according to the Standard of Yang~([3]),and the intraocular pressure(IOP) was measured 1,3,and 7 days after surgery.Written informed consent was obtained from each patient before the initiation of any study protocol.Results The numbers of eyes with severe anterior uvetitis were significantly decreased 1,3,7 days after operation in the TA group in comparison with the Dex group (χ~2=10.857,P=0.028;χ~2=8.467,P=0.037;χ~2=11.286,P=0.004 ).No significant differences were found in IOP values between the TA group and the Dex group 1,3,7 days after operation (19.12±3.27 versus 19.49±3.23mmHg,t=0.469,P=0.644;17.91±1.95 versus 17.06±2.90mmHg,t=1.257,P=0.223;13.67±1.68 versus 13.05±1.66mmHg,t=1.201,P=0.237,respectively).Mild edema of corneal endothelium was seen in the early stage after operation and dissipated 4 days later in both groups,no significant differences were found between the TA group and the Dex group (P>0.05).Conclusion TA can effectively inhibit the inflammatory reaction of anterior uvea after cataract combination with anti-glaucoma surgery.No severe adverse effect is found after anterior chamber injection of TA.
6.Effect of follow-up by phone call on medication compliance of patients with cancer pain

Objective To explore the effect of follow-up by telephone calls on the medication compliance of patients with cancer pain.Methods Two hundred and forty hospitalized cancer patients suffering from moderate to severe cancer pain were included in the study. All the patients were provided with education on pain treatment before discharge.Then they were randomly assigned to receive telephone follow-up or routine nursing after discharge.The medication compliance were compared 2 months after discharge.Result The medication compliance in the observation group was significantly better than that in the control group.Conclusion Telephone follow-ups can improve medication compliance in patients with cancer pain.
7.Silent information regulator 1 and neuroprotective effects of resveratrol in cerebral ischemia

Resveratrol has several neuroprotective effects in cerebral ischemia, including reductions of oxidative stress, inflammatory responses and apoptosis. Recent studies have shown that these neuroprotective effects of resveratrol may be involved in activation of silent information regulator 1.
8.Glibenclamide for the prevention of cerebral edema and hemorrhagic transformation in ischemic stroke

Cerebral edema and hemorrhagic transformation(HT) are the common complications of ischemic stroke. Ischemic cerebral edema is the primary cause of death in patients w ith large infarction. The sulfonylurea receptor 1 (Sur1) transient receptor potential M4 (Trpm4) channel plays an important role in focal cerebral ischemia. Both animal experiment and clinical studies have show n that the Sur1 selective inhibitor glibenclamide provides neuroprotection for ischemic stroke, including infarct volume reduction and improvement of neurological function, especial y in attenuating cerebral edema and reducing the incidence of HT. This article review s the advances in research on glibenclamide for the prevention of cerebral edema and HT in patients w ith ischemic stroke.
9.Effect of Electroacupuncture plus Early Rehabilitation Training on Function Recovery after Surgery for Cervical Spinal Cord Injury

Objective To investigate the effect of electroacupuncture plus early rehabilitation training on the function recovery after surgery for cervical spinal cord injury. Methods Sixty patients with cervical spinal cord injury undergone anterior cervical decompression and internal fixation were randomized into a treatment group and a control group, 30 cases in each group. The treatment group was intervened by electroacupuncture plus rehabilitation training, while the control group was by rehabilitation training alone. The Frankel’s grading (FG), Barthel Index (BI), and Functional Independence Measure (FIM) were adopted for evaluation before and after intervention. Results The FG was not significantly changed after intervention in both groups (P>0.05). After intervention, there was no significant difference in comparing FG between the two groups (P>0.05). The BI and FIM scores were significantly changed after intervention in both groups (P<0.01). There were significant differences in comparing BI and FIM scores between the two groups (P<0.05). Conclusion Electroacupuncture plus early rehabilitation training can promote the function recovery after surgery for cervical spinal cord injury, and improve the quality of life.
10.Research progresses on relationship between Wnt inhibitory factor-1 and tumor

WIF-1 is an important antagonist in the Wnt/?-catenin pathway.As a tumor suppressor, WIF-1 is down-regulated in majority cancers but when it is correlated with WIF-1 promoter hypermethylation and treated with demethylation drug, its expression can be restored.Functional loss of WIF-1 due to promoter hypermethylation plays an important role in a tumor's pathogenesis, possibly mainly through aberrant canonical Wnt/?-catenin signal activation.The correlation of the expression of WIF-1 with clinicopathological features in different cancers is not identical.Detecting WIF-1 and promoter hypermethylation may offer help for early diagnosis, therapy and predicting prognoses.
