1.Inhibitory effect of 5 chemotherapeutic agents on KBV200 cell line and reverse effect on multidrug resistance of Matrine

Objective To observe the inhibitory effect of 5 chemotherapeutic agents including vincristine,adriamycin,pingyangmycin,cisplatin,cyclophosphamide on the KBV200 cell line and to study the reverse effect on the multidrug resistance of Matrine.Methods MTT assay was used to investigate the inhibition ability of 5 drugs on the cell line and the reverse effect on the multidrug resistance of Matrine.Results Vincristine,pingyangmycin,cyclophosphamide had less inhibitory effects than adriamycin,cisplatin on the KBV200 cell line.Matrine had the ability to reverse the multidrug resistance of the cell line to vincristine,adriamycin.Conclusion KBV200 cell line is multidrug-resistant to vincristine,pingyangmycin,cyclophosphamide.Matrine can enhance the sensitivity of KBV200 to the vincristine,adriamycin.
2.Investigation of the effect of verapamil and chemotherapeutic agents on the P-glucoprotein of the KBV200 cell line

Objective: To observe the effect of verapamil and 6 chemotherapeutic agents vincristine,adriamycin,bleomycin,cyclophosphamide,5-Fu,cisplatin on the P-glucoprotein(P-gp) of the KBV200 cell line.Methods: Flow cytometer(FCM) assay was used to investigate the expression of P-gp on the KBV200 cell line in the different conditions.Results: Verapamil act with vincristine,adriamycin,pingyangmycin and cyclophosphamide separately decreased expression of P-gp on KBV200 cell line by 80.17%,89.69%,19.19% and 53.48% Among all the combinations,the synergistic effects of varapamil act with vincristine and adriamycine show a better effect.Conclusion: Verapamil act with vincristine,adriamycin,pingyangmycin and cyclophosphamide separately can decrease expression of P-gp on KBV200 cell line significantly,and can reverse the drug resistance of KBV200 cell line.
3.Investigation of verapamil reverse the drug resistance of KBV200 cell line

Objective: To make a study of KBV200 cell line on its multidrug resistance and provide experimental data for clinical individual chemotherapy.Methods:MTT reduction assay was used to investgate the inhibition of 8 common chemotherapetic agents on KBV200 cell line.Result:Vincristine、Adriamycin、Bleomycin、Cyclophosphamide and Arabinosyl Cytosine have poorer inhibition than Harrihgtonine、Cicplatin and 5-Fu on KBV200 cell line.Conclusion:KBV200 cell line is multidrug resistance to Vincristine、Adriamycin、Bleomycin、Cyclophosphamide and Arabinosyl Cytosine.
4.Bioequivalence of Ambroxol Hydrochloride Orally Disintegrating Tablets

OBJECTIVE:To study the pharmacokinetic profiles and bioequivalence of test ambroxol hydrochloride vs.refe-rence preparation.METHODS:18 healthy volunteers were administered with a single oral dose of test(ambroxol hydrochloride orally disintegrating tablets)and reference ambroxol hydrochloride(60 mg)by randomized crossover way.The plasma concentrations of ambroxol hydrochloride were determined by LC-MS.The pharmacokinetic parameters and the bioavailability were computed using DAS 1.0 program.RESULTS:The main pharmacokinetic parameters of test and reference ambroxol hydrochloride were as follows:Cmax:(84.7?29.3)and(81.2?28.2)ng?mL-1;tmax:(1.7?0.5)and(1.8?0.4)h;t1/2:(7.9?0.9)and(8.3?1.0)h;AUC0~24:(638.7?180.8)and(591.4?148.3)ng?h?mL-1;AUC0~∞:(717.5?211.1)and(671.1?173.2)ng?h?mL-1,respectively.The relative bioavailability of test ambroxol hydrochloride orally disintegratiog tablets was(108.2?16.5)%.CONCLUSION:The test and reference preparations of ambroxol hydrochloride were bioequivalent.
5.Theefficacy of of nimodipine combined with gangliosides in the treatment of cerebral hemorrhage and its effects on the expression of serum resistin and oxidized low-density lipoprotein

Objective:To explore the efficacy of nimodipine combined with ganglioside in the treatment of cerebral hemorrhage and its effect on the expression of serum resistin and oxidized low-density lipoprotein (ox-LDL).Methods:According to the random number table method, 100 patients with cerebral hemorrhage admitted in Central Hospital of Zibo from February 2018 to February 2020 were divided into the single treatment group and the combined treatment group, with 50 cases in each group. In addition to conventional treatment, the single treatment group was additionally treated with nimodipine, and the combined group was additionally treated with nimodipine combined with ganglioside. After 2 weeks of intervention, the efficacy of the two groups was evaluated, and the National Institutes of Health Stroke Scale (NIHSS) scores, Ability of Daily Living (ADL) scores, serum inflammatory factors, resistin and ox-LDL levels were evaluated and recorded between the two groups before and after treatment.Results:The total effective rate of treatment in the combined treatment group was higher than that in the single treatment group: 90.0%(45/50) vs. 74.0%(37/50), and the difference was statistically difference ( P<0.05). After 2 weeks′ treatment, the scores of NIHSS was decreased and the scores of ADL was increased in two groups, and the scores of NIHSS in combined treatment group was lower than that in the single treatment group: (9.41 ± 1.27) scores vs. (12.10 ± 1.65) scores; the scores of ADL in the combined treatment group was higher than that in the single treatment group: (67.82 ± 9.04) scores vs. (59.10 ± 8.75) scores, and the differences were statistically differences ( P<0.05). After 2 weeks′ treatment, the levels of serum high-sensitivity C-reactive protein (hs-CRP), interleukin-6(IL-6), tumor necrosis factor-α(TNF-α), resistin and ox-LDL in the two groups were decreased, and the levels of above index in the combined treatment group were lower than those in the single treatment group: (4.81 ± 1.03) mg/L vs. (6.10 ± 1.73) mg/L, (40.96 ± 9.21) μg/L vs. (56.74 ± 8.93) μg/L, (33.20 ± 7.96) μg/L vs. (44.76 ± 8.43) μg/L, (0.29 ± 0.09) μg/L vs. (0.45 ± 0.13) μg/L, (336.25 ± 69.74) mg/L vs. (372.18 ± 68.52) mg/L, and the differences were statistically differences ( P<0.05). There was no significant difference in adverse reactions between the two groups ( P<0.05). Conclusions:Nimodipine combined with ganglioside has a clear efficacy in the treatment of cerebral hemorrhage. It can effectively inhibit inflammation indicators, reduce serum resistin and ox-LDL levels, promote the recovery of nerve defects, and improve their living ability.
6.Clinical Observation on Tiotropium Bromide Combined with Salmeterol Xinafoate and Fluticasone Propio-nate in the Treatment of Medium and Severe COPD

OBJECTIVE:To observe the clinical effect of tiotropium bromide combined with salmeterol xinafoate and fluticasone propionate in the treatment of medium and severe COPD. METHODS:119 patients with medium and severe COPD were divided into control group(54 cases)and observation group(65 cases)according to admission order. Control group was treated with Salmeterol xi-nafoate and fluticasone propionate powder for inhalation 50 μg,bid;observation group was additionally given Tiotropium bromide powder for inhalation 18μg,qn. Both groups received 1 month of treatment. Lung function indexes as FEV1,FVC and IC,blood gas indexes as SaO2,PaCO2 and PaO2 and inflammatory factors as IL-8 and TNF-α,COPD assessment test(CAT)score were observed in 2 groups,and the occurrence of ADR was also observed. RESULTS:Before treatment,there was no statistical significance in above indexes between 2 groups(P>0.05). After treatment,FEV1,EVC,IC of observation group were increased significantly,SaO2 and PaO2 of observation group were increased significantly,while PaCO2,IL-8 and TNF-αlevels was decreased significantly;those index-es of observation group were significantly better than those of control group,with statistical significance(P<0.05). CAT score of 2 groups were decreased significantly compared to before treatment,and the observation group was significantly lower than control group,with statistical significance(P<0.05). No obvious ADR was found in 2 groups. CONCLUSIONS:Tiotropium bromide com-bined with salmeterol xinafoate and fluticasone propionate shows good clinical efficacy and safety in the treatment of medium and se-vere COPD.
7.Inhibition of human lung carcinoma-associated antigen gene (ALT04-AG) expression on human lung carcinoma cell line growth and gene expression profile

Objective To study the effect of inhibition of human lung carcinoma-associated antigen gene(ALT04-AG) expression on the growth characteristic and associated gene expression of human lung carcinoma cells(L78).Methods L78 cells were transfected with recombinant plasmid vector expressing(ALT04-AG) antisense RNA mediated by liposome reagent,and treated with DFMO respectively. The growth characteristics were studied by MTT,FCM analysis.The associated gene expression was detected by immunocytochemical staining,Northern blot and gene chip analysis.Results The recombinant plasmid expressing ALT04-AG antisense RNA pALT04-AG as constructed.It was shown that the down-regulation of ALT04-AG gene expression was observed not only in pALT04-AG as transfected L78 cells,but also in DFMO-treated L78 cells,which resulted in growth inhibition of these cells.Gene chip analysis indicated that inhibition of(ALT04AG) gene expression by ALT04-AG antisense RNA regulated some proliferation associated gene expression,while inhibition of polyamine biosynthesis by DFMO regulated proliferation associated genes and also some apoptosis associated genes.Conclusion It is suggested that human lung carcinoma associated antigen gene ALT04-AG plays an essential role in maintaining malignant phenotype of cancer cells.
8.Clinical observation of injection of triamcinolone acetonide into the anterior chamber for anterior uveitis after cataract combined with anti-glaucoma surgery

Background Anterior uveitis is a common complication following the combination procedure of cataract extraction and anti-glaucoma surgery.More and more researches demonstrated the anti-inflammatory effectiveness of intraocular injection of triamcinolone acetonide (TA).Objective This study aims to observe the anti-inflammatory effectiveness of anterior chamber injection of TA for anterior uveitis following cataract extraction combined with anti-glaucoma surgery.Methods This is a case-control study.The clinical data from 42 eyes of 42 cases underwent the combination surgery of catarat extraction and anti-glaucoma was retrospectively reviewed and analyzed.The operation was performed by the same ophthalmologist on all patients.Twenty-one eyes (21 case) receiving conjunctiva injection of dexamethasone (Dex) in intraoperation served as the control group and the matched cases who received anterior chamber injections of TA in intraoperation were regarded as the TA group.The anterior uveitis was graded according to the Standard of Yang~([3]),and the intraocular pressure(IOP) was measured 1,3,and 7 days after surgery.Written informed consent was obtained from each patient before the initiation of any study protocol.Results The numbers of eyes with severe anterior uvetitis were significantly decreased 1,3,7 days after operation in the TA group in comparison with the Dex group (χ~2=10.857,P=0.028;χ~2=8.467,P=0.037;χ~2=11.286,P=0.004 ).No significant differences were found in IOP values between the TA group and the Dex group 1,3,7 days after operation (19.12±3.27 versus 19.49±3.23mmHg,t=0.469,P=0.644;17.91±1.95 versus 17.06±2.90mmHg,t=1.257,P=0.223;13.67±1.68 versus 13.05±1.66mmHg,t=1.201,P=0.237,respectively).Mild edema of corneal endothelium was seen in the early stage after operation and dissipated 4 days later in both groups,no significant differences were found between the TA group and the Dex group (P>0.05).Conclusion TA can effectively inhibit the inflammatory reaction of anterior uvea after cataract combination with anti-glaucoma surgery.No severe adverse effect is found after anterior chamber injection of TA.

10.Comparison of total flavone between root-extract and stem-extract from Acanthopanax

AIM:To understand the differences between the extraction parameters of different parts from Acanthopanax. METHODS: The orthogonal design method was used in experiment arrangement; the influence caused by the different factors on the extraction of total flavone from the root and the stem of Acanthopanax senticosus Harms was obtained. RESULTS: The optimum extracting technical parameters for the root were as follows: 20 times of 50% ethanol to Acanthopanax senticosus Harms, refluxing for 3 h at 60 ?C . The optimum extracting technical parameters for the stem were as follows: 20 times of 50% ethanol to Acanthopanax senticosus Harms, refluxing for 4 h at 70 ?C. CONCLUSION: The optimum extracting technical parameters for the root were different from the stem, which is related to difference in the biological structures between the root and the stem of Acanthopanax senticosus Harms.
