1.Research and development of pulmonary stem cell markers

By incorporation of tritiated thymidine or 5-bromodeoxyuridine and using them as the markers,we found that the basal cells,Clara cells and type II alveolar epithelial cells in the pulmonary tissues may in some way behave like stem cells and can eventually differentiate into respiratory tract epithelium.The slow progress of researches on pulmonary stem cells up to date has its root in the complex structure of pulmonary tissues and respiratory tracts,which is demonstrated by the facts that the respiratory system is composed of at least 40 types of cells,plus that the growth,renewal,and regeneration rate or capacity of respiratory epithelium are extremely limited.Mean while,there have been no specific stem cell markers for pulmonary and lung cancer,so researchers rely on stem cell markers borrowed from other systems to search for stem cells in pulmonary system.At present,the studies of lung stem cell markers were focused on Sca-1,ABCG2 /Bcrp1,cell retaining marker,surface marker,cell keratin and CCSP.Recently the lung stem cells have been shown to be the targets of transformation during lung carcinogenesis,which has provide more thoughts and direction for the occurrence and treatment of lung cancer.
2.Construction of academic native resource repository platform

4.Study on iron overload in thalassaemia minor patients

Objective To assess of iron overload in thalassaemia minor patients and provide basis for clinical intervention.Methods A total of 458 thalassaemia minor patients confirmed by genetic tests were divided into two groups(α-thalassaemia group and β-thalassaemia group),and 120 healthy individuals were recruited into control group.Serum iron and serum ferritin were measured,correlations between serum ferritin and age,genotypes,hemoglobin were evaluated.Results There was no significant difference in serum iron among the three groups(P>0.05),but the differences of serum ferritin was statistically significant(P<0.05),and serum ferritin in the β-thalassemia group was higher than that of the α-thalassemia group(P<0.05).Serum ferritin also had significant differences between different age groups(P<0.05),and which was higher in young patients(≤20) and older patients.Besides that,in patients older than 20 years,the level of ferritin was positively correlated with age.Moreover,we found serum ferritin had statistically differences between genotypes in α-thalassaemia patients(P=0.006),and which had poor negatively correlation with hemoglobin in β-thalassaemia patients(r=-0.252,P=0.001).Conclusion Serum ferritin was a sensitive indicator to assess iron overload in thalassaemia minor patients,and which was positively correlated with age in thalassaemia patients 20 or more than 20 years old.Our research also indicates different iron overload traits between α-thalassaemia patients and β-thalassaemia patients.
5.Simultaneous Determination of Four Compounds in Zhizhu Pills by HPLC

Objective To develop a method for simultaneous determination of 4 compounds in Zhizhu Pills by HPLC.Method Reversed-phase HPLC was used to determine the content of the compounds such as hesperidin,naringin,neohesperidin and atractylenolideⅠ.The column was symmetry C18(250 mm?4.6 mm,5 ?m) with temperature of 30 ℃.Methanol-0.095 %phosphoric acid solution served as the mobile phase by gradient elution.The wavelength of detection was set at 283 nm and 224 nm.The injection volume was 10 ?L and the flow rate was 1.0 mL/min.Results Hesperidin,naringin,neohesperidin and atractylenolideⅠin Zhizhu Pills can be isolated completely.Three batches of samples were determined and the recoveries of naringin,hesperidin,neohesperidin and atractylenolideⅠwere 98.81 %(RSD=1.11 %,n=6),100.63 %(RSD=1.90 %,n=6),99.32 %(RSD=1.44 %,n=6) and 99.65 %(RSD=1.53 %,n=6) respectively.Conclusion This method is accurate,reliable and with good reproducibility.It can be used for the quality control of Zhizhu Pills.
6.THE EFFECT OF ZINC DEFICIENCY ON THE PHENOTYPES AND SUBSETS OF PERIPHERAL BLOOD LYMPHOCYTES IN GROWING RATS

Objective: To observe the effects of zinc deficiency on peripheral blood lymphocyte phenotypic distribution and the balance between Th1 and Th2 cells in growing rats. Methods: Two-color cytofluorometric analysis was used. Results: Compared with zinc adequate (ZA) and paired-fed (PF) groups, peripheral blood lymphocytes in zinc deficient (ZD) rats had higher percentage of CD8+ cells (cytolytic T cells), lower percentage of CD4+ cells (T helper cells) and lower percentage of CD45RA+ cells (pure B lymphocytes).In addition, zinc deficiency selectively caused reduction of Th1-like CD45RC+CD4+ cells , but Th2-like CD45RC-CD4+cells had no significant changes in weanling rats. Zinc supplementation could reverse these changes. Conclusion: Dietary zinc deficiency leads to changes of lymphocyte phenotypic distribution and the imbalance of Th1 and Th2 subsets. These changes are reversible.
7.Assessment of journals according to their weighted citations

A method to assess library-holding journals according to their weighted citations was proposed in light of the actual needs of users for core journals, the use of library-holding journals and references by authors in different orders of precedence.The Western journal of rheumatology, highly cited by authors of Second Military Medical U-niversity, were assessed using this method, showing that this method is better than the citation analysis-based tra-ditional method in assessment of journals.
8.Effects of cell density on proliferation and osteogenic differentiation of human mesenchymal stem cells during subculturing in vitro

BACKGROUND: Rapid development of tissue engineering is the new hope of repairing bone defect so as to obtain complete bone regeneration,and rapid proliferation of seed cells is one of the important factors in it.OBJECTIVE: To evaluate the proliferation and osteogenic differentiation of human mesenchymal stem cells (MSCs) plated at different cell density during subculturing period.DESIGN: Repetitive measurement.PARTICIPANTS: The experiment was conducted in the Trauma, Burn and Combined Injury Laboratory of the Third Military Medical University of Chinese PLA between January 2003 and March 2004. MSCs were obtained from iliac bone bone marrow of the 5 healthy male volunteers by separation and proliferation.METHODS: The MSCs of second generation were inoculated at 8×103/cm2,3×103/cm2, and 8×102/cm2, respectively, and then the growth curve was drawn and analyzed. After 18 days of proliferation and culture, the increased amount of MSCs was recorded to analyze the post-proliferation growth curve and osteogenic ability.MAIN OUTCOME MEASURES: ①Growth curve of cells plated at different cell density; ②Results of alkaline phosphatase(ALP) staining and immunohistochemical staining of osteocalcin.RESULTS: Human MSCs from bone marrow were CD34 negative and expressed low level of ALP. Population doubling time was 40 hours after MSCs were plated at 8×103/cm2 and the number of the cells was expanded (51±13) times after 18 days. MSCs were expanded (28±6) times 18 days after plated at 3×103/cm2. While plated at 8×102/cm2, the number of the cells was only expanded (5±3) times. Proliferative ability of cells cultured at a low density was stronger that that at a high density. MSCs could be induced to differentiate into the osteocytic lineages after plated at not only 8×103/cm2 but also 8×102/cm2.CONCLUSION: Cell density plays an evident role in the proliferation of MSCs. Cell density of 8×103/cm2 is proper to the rapid proliferation of MSCs as seed cells in bone tissue engineering.

0.900.Comparison of the HPLC profiles of the total flavonoids material and the effective parts of the total flavonoids was established and the similarity with the corresponding Citrus Aurantium L.var daidai Tanaka fruits indicated that they were closely related to each other.Conclusion:The HPLC fingerprints of the medicine material and the effective parts of the total flavonoids with high specificity and can be used to control its quality.
10.Effects of different doses of dexmedetomidine administered intranasally on median effective concentration of sevoflurane inhibiting responses to laryngeal mask airway insertion in pediatric patients

Objective To evaluate the effects of different doses of dexmedetomidine administered intranasally on the median effective concentration (EC50) of sevoflurane inhibiting responses to laryngeal mask airway (LMA) insertion in the pediatric patients.Methods American Society of Anesthesiologists physical status Ⅰ or Ⅱ pediatric patients of both sexes, aged 1-3 yr, with body mass index of 20-25 kg/m2, scheduled for elective surgery under general anesthesia, were randomly divided into 3 groups: control group (group C) , dexmedetomidine 1 μg/kg group (group D1) , and dexmedetomidine 2 μg/kg group (group D2).In D1 and D2 groups, dexmedetomidine 1 and 2 μg/kg (in 1 ml of normal saline) were administered intranasally at 1 h before induction of anesthesia, respectively.Group C received the equal volume of normal saline administered intranasally.The children were separated from their parents at 45-60 min after intranasal administration and admitted to the operating room.The children inhaled 8% sevoflurane until they lost consciousness.After loss of consciousness, the end-tidal concentration of sevoflurane was determined using the modified Dixon's up-and-down method.The initial end-tidal concentration of sevoflurane was set at 2%.The concentration was increased/decreased by 0.2% in the next patient according to the response to LMA insertion.The patient's sedation status and with LMA acceptance were evaluated, and the patient's satisfaction with sedation and with LMA acceptance was recorded.Probit analysis was used to calculate the EC50 and 95% confidence interval of sevoflurane inhibiting responses to LMA insertion.Results The patient's satisfaction with sedation and with LMA acceptance was significantly higher in D1 and D2 groups than in group C, and in group D2 than in group D1 (P<0.05).The EC50 (95% confidence interval) of sevoflurane inhibiting responses to LMA insertion was 2.03% (1.91%-2.15%), 1.76% (1.65%-1.87%) and 1.63% (1.53%-1.73%) in C, D1 and D2 groups, respectively.The EC50 was significantly lower in D1 and D2 groups than in group C, and in group D2 than in group D1 (P<0.05).Conclusion Dexmedetomidine 1 and 2 μg/kg administered intranasally can decrease the EC50 of sevoflurane inhibiting responses to LMA insertion in the pediatric patients, and 2 μg/kg produces better efficacy.
