To study the effect of urokinase on platelet activation and aggregation in the patients with glomerulonephritis. Platelet counts(PC),mean platelet volume (MPV), and platelet distribution width (PDW) were determined with automatic blood cell analytic apparatus, the expressive levels of platelet granular membrane protein (GMP 140) and fibrinogen receptor(GPⅡb Ⅲa) on the surfaces of platelets were assayed with flow cytometry, and the levels of fibinogen and fibrin degradation products (FDP) in blood were measured with ELISA in 102 children with glomerulonephritis. The results showed that urokinase led to PC decreas,MPV increase and PDW exceeding normal value in 102 children with glomerulonephritis, including micro change (MC),glomerulosclerosis (GS),membranous nephropathy (MN), and proliferative glomerulonephritis (PGN).The expressive levels of GMP 140 after the treatment with urokinase were significantly higher than that before the treatment and controls( P 0 01). The results suggested that urokinase used in the patients with glomerulonephritis increased platelet activation function and decreased platelet aggregation function.

BACKGROUND: The combination of electric acupuncture and clomipramine is the behavior attempt in the integration of Chinese and western medicine in the treatment of obsession.OBJECTIVE: To probe into the long-term therapeutic effects of the combination of acupuncture and medication on obsession and its influence on social functional recovery.DESIGN: Controlled study with observation in which the patients were taken as the objects.SETTING: Wards of a municipal mental hygiene center PARTICIPANTS: The research was performed in the Fifth People's Hospital of Zibo. Totally 30 inpatients were selected from January 1997 to October 2003 in the hospital, aged varied from 18 to 57 years, at the average of (33 ± 12) years. Thirty patients were divided randomly into two groups,named as experimental group, 15 cases with 8 males and 7 females; and the control group, 15 cases with 7 males and 8 females.METHODS: In the experimental group, electric acupuncture was applied on neiguan, renzhong and chengjiang, together with clomipramine orally. In the control group, only clomipramine was applied. The therapeutic effects and side effects were evaluated with Y-Bocs, TESS and clinical evaluation standards successively before the treatment, at the 4th, 6th and 8th weekends after treatment and by 6 months following visits. SDSS evaluation was carried on at the 8th week of the treatment.MAIN OUTCOME MEASURES: Comparisons of evaluating results with Y-Bocs, TESS and SDSS.RESULTS: At the 4th, 6th and 8th weekends of the treatment, the score-reducing rates of Y-Bocs in the experimental group were higher than those in the control group, but the difference was not significant ( P ＞ 0.05). The scores of TESSin the control group were remarkably higher than those in the experimental group( P ＜ 0. 01 ) . Factor scores of "profession and work" in SDSS in the experimental group were lower than that in the control group ( P ＜ 0.05 ).CONCLUSION: Electric acupuncture on neiguan, renzhong and chengjiang reduces the side effect of clomipramine in the treatment of obsession and improve the living quality of the patients with obsession.

Objective To investigate the lonely behavior characteristics and related factors of chronic obstruetive pulmonary disease(COPD)patients.Methods 99 eases with stable patients with COPD and 90 healthy subjects were surveyed by SDS,ESLI.Relevant factors were analyzed.Results Patients with COPD showed difierent degrees of emotional isolation[(9.90±3.97),(6.15±5.22),F=82.539,P＜0.01)]and social intercourse isolation[(8.00±3.53),(4.31±2.46),F=73.540,P＜0.01)].Emotional isolation was mainly.Moderate emotional isolation and general social intercourse isolation were predominant,and emotional(r=0.399,t=3.476,P＜0.01)and social intercourse isolation(r=0.372,t=3.154,P＜0.01)were related to depression.Conclusion Patients with COPD presented obvious emotion and social intercourse isolation behavior.

Topical photodynamic therapy (PDT),a highly effective,safe and non-invasive therapeutic method,exerts good therapeutic effects on various diseases in dermatology,such as actinic keratosis,condyloma acuminatum,acne,and so on.The major drawback of PDT is pain during the treatment.Some patients have to interrupt or terminate the treatment because of pain intolerance.Some progress has been made in the management of PDT-related pain.It has been revealed that multiple factors are associated with PDT-related pain,including individual factors,lesion characteristics,therapeutic parameters (such as light sources,photosensitizers).Many methods can be used to manage PDT-related pain,including local cooling,oral analgesics,two-step irradiance protocol,topical anesthesia,injection anesthesia,inhalation anesthesia,transcutaneous electric nerve stimulation,hypnosis.

Objective: To study the protective effect of ginkgolide on rat's renal failure induced by adenine.Methods: The chronic renal functional failure model of rat was established with adenine. Its urea nitrogen (BUN), reatinine (Crea), Cholesterin (CH), total protein (TP), and albumin (ALP) in blood were determined. The pathologic changes were also observed. Results: Ginkgolide has effects on renal failure caused by adenine. Low and high dose groups decrease BUN. Crea, low dose group decrese CH, high dose group increase numbers of kindney glomerulus.Conclusion: Ginkgolide has protective effect on rat's renal failure induced by adenine.

In order to improve the diagnosis and treatment of severe hemolytic uremic syndrome (SHUS),we summarized the rescuing experiences in 3 children with SHUS.Therefore,close observation of gastro intestinal symptoms,collaborated with repeated examinations of the blood,renal function,urinary output,blood pressure,and manifestations of the central nervous system could help predict to certain extent the occurrence of further hemolysis and renal failure in these patients.Energetic treatment of malignant hypertension and relieving the symptoms of center nervous system could successfully improve renal functions.If urinary output decreased quickly,creatinine (Cr) and blood urea nitrogen (BUN) increased,or severe infection occurred in SHUS patients,hemodialysis should be undertaken as early as possible to prevent caute renal failure and multiple organ failure syndrome (MOFS).Therefore,the treatment of SHUS in children has its peculiarity and complexity.

Objective To observe the changes of neuron-specific enolase(NSE),S-100 protein and TBC in the blood of newborn rats at early stage of bilirubin encephalopathy,and the protective effect of mild-hypothermia on the brain.Methods 42 Wistar rats(7-day postnatal) were divided randomly into the group C(control group,n=10),group M0(normal-temperature group,n=17) and group M1(mild-hypothermia group,n=15).The rats of the group C received physiological saline 0.5 ml,the rats in the groups of M0 and M1 were injected with bilirubin intraperitoneally(200 mg/kg) to establish the model of bilirubin encephalopathy.The changes of the content of NSE and S-100 protein in the blood of newborn rats,and the protective effect of mild-hypothermia on brain were observed.Results The animals with established bilirubin encephalopathy shown significant changes of neurobehaviour and pathological examination.Values of NSE and S-100 protein of the group M1 decreased after the treatment of mild-hypothermia,and there was a significant difference compared with group M0( P<0.01).Conclusion The mild-hypothermia has protective effect on brain of newborn rats with bilirubin intraperitoneally.

Objective To study the effects of short hairpin RNA (shRNA) targeting STAT3 gene on the proliferation and apoptosis of A375 human malignant melanoma cells.Methods Sense and antisense oligonucleotides with small hairpin structures targeting STAT3 gene were designed,synthesized and cloned into the plasmid vector psiRNA-hHlneo after annealing.Cultured A375 cells were divided into 3 groups: control group receiving no treatment,psiRNA-H1 group transfected with empty plasmid,and psiRNA-H1/STAT3 group transfected with the recombinant plasmid containing the shRNA.After additional culture for different durations,reverse transcription PCR and Western blot were performed to detect the expression of STAT3 mRNA and protein,methyl thiazolyl tetrazolium (MTT) assay to evaluate cell proliferation,flow cytometry to assess cell cycle and apoptosis.Results The expression level of STAT3 mRNA and protein in A375 cells in psiRNA-H1/STAT3 group (0.2136 ± 0.0626,0.8214 ± 0.043,respectively) were significantly lower than that in the control group (0.7826 ± 0.0701,3.1693 ± 0.0846,respectively,both P ＜ 0.01) and psiRNA-H1 group (0.8518 ± 0.0783,3.218 ± 0.078,respectively,both P ＜ 0.01 ).The inhibition rates of cell proliferation at 24,48 and 72 hours were 21.35％ ± 2.12％,32.52％ ± 2.64％ and 40.4％ ± 3.08％ respectively in psiRNA-H1/STAT3 group,statistically different from those in the control group (1.39％ ± 0.53％,3.05％ ± 1.16％,4.41％± 1.42％,respectively,all P ＜ 0.01) and psiRNA-H1 group (2.63％ ± 1.38％,5.84％ ± 2.47％,10.32％ ±2.48％,respectively,all P ＜ 0.01).Flow cytometry showed a statistical increase in cell apoptosis rate in psiRNA-H1/STAT3 group compared with the control and psiRNA-H1 group (81.06％ ± 2.10％ vs.26.28％ ±0.47％ and 27.31％ ± 1.05％,both P ＜ 0.01 ).The psiRNA-H1/STAT3 group exhibited a higher percentage of cells at G0/G1 phase (68.43％ ± 4.00％) but a lower percentage of cells at S phase (17.4％ ± 2.05％) compared with the control group (60.07％ ± 2.47％,P ＜ 0.05; 28.40％ ± 2.09％,P ＜ 0.01 ) and psiRNA-H1 group (60.29％ ± 2.26％,27.34％ ± 3.63％,both P ＜ 0.05 ).Conclusions The small interference RNA targeting STAT3 gene can specifically down-regulate the expressions of STAT3 mRNA and proteins in,inhibit cellular proliferation of,and induce apoptosis in,A375 cells.
