Modern Clinical Nursing 2015;(8):8-11
Objective To investigate the effect of abdominal vibration therapy on chronic obstructive pulmonary disease (COPD) patients with constipation. Methods In the respiratory department from August 2013 to July 2014, eighty-three COPD patients with constipation were divided into the experiment group (n=35) and the control group (n=48) using random digit table. The control group were treated with conventional western medicine and the experiment group received impingement abdomen vibration therapy on the basis of western medicine treatment. The two groups were compared in terms of symptoms of constipation and curative effect. Results The curative effect of the experiment groups was significantly better than that of the control group (P<0.05). The score on constipation in the experiment group was significantly lower than that of the control group (P<0.05). Conclusion The abdominal vibration therapy is effective in the treatment of COPD patients with constipation. It can improve their quality of life and be worthy of clinical application.

Wild-type p53 has been shown to be involved in several aspects of cell growth control. In order to study the role of wild-type p53 in the growth of lung adenocarcinoma cell line GLC-82, recombinant p53 adenovirus vector and the control LacZ gene were constructed. Infection conditions were detected by biochemistry staining for LacZ s expression product ?-gal, immunohistochemical analysis for p53 protein expression, and PCR technique for the infected cells. The cell growth rate and cell cycle were analysed with flow cytometry. The results showed that the constructed recombinant adenovirus vector could infect cells with high level expression of (?-gal and p53 protein and p53 cDNA could be found in the infected cells. The wild-type p53 could inhibit GLC-82 cell proliferation and cause cellular death. These results indicated that wild-type p53 gene mediated by adenovirus vector might be used in the treatment of lung adenocarcinoma.

Objective In order to research the best fermentation conditions of recombinant bacteria pThioHisA-B1/DH5? which contains gene of esterase B1.Methods Recombinant bacteria pThioHisA-B1/DH5? was induced in different culture media, induction temperatures, different inductor concentrations, different induction time in order to obtain the best expression effect. The expression products of different conditions were detected by SDS-PAGE and thin-layer gel scanning analysis. Results Based on the experiments, the optimized fermentation condition were that the culture medium was TB, inducing temperature was 28 ℃ and inducing time was 6 h,the concentration of IPTG was 0.4 mmol/L.The expression quantity of target protein attained 40.3% in total bacterial protein, and the soluble fusion protein accounted for 66.2% in total target proteins at the optimum conditions. Conclusion The optimized fermentation conditions of expressing fusion protein in recombinant bacteria have been found and in the conditions, soluble esterase B1 fusion protein of high yield can be obtained.

Idiopathic pulmonary fibrosis(IPF), with unknown pathogeny, is an interstitial lung disease. The pathological features are diffuse epithelial-cell lesion, fibroblast proliferation, myofibroblast differentiation and excessive extracellular matrix deposition.CXCR4 is the predominant chemokine receptor on fibrocytes;CXCL12 is the only ligand of CXCR4.A large number of studies have shown that CXCL12/CXCR4 biological axis plays an important role in the pathogenesis of idiopathic pulmonary fibrosis.Under the regulation of hypoxia, HIF-1α and PI3K-Akt-mTOR path, CXCL12/CXCR4 biological axis promotes lung fibroblast proliferation, myofibroblast differentiation and extracellular matrix deposition, resulting in development and progression of IPF.

Objective To investigate the clinical efficacy of postoperative three-dimensional conformal radiotherapy concurrent with paclitaxel +TS-1 chemotherapy in the treatment of advanced gastric carcinoma. Methods A total of 60 patients with advanced gastric carcinoma were randomly divided into treatment group and control group. In treatment group,30 patients received simultaneous three-dimensional conformal radiations therapy combined with paclitaxel +TS-1 chemotherapy. In control group,30 patients received paclitaxel +TS-1 chemotherapy. The long term survival rate, local recurrence rate and side reactions were observed in two groups. Results The 1,2-year survival rate in treatment group [93.1% (27/29),75.9 % (22/29)] and control group [72.4 % (21/29),48.3 % (14/29)] had significant difference (P < 0.05).The 1,2-year local recurrence rate in treatment group [6.9% (2/29),13.8% (4/29)] and control group [31.0% (9/29),44.8 % (13/29)] had significant difference (P < 0.05 or < 0.01).The side reactions in two groups were mainly manifested as leukopenia, thrombocytopenia, anemia, nausea, vomiting, liver function damage and oral mucositis. The incidence rate of nausea, vomiting in treatment group was significantly higher than that in control group [83.3% (25/30) vs.53.3% (16/30)] (P =0.012).Conclusions Postoperative threedimensional conformal radiotherapy concurrent with paclitaxel +TS-1 chemotherapy in the treatment of advanced gastric carcinoma may improve the long-term survival rate, lower local recurrence rate. But the adverse reactions are increased.

Aim To investigate whether EGCG regu-lates ABCA1 expression by influencing the expression of miR-33 a to promote cholesterol efflux from macro-phages. Methods THP-1 macrophage-derived foam cells were established by co-incubated with oxLDL, and then treated with EGCG, and miR-33a expression was detected with Real-time PCR. THP-1 macrophage-derived foam cells were randomly divided into the fol-lowing groups: control group, 50 μmol · L-1 EGCG treatment group, 50 μmol · L-1 EGCG +80 nmol · L-1 miR-33a mimic treated group. Real-time PCR and Werstern blot was used to detect ABCA1 mRNA and protein expression, Oil Red O staining and high per-formance liquid chromatography were used to detect in-tracellular lipid content, and [3H] assay was used to determine cellular cholesterol efflux. Results EGCG could downregulate miRNA33 a expression in a dose-and time-dependent fashion within safe doses. EGCG significantly upregulated ABCA1 mRNA and protein expression, which could be inhibited after miRNA33 mimic transfected into cells, however. EGCG may re-duce lipid accumulation in THP-1 macrophage-derived foam cells, and this effect could also be weakened after miRNA33 mimic transfected. EGCG could reduce the accumulation of intracellular cholesterol, which was re-lated with EGCG promoting intracellular cholesterol ef-flux , and excess miRNA33 a transfected into cells could inhibit intracellular cholesterol efflux. Conclusion EGCG may upregulate ABCA1 expression by reducing miRNA33a generation, resulting in the promotion of cholesterol efflux from macrophages, which may be one of the molecular mechanisms of EGCG anti-atheroscle-rotic effect.

Objective To investigate the values of CD21 and CD43 proteins in the differential diagnosis of mucosa-associated marginal zone B-cell lymphoma (MALToma) from benign lymphadenosis. Methods The expression of CD21 and CD43 proteins in the tissues of 25 MALToma (case group) and 25 benign lymphadenosis (control group) was detected by immunohistochemistry. Results Abnormal CD21+follicular dendritic cells (FDC) meshes were found in all patients of case group. Most of the FDC meshes were sparse and broken, and a few were enlarged or fused into pieces. Intact CD21+FDC meshes were all found, and abnormal FDC meshes were not found in control group. The positive rate of abnormal FDC meshes in case group was significantly increased compared with that in control group (χ2 = 46.080, P= 0.000). The expression rate of CD43+in CD20+cells was 24 % (6/25) in case group, but it was negative in control group (χ2=4.375, P=0.030). Conclusions Abnormal CD21+FDC meshes and CD43+expression in CD20+cells are useful in the differential diagnosis between MALToma and benign lymphadenosis. The abnormal FDC meshes of MALToma are enlarged or fused in the minority of cases.
