1.Clinical features and imaging diagnosis of multiple osteochondroma

Objective To explore the etiology,clinical features,and imaging features of multiple osteochondroma.Methods A total of 21 patients of multiple osteochondroma with completely clinical and imaging data were brought into this study.All these patients accepted X ray DR examination, and 15 cases of them accepted CT scanning and 4 cases of them accepted MRI examination.The clinical and imaging data were analyzed retrospectively combined with the relative literature.Results All the 21 patients with multiple osteochondroma had family history,and it's likely to be found in the metaphysis of limb long bone.Patients with multiple osteochondroma usually had bony lump with cartilage cap in long bones' metaphysis.It often occured in double sides but not symmetry,and it was more likely to be found in the lower limbs.Bone around the knee-joint was the most common position of lesion,and the growth direction was usually deviating from the joints.Conclusion Multiple osteochondroma has the characteristics of internalization bone cartilage defects and osseous vegetations formation,and it's usually combined with bone bending,shortening deformity and adjacent joint activities obstacles.It can be diagnosed according to the typical clinical features,X-ray and CT imaging features.
2.Expressions of inflammatory factors in thyroid papillary carcinoma tissue and their clinical significances

Objective To investigate the expressions of inflammatory factors in papillary thyroid carcinoma tissue and their relationships with the clinical pathological characteristics,and to provide an experimental basis for guiding clinical treatment and prognosis.Methods 74 patients with papillary thyroid carcinoma dignosed by puncture were selected as experimental group,and 26 cases of healthy people were used as normal control group.The fasting venous blood samples from the subjects in two groups were collected,and the postoperative specimens from cancer tissue,adjacent normal tissue and the fasting venous blood 7 d after operation of the patients in experimental group were collected again.The interleukin 1α(IL-1α),interleukin 1β(IL-1β)and cyclooxygenase-2 (COX-2)protein expression levels in serum,cancer adjacent normal tissue and cancer tissue were detected by ELISA;the gene and protein expression levels of IL-1α,IL-1βand COX-2 were examined by real-time fluorescent quantitative method and immunohistochemistry;the relationships between their expressions and clinical stages, pathological types,lymph node metastasis of thyroid papillary carcinoma were analyzed.Results The expression levels of serum IL-1α,IL-1βand COX-2 protein of the patients in experimental group were significantly higher than those in normal control group (P<0.01);the gene and protein expression levels of IL-1α,IL-1βand COX-2 in thyroid papillary carcinoma tissue were significantly higher than those in adjacent tissue (P<0.01).There were positive correlations between the expressions of IL-1αand IL-1β,IL-1αand COX-2,IL-1βand COX-2 in thyroid papillary carcinoma tissue (r=0.64,P=0.035;r=0.71,P=0.042;r=0.69,P=0.038).Conclusion The expression levels of IL-1α, IL-1βand COX-2 in thyroid papillary carcinoma tissue are increased, suggesting that inflammation is involved in metastasis and invasion of thyroid papillary carcinoma,its application is expected to become a new means of cancer treatment strategies.
3.Improvement of the analytical sensitivity of domestic HBV DNA kit based on magnetic beads nucleic acid extraction method

Objective To establish a method of nucleic acid extraction and enrichment based on magnetic nanoparticle as medium for elevating the analytical sensitivity of domestic HBV real-time PCR kit and detection of the trace amount HBV DNA. Methods After receiving antiviral treatment, the serum samples of 50 hepatitis B patients with HBV DNA concentration ≤1×104 IU/ml were collected. The WHO HBV DNA calibrator was used as the standard material. Nanometer magnetic beads were used to adsorb and enrich the HBV nucleic acid and increase the concentration of the extracted HBV nucleic acid template. Compared with Roche HBV DNA detection reagent and four domestic reagent with conventional nucleic acid extraction and detection method, the improvement effect of this method on domestic nucleic acid detection reagent was evaluated. Results After application of nanometer magnetic extraction method to domestic regent, the analytical sensitivities of the domestic reagent reached 10 and 50 IU/ml, respectively,which was about the same detection level to 12 IU/ml of the imported Roche reagent. The positive rates of the detection of serum trace amount HBV DNA of hepatitis B patients with four kinds of domestic extraction reagent were 64% ( 32 ), 56% ( 28 ), 62% ( 31 ) and 58% (29), respectively. There were significant statistical differences between Roche reagent and four domestic extraction reagent kits(x2 = 7. 895, 12. 698,9. 013 and 11. 416 ,P <0. 05 ). With nanometer magnetic extraction method combined with domestic reagent kits, the detection rates were 88% (44), 88% (44), 88% (44) and 86% (43) ,respectively. There was no significant difference compared with the imported Roche reagent (x2 = 0. 000, 0. 000, 0. 000 and 0. 088,P >0. 05). Moreover, when the HBV nucleic acid concentration was 101-103 IU/ml, the logarithm value of viral nucleic acid concentration was in reverse correlation to Ct value, but the correlation decreased in the concentration range of 103-106 IU/ml. Conclusions The nucleic acid extraction method based on magnetic nanoparticle as medium can significantly improve the analytical sensitivity of domestic HBV DNA detection reagent, which can be used to monitor the trace amounts HBV DNA in the sera of the hepatitis B patients.
4.Effect of Red Ginseng on Metabolic Syndrome:A Meta-Analysis

Objective To systematically evaluate the effect of red ginseng on improving metabolic syndrome,and to provide basis for clinical practice.Methods Retrieval from PubMed,Embase,The Cochrane Library,Web of Science,Clinical Trials Gov,WanFang Data and CNKI databases,articles of RCTs about red ginseng on improving metabolic syndrome were included.The searching range was from the establishment of the database to May 2022.The articles were screened according to inclusion and exclusion criteria,and then the inclusion study was analyzed through RevMan 5.4.1 software.Results Totally 17 randomized controlled trials involving 957 patients were included.Compared with the control group,red ginseng exhibited significant effect on fasting blood glucose,total cholesterol,low-density lipoprotein,systolic blood pressure and diastolic blood pressure(P<0.05),and its effect was related to the disease,age,duration of administration and dosage.Conclusion Red ginseng could significantly improve the symptoms of hyperglycemia,hyperlipidemia and hypertension in metabolic syndrome,which provides a theoretical basis for clinical use of red ginseng in treating metabolic syndrome.
5.Role of hsa_circ_0008039 and miR-484 in oxygen-glucose deprivation/reoxygenation injury in SK-N-SH cells: relationship with Fis1

Objective:To evaluate the role of has_circ_0008039 and miR-484 in oxygen-glucose deprivation/reoxygenation (OGD/R) injury in SK-N-SH cells and the relationship with Fis1.Methods:SK-N-SH cells were cultured in vitro to logarithmic growth stage and divided into 5 groups ( n=25 each) according to the random number table method: control group (group C), OGD/R group, has_circ_0008039 siRNA group (group S), hsa_circ_0008039 over-expression group (group E) and has_circ_0008039 siRNA plus miR-484 inhibitor group (group S+ I). Cells were cultured in normal condition in group C. In S, E and S+ I groups, after the cells were transfected with hsa_circ_0008039 siRNA, has_circ_0008039 over-expression vector, hsa_circ_0008039 siRNA and miR-484 inhibitor, the cells were subjected to oxygen-glucose deprivation for 12 h followed by 24 h restoration of O 2-glucose supply to develop the OGD/R model.At 24 h of restoration of O 2-glucose supply, the cell viability and amount of lactic dehydrogenase (LDH) released were measured using CCK-8 assay, the expression of hsa_circ_0008039, miR-484 and Fis1 mRNA was detected using real-time polymerase chain reaction, and the expression of Fis1 was detected by Western blot.A dual-fluorescein experimental report was used to verify the targeting relationship between hsa_circ_0008039 and miR-484. Results:Compared with group C, the cell viability was significantly decreased, and the amount of LDH released was increased in the other 4 groups, the expression of hsa_circ_0008039 and Fis1 was significantly up-regulated, and the expression of miR-484 was down-regulated in OGD/R and E groups, the expression of hsa_circ_0008039 and Fis1 was significantly down-regulated, and miR-484 was up-regulated in group S, and the expression of hsa_circ_0008039 and miR-484 was significantly down-regulated, and the expression of Fis1 was up-regulated in group S+ I ( P<0.05). Compared with group OGD/R, the cell viability was significantly decreased, and the amount of LDH released was increased in E and S+ I groups, the cell viability was significantly increased, and the amount of LDH released was decreased in group S, the expression of hsa_circ_0008039 and Fis1 was significantly up-regulated, and the expression of miR-484 was down-regulated in group E, the expression of hsa_circ_0008039 and Fis1 was significantly down-regulated, and the expression of miR-484 was up-regulated in group S, and the expression of hsa_circ_0008039 and miR-484 was significantly down-regulated, and the expression of Fis1 was up-regulated in group S+ I ( P<0.05). Compared with group S, the cell viability was significantly decreased, the amount of LDH released was increased, the expression of miR-484 was down-regulated, and the expression of Fis1 was up-regulated in group S+ I ( P<0.01). The dual-fluorescein experimental report verified that miR-484 was the target of hsa_circ_0008039 which binded to miR-484 specifically. Conclusions:has_circ_0008039 is involved in OGD/R injury in SK-N-SH cells by targetedly binding to miR-484, which is associated with up-regulation of Fis1 expression.
6.Traditional Chinese Medicine in Treatment of Constipation Based on 5-HT Signaling Pathway: A Review

Chronic constipation (CC) is one of the most common functional gastrointestinal diseases. At present, the overall therapeutic effect of CC is still not satisfactory worldwide, which seriously affects the quality of life and social function of patients. The etiology and pathophysiological mechanism of constipation are still unclear. It involves comprehensive factors such as heredity, social psychology, diet, intestinal flora imbalance, intestinal motility disorder, visceral sensitivity change, pelvic floor muscle group dysfunction and enteric nervous system (ENS) disorder. Among them, the abnormal factors of the brain-gut-microbiome axis are particularly significant. The brain-gut-microbiome axis is a complex network of interactions between the intestine and the brain, integrating and coordinating the physiological functions and pathological processes of the gastrointestinal tract. Microorganisms in the intestinal lumen play an important role in it, and can communicate with the intestinal tract and the central nervous system through nerve, endocrine and immune pathways. As a key brain-gut peptide in the regulation pathway of the brain-gut-microbiome axis, 5-hydroxytryptamine (5-HT) is involved in the regulation of gastrointestinal motility, sensation and secretion. The abnormal conduction of the 5-HT signaling pathway is closely related to the occurrence and development of constipation. Traditional Chinese medicine(TCM) is a unique precious resource in China, which has good curative effects and safety. In recent years, it has received extensive attention in the treatment of constipation. TCM and active ingredients, acupuncture and massage specifically regulate 5-HT signal transmission, so that the expressions of related molecules tend to be suitable for individual disease state levels to effectively improve constipation symptoms, with unique advantages. Therefore, this study used ''constipation'', ''intestinal flora'', ''5-HT'', and ''traditional Chinese medicine'' as the keywords to search PubMed, China National Knowledge Infrastructure (CNKI) and other literature databases. The correlation between 5-HT and constipation as well as brain-gut-microbiome axis and the research progress of TCM intervention in the 5-HT signaling pathway in the treatment of constipation were reviewed in order to explore the potential therapeutic value of 5-HT system in this disease and provide references for subsequent research.
7.Regulation of Constipation-related Signaling Pathways by Traditional Chinese Medicine: A Review

The pathophysiological mechanism of constipation has not been fully elucidated, and studies have mainly focused on the dysfunction of the enteric nervous system (ENS), abnormalities of Cajal interstitial cells (ICCs), alterations in mucosal immune function (low-grade intestinal inflammation, intestinal barrier damage, and immune activation), abnormalities of gastrointestinal hormones, dysregulation of intestinal flora, and dysfunctions of intestinal dynamics. Traditional Chinese medicine (TCM) is a unique and precious resource in China with good efficacy and safety. TCM and its active ingredients can effectively treat constipation, and its mechanism of action needs to be further investigated. Signaling pathways are involved in the development of constipation, which is one of the main targets in the study of pathological mechanisms of constipation and the pharmacological study of therapeutic drugs. Therefore, the authors searched PubMed, China National Knowledge Infrastructure (CNKI), and other literature databases with the keywords of ''constipation'', ''signaling pathway'', and ''traditional Chinese medicine'' to systematically summarize the signaling pathways related to constipation regulated by TCM. The comprehensive analysis shows that TCM mainly plays a role in regulating autophagy, apoptosis, and proliferation and ensuring anti-oxidative stress, anti-inflammation, and protection of intestinal barrier of ICCs by modulating signaling pathways such as stem cell factor/tyrosine kinase receptor (SCF/c-kit), phosphatidylinositol 3-kinase/protein kinase B (PI3K/Akt), 5-hydroxytryptophan (5-HT), mitogen-activated protein kinases (MAPK), and Toll-like receptors (TLRs). TCM regulates neurotransmitter secretion and intestinal water metabolism to improve constipation. This demonstrates the advantages of multi-level, multi-target, and multi-level effects of TCM, and in-depth research can provide new ideas for exploring the pathogenesis of constipation and developing new drugs.
