1.Role of EP_2 in synaptic transmission in dentate gyrus of hippocampus
Journal of Xi'an Jiaotong University(Medical Sciences) 2010;31(1):59-62
Objective To evaluate the role of EP_2 in synaptic transmission in dentate gyrus of hippocampus. Methods Field excitatory post-synaptic potentials (fEPSP) were recorded at the perforated path-granule neurons in dentate gyrus in vitro. Results ① Butaprost, an agonist of EP_2, enhanced the synaptic transmission in dentate gyrus and decreased the paired-pulse ratio, and these effects were reversed by bath application of AH6809 (EP_2 antagonist). ② Application of Forskolin alone or with AH6809 elevated the slope of fEPSP. ③ The Butaprost-induced responses were mediated via PKA, ERK and IP_3 signal pathways. Conclusion Multiple signal pathways were involved in the EP_2 activation-mediated enhancement of synaptic transmission.
2.COX-2 modulates Δ~9-THC-induced inhibition of LTD in CA1 area
Journal of Xi'an Jiaotong University(Medical Sciences) 2009;30(6):701-704
Objective To explore the role of cyclooxgense-2 (COX-2) in tetrahydrocannabinol (THC)-induced inhibition of long-term depression (LTD) in CA1 area in vitro. Methods The hippocampal slices were prepared at 24 h after intraperitoneal injection of Δ~9-THC (10 mg/kg) or COX-2 inhibitor NS398 (10 mg/kg). Field excitatory post-synaptic potentials (fEPSP) were recorded in the stratum radiatum of the CA1 area in vitro to observe the effect of NS398, a selective inhibitor of COX-2, on the THC-induced inhibition of LTD and THC's effect on membrane excitability in pyramidal neurons by using the whole-cell patch clamp technique. Results ① Low-frequency stimulation (1 Hz, 15 min) induced-LTD in CA1 area was significantly attenuated by Δ9-THC. ② Δ~9-THC did not affect the basal synaptic transmission and membrane excitability (including membrane resting potentials, input resistance and firing property). ③ THC-induced inhibition of LTD was reversed by NS398. ④ THC-induced inhibition of LTD was robustly impaired in COX-2 knockout mice. Conclusion THC-induced inhibition of LTD in CA1 area was mediated via COX-2.

7.Investigation of topical ofloxacin liposomes in the treatment of experimental pseudomonas keratoconjunctivitis in rabbits
Ophthalmology in China 1994;0(02):-
Pseudomonas aeruginosa was inoculated into corneal stroma of rabbits and the model of keratoconjunc tivitis was established. Empty liposomes were used in a control, group ofloxacin liposome and ofloxacin eyedrop groups as experimental ones. After treatment for six times conjunctival and corneal lesions were examinated. Swabs of conjunctival sac were cultured. Result: in conjunctiva, lesions in the two experimental groups were all significantly less than those in the control group on the 6th, 8th and 11th day. Lesions in the ofloxacin liposomes group were also significantly less than those in the ofloxacin eyedrop group on the6th day. In cornea, lesions in the ofloxacin liposomes group were significantly less than those in the control group on the 6 th, 8th and 11th day. Lesions in the ofloxacin eyedrop group were significantly less than those the control group on the 11th day. In the result of bacterium culture, the culture's positive rate of two experimental gronqs and control group had significant differences on the 6th day. On the 11th day, the positive rate the ofloxacin liposome groups and ofloxacin eyedrop groups increased, and in the ofloxaein liposomes group had a less increase. Conclusion: ofloxacin liposomes and ofloxacin eyedrops were effective in treatments of pseudomonas keratoconjuntivitis in the rabbit model, the effects of ofloxacin liposomes were better than that of ofloxacin eyedrops.
8.Study on influence of AEG-1 siRNA on proliferation and apoptosis of cervical carcinoma SiHa cells
Chongqing Medicine 2014;(17):2139-2141
Objective To investigate the effect of siRNA-down-regulating astrocyte elevated gene-1(AEG-1)on the proliferation and apoptosis of cervical carcinoma cells. Methods The designed and synthesized AEG-1 siRNA was transfected to the cervical car-cinoma SiHa cells. The influence of AEG-1 siRNA on the expression of AEG-1 gene was detected by real-time fluorescent quantita-tive-polymerase chain reaction (RT-PCR);the inhibiting effect of AEG-1 siRNA on the proliferation of cervical cancer SiHa cells was detected by the MTT method;the influence of AEG-1 siRNA on apoptosis and cell cycle of the cervical cancer SiHa cellswas determined by the flow cytometry. Results Compared with the control group,AEG-1 siRNA transfecting human cervical cancer Si-Ha cells could evidently down-regulate the expression of AEG-1 gene with statistical difference (P<0.01);at the same time, the dwn-regulation of AEG-1 gene expression could significantly inhibit the proliferation of the cervical cancer SiHa cells (P<0.01), promote the apoptosis of SiHa cells (P<0.01)and arrest the cell cycle in G0/G1 phase. Conclusion AEG-1 siRNA can down-regulate the expression of AEG-1 gene in the cervical carcinoma SiHa cells, inhibit the proliferation of the cervical carcinoma SiHa cells and promote their apoptosis.
