The purpose of this study is to demonstrate if Gadolinium-enhanced MRI can detect early reversible ischemia of the femoral head epiphysis caused by hip hyper-abduction in piglets. Between 3 and 6 h consistent hyper-abduction, gadolinium-enhanced MRI was performed in 20 femoral heads of 10 piglets. After completion of MRI scan, the piglets were allowed to ambulate freely for 1 or 7 days and re-imaged. The enhanced-MRI results of epiphyseal and physeal cartilage and the secondary center of ossification were observed. MRI appearances and histological findings were compared. On Gadolinium-enhanced MRI, decreased or absent enhancement was seen in 14 cartilaginous epiphyses of all 20 femoral heads. Reperfusion was completed in 10 of 14 femoral heads after one day of ambulation and in the rest 4 after 7 days of ambulation. Gadolinium-enhanced MRI can identify early ischemia and its reversal of the capital femoral epiphysis induced by hip hyper-abduction.

This study is to investigate the inhibitory effect of lidamycin (LDM) and its combination with methotrexate (MTX) on lung metastasis of fibrosarcoma by bioluminescence imaging in athymic mice. A stable luciferase transfected HT-1080 cell line was constructed and the capability to establish experimental lung metastasis in athymic mice was confirmed. The optical imaging system was applied to evaluate the formation of lung metastasis in vivo. In addition, metastatic nodules were counted for the evaluation of inhibition rates. As shown, the fluorescent intensity of luciferase-transfected HT-1080 cells was colinear with the cell population and the minimal detected cell population was 100 cells/well. Optical imaging showed that the fluorescent intensity of treated group was apparently lower than that of the control. The inhibition rates of lung metastasis by LDM alone at 0.025 mg x kg(-1) and 0.05 mg x kg(-1) were 53.9% and 75.9%, respectively, while that of MTX alone at 0.5 mg x kg(-1) was 70.2%. The combination of LDM at 0.025 mg x kg(-1) and MTX at 0.5 mg x kg(-1) showed an inhibition rate of 88.7%. The coefficient of drug interaction (CDI) was 0.82. The results herein demonstrated that LDM alone had strong anti-metastasis effect on human fibrosarcoma HT-1080 and the inhibition efficacy is strengthened when combined with MTX.

ObjectiveOptic nerve stimulation with penetrating electrode.is a new method for developing visual prosthesis.A simulation system was developed with computer software MATLAB to investigate its mechanism.MethodsVolume conductor model of optic nerve and optic nerve fiber model were developed. With the stimulation of monopolar penetrating electrode, the excitation thresholds of fibers at different depth were calculated. The activating function and activation region were employed to characterize the external stimulation effect. The impact of different parameters on stimulation effects were explored by changing the fiber diameter and the stimulation pulse width. Results Excitation thresholds increased as well as the percentage of activated fibers with the increase of electrode-fiber distance.Excitation threshold of the same depth fiber decreased as the fiber diameter increased, and the longer the electrode-fiber distance was,the more significant the drop was.Excitation threshold of the same depth fiber decreased as the pulse width of monophasic rectangle wave increased. The excitation threshold of the fiber at the same depth hardly changed for pulse durations greater than 0.5 ms.ConclusionThe simulation results indicates that optical nerve stimulation can be realized with penetratingelectrode, and different stimulation parameters can produce different activation effects. The present results providetheoretical guidance for the design of experiments.
