The establishment of cerebral collateral circulation is a secondary compensatory mechanism after cerebral ischemia.The expression of endogenous angiogenic growth factors is up regulated,angiogenesis and arteriogenesis are initiated,and the establishment of collateral circulation is accelerated after cerebral ischemia.Some drugs may promote the establishment of collateral circulation after ischemic stroke and improve the prognosis of patients.
2.Invesitigation of the effects of Compound Danshen on endothelins and 6-Keto-PGF_(1?) of patients with pulmonary heart disease

Objective To observe the changes of endothelin and 6 Keto PGF 1? of patients with pulmonary heart disease and effects of Compound Danshen,to study the role of ET 6 Keto PGF 1? in pulmonary heart disease and pulmonary artery hypertension,and explore a safe and effective drug for pulmonary artery hypertension Methods 40 patients with CPHD and 30 patients with COPD as well as 20 healthy persons as control whose levels of plasma ET and 6 Keto PGF 1? were measured by radioimmunization,meanwhile,PaO 2 was measured by blood gas analyze instrument Results The level of ET in CPHD was higher than that in COPD and healthy group,company with the lower level of 6 Keto PGF 1? After treatment with Compound Danshen,the level of ET was reduced and 6 Keto PGF 1? was elevated Conclusions The balance between ET and PGI is important in modulating pulmonary circulation Compound Danshen can reduce pulmonary artery tension through protecting endothelium,decreasing ET,and increasing PGI 2
3.A prospective study of tumor prevention: effects of synthetic peptides on in vitro invasive ability of highly metastatic human lung giant-cell carcinoma cell line

BACKGROUND: The invasion of highly metastatic carcinoma cells on reconstituted basement membrane component is considered as a common model for in vitro experimental studies of invasion and metastasis of malignant tumors. It has been reported that synthetic peptide influences some tumor cells. However, the effect of synthetic peptide on highly metastatic human lung giant-cell carcinoma cell(PG) has been rarely studied.OBJECTIVE: To study the effects of synthetic peptide RGD, YIGSR and their derivatives on invasive ability of PG in vitro.DESIGN: A completely randomized controlled trial was conducted.SETTING and MATERIALS: The human lung highly metastatic tumor and NIH3T3 cell line were both supplied by the Department of Pathology of Beijing Medical University. The synthetic peptides were provided by the Synthetic Laboratory of the Institute of Materia Medica of the Chinese Academy of Medical Sciences and Chengde Medical College. Transwell invasion chamber was purchased from Costor Company. The whole experiment was conducted at the Department of Biochemistry of Chengde Medical College.INTERVENTIONS: NIH3T3 cell line was used to prepare chemotactic factor. PG cells were seeded in Transwell invasion chamber. Under the effect of chemotactic factor, the effects of synthetic peptides on PG invasive ability were examined.MAIN OUTCOME MEASURES: PG cell number of passing through the reconstituted basement membrane was used as an index to evaluate the invasive ability of tumor cells.RESULTS: An appropriate number of PG cells were cultured for 10 hours with chemotactic factor, and the number ofinvasive cells reached a number range that can be applied to statistical analysis(50 to 100 invasive cells). After 100 mg/L of synthetic peptides RGD, YIGSR were incubated for 48 hours with PG cells,the rate of inhibition on invasive ability was 53.63% and 36.86% respectively, and 0% and 6.48% respectively after incubation for 10 hours.CONCLUSION: The PG cells were the proper cells for in vitro experimental studies of invasive ability of cancer cells for the purpose of screening of anti-tumor medicine. RGD and Juncha/Zacha-peptide YIGSR showed a strong activity in inhibiting tumor invasion and a potential value in prevention and treatment of malignant tumor development.
4.Subchronic Toxicological Assessment of Diosgenin in Kunming Mice

This study was aimed to evaluate the subchronic toxicity of diosgenin in mice. A total of 80 mice were divided into 4 groups, which were 0 (control), 100, 200, and 400 mg·kg-1 by the random number table. Intragastric administration was given once a day for 90 days in the assessment of subchronic toxicity of diosgenin among mice. The observed indexes contained body weight, fur color, diet, feces, and etc. The detected indexes contained blood routine analysis, blood biochemistry and pathological examination. The results showed that compared with the control group, the body weights of mice in the male medication group were slight reduced. There were no significant hematologic and pathologic abnormalities. It was concluded that the subchronic toxicity of diosgenin with no observed adverse effect dose level was more than 400 mg·kg-1. The oral administration was relatively safe.
5.Experimental Study on Anti-tumor Prescription Screening and Therapeutic Effects of Shengma Combined with Zao Jiao-ci

Objective To screen the anti-tumor prescription (Shengzao Fang) of Shengma (Rhizoma Cimicifugae) combined with Zao Jiao-ci (Spina Gleditsiae), and evaluate its anti-tumor effects. Methods The lung metastatic model of 4T1 breast cancer and the 4T1 subcutaneous transplanted model were established, both models of mice were randomly divided into model group, Shengma group, Zao Jiao-ci group, Shengzao Fang first group, Shengzao Fang second group and Shengzao Fang third group, 10 mice for each group. The proportion of Shengzao Fang was ascertained by their anti-tumor effects. MTT assay was used to investigate the effects of Shengzao Fang on 4T1 breast cancer. The concentration of type 1 collagen (Col-Ⅰ), platelet-activating factor (PAF), tissue inhibitor of metalloproteinase 1 (TIMP-1), tyrosine kinase receptor A (TrkA) and tyrosine kinase receptor B (TrkB) in serum from tumor-bearing mice was tested by enzyme-linked immunosorbent assay (ELISA). The effects of Shengzao Fang on matrix metalloproteinase (MMPs) was examined by Gelatin matrix method. Results Compared with the model group, the prescription of Shengma∶Zao Jiao-Ci=2∶1 significantly suppressed lung metastasis of 4T1 breast cancer, showing fewer lung nodes, lower lung metastasis rate and highest tumor inhibitory rate to 4T1 subcutaneous transplanted model. Although the concentration of serum Col-Ⅰ, PAF, TIMP-1, TrkA and TrkB was decreased in all treated group, the prescription of Shengma∶Zao Jiao-ci=2∶1 had the strongest activity, and its inhibitory effect on the expression of MMP-2 and MMP-9 was strongest, too. Conclusion The prescriptions of Shengma combined with Zao Jiao-ci had different antitumor activity, the strongest activity was exhibited when shengma∶Zao Jiao-ci was 2∶1, suggesting that 2∶1 was optimization for Shengma combined with Zao Jiao-ci.
6.Value of model for end-stage liver disease combined with serum sodium in predicting the prognosis of cirrhosis

Objective To evaluate the capability of model for end-stage liver disease (MELD)combined with serum sodium (MELD- Na,MELDNa and MESO scores) in predicting the prognosis of patients with decompensated liver cirrhosis in 6 and 12 months.Methods One hundred and nineteen patients with decompensated liver cirrhosis and completed follow-up data were retrospectively studied.The MELD,MELD- Na,MELDNa and MESO scores were calculated according to the clinical data of each patient.Receiver operating characteristic (ROC) curve and the area under the curve (AUC) was used to measure the values of the four models in predicting the 6 and 12 months survival,and Z-test was used to compare their predictive values.Results MELD,MELD-Na,MELDNa and MESO scores were significantly different between patients who survived and those who died within 6 and 12 months follow-up.The AUC for the MELD- Na,MELDNa and MESO scores were all more than 0.8 in predicting 6 and 12 months survival.However the differences of the AUC between the MELD score and MELD-Na,MELDNa,MESO scores were not significant in predicting 6 and 12 months survival.Conclusion The model for MELD combined with serum sodium can accurately predict the prognosis of patients with decompensated liver cirrhosis in 6 and 12 months,while these scores are not superior to MELD score.
7.Proliferations of T cell lines towards neural myelin sheath components in multiple sclerosis induced by neural myelin sheath and delipidated neural myelin sheath

BACKGROUND: Multiple sclerosis(MS) is a chronic autoimmune disease induced by the interaction between genetic and environmental factors. Its pathogen and the mechanism of the relapse and remission m the course of the disease are still unknown. Most of the MS research centers are looking for the pathogenic polypeptide epitope in proteolipid protein(PLP), myelin sheath basic protein (MBP) and oligodendrocyte glycoprotein (MOG) OBJECTIVE: To compare the proliferation of T cell lines(TCL) in MS induced by myelin sheath and delipidated myelin sheath towards 11 components of myelin sheath to mainly search the possible pathogenic polypeptide epitope in PLP, and investigate the possible effects of abnormal dcgrease in myelin sheath.DESIGN: A case-controlled trial.SETTING: Department of neurology in a hospital of a university.PARTICIPANTS: Mononuclear cells(MNC) of 16 MS cases(clinical relapsing-remitting type, patients did not receive any immunosuppresant for at least 3 months when their peripheral blood samples were taken) and 12 HLA-DR15 healthy volunteers were furnished by Dr. Trotter JL of MS Research Center of Washington University from the cell database.INTERVENTIONS: MS-TCL and normal TCL were induced twice by stimulation with myelin sheath and delipidated myelin sheath in vitro by cell culture in vitro. TCL proliferation was tested by 11 antigens including PLP,MBP, M87-106, P30-49, P40-60, P89-106, P95-117, P117-137,P139-151, P178-191, and P185-206.MAIN OUTCOME MEASURES: Difference of scintillation counting in every minute of every well, and the stimulative index of each well were calculated, and the mean wells with positive proliferation of TCL towards each antigen were confirmed as well.RESULTS: The general specific proliferation towards myelin sheath antigens was bigger in MS group than control group 5.49 ±5.31 to 3.10 ± 3. 17, and delipidated myelin sheath-induced TCL was bigger than myelin sheath-induced one 5. 49 ± 5.31 to 3.41 ± 4. 83 . Delipidated myelin sheath significantly changed the immune responses of MS group,especially the changes of responses towards P30-49, P40-60, P89-106,P117-137, P139-161, and P185-206 were significant compared with that the control group only responded to two polypeptides, which indicated that the antigen epitope of MBP, PLP, M87-106, P95-117, P40-60, and P185-206 might have significance in the triggering of MS autoimmune responses.CONCLUSION: TCL induced by MS myelin sheath has different proliferation towards antigen components of myelin sheath from control group. Delipidated myelin sheath significantly increases TCL proliferation in MS group, which suggests that if MS patients developed abnormal degrease in myelin sheath, TCL would produce autoimmune response towards self-myelin sheath, MBP, PLP and its polypeptide segments all can trigger MS or aggravate the state of the illness. Our finding supports the hypothesis of MS autoimmune pathogenic mechanism.
8.Investigation on the status of human resources for AIDS control at city and county level in Hebei

Objective To understand the status of human resources for AIDS control at city and county level in Hebei Province,and to improve the outfit of human resources.Methods General investigation and self-designed questionnaire were used by the trained investigators.Epi Data 3.0 was used to establish a database,and SPSS 13.0 was used to analyze the data.Results There were 1 347 missionaries at the city and county level,547 of whom were full-time staff.Significant differences existed in academic qualifications and titles between city and county level.A lack in technical staff in county level appeared.Conclusions The number of staff for HIV control in Hebei is not sufficient.Therefore human resources in these places should be strengthened and rationally allocated for AIDS control.
9.Cost-effectiveness Analysis of Xueshuantong for Injection versus Ginkgo Leaf Extract and Dipyridamole In-jection in the Treatment of Ischemic Stroke

OBJECTIVE:To compare pharmacoeconomic and effect of Xueshuantong for injection and Ginkgo leaf extract and dipyridamole injection in the treatment of ischemic stroke. METHODS:Retrospective study was conducted. Totally 404 inpatients with ischemic stroke were divided into Xueshuantong group(271 cases)and ginkgo leaf extract and dipyridamole group(133 cas-es) according to clinical treatment programs. Based on the conventional treatment,patients in 2 groups were given Xueshuantong for injection and ginkgo leaf extract and dipyridamole injection,respectively. The average treatment course was 10 d. Cost-minimi-zation analysis was performed with the determination index of total effective rate. RESULTS:The total effective rates in Xueshuan-tong group and ginkgo leaf extract and dipyridamole group were 90.77% and 88.72%,respectively,the difference was not statisti-cally significant(P>0.05). The costs in 2 groups were 12 860.21 yuan and 13 155.40 yuan,respectively,and xueshuantong group had lower than ginkgo leaf extract and dipyridamde group. CONCLUSIONS:Both Xueshuantong for injection and Ginkgo leaf ex-tract and dipyridamole injection are effective in the treatment of ischemic stroke. However,the economy of Xueshuantong for injec-tion is superior to the other one.
10.Effect on Discornin Tablets of Nuclear Transcription Factor NF-κBp65 in RSC-364 Cells

This study was aimed to observe the influence of Discornin Tablets on activation nuclear transcription factor NF-κBp65 of rheumatoid arthritis (RA) cell model as well as the expression of MMP-9, VEGF and tumor necrosis factor-α (TNF-α). Interleukin-17 (IL-17) and TNF-α were used for stimulating RSC-364 cells. Discornin Tablets at different concentrations were used for intervention. The influence of Discornin Tablets in different concentrations on cell viability was detected by MTT method. Expressions of NF-κBp65 and its inhibitory protein (IκB-α) in each group were detected by western blot method. Changes in VEGF, MMP-9 and TNF-α protein levels in cell broth supernatant were checked by ELISA. The results showed that Discornin Tablets can promote the expression of κB inhibitory pro-tein, reduce the high expression of NF-κB protein level, and inhibit the cellular secretion of VEGF, MMP-9 and TNF-α. It was concluded that Discornin Tablets had negative regulation effect on nuclear transcription factor κB of RSC-364 cells. It can increase the expression of IκB-α, as well as reduce the secretion of inflammation factors and blood vessel newborn factors. It suggested that Discornin Tablets may have the potential regulation effect on RA.
