1.Research on the clinical efficacy of standard large trauma craniotomy for intracranial hematoma ;caused by severe traumatic brain injury

Objective To investigate the effects of standard large trauma craniotomy on neuron-specific enolase(NSE)inflammatory factors in patients withintracranial hematoma caused by severe trau-matic brain injury(sTBI).Methods A total of 64 cases of sTBI patients were randomly divided into con-ventional surgery group (conventional group)and standard large trauma craniotomy group (standard group),with 32 cases in each group.The postoperative Glasgow Outcome Scale(GOS)score,preoperative and postoperativeserum inflammatory factors(IL-8,IL-6,TNF-α,ICAM-1 ,and IL-1 0),NSE levels,and postoperative complications were compared.Results The GOS score of the standard group was signifi-cantly superior tothe conventional group(P <0.05);postoperativeIL-6,TNF-α,ICAM-1 ,and NSE levels in the conventional group were significantly reduced(P <0.05);postoperativeIL-8,IL-6,TNF-α,ICAM-1 ,and NSE levels were significantly reduced and IL-1 0 levelswassignificantly improvedin the standard group(P <0.05);there were3 cases of postoperative cerebrospinal fluid leakage and four cases of inci-sional hernia in the conventional group,and two cases of postoperative cerebrospinal fluid leakage and three cases of incisional hernia in the standard group;there were no significant differencesin the incidence of postoperative complications in both groups(P >0.05 ).Conclusion Inflammatory factors and NSE both playimportant roles in sTBI.Standard large trauma craniotomycan control these indicators effectively and reduce the severity of the patient's illness.
2.Clinical Research on Cervus and Cucumis Polypeptide Combined with Duhuo Jisheng Decoction in the Treatment of Lumbar Intervertebral Disc Herniation

Objective To observe the clinical efficacy of cervus and cucumis polypeptide combined with Duhuo Jisheng Decoction Jiaji pain point injection in the treatment of lumbar intervertebral disc herniation. Methods Totally 68 cases of patients with lumbar intervertebral disc herniation were randomly divided into treatment group (34 cases) and control group (34 cases). Both groups were given Duhuo Jisheng Decoction orally besides basic treatment, and the treatment group received cervus and cucumis polypeptide Jiaji pain point injection additionally. Both groups were in a course of 10 days and treated for two courses. The clinical symptoms, visual analog pain scale (VAS), and Oswestry disability index (ODI) of the two groups were observed and compared among those before treatment, after one and two courses of treatment. Results Compared with the pretreatment of the same group, the VAS and ODI scores were lower after the first and second courses of treatment (P<0.05). After one and two course of treatment, VAS and ODI of the treatment group were lower than the control group (P<0.05, P<0.01). After one course of treatment, the total efficiency of treatment group was 91.2%(31/34), while the control group was 70.6%(24/34);after two courses, the total efficiency were respectively 100%(34/34) and 85.3%(29/34);the treatment group was better than control group during the same period (P<0.05). Conclusion Cervus and cucumis polypeptide combined with Duhuo Jisheng Decoction Jiaji pain point injection has good clinical efficacy in the treatment of lumbar intervertebral disc herniation.
4.The cytotoxicity, biocompatibility and morphological observation of bone formation traced in porous tantalum

Objective To investigate cytotoxicity,biocompatibility and new bone formation traced of Chinese porous tantalum,and provide experimental strategies for further clinical application.Methods The physical properties of the porous tantalum were observed by the SEM.The osteoblasts were isolated from rabbit embryo.The extract fluid from tantalum was made.The cytotoxicity and proliferation of osteoblasts compounded with porous tantalum in vitro were detected by the MTT assay.The osteoblasts were co-cultured with extract of tantalum in vitro and the morphological changes,proliferation and adhesion were observed under SEM.A total of 24 New Zealand rabbits were used to establish the model of femoral condyles with porous tantalum bars implanted.Among which 4 of them was injected with calcein and alizarin on the 5th day and the 19th day and sacrificed at 10 week postoperatively.The specimens were observed with LSCM at 488 nm and 543 nm wavelength respectively.The remained 20 animals were sacrificed successively at 2,4,8,12 weeks of implantation,then were examined by histological observation.Results The SEM showed that the pore of porous tantalum were three-dimensional connected morphology.MTT assay showed that the osteoblasts grew well in extract and no significant difference between experimental and control groups.The osteoblasts grew and spread extensively on porous tantalum.Early on co-culture,the osteoblasts attached to the surface and inner walls of material,in the later stage,the osteoblasts excreted bone matrix over the surface of porous tantalum.The animal model showed that porous tantalum was bonded closely with host bone.Hard slicing showed that new bone and capillary regenerated on tantalum-bone interface at 2,4 weeks postoperatively.The pores were full with bone tissue at 8,12 weeks.The LSCM indicated that the green and red fluorescence-labeled new bone was displayed on tantalum-bone interface,while the red zone located around the green zones.They appeared to be discontinuous at early stage,but connected with each other at the end.Conclusion The Chinese porous tantalum has good biocompatibility and no cytotoxicity.The contact osteogenesis and bone conduction exist in tantalum-bone interface,and in a time-dependent manner.
5.Pharmacokinetic Study on Ferulic Acid in Plasma of the Healthy Female Volunteers after Oral Administration of Shenghua Decoction

Objective To study the pharmacokinetics of ferulic acid (FA) in plasma of the healthy female volunteers after oral administration of Shenghua decoction.Methods Using p-hydroxybenzaldehyde as the internal standard, the plasma concentration of FA was determined by RP-HPLC. Plasma samples were extracted and treated with boiling water and 3P97 programme was used to calculate the pharmacokinetic parameters. Results The main pharmacokinetic parameters of FA were as follows: T1/2?=18.72 min ,T1/2?=79.21 min,T1/2 Ka=11.19 min,AUC=18004.87 ?g?min?L-1,CL=0.17L?min-1?kg-1,Cmax=206.30 ?g?L-1, Tpeak=22.78 min.Conclusion After oral administration of Shenghua decoction, FA could be absorbed and eliminated rapidly and pharmacokinetics of FA conforms to a two-compartment open model.
6.Toxic experimental study of Antineoplastic agents-Fotemustine

Objective To evaluate the safety of Fotemustine. Methods The studies had been done according to the experimental method of injection agents in pharmacodynamics guide principle on toxic test methods of application. The vascular stimulative assay of fotemustine injection on rabbit, the hemolyzation or agglutination of it in vitro, and the allergic reaction on guinea pig were performed. Results It was showed that the Fotemustine injection had no vascular irritation and injury effect on rabbit vascular endothelial cell. Compared with that of the control group, there was no difference. Conclusion No evidences of hemolyzation and aggregation on rabbit erythrocyte, and no allergic reaction on guinea pig were observed.
7.Effects of cyclooxygenase-2 inhibitor celecoxib on the phenotypes and function of glioma dendritic cells

ObjectiveTo investigate the effects of glioma microenvironment and COX-2 inhibitor celecoxib on the phenotypes and function of dendritic cell (DC).MethodsThe expression of cyclooxygenase-2 (COX-2) and prostaglandin E2 (PGE2) production were detected in glioma C6 cells treated with different concentration of celecoxib.Monocytes were isolated from human peripheral blood and cultured with 200 ng/ml rhGM-CSF and 50 ng/ml rhIL-4,either C6 tumor cells supernatant (TSN) or TSN from C6 cells treated with celecoxib to generate DCs.Cell morphology was observed.Cell phenotype including CD1a,CD80,CD83 and D86 were analyzed on a FACScan.Production of IL-12 in DC supernatant and the potential to stmiulate allogeneic T cell proliferation were detected.ResultsThe expression of COX-2 and PGE2 production in C6 cells decreased after treated with celecoxib in a concentration dependant manner.Typical DCs were induced in all groups and the expression of CD1a ((75.56±2.40)%,(75.09±3.67)%,(76.03 ±3.43)%),CD83((72.04±3.45)%,(71.44±3.78)%,( 73.63 ± 3.31 ) % ) had no difference (P > 0.05 ).Expression of CD80 ( ( 58.41 ± 3.85 ) % ),CD86 ( ( 58.22 ±3.25)% ) in DC with TSN obviously decreased compared with normal group( (70.36 ± 2.91 )%,(69.31 ±4.29 ) %,P < 0.01 ) as well as the IL-12 production ( ( 137.88 ± 5.33 ) pg/ml,( 186.04 ± 4.76 ) pg/ml) and the potential to stmiulate allogeneic T cell proliferation ( P < 0.01 ).Celecoxib increased the expression of CD80,CD86 (66.83 ± 2.51,63.51 ± 5.47,P< 0.01 ) in DC and the same as IL-12 production( ( 170.31 ± 3.46) pg/ml,P < 0.01 ) and the potential to stmiulate allogeneic T cell proliferation ( P < 0.01 ),which were lower than the normal level.ConclusionGlioma microenvironment may induce the celecoxib can inhibit the expression of COX-2 and PGE2 in gliomas cells and improve the phenotypes and function defect of DCs.
8.Polysaccharide of spirulina on the prophylaxis and treatment of myelosup pression of mice caused by cyciophosphamide

Aim The effect of polysaccharide of spirulina on mouse myelosuppression induced by cyclophosphamide was studied. Methods Mice in the model, treatment groups Ⅰ and Ⅱ were given cyclophosphamide(ip 100 mg · kg- 1) for there days. At the same time mice in the treatment groups Ⅰ and Ⅱ were given ig spirulin 30 mg · kg-1 and 60 mg · kg-1 for ten days, while mice in the control group and model group were given ip 20 mg · kg-1 of NS. Peripheral blood and marrow cytometry, micromcelsus and chromosome aberration were analyed. Results In the dosages of 30 and 60 mg · kg-1 polysaccharide of spirulina, the polysaccharide of spirulina had perfect action on the perihemateikon and the function of bone hematoplast induced by cyclophosphamide(P <0.01) and antagonistic action on increase of bone cell micronuclsus(P<0.05); The ratio of chromosome aberration in the treated group was lower compared with that in the model group. The inhibitory ratio was 51.2% and 62.5% in the treated groups Ⅰ and Ⅱ .Conclusion Polysaccharide of spirulina can perfect the hematoplast function of mice injured by cyclophosphamide.
9.Effects of cistanche desertica polysacchrides on the constitution of protein and anti-oxidative capacity of lune in aging mice

AIM The relation between lung collagen,lung elastin and anti-oxidative capacity was investigated in the experiment, and the aging model of mice was induced by D-galactose. METHOD The aging mice treated with cistanche desertica polysacchrides (50,100 mg*kg-1*d-1) and determinate the modification of SOD、GSH-Px、Vit E、MDA in blood and lung. RESULTS Cistanche desertica polysacchrides (CDP), which was isolated from Cistanche desertica cultivated in Xinjiang could enhance lung anti-oxidative capacity and inhibit the modification of lung elatic constitution induced by D-galactose. CONCLUSION Cistanche desertica polysacchrides have anti-oxidative injury and anti-aging effects on lung.
10.The expremental research of sodium ferulate on asthmatic guinea pig

Aim To evaluate the pharmacological effects of sodi um ferulate on asthmatic guinea pigs. Methods Guinea pigs were sensit ized by ovalbumin. Two weeks later,ment of Sodium ferulate were injected every d ay five minutes before inhale, the guinea pigs were killed 7 days later. Granul ocyte-macrophage colony-stimulating factor (GM-CSF), interleukin-4 (IL-4), interleukin-5(IL-5), Eosinophil cationic protein (ECP) in serum were measured by Enzyme-linked immunosorbent assay(ELISA) and fluoroimmunoassay method, the e osinophils (EOSs) were counted and the phosphodiesterase (PDE) in the bronchoalv eolar lavage fluid(BALF) were determined by inverse high-performance liquid chr omatography (HPLC) method. Result After treatment with sodium f erulate for one week at the dose of 25,50,100 mg?kg -1 respectively, th e GM-CSF, IL-4, IL-5 contents were inhibited as compared with the untreated g roup in a dose dependent manner.The count of eosinophil was decreased.The activi ty of PDE of cell in BALF and the level of ECP were decreased.Conclusion Sodium ferulate attenuates the GM-CSF, IL-4, IL-5 in serum and eosi nophils and the activity of PDE and ECP and has protective effect in asthmatic g uinea pig.
