1.Effects of Danshen Decoction on experimental gastric ulcer in rats and mice

OBJECTIVE: To investigate the preventive and therapeutic effects of Danshen Decoction on gastric ulcer in experimental animal models. METHODS: The model of water immersion restraint stress ulcer in mice, and the models of acetic acid impaired gastric ulcer and pyloric ligation gastric ulcer in rats were established. The total gastric acid, the activity of pepsin and the amount of gastric wall binding mucus were detected. RESULTS: Danshen Decoction reduced the acute gastric mucosal lesion. The effect of 5.0 or 10.0 g/kg group was obviously better than that of the normal saline control group (P<0.05 and P<0.01 respectively). Danshen Decoction accelerated the healing of ulcer induced by acetic acid significantly, and increased the content of gastric wall binding mucus (P<0.01); and it also inhibited the formation of gastric ulcer induced by pyloric ligation, while having no influence on the secretion of gastric acid and the activity of pepsin. CONCLUSION: Danshen Decoction has a significant anti-ulcerative effect, which may be related to improving the gastric mucosal defensive function.

3.Therapeutic effect of syringin on adjuvant arthritis in rats and its mechanisms.

This study is to investigate the therapeutic effect of syringin on adjuvant arthritis (AA) in rats and its mechanisms. Complete Freund's adjuvant (FCA) was used to induce AA in rats. Secondary paw swelling of AA rats was measured with volume meter. Pain response and polyarthritis index were scored. Meanwhile, splenic lymphocyte proliferation response induced by concanavalin A (ConA) or lipopolysaccharide (LPS) was examined with MTT assay. IL-2 production of splenic lymphocytes and IL-1 beta, TNF-alpha production of peritoneal macrophage (PM phi) were estimated by enzyme linked immunosorbent assay (ELISA). The secondary inflammation of AA rats appeared on the 14th day after injection of FCA. Syringin and tripterygium glycosides (TG) were given by intragastric administration for 16 days from the 14th day. Treatment of AA rats with syringin and TG from the 22th day significantly attenuated the secondary hind paw swelling, as well as relieved the pain response and the polyarthritic symptoms of the whole body as compared with that of the AA model group. The suppressed lymphocyte proliferation and IL-2 production of splenic lymphocytes in AA rats were reversed by treatment with syringin. Meanwhile, syringin remarkably down-regulated IL-1 beta, TNF-alpha productions from PM phi. These results indicate that anti-inflammatory effects of syringin on AA rats are mediated by modulating the immune function of abnormal cells and the balance of cytokines.
4.Effect of Echinacoside on immune function and mitochondrial DNA relative content of aging mice

Aim To study the effect of Echinacoside on immune function and the hepatocyte mitochondrial DNA relative content of experimentally aging model mice. Methods Mice were given sc 10%D-galactose 10 ml?kg-1,once daily for 8 weeks to establish the model of aging mice. Vitamin E group ,and ECH groups were given ig Vitamin E 40 mg?kg-1 and ECH 20,40,60 mg?kg-1,respectively at one time. The interleukin-2(IL-2) and interleukin-6(IL-6) content in peripheral blood was measured by ELISA method.Phagocytosis of peritoneal macrophages was tested by neutral red test. Proliferation and transformation of spleen lymphocytes induced by ConA were determined by MTT method. The mitochondrial DNA relative content of liver cells was measured by the means of SDS alkali decomposition from liver.Results The IL-2 content,phagocytosis of peritoneal macrophages and proliferation of spleen lymphocytes were obviously decreased in the aging model group as compared with those in the normal control group.The IL-6 content and mitochondrial DNA relative content of liver cells were significantly increased. ECH 20,40,60 mg?kg-1 and Vitamin E 40 mg?kg-1 could improve the IL-2 content,phagocytosis of peritoneal macrophages and transformation of spleen lymphocytes,and reduce the IL-6 content and mitochondrial DNA relative content of liver cells. Conclusion ECH can enhance immune function and reduce the mitochondrial DNA relative content of liver cells,which may be one of the mechanisms of anti-aging.
5.Experimental Study of Xiaomin Granules on Antagonizing Type Ⅰ Hypersensitivity

Objective To investigate the effects and mechanism of Xiaomin granules on type Ⅰ hypersensitivity.Methods The experimental animal model was made for type Ⅰ hypersensitivity.The mice were divided into five groups named vacant control group,Xiaomin granules(low-,middle-and high-dose) group,chlorpheniramine group randomly and were given homologous medication respectively.Results Xiaomin granules can inhibit capillary penetrance caused by histamine and 5-HT in mice(P 0.05).Xiaomin granules could inhibit the mice hind paw edema degree caused by histamine.The further study found that Xiaomin granules could decrease anaphylactic shock of the mice and decrease the death rate.Conclusions Xiaomin granules could restrain type Ⅰ hypersensitivity by inhibiting inflammatory mediators.
6.The dynamic expression of EGF and NT-4/5 in the frontal lobe,hippocampus and thalamus of rats after cerebral ischemia-reperfusion

Objective:To investigate the content change of EGF and NT-4/5 in the the frontal lobe,hippocampus and thalamus of rats after cerebral ischemia-reperfusion,in order to reveal the dynamic protection of EGF and NT-4/5 to cerebellum ischemia-reperfusion injury.Methods:Wistar senile rats were divided into the control group and the experiment groups at random.The control group included 5.The experimental groups were divided into cerebral ischemia 15 min group and ischemia-reperfusion 1 h,6 h,2 d,4 d,9 d groups after 15 min ischemia,5 rats per group.Results:There was all a little expression of EGF and NT-4/5 in the frontal lobe,hippocampus and thalamus of normal senile rats.The frontal lobe was shown evident increase of EGF expression in ischemia-reperfusion 2 d only.The hippocampus was shown continued increase of EGF expression during ischemia-reperfusion 6 h to 4 d and the EGF expression was shown a peak in ischemia-reperfusion 4 d.The increase of EGF expression in thalamus continued during ischemia-reperfusion 2-9 d too.The NT-4/5 expression of the frontal brain was clearly reduced after ischemia-reperfusion and returned to normal in ischemia-reperfusion 9 d.The NT-4/5 expression of hippocampus was shown one-off reduction after ischemia-reperfusion,but the expression quickly returned to normal and shows a little increase.The thalamus had only one-off reduction of NT-4/5 expression.Conclusion:The frontal lobe lacks of fast neuroprotective mechanism of EGF in early stage of ischemia-reperfusion and it has a limited neuroprotective effect of EGF in middle and advanced stage.The hippocampus has a fast and long-lasting neuroprotective mechanism of EGF.The thalamus has a delayed neuroprotective mechanism of EGF.The frontal brain and hippocampus have a delayed neuroprotective mechanism of NT-4/5.The neuroprotective mechanism of NT-4/5 is weaker in thalamus.
7.Effects of recombinant human interleukin-1 receptor antagonist on guinea-pig isolated airway smooth muscle

AIM To study the effects of recombinant human IL 1 receptor antagonist (IL 1ra) on isolated airway smooth muscle tissue. METHODS Changes of isolated organ's tension were monitored with force displacement transducer and MedLab recording system. RESULTS ①IL 1ra haddirect relaxant effect in normal and ovalbumin sensitized pulmonary as well as tracheal smooth muscle. The mean EC 50 values were 1 29?10 -8 mol?L -1 and 8 06?10 -8 mol?L -1 in normal pulmonary and tracheal smooth muscle, but the mean EC 50 values were 2 61?10 -7 mol?L -1 and 5 88?10 -7 mol?L -1 in ovalbumin sensitized pulmonary and tracheal smooth muscle. Objective To study the chemical constituents from Onychium japonicum.Methods Che-mical constituents were isolated by repeated column chromatography and preparative HPLC,and their structures were elucidated on the basis of spectroscopic method.Results Ten compounds were identifed as:4,3',4'-trihydroxy-2,6-dimethoxychalcone(1),chrysoeriol(2),luteolin(3),butin(4),protocatechuic acid(5),3,4-dihydroxy-acetophenone(6),caffeic acid(7),vanillic acid(8),2,4-dihydroxybenzaldehyde(9),syringic acid(10),and ?-sitosterol(11).Conclusion Compound 1 is a new product named japonicone D,compounds 2-10 are isolated from the plants of Onychium Kaulf for the first time.
9.The effects of anticipatory and post-event information processing on perception and memory of the symptom in social anxiety disorder

Objective To study the influence of anticipatory and post-event information processing on the memories and perception of the symptom in patients with social anxiety disorder (SAD). Methods A group of 32 SAD patients and a control group of 35 healthy indivisuals were included. Instruments including Self-Rating Depression Scale, Subjective Units Discomfort Scale( SUDS ), Rumination Questionnaire ,Open-ended Recall and Body Sensations Questionnaire were adopted in both groups. Results In the group SAD, no significant difference was identified between the experimental group( (49.68 ± 17.68), ( 19.00 ± 1.25), (0. 54 ±0. 17) ) and the experimental control group( ( 50.43 ± 20.72 ), ( 18.68 ± 1.25 ), ( 0.52 ± 0.17 ) ) when the ratings the memories of body sensations, rumination, and negative self-information score were compared (P> 0.05 ). There was a significant positive correlation between the level of rumination ,SUDS, the memories of negative self-information and the body sensations scores( r= 0.72; r= 0.94; r= 0.70, P< 0.01 ). The scores of rumination explained 64% of the variation in SUDS scores(β=0.82, P<0. 01 ). Conclusion This study suggest that social anxiety is affected directly by rumination which can result in more memories of negative self-information and the body sensation. Symptoms are maintained by post-event information processing.
10.Changes of IL-18 and Th1/Th2 in serum of lung cancer patients and their relationship with tumor staging
