1.Application of proteomics in research for gastrointestinal tumors

Proteomics is a technology to study all the proteins encoded by gene.It has become a useful tool for analyzing the expression changes of proteins,and has been widely used in research of gastrointestinal tumors in recent years.Through kinds of techniques such as two-dimensional gel electrophoresis,some differentially expressed proteins correlated with patients prognosis were found,which may served as the possible biomarkers of gastroin testinal tumors in future.It helped to find some certain proteins related to tumor cell nutritional intake,cell metabolism,cell adhesion,cell migration and cytoskeleton remodeling which may play the important roles in development of gastrointestinal tumor.It also helped to explain the drug resistance mechacism of chemotherapy and molecular targeted theraphy for gastrointestinal tumors.Proteomics technology plays an increasingly important role in the basic research and clinical services for gastrointestinal tumors.

Itraconazole has potent anticancer activity at the standard therapeutic doses,which is proved in vitro and in vivo preclinical studies,even in active clinical trials.The precise mode of action of itraconazole for its anticancer activity has not been elucidated,however multiple putative mechanisms are proposed,such as anti-angiogenesis,inhibition of Hedgehog pathway,autophagy induction and reversal of multi-drug resistance.Its high efficacy,known toxicity and well-estabhshed pharmacokinetics make this generic drug a strong candidate for repurposing as an oncological treatment.

4.Study of the effect of methotrexate and cyclophophamide on cell cycle and cyclin DI of lymphocytes in the periphery blood and bone marrow in rats

Objective To investigate the synergistic effect of methotrexate (MTX) and cyclophos phamide (CTX) on cell cycle and cyclin D1 of periphery blood lymphocytes (PBLs) and bone marrow byflow cytometry. Methods Wistar rats were randomly divided into four groups including normal control, MTX and cyclophosphamide combination group, MTX and CTX only treatment groups respectively. PBLs were isolated for flowcytometry analysis for the changes of cell cycle and the expression of cyclin D1 at week 0, week 3,week 9, week18 and week 27. Mice were dissected and the changes of lymphoeytes cell cycle and the expressions of cyclin D1 in the bone marrow were measured at week 0, week 3, week 9, week 18 and week 27 increased and the ratio of phase S cells was decreased (P>0.05). In the CTX treatment group, there was no statistical difference in ratios of each phase. In the MTX and CTX combination treatment group, the proportion of phase G0/G1 cells decreased significantly and the percentage of phase S cells increased in both PBLs and bone marrow ceils (P<0.05). And there was no statistical significant difference in different time points after marrow between different groups or different dissecting time points. Conclusion MTX combined with CTX has been shown to have antagonistic effect on cell cycle. However, this effect is not via the cyclin DI pathway.
5.Effect of different anesthetic techniques on perioperative changes in T-lymphocyte subsets in patients with cardia cancer

Objective To observe the effects of different anesthetic techniques on T-lymphocyte subsets in patients with cardia cancer. Methods Thirty-two patients undergoing elective radical operation for cardia cancer who had been average assigned into two groups by random digits table, group Ⅰ :general anesthesia;group Ⅱ :epidural anesthesia combined with general anesthesia (16 cases in each group).Peripheral blood T-lymphocyte subsets were measured before induction, after anesthesia, end of operation, 1d,3 d after operation. Results CD3,CD4,CD8 and CD4/CD8 all decreased in two groups after anesthesia,end of operation and 1 d after operation than before induction (P < 0.05). The index almost returned to the baseline values at 3 d after operation in group Ⅱ [ (60.75 ± 4.22 )%, (39.65 ± 3.64)%, (25.90 ± 1.17 )%,1.57 ±0.15](P >0.05),while in group Ⅰ still lower [(55.83 ±5.20)%, (35.15 ±5.65)%, (23.00 ±1.03 )%, 1.47 ± 0.35 ](P< 0.05 ). The two groups ontrast had significant deviation at 3 d after operation (P<0.05). Conclusion Epidural anesthesia combined with general anesthesia can reduce depression of Tlymphocyte subsets induced by surgical trauma and anesthesia.
6.Significance of β-catenin and Cyclin D1 express in glioma

AIM: To investigate the expression and the significance of β-catenin and Cyclin D1 in gliomas. METHODS: The SABC immunohistochemistry were used to detect the expression of β-catenin and Cyclin D1 in 49 brain gliomas and 5 normal brain tissues. RESULTS: The β-catenin positive ratio in normal brain tissues and glioma samples are 2/5 and 38/49(P<0.01) respectively, and the Cyclin D1 positive ratio in normal brain tissues and gliomas are 1/5 and 42/49(P<0.01); Compared with the normal brain tissue, both the immunohistochemical expression of β-catenin and Cyclin D1 were up-regulated(P<0.05), and the expression of β-catenin and Cyclin D1 between the normal brain tissues and gliomas exhibited significantly difference (P<0.01). CONCLUSION: β-catenin and Cyclin D1 increased in gliomas may be precipitate the tumorigenesis of glioma.
7.Superiority, Confusion and Countermeasures to DCD WeChat Ethical Review

DCD WeChat ethical review had its necessity and superiority but problems and confusion as well. For instance, WeChat review time was urgent and limited;WeChat review discussed insufficiently even hard to car-ry out;the methods of WeChat review were limited;the authenticity of audit opinion was difficult to grasp;the re-view data privacy existed hidden danger;the review quality was difficult to guarantee;WeChat conference content file archive was difficult, and so on. As to this, the author put forward the following countermeasures:giving war-ranty on the ethical review time as far as possible;initiating the committee review by the referee and uploading the review information to WeChat group, then ethical reviewers expressing their opinions in real names to ensure data confidentiality and privacy protection; strengthening the ethics committee education for organ transplantation; im-proving the quality of the review committee members and moral cultivation;archiving the WeChat conference infor-mation.
8.ECM1 Gene Mutations in a Family with Lipoid Proteinosis

Objective To study mutations in the extracellular matrix protein 1 (ECM1) gene in a family with lipoid proteinosis (LP). Methods Bi-directional DNA sequencing was used to detect the ECM1 gene in patients with LP as well as among their parents, siblings and children. Results Sequencing from the affected individuals revealed a new compound heterozygote of missense/nonsense mutations, C220G/R476X, which were not found in the control group. The father was a carrier of the missense mutation C220G and the mother was a carrier of the nonsense mutation R476X, both on one allele. The other siblings, the patients' children and their siblings' children were carriers of either C220G or R476X mutations. Conclusion A new compound heterozygous mutation of ECM1 gene was identified in this LP family.
9.Assessment of fatigue in patients with rheumatoid arthritis: relation with disease activity and healthrelth-related quality of life

Objective To investigate the prevalence of fatigue in rheumatoid arthritis (RA) and its relationship with other clinical and functional parameters used for the evaluation of disease activity and health related quality of life. Methods The fatigue was assessed in 230 patients with RA using visual analogue scale (VAS). The correlation between fatigue and the clinical disease activity including morning stiffness, pain, PGA, physician's global assessment, TJC, TJI, SJC, SJI, DAS28, HAQ and health-related quality of life were assessed. Results The prevalence of fatigue was 85.7% and the fatigue score of 51.7% patients was higher than 50 mm. After controlled for the possible confounding factors such as age, gender and disease duration, it was found that fatigue was highly correlated with pain, disease activity, functional disability, physical health and mental health. Conclusion Fatigue is an important symptom of RA and is correlated with pain, disease activity, functional disability and health-related quality of life.
10.Distribution of six genes of essential hypertension In Yunnan Han healthy population

Objective To explore the distribution of RAS,AGT,ACE,eNOS,ET-2,ANP and NPRC of es-sential hypertension in Yunnan Han healthy population.Methods Gene chip technology was used to detect the pol-ymorphism of AGT M235T (MM, MT,TT), ACE I/D (II, ID, DD ), eNOS Glu298Asp (EE, ED, DD), ET-2 A985G (AA,AG,GG) ,ANP T2238C(TT,TC,CC) and NPRC A-55C(AA,AC,CC) in 97 health subjects.Results The MM,MT and TT genotype frequency of AGT M235T was 0.052,0.381 and 0.567 ,alle frequency of M and T was 0.242 and 0.758 in 97 healthy subjects of Yunnan population;The II, ID and DD frequency of ACE I/D was 0.340, 0.598 and 0.062, alle frequency of I and D was 0.680 and 0.320 in 97 healthy subjects of Yunnan population ;EE, ED and DD frequency of eNOS Glu 298 Asp was 0.845,0.144 and 0.011 ,alle frequency of E and D was 0.918 and 0.082 in 97 healthy subjects of Yannan population;AA,AG,GG frequency of ET-2 A985G was 0.020,0.258 and 0.722, alle frequency of A and G was 0.149 and 0.851 in 97 healthy subjects of Yunnan population;TT and TC fre-quency of ANP T2238C was 0.959 and 0.041 ,and CC was not detected;alle frequency of T and C was 0.979 and 0.021 in 97 healthy subjects of Yunnan population;AC and CC frequency of NPRC A-55C was 0.763 and 0.237, and no AA was detected,alle frequency of A and C was 0.381 and 0.619 in 97 healthy subjects of Yunnan popula-tion.Conclusion The polymorpbism of ACT M235T,ACE I/D,eNOS Glu298Asp,ET-2 A985G,ANP T2238C and NPRC A-55C is locally distributed in Yunnan Han healthy population.
