1.Influence of modified oxygen supply device on oxygen inhalation effect of patients receiving oxygen therapy

Objective To discuss the effect of the modified structure of oxygen equipment on oxygen inhalation effect of patients receiving oxygen therapy.Methods The structure of oxygen equipment was modified.and 80 patients who required oxygen therapy were divided into the modified group and the non-modified group according to odd and even number of hospital admission sequence randomly with 40 patients in each group.The modified group used the modified oxygen equipment,the non-modified group used traditional one.The differences of respiration,heart rate and blood oxygen saturation rate between the two groups before oxygen inhalation,30 minutes and 2 hours after oxygen were compared.Results There was no significant difference in the respiration,heart rate and blood oxygen saturation rate between the two groups.Conclusions The modified oxygen equipment doesn't affect the oxygen therapy effect of patients,besides,it avoids the problems of pollution and the complicated disinfection procedure of the structure of the humidifier bottle,fluid and breathing vessel of the traditional equipment.
2.Detection of Phenotype and Genotype from Vancomycin-resistant Enterococci

OBJECTIVE To explore the infection rate with identifying test of vancomycin-resistant enterococci(VRE) from clinical samples,that is 3.7 percent and to genotype VRE. METHODS vanA,vanB and vanC were detected by PCR in six isolates of VRE which were identified by the broth microdilution susceptibility test and Etest.The one of vanB was further analyzed with DNA sequence and ermB,qacE△1-sul1 gentypes were detected. RESULTS Seventeen isolates of enterococci(MIC≥4 ?g/ml) were obtained of 160 isolates of enterococci which came from Jiangxi Children′s Hospital by microdilution methods.while 6 isolates were gotten by Etest.It demonstrated that susceptibilities of VRE were different in four in vitro susceptibility testing methods.VRE showed resistance to erythromycin(10/17),disinfectant/sulfanilamide(0/17). CONCLUSIONS VRE screening test and the determination of MIC are reliable in finding VRE.VRE genotype is valuable on further research and epidemiological survey of our province.
3.Promotion of rat skin healing by using natural polymer chitosan derivatives.

We in the present study observed the effect of N-fructose modified chitosan quaternary ammonium derivatives on on rat skin wound healing through animal experiments. Forty rats were randomly divided into eight groups (5 in each group). Four groups among the all 8 groups were the experimental groups, while the other 4 groups were the control groups. Next to the skin along the back of the spine, 1.50 cm x 2.00 cm x 0.16 cm full-thickness skin was cut to make an excision wound model for every rat. Those in the experimental groups were treated with the N-fructose-modified chitosan quaternary ammonium derivatives ointment dressing the wound, while those in the control groups with sterile medical vaseline processing. We dressed the wounds twice a day to observe the wound healing of all rats in different groups. We then observed the wound healing and wound pathology after 3, 7, 10, 15 days re spectively in different groups. Results showed significant differences of the time of wound healing, area of wound healing and volume of wound healing between the experimental groups and control groups (P < 0.05). It can be well concluded that N-fructose-modified chitosan quaternary ammonium derivatives does not harm the skin, but could promote skin healing, so that they could be suitable skin repair materials and ideal raw materials for medical dressing.

4.Protective Effects of Folic Acid on Cardiac Development Related Genes GATA-4 and NKx2.5 Expression of Offspring Rats with Maternal Coxsackievirus B3 Infection

Objective To investigate the effects of oral folic acid on cardiac development related gene expression of offspring in an experimental model of coxsackievirus B3(CVB3) infection of pregnant rats. Methods SD female rats were randomized into control group, folic acid group, CVB3 group and CVB3+folic acid group. The female rats were given folic acid by gavage for 2 weeks before pregnancy in folic acid group and CVB3+folic acid group. After conception for 7 days, rat model was established by intraperitoneal injection of CVB3 for 5 days in CVB3 group and CVB3+folic acid group. After nat-ural childbirth neonatal heart was taken and stored in liquid nitrogen. The morphological changes of neonatal rat myocardial tissues were observed by HE staining. The expressions of GATA-4 and NKx2.5 mRNA were detected by RT-PCR and West-ern blot assay. Results There was significant myocardial injury, such as myocardial fiber disarray and myocardial fiber breakage, in neonatal rats in CVB3 group. These damages were improved in CVB3+folic acid group. The expression levels of GATA-4 and NKx2.5 genes in myocardial tissues were significantly lower in CVB3 group than those of control group (P<0.05). The expression levels of GATA-4 and NKx2.5 proteins in myocardial tissues were significantly higher in CVB3+folic acid group than those of CVB3 group (P<0.05). Conclusion CVB3 infection in the early pregnancy inhibited the expres-sion of neonatal rat cardiac development factor. Folic acid supplementation has obvious protective effects on the neonatal rat cardiac development.
5.Protective Effect of Kidney-tonifying Herbal Medicine on the Changes of Female Rat Genital System Induced by Tripterygium wilfordii Hook

Objective To observe the effect of kidney-tonifying herbal medicine (KTHM) on the changes of female rat genital system induced by Tripterygium wilfordii Hook(TWH).Methods Thirty female rats with normal oestrus cycle were randomly divided into 3 groups: control group,TWH group and TWH+KTHM group. The changes of genital system in all rats were examined after 90-day feeding. Results Compared with the TWH group,oestrus cycle was normal, estrogen and progestogen level and the weight of reproductive organs increased, the ovary was big,follicle grew well with more corpus luteum and good blood supplying, endometrium was thick with hyperplastic uterine gland,and vaginal epithelium became thick and cornificated in TWH+KTHM group. Conclusion Kidney-tonifying herbal medicine can antagonize the toxic and side effects of Tripterygium Wilfordii on the genital system of female rat.
6.Advances of human papillomavirus therapeutic vaccine

Human papillomavirus continuous (HPV) infection is an essential factor to induce cervical intraepithelial neoplasia and cervical cancer.Treating HPV infection is considered as a starting point to develop an effective therapeutic vaccine, which is a new strategy for prevention and treatment of cervical cancer.In recent years, development and trials of therapeutic HPV vaccine have made great progress.Selection of vectors, utilization of adjuvants, synthesis of fusion proteins and chimeric proteins have been widely applied to research to enhance vaccine immunogenicity, to increase vaccination safety, to reduce the side effect and so on.The clinical trial results are encouraging: various types of vaccines can induce a specific immune response with good tolerance.However, numerous studies are still required to obtain further success.In addition, HPV exists in various forms, thus it is also the focus of study to expand the range of action and reduce immune escape.

8.Meta-analysis of insertion/deletion genetic variation of ACE gene and onset riskof type 2 diabetic nephropathy in Chinese population

Objective To systematically assess the relation between angiotensin-I converting enzyme(ACE) gene insertion/deletion (I/D) variation and type 2 diabetic nephropathy (T2DN) onset risk among Chinese population.Methods The related literatures were retrieved from the China National Knowledge Infrastructure (CNKI) and Wanfang Data until June 1st,2016.The RevMan 5.0 was used to conduct the statistical analysis.The merge OR value and corresponding 95% confidence interval(95%CI) were used to assess ACE gene I/D polymorphism and T2DN onset risk.Results Totally 29 papers with 4 357 subjects were included according to the inclusion and exclusion standard,including 2 208 cases of DN and 2 149 cases of T2DM without DN.Meta analysis showed that compared with ACE gene I/D polymorphism I allele,D allele could significantly increase the risk of T2DM patients suffering from DN,the OR value and corresponding 95%CI were 1.44(1.25,1.66);the gene analysis showed that ACE gene I/D polymorphism loci were significantly correlated with DN onset risk in the Asian population.The corresponding relative onset risk OR and 95%CI were 1.42(1.15,1.76) and 1.75(1.46,2.10) in the dominant and recessive genetic model.The Begg′s test showed that the included data had no obvious publication bias existence.Conclusion ACE gene I/D polymorphism is closely correlated with the onset risk of T2DN,and D allele might be a risk genetic factor for DN occurrence in the patients with T2DM.
9.The clinical value of contrast-enhanced ultrasound in superficial lymphadenopathy qualitative diagnosis

Objective To evaluate the value of contrast-enhanced ultrasound in superficial lymphadenopathy. Methods Ninty-four superficial enlarged lymph nodes were studied by 2-dimensional, color Doppler ultrasound, and contrast-enhanced ultrasound. Then the contrast-enhanced images were analyzed by Philips Q-LAB software. All the results were compared with pathological diagnosis. Results For the 94 superficial lymph nodes examined,44 were benign,33 were metastases and 17 were lymphomas. The sensitivity, specificity,and accuracy of contrast-enhanced images were 84% ,74% and 790//oo respectively. Contrast-enhanced ultrasound examination showed intense homogeneous enhancement in 39 of 44 benign lymph nodes; high or low homogeneous enhancement in 25 of 33 and 7 of 33 in metastases respectively;intense homogeneous, and scarce enhancement in 6 of 17 and 9 of 17 in lymphomas respectively. Time-intensity curves showed that compared with metastasis lymph nodes and lymphomas, benign lymph nodes had higher peak intensity and larger area under the curve (P<0.01). Conclusions The diagnosis accuracy was significant increased when contrast-enhanced ultrasound was test against conventional ultrasound. The character of contrast agent enhancement and Q-LAB time-intensity curves provide valuable diagnosis information for differential diagnosis of benign,metastasis lymph nodes and lymphomas.
10.Preparation and identification of polyclonal antibody against protein H1b: the variant of major subunit of human ASGPR

AIM: To prepare and identify mouse polyclonal antibody against protein Hlb, which is the variant of major subunit of human ASGPR. METHODS: Hlb specific peptide was synthesized and coupled with keyhole limpet hemocyanin (KLH) for immunization. Then H1b-KLH conjugation was injected into mouse subcutaneously to produce polyclonal antibody. ELISA assay was used to detect the titer of the antibody. Antibody was also identified by Western blot and immunohistochemistry assays. RESULTS: Mouse antibody against Hlb was prepared after injection of H1bKLH conjugation. The titer of H1b antibody was about 1:10~5.Western blot confirmed its high specificity. This antibody could also be used for immunohistochemistry analysis. CONCLUSION: The successful preparation of the polyclonal antibody against protein H1b, which can discriminate the two variants of the major subunit of ASGPR with high specificity, will provide an efficient reagent for further study of the physiologic functions of H1b and its role in the pathogenesis of human disease.
