China Pharmacy 2005;0(16):-
OBJECTIVE:To probe into the safety of influenza A/H1N1 vaccine. METHODS:1 200 medical staff who were vaccinated influenza A/H1N1 vaccine on Oct. 27 th of 2009 were followed up on next day. Those cases of severe hematuria and proteinuria were analyzed and reported. RESULTS:A 32 years old woman developed abdominal pain, lumbago, accompanied by frequent urination, urgency of urination and odynuria in 20 hours after vaccinated influenza A/H1N1 vaccine. Routine urine tests showed dark red turbid urine,RBC(+ + +),proteinuria(+ + +). 20 days later, the index of routine urine test returned to normal. CONCLUSION:Hematuria and proteinuria caused by influenza A/H1N1 vaccine shows rare adverse reaction while great importance should be attached to it.

ObjectiveTo investigate the effects of Tangmaikang on oxidative and carbonyl stress in streptozocin-induced diabetic rat kidneys, and to also explore their reno- protecting mechanisms in diabetic rats.MethodsRats were randomly divided into three sub-groups:normal control (group A),diabetic control (group B) and diabetic with Tangmaikang (group C).Blood glucose, blood lipid,HbAlC,kidney to body weight ratio and urinary protein excretion were measured after 12 weeks.Malonyldialdehyde (MDA)level, superoxide dismutase (SOD) activity, glutathione peroxidase (GSH-Px) activity, total sulfhydryl group and protein carbonyl levels in renal tissue were also determined.ResultsCompared to group A,kidney to body weight ratio, urinary protein excretion,MDA,and protein carbonyl levels in group B were significantly higher, while SOD and GSH-Px activities and total sulfhydryl group level were significantly lower than those in group A.Kidney to body weight ratio, urinary protein excretion,MDA,and protein carbonyl level in group C were significantly lower than those in group B.SOD and GSH-Px activity in group C were higher than those in group B.ConclusionTangmaikang can prevent renal hypertrophy and decrease urinary protein excretion in diabetic rats. The renoprotective effects of this drug may be related to his ability to inhibit oxidative and carbonyl stress.

Objective To study the anti-inflammatory, antipyretic and analgesic effects of Rhizoma Zingiberis alcoholic Extract (RZE) and to explore its in-vitro antibacterial actions. Methods Auricular swelling method in mice, typhoid and paratyphoid vaccines-induced fever in rabbits and bodytwisting test in mice were used respectively to observe the anti-inflammatory, antipyretic and analgesic effects of RZE. Results RZE could inhibit xylene-induced auricular swelling and bodytwisting response reaction induced by acetic acid in mice; RZE also reduce the vaccines-induced fever in rabbits. MIC of RZE for 8 bacterial strains ranged 13.5 mg/mL to 432 mg/mL. Conclusion RZE has a significant anti-inflammatory, antipyretic and analgesic effects and exert in-vitro antibacterial action.

Acute bulbar palsy is considered an extremely rare manifestation of neuromuscular disorder in thyrotoxicosis. It happens abruptly and progresses quickly, sudden onset of dysphagia is common. It will endanger patient's life without proper treatment. The definite treatment of acute bulbar palsy in hyperthyroidism is to restore the euthyroid state. The dysphagia usually resolves within 3 to 4 weeks. In this article a case of thyrotoxicosis with acute bulbar palsy in reported herewith to call attention to the diagnosis and treatment of this disorder.

Objective To investigate the effects of advanced glycation end products (AGEs) on NADPH oxidase expression in rats′ pancreatic cells. Methods Isets of rats were isolated and intervened with AGEs for certain times (2, 12, 24, 48 h). The Nox2 mRNA expression was assayed by reverse transcription polymerase chain reaction (RT-PCR) and Real-time PCR. NADPH oxidase activity was detected by lucigenin-enhanced chemiluminescence. Results Nox2 mRNA level was 1.75, 2.31-fold higher in AGEs intervened group than control at 12 h and 24 h respectively. The NADPH oxidase activity was increased by AGEs stimulation in a time dependent manner. A distinct peak was at 24 hours and then with a decline , which is still higher than that of the control. Conclusion Exposure of rat islet to AGEs induces NADPH oxidase expression and leads to injury of islet.

Objective To observe the role of mulberry flavone on the expression of insulin receptor substrate(IRS)in HepG2 cells with insulin resistance, and to explore the possible molecular mechanism that mulberry flavone improves insulin resistance. Methods HepG2 cells were cultured to establish insulin-resistance cell model in high concentration of insulin, and then incubated with mulberry flavone. Effect of mulberry flavone on the HepG2 cell model of glucose incorporation rate was observed. Western blot was used to observe the variety of IRS protein expression. Results Mulberry flavone increased the glucose incorporation rate of (33. 9 ± 1.0)higher than that of Conclusions Mulberry flavone can improve insulin resistance. Furthermore, IRS protein expression increasing is the possible molecular mechanism that mulberry flavone improves insulin resistance.

Objective To evaluate reverse effect of recombinant human Endostatin on drug-resistance of A549/DDP cells to cisplatin (DDP). Methods Lung adenocarcinoma cell line A549 and its DDP-resistant cell line A549/DDP were treated with DDP and recombinant human Endostatin. Difference in drug resistance was analyzed between different regimens ( DDP, Endostatin and combination) and between different cell lines ( human lung adenocarcinoma A549 and drug resistant A549/DDP), after a 72h-treatment in vitro. Reverse effect of recombinant human Endostatin on drug-resistance of A549/DDP to DDP was tested by MTT assay. Results The observed 50% inhibitory concentration ( IC50 ) was (0.72 ± 0.05 ) ug/ml against A549 and ( 11.54 ± 0.64)against A549/DDP in DDP, and (2.0 ± 0.1 ) μg/ml against A549/DDP in rh-Endostatin- DDP combination respectively, with a reversal fold (RF) of 5.77 and a relative reversal rate of 88. 2%. Conclusion rh-Endostatin may reverse drug-resistance of A549/DDP cells to DDP.

Journal of Chinese Physician 2010;12(2):169-171
Objective To evaluate the clinical value of CHO-hTSHR cells in detecting thyroid stimulating antibody (TSAb). Methods The cAMP production and TSAb activity were measured and cal-culated by stimulating CHO-hTSHR cell line with IgGs of normal control group and Graves" disease ( GD )group. TSAb positive standard was set to more than the mean + 2SD of TSAb activities in control subjects. The positive percentage of TSAb activity in GD group was calculated. Results The cAMP production and TSAb activities of GD group were higher than those of normal group[( 353. 65±126. 34 ) pmol/L vs (237.21±77. 15)pmol/L, ( 149. 08±53. 26)% vs ( 100±32. 52)%, P <0. 05] . The value that higher than 165% was set to be positive for TSAb. The positive percentage of TSAb in GD group was 50% ( 14/28). Conclusion CHO-hTSHR cell line constructed by our group is suitable for detecting TSAb activity in the sera of patients with GD.
