This study was aimed to investigate effect of aconite alkaloids on proliferation and apoptosis of hu-man gastric cancer cell line SGC-7901 . Effects of different concentrations of aconite alkaloids on proliferation of human gastric cancer cell line SGC-7901 were investigated with MTT assay; induced apoptosis and cell cy-cle blocking were detected with flow cytometer ( FCM ) . The results showed that the IC50 of aconite alkaloids on human gastric cancer cell line SGC-7901 was 0 . 2318 ± 0 . 0022 , 0 . 1601 ± 0 . 0227 , 0 . 1031 ± 0 . 0231 mg/ml at 24 h , 48 h and 72 h , respectively , compared to the control group with significant difference ( P <0.01). When the aconite alkaloids concentration was 0.8 mg/ml, it appeared with obvious apoptosis. The apop-tosis rate was ( 59 . 38 ± 5 . 05 )%. The FCM detection showed that compared with control group , the percentage of S-phase cells increased in the treatment group . And a typical sub-diploid peak appeared before G0 / G1 phase . It was concluded that aconite alkaloids can inhibit the proliferation of human gastric cancer cell line SGC-7901 in vitro, induce the apoptosis of cells and make SGC-7901 cells remain in S phase.

According to the super-position principle of the reinforcement of biological activities, a series of novel E-substituted 2, 3-diaryl propenoic acyloxy phosphonate derivatives were designed and synthesized. And the structures of the target compounds were confirmed by IR, 1H NMR, 13C NMR and elemental analysis. Furthermore, the cytotoxicities of all compounds on A-549, SGC-7901 and EC-109 in vitro were evaluated by MTT assay, and some of them showed good antitumor activity. Among the active compounds, especially, the IC50 value of compound 3e was (12.7 ± 1.9) μmol x L(-1) against A-549 cells, similar to cisplatin [IC50 = (8.0 ± 1.5) μmol x L(-1)], compounds 3g and 3k had better inhibition effect on EC-109 cells growth, with the IC50 values of (9.5 ± 1.8) μmol x L(-1) and (11.5 ± 0.9) μmol x L(-1) respectively, and compounds 3i and 3k exhibited good cytotoxic property on A-549, SGC-7901 and EC-109, which were worth further investigation.

Objective To explore the effect of two kinds of combined biliary drainage in the treatment of malignant obstructive jaundice (MOJ) caused by Bismuth Ⅳ Klatskin tumor.Methods Data of 46 MOJ patients caused by Bismuth Ⅳ Klatskin tumor who received combined biliary drainage were retrospectively analyzed.The patients were divided into bilateral percutaneous transhepatic biliary drainage (PTBD) group (n =19) and endoscopic nasobiliary drainage (ENBD) combined with unilateral PTBD group (n=27).Internal and external drainage of the biliary tract were performed.The clinical effect of drainage,survival time and complication rate after drainage were compared between the two groups.Results The technical success rates of drainage in both groups were 100％.There was no statistically significant difference in the effective rate of drainage between the two groups (89.47％ [17/19] vs 92.59％ [25/27];x2=0.02,P=0.831).Compared with ENBD combined with unilateral PTBD group,patients in bilateral PTBD group showed longer survival time ([318.37±167.39] days vs [267.57±145.21] days;t=7.31,P=0.007) and lower complication rate (10.52 ％[2/19] vs 14.81％ [4/27];x2 =1.92,P=0.028) after drainage.Conclusion Combined biliary drainage techniques can effectively alleviate MOJ caused by Bismuth 1Ⅳ Klatskin tumors and improve liver function.Bilateral PTBD is more beneficial to prolong patients;survival time than ENBD combined PTBD.
