1.Application study of optical method in measuring wall thickness of dental impression.

Dental impressions are widely used in the field of oral restoration. The materials are used for making impression in oral cavity. In order to measure the thickness of impression for reference in clinic, the real impressions are taken as the object for studying. Through optical method, charge-coupled device (CCD) is used for collecting the grey image of cutting section of the impressions which are located in the same plane with steel dividing ruler. According to convert relationship between dividing ruler and pixels collecting grey image, the thickness of impression specimens can be obtained. The results show that the optical method used for measuring thickness is feasible to the task and the precision can reach micro dimension. The experiment method and technique can also be provided for measuring thickness of similar tissue engineering materials.

2.Establishment and characterization of a metastatic cell line from spinal cord metastasis induced by injection of Mc3 cells in nude mice

Objective: To establish a metastatic cell line from distant organ metastasis using Mc3 cell line in nude mice. Methods: Tail vein injection of Mc3 cells and cell culture technic were employed to induce metastasis in distant organ . Cell counting and flow cytometry were used to study the cell growth. Karyotype analysis and histopathological observation were used to study the morphological features with light and electron microscopy. Results: Paralized nude mouse was observed in 1 out of 50 experimental nude mice. The cells derived from the spinal cord were cultured and transferred for more than 50 passages. The cells were proved to be of mucoepidermoid carcinoma from human being by the morphology, histopathology and karyotype of the cells. The population doubling time and S-phase cell of the cells were 43 h and 22.7% respectively. The cell line was named Ms. Conclusion: Ms is a metastatic cell line of spinal cord metastasis in nude mouse derived from human mucoepidermoid cacinoma cells.
3.Expression and significance of MMP-3 and TIMP-3 in transitional cell carcinoma of bladder

Objective To study the expressions and significances of matrix metalloproteinases-3 (MMP-3) and tissue-inhibitor of matrix metalloproteinase-3 (TIMP-3) in transitional cell carcinoma of bladder.Methods Immunohistochemistry was used to detect the expressions of MMP-3 and TIMP-3 in 48 cases of transitional cell carcinoma of bladder and 20 normal bladder tissues.Results MMP-3 and TIMP-3 in normal bladder tissues did not express.The more grading and staging,the more MMP-3 expression,but no statistical significance(P >0.05),by contrast,TIMP-3 expression decreased and has statistically significant in the tumor grading (P < 0.05).Conclusions High expression of MMP-3 and low expression of TIMP-3 is related to the malignancy of transitional cell carcinoma of bladder and may play an important role in the development of transitional cell carcinoma of bladder,as an important reference to judge the degree of malignancy of transitional cell carcinoma of bladder and prognostic indicator.
4.Application of dynamic contrast-enhanced perfusion MR imaging in intracranial tumors

Dynamic contrasted-enhanced perfusion MR imaging can provide noninvasive and physical maps of cerebral microcirculation. The tumor vascularities are assessed by calculating cerebral blood volumes of the normal cerebral tissue and tumor tissue from imaging signal intensities. This technique is helpful to better evaluate the histopathological grade of gliomas, determine the biopsy site for tumors. And it is very useful for the preoperative diagnosis of gliomas, solitary metastases, cerebral lymphomas, as well as differentiating tumor recurrence after surgical resection and radiotherapy from radiation necrosis. But there are some limitations in evaluating extraaxial tumors and cerebral tumors with severe breakdown of blood-brain barrier.
5.Establishment of the assessment system for blood transfusion and discussions on the method of clinical transfusion monitor

Objective Discuss the methodology and significance of clinical transfusion assessment,establish an effective evaluation system for blood transfusion,identify rational indexes for the evaluation,and promote quality of clinical transfusion.Methods Development of the clinical blood transfusion assessment regulations,tightened blood transfusion approval system,enhanced medical record check for blood transfusion,better statistics for the data collected,and analysis of data in 2012 to compare changes in blood volume before and after tighter management in place.These efforts aim at improving the assessment system for optimal clinical blood transfusion.Results Compared to Jan.-June in 2012,patients discharged and surgical cases in Dec.of the same year dropped 2.15% and 0.73% respectively.However,the volume of blood transfusion decreased 22.7%,the percentage of blood transfusion for inpatients decreased from 8.78% to 7.17%,and the average use of blood for inpatients decreased from 0.73U to 0.57U.Conclusion Reasonable and scientific assessment for blood transfusion and better clinical blood use management can improve blood transfusion therapeutic efficacy and save blood resources.
6.The effects of mouth rinse containing 1 g/L cetylpyridinium chloride in the treatment of simple gingivitis

7.Diagnosis of one case with 7p15.3p22.1 microdeletion by applying array-based comparative genomic hybridization

Objective To investigate the diagnosis of a case with 7p15.3p22.1 microdeletion by applying array-based comparative genomic hybridization (array-CGH) and to analyze the relationship between the clinical manifestations and 7p15.3p22.1 microdeletion. Method Array-CGH technique was used to detect genomic copy number variations (CNVs) in an infant with normal karyotype after conventional chromosomal karyotyping. Results Array-CGH detected 7p15.3p22.1 deletion (chr7: 6777262-23981753), which was confirmed as pathogenic CNV after comparative analysis with database. Conclusion Array-CGH could serve as a useful complement for G-banding to be used in the clinical cytogenetic diagnosis.
8.The effect of co-inoculating human lymphatic endothelial cells on growth and metastasis of breast cancer cell and of osteosarcoma cell in nude mice

Objective To investigate the effects of lymphangiogenesis on cancer growth and metastasis.Methods Human lymphatic endothelial cells(HLyECs)were isolated and purified from human lymph node by magnetic beads coated with antibody against human VEGFR3.Human breast cancer cell line(MDA-MB-435)or human osteosarcoma cell line(MG-63)was inoculated alone or co-inoculated with HLyECs subcutaneously into nude mice.The weight of tumor and lung surface metastasis were detected;peri-tumor lymphatic vessels were shown by Evans blue,intra-tumor lymphatic vessels were shown by immunohistochemistry for human and mouse PDPN.Conditioned medium of tumor on proliferation of HLyECs was evaluated by MTT assay.Results Compared with inoculating tumor cells alone,the growth and metastasis of MDA-MB-435 were promoted by co-inoculating HLyECs.The peritumoral and intra-tumoral lymphatic vessel density of MDA-MB-435 were increased by co-inoculating HLyECs.Both hPDPN-and mPDPN-positive lymphatic vessels were found.But co-inoculating HLyECs had no effect on lymphatic vessels density of MG-63.Neither intra-nor peri-tumor lymphatic vessels were found.The proliferation of HLyECs was increased by conditioned medium of MDA-MB-435 but not by MG-63.Conclusion Growth and lymphangiogenesis of breast cancer can be enhanced by co-inoculating lymphatic endothelial cells.
9.Study of copy number variations in children with unexplained mental retardation/brain development delay

Objective By using array-based single nucleotide polymorphisms comparative genomic hybridization (SNP-aCGH) to detect and fine mapping copy number variations (CNVs) in children with unexplained mental retardation/brain development delay(MR/BD),then the CNVs were analyzed to determine diagnosis and offer genetic counseling,finally to discuss the application of SNP-aCGH in genetic diagnosis of MR/BD with unknown causes.Methods Ninety-two children with unexplained MR/BD were recruited.SNP-aCGH was used to get CNVs from the whole genome-wide,and the correlation of CNVs and phenotype was analyzed to definit disease genes or pathogenic fragment.Statistics was performed to analyze the common phenotype between positive cases (case with CNVs) and negative cases.Results (1) The CNVs were detected in 10 cases with a detection rate of 10.86%,from which 8 cases showed subtelomeric aberration,5 cases without subtelomeric aberration,and the rate was 8.70%,5.40%,respectively.The CNVs related to MR/BD involved 10 different subtelomeric regions (9p,21q,3p,2p,15q,4p,12p,22q,16p,17p),and 7 different regions without subtelomeric (1p,4q,2p,14q,15q,12q,22q).The deletions involved 11 zones (size:1.05-8.80 Mb),and duplications referred to 8 zones (size:1.33-31.25 Mb).(2) One case was diagnosed as 9p duplication syndrome,for candidate genes:DOCK8,VLDLR.A case was detected with a gene fracture (CRBN).One case was diagnosed as Coffin-Sirrs syndrome combined with a deletion of 15q26.3-qter,for candidate genes:SOX11 and LINS1,respectively.One case referred to 12p13.3 deletion syndrome,for candidate genes:ELKS,ERC1.One case referred to 22q13.2-qter deletion,for candidate genes:SHANK3.Two cases were diagnosed as ATR-16 syndrome with 17p13.3 deletion syndrome,their candidate genes:HBA1,HBA2,SOX8 for the former,YWHAE,LIS1 for the latter.(3)There were statistically significant differences in comparison of positive cases to the negative ones for growth delay,internal organs deformity,low birth weight infant(LBW) and premature infant (all P <0.05).Conclusions (1) Besides MR/BD in different degrees in all the positive cases,they also showed growth delay,a portion of them with internal organs deformity,low birth weight infant and premature infant.(2) Subtelomeric aberrations are related to MR/BD,while the submicroscopic rearrangement in regions without subtelomeric is suspiciously pathogenic,and need to be further studied.(3) SNP-aCGH can fine mapping the region of CNVs by high resolution from the whole genome-wide,which does contribute to limit the zones for finding pathogenic region and candidate genes,as well as to offer a technology platform for investigating about the correlation of phenotype and genes or CNVs.
10.The analysis of sacrococcygeal microcirculation in complete spinal cord injury patients

Objective To assess the sacrococcygeal microcirculation and the change trend of complete spinal cord injury patients,and in order to provide evidence for turn-over time of spinal cord injury patients.Methods The spinal cord injury patients that met the inclusion criteria were set as the experimental group,patients with orthopedic trauma were named as the similar control group.And certain healthy volunteers were named as the healthy control group.The sacrococcygeal microcirculation of supine position for 1h,and lateral position for 0.5 h was monitored using Doppler blood flow monitor system,then take load period 0min、15m in 、30min、45min、60min,recovery period 0min、15min、30min as research points,the blood perfusion value and the change trend were monitored and analyzed.Results The sacrococcygeal microcirculation in SCI patients were significantly less than two control groups after 30min in supine position,and the blood flow increased at first and then decreased within one hour.While the healthy subjects increased gradually within one hour.Conclusions The time of position change for SCI patients should be reassessed,the interval should be controlled in one hour.
