1.Advancement of hybrid procedures in congenital heart disease

Hybrid procedure is an method that combines surgery and intervenfional catheterization. As collaboration between surgeon and interventionalist has improved, the procedures for complex congenital heart disease is progressing and developing remarkably. Combining echocardiography monitoring in hybrid surgical procedure of complex congenital heart disease not only reduces side effect of extracorporeal circulation and operation time, but also retrieves blind spot of interventional catheterization for infant and low weight baby. Currently it is applied in the field of atrial septal defect, muscular ventricular septal defect, pulmonary stenosis and post-operation residual shunts or stenosis.
2.Serum levels of uric acid in the population of health check-up

Total 11 008 subjects underwent routine health check-up at Tianjin General Hospital in 2008. The results showed that the rate of hyperuricemia was 11.56% ( 1272/11 008 ) in this population, 17. 17% ( 1216/7082 ) for male, and 1.43% (56/3926) for female (P <0. 05). The serum uric acid levels were positively correlates with body mass index ( r = 0. 346), waist circumference ( r = 0. 449), systolic blood pressure ( r = 0. 193 ), diastolic blood pressure ( r = 0. 240), total cholesterol levels ( r = 0. 240) and triglyceride ester levels ( r = 0. 284 ). The factors including overweight, hypertension, hyperlipidemia and hyperglycosemia were clustered in subjects with hyperuricemia.
3.Effect of WWOX gene on the attachment and adhesion of ovarian cancer ceils

Objective To explore the relationship between WWOX gene and attachment and adhesion of ovarian cancer. Methods The expression of WWOX mRNA was detected by RT-PCR, the expression of the WWOX protein was evaluated by western blot in WWOX-transfected PEO1 cells (H6, H7, H8 cell) and vector-transfected control cells (vec-1, vec-2 cell). Attachment assay was used to assess the adhesion of the tranafection in PEOI cells via culturing the cells on the pre-coated fibronectin wells. RNA interference (RNAi) was used to knockdown the endogenous expression of WWOX in the A2780 ovarian cancer cell line by liposome. Attachment assay was detected the adhesion to fibronectin after gene silencing. Restdts RT-PCR showed that expression of mRNA WWOX in exon9 was in all transfection cells (H6, H7, H8, vec-1, vec-2 cell ). Western blot showed that expression of WWOX protein was in the WWOX-transfected cells (H6, H7, H8 cell ), but not in the vector-transfected cells (vec-1, vec-2 cell ). Attachment assay showed that H6, H7, H8 cell (0.098±0.003, 0.091±0.004, 0.099±0.003) adhered more slowly to fibronectin than vec-1, vec-2 cell (0.185±0.003, 0.175±0.006) and non-transfected PEO1 cell (0.211±0.007), and demonstrated significantly reduced adhesion after 2 hours (P < 0.01). A2780 adhesive cells that WWOX gene be knockdown was 0.059±0.005, adhered more significantly rapid than those untreated cells that was 0.029±0.003 after treated 30 minutes (P < 0.05 ). Conclusions WWOX gene can suppress adhesion to fibronectin in ovarian cancer cells. This suggests an important role for loss of WWOX gene in promoting attachment and adhesion of ovarian cancer cells on loco-regional peritoneum, and further resulting in enhancing loco-regional peritoneal tumor invasiveness and spread.
4.Search for proofs of TCM literature review for breast cancer in ancient times

Breast cancer belongs to high morbidity malignant tumor for women. Some known aspects to breast cancer were etiological factor, pathogenesis and prescription based upon literature review of Traditional Chinese Medicine (TCM) in ancient times, and the valuable references to modern clinical medicine (CM) was offered. Some animal studies and modern clinical trials indicated that curative effect of integration of Chinese and Western Medicine in experimental group was better than that of Western Medicine treatment alone in control group. It aims to offer open-minded idea under syndrome differentiation for professional staff in clinic by arranging, inducing-and summarizing related records on breast cancer.
5.Estradiol activates MAPK signaling pathway by estrogen induced VEGF and bFGF in endometrial cancer cells

Objective To explore the effects of mitogen-activated protein kinase (MAPK) pathway by estradiol induced vascular endothelial growth factor (VEGF) and basic fibroblast growth factor (bFGF) in endometrial cancer Ishikawa cells.Methods The experiments were divided into 4 groups:E2 group (Ishikawa cells treated with 1 p mol/L estradiol for 30 minutes); inhibitor group:including Ishikawa cells treated with 10 μmol/L Bibf1 120 (Bibf1 120 group),or treated with 2.5 μmol/L Ponatinib (Ponatinib group),or treated with 10 p mol/L U0126 (U0126 group) for 60 minutes; inhibitor + E2 group:including Ishikawa cells treated with 10 μmol/L Bibf1120 (Bibf1120 + E2 group),or treated with 2.5 μmol/L Ponatinib (Ponatinib + E2 group),or treated with 10 μmol/L U0126 (U0126+ E2 group) for 60 minutes following incubation with 1 μmol/L estradiol for 30 minutes; control group:only adding the culture medium without serum DMEM.(1) Western blot analysis was used to detect phosphorylation extracellular signal-regulated kinase 1/2(p-ERK 1/2) protein expression with stimulation in different concentrations of estradiol (0.01,0.1,1,10,100 μmol/L).(2) Quantitative fluorescent reverse transcription (qRT)-PCR and western blot analysis was used to test the level of mRNA and protein of VEGF,bFGF,MAPK kinase 1/2 (MEK1/2),extracellular signal-regulated kinase 1/2 (ERK1/2),p-ERK1/2 and phosphorylation MEK1/2 (p-MEK1/2).Flow cytometry were used to examine the cell cycle,and transwell chamber assay were used to detect the cell migration in different groups.Results The expression of the p-ERK1/2 protein at 0.01,0.1,1,10,100 μ mol/L were 0.16±0.03,0.10±0.03,0.41 ±0.04,0.19±0.03,0.19±0.03,there were significantly higher than that in control group(0.05±0.00,P<0.05),and which was more obvious at the concentration of 1 μmol/L estradiol.The expression level of VEGF,bFGF mRNA and protein in E2 group were higher than those in the control group (P<O.05).VEGF mRNA and protein in Bibf1120+E2 group were higher than those in E2 group.The expression of MEK1/2,ERK1/2 mRNA protein in E2 group were higher than those in control group (P<0.05).The expression of MEK1/2,ERK1/2 mRNA or p-MEK1/2,p-ERK1/2 protein in Bibf1120 + E2 group,Ponatinib+E2 group or U0126+E2 group were lower than those in E2 group(all P<0.05).Percentage of G1 phase [(53.6±3.2)%] and S phase[(29.2±4.2)%] in E2 group was significantly different with those in control group respectively(P<0.05).Percentage of G1 phase[(66.8±2.6)%,(63.1±2.6)% and (63.3±0.4)%] and S phase [(25.4±1.9)%,(25.0±3.8)% and(23.8±0.5)%] in U0126+E2 group,Bibf1120+E2 group or Ponatinib +E2 group was also significantly different with those in control group (all P<0.05); percentage of G1 phase and S phase in U0126+E2 group was significant difference with those in Bibf1120+E2 group or ponatinib+E2 group (P<0.05).The number of cell colony in E2 group (110± 17) was more than those in control group (65±8) ;the number of cell colony in U0126+E2 group(28±4),Bibf1120+E2 group(38±5) or Ponatinib+E2group(42±6) were significant different with those in E2 group (P<0.05),the number of cell colony in U0126+E2 group was significant difference with those in Bibf1 120+E2 group or Ponatinib+E2 group (all P<0.05).The results shown that the abilities of proliferation and cell migration were significantly increased in cells after estradiol stimulation.Conclusion Estradiol inducing the production of VEGF and bFGF could activate MAPK pathway through ER-independent manner,further promote development.
6.Application of multimodal imaging in stem cell research

In recent years,stem cell research has been developing quickly in biological science.As the key of regenerative medicine,stem cell therapy becomes another innovative treatment following drug therapy and surgery.In vivo stem cell tracking,including optical imaging,radionuclide imaging and MRI,can trace the viability,distribution and function of engrafted cells.Multimodal imaging integrates two or more types of imaging techniques to obtain the combined advantages of each technology,and therefore is able to accurately and effectively trace stem cell in vivo,hopefully promoting its clinical transformation.This paper reviews the application of multimodal imaging in stem cell research.
7.Screening of risk factors for type 2 diabetes in general population

Total 10 450 people received health check-up in our hospital from March 2009 to December 2010.The subjects were classified as high-risk for type 2 diabetes mellitus ( DM ) group ( n =9016) and non-high-risk group (n =1434).The rate of new DM was 3.36% (303/9016) and the rate of impaired glucose regulation (IGR) was 12.38% (1117/9016) in high-risk group; while in non-high-risk group there was no new DM cases,and IGR rate was 2.58% (37/1434).The detection rate of IGR and DM significantly increased with the increasing rate of risk factor and numbers (P < 0.01 ).Logistic regression analysis revealed that the abnormal glucose metabolism was associated with age,male,positive family history,heart rate,hypertension,abnormal lipid metabolism and hyperuricacidemia.The results suggest that subjects with multiple risk factors should be the key population for prevention and screening of type 2 diabetes.
8.Blood uric acid levels of routine health check-up receivers in Tianjin City

Objective To investigate blood uric acid levels in health check-up receivers in Tianjin City. Methods Blood uric acid levels in 28 661 health check-up participants were tested and the incidence of hyperuricaemia was analyzed. Blood pressure, body mass index, blood sugar, cholesterol and triglyceride were measured and compared. Results The average level of blood uric acid was (349±73 ) μ mol/L in male,and (256±59) μmol/L in female. The peak value in male was found in the 30 to 39 years old group,while in female the age was increased to 70 years old. The incidence of hyperuricaemia was higher in male ( 15. 2% vs 6. 3% ,P<0.01). The level of blood sugar,triglycerides and total cholesterol was higher in the hyperuricaemia group. Also in the hyperuricaemia group, the incidence of overweight/obesity,hyperglycaemia, hyperlipidemia and hypertension was higher. Conclusions The incidence of hyperuricaemia in routine health check-up individuals in Tianjin City could be 11.3%. Blood uric acid may be significantly associated with metabolism.
9.Prevalence and risk factors of impaired glucose regulation in health check-up populations

Objective To investigate the prevalence and risk factors of impaired glucose regulation (IGF) in health check-up populations in Tianjin.Methods A total of10126 individuals were assigned to group according the1999 World Health Organization (WHO) criteria for diabetes mellitus ( DM ).Clinical characteristics and the prevalence of DM were compared between the groups. Student's t test,Chi-square test and logistic regression were used for data analysis.Results The incidence of IGR was much higher in male than in female (x3 =136.2,P <0.05).Abnormal glucose metabolism showed an increasing trend with age ( P<0.05 ).In logistic regression analysis,age,male,family history of DM,overweight/obesity,hypertension,hypertriglyceridemia and hyperuricemia were risk factors of IGR.Conclusion Age,male,overweight/obesity,family history of DM,hypertension,hypertriglyceridemia and hyperuricemia could be risk factors of IGR.Early screening and intervention may help to prevent the development of DM.
10.Synthesis and biocompatibility of ethynylated open ring derivatives of polyasparamide

Objective To synthesize a new ethynylated open ring derivatives of polyasparamide as functional drug carrier.Methods L-phenylalanine methyl ester hydrochloride was prepared using L-phenylalanine and then was used for ring opening reaction of polysuccinimide.To synthesize the target product of PSI-Phe-OMe-PA, the obtained polyasparamide-g-phenylalanine derivatives ( PSI-Phe-OMe) was further ring opened by propargylamine.The structure of PSI-Phe-OMe-PA was confirmed by 1 H NMR.The biocompatibility of PSI-Phe-OMe-PA was evaluated by MTT method, inverted microscope observation and cell cycles analysis ( propidium iodide staining ) .Results The ring-opening rate of polyasparamide by L-phenylalanine methyl ester and propargylamine was 40%and 100%, respectively.All results of biocompatibility studies indicated that PSI-Phe-OMe-PA may be a good candidate for functional drug carrier.Conclusion Based on the ring-opening capability of amino-group and the specificity of click reaction, L-phenylalanine methyl ester hydrochloride and propargylamine were used successively to react with polyasparamide.PSI-Phe-OMe-PA is a biocompatible functional drug carrier.
