1.THE EXPENMENTAL STUDY OF THE EFFECT OF DEXAMETHASON ON EXPERIMENTAL AUTOIMMUNE SENSORINEURAL NEARING LOSS

In this experiment, Animal model of guinea pig's experimental Autoimmune sensorineural hearing Loss was established by injecting crud inner ear antigen (CIEAg), Dexamethason was used to tret the deaf guinea pigs for one week, the ABRT(Acoustic Brain—stem Reaction Threshold) of guinea pigs was measured and the morphological change of the guinea pigs inner ear, tissue was observd. We found that the deaf guinea pig's ABRT rose distinctly to 60. 8?5.31 dBspl and inner ear hair cell showed severe damage. The number of the hair cell of the third cochlear turn lost 111?55.8. After one week treatment with Dexamethason,the treated group's ABRT reduced to 48. 8?6. 69dBspl, the number of the outer—hair—cell of the third cochlear turn is only 57. 254?18. The results indicate that Dexamethon has effect on the AISNHL,it can clearly lighten the damage of inner ear tissue and significantly improve the ABRT. The mechanism still remains unknown.
2.Advancement in the tracing technique of seed cells for tissue engineering

The development of tissue engineering has provided new methods for organ replacement and disease treatment.The research of seed cells includes cell selection,culture,transplantation,tracing and so on.The tracing technique of seed cells involves molecular biology,molecular immunology,molecular imaging and many other subjects,making it difficult to select a label for seed cells.This review summarizes the recent research progress in the tracing technique of seed cells.
3.Effect of insulin detemir combined with insulin aspart on islet cell functionin in type 2 diabetes

Objective To explore modification of islet cell function in type 2 diabetes by the treatment of insulin detemir and insulin aspart.Methods A total of 68 patients with diagnosed type 2 diabetes were randomized into two grouops,the control group (insulin glargine combined with insulin aspart)and observation group (insulin detemir combined with insulin aspart),each group had 34 cases.Before and after treatment,fasting insulin and 2 h postprandial insulin,glycated hemoglobin (HbA1c),C peptide and fructosamine (FMN)levels were compared in two groups,and the treatment safety was observed.Results After treatment,fasting insulin and C -peptide levels in the two groups were significantly increased than before treatment(P <0.05 ).In observation group,2 h postprandial insulin level (29.97 ±9.18)mIU /L was significantly higher than before treatment (20.79 ±7.49)mIU /L(t =4.52,P =0.00), which was better than the control group (24.67 ±7.69)mIU /L,the difference was significant (t =2.58,P =0.01). After treatment,postprandial 2h C peptide levels in the two groups had no significant differences compared with before treatment (P >0.05).In the observation group,except for 2h postprandial insulin levels,other indicators had no significant differences compared with the control group (P >0.05 ).After treatment,FMN and HbA1c in the two groups decreased significantly compared with before treatment(P <0.05 ),but there was no significant difference between the two groups (P >0.05).During treatment,both two groups observed hypoglycemia,but there were no severe adverse reactions.Conclusion The clinical application of insulin detemir combined with insulin aspart in the treatment of type 2 diabetes can significantly improve the insulin -producing cells and accelerate functional recovery, and the clinical effect is better than the united aspart insulin glargine,with no significant adverse reactions,it can be recommended as a clinical type 2 diabetes treatment drug of choice.

5.Comparison of the effects of autotransfusion and homologous transfusion on milieu interieur and secretion of erythropoietin

Objective To compare the effects of autotransfusion and homologous transfusion on milieu interieurand secretion of erythropoietin (EPO) .Methods Thirty patients aged 8-21 yr undergoing surgical correction of kyphoscoliosis were randomly divided into two groups of fifteen: group A and group B. In group A patients received homologous transfusion during operation, while in group B shed blood was collected from operative field and after filtration RBCs were separated by a differential centrifugation and washed in normal saline and reinfused. Anesthesia was induced with fentanyl 2 ?g? kg-1 , propofol 1 mg ? kg-1 and vecuronium 0.1 mg ? kg-1 and maintained with inhalation of isofiurane and 50% N2O in oxygen and intermittent iv boluses of vecuronium and fentanyl. Blood routine, blood gases, electrolytes, plasma osmotic pressure and activated clotting time (ACT) were measured before operation, 2 h after skin incision and at the end of surgery. Blood EPO concentration was measured before operation, at the end of surgery and 24 h after operation. Results MAP and HCO3- were significantly lower during operation in group A than those in group B. Platelet count was significantly lower at the end of operation in group B than in group A but still within normal range. The EPO level increased at the end of operation and returned to preoperative level 24 h after operation in both groups, and there was no significant difference in EPO levels between the two groups. Conclusion Autotransfusion of the washes RBCs is helpful in maintaining the patients milieu interieur. There is no difference in EPO secretion and erythropoiesis between autotransfusion and homologous transfusion.
6.THE SCAVENGING ROLE OF OXYGEN FREE RADICALS BY SALVIA MILTIORRHIZA

Using Xanthine-Xanthine Oxidase system to generate superoxide ani-on, using H2O2 -Fe2 + system to generate hydroxyl radical, the role of scavenging oxygen free radical by Miltiorrhiza was studied by electron paramagnetic resonance and spin trapping. The result suggested that the scavenging rate of hydroxyl radical by Salvia Miltiorrhiza injection is 65%, and that of superoxide anion is 100%. This result suggests that scavenging cytotoxic oxygen free radicals may be one of important pharmacological mechanism of Salvia Miltiorrhiza.
7.Blood purification therapy of gestational hyperlipidemic pancreatitis:A case report

8.Construction of expression vector for NT4-ADNF-9 fusion gene

Objective To construct the prokaryotic expression vector bearing fusion gene NT4-ADNF-9 and lay foundation for further study on genetic therapy of neuraseusory deafness. Methods By means of asymmetrical prince/ template, double stranded eDNA of activity dependent neurotrophic factor-9 (ADNF-9) was obtained, which included restriction enzymes sites on the two extremities. ADNF-9 eDNA was ligated to the signal and leader peptides of nenrotrophin 4 (NT4), and the fusion gene was named NT4-ADNF-9. Then it was suheluned into prokaryotic expression vector pBV220, and called pBV220/ NT4-ADNF-9. Results Evidences of DNA sequence analysis and restrtction enzymes digestion showed that we recombined ADNF-9 eDNA to the 3'terminal of the signal and leader peptides of NT4, and the fusion gene was subcluned into pBV220 successfully. Bioactivity of the products was proved that it could support the cell survival and neurite growth in the primary cultures of dorsal root ganglia (DRG) of embryonic day-8 cbicken neurons as compared to the control. Conclusion Prokaryotic expression vector pBV220/NT4-ADNF-9 can be constructed successfully and the bioactivtty is satisfactory.
9.Chitosan-coated ophthalmic submicro emulsion for pilocarpine nitrate.

The study is to design chitosan-coated pilocarpine nitrate submicro emulsion (CS-PN/SE) for the development of a novel mucoadhesive submicro emulsion, aiming to prolong the precorneal retention time and improve the ocular absorption. CS-PN/SE was fabricated in two steps: firstly, pilocarpine nitrate submicro emulsion (PN/SE) was prepared by high-speed shear with medium chain triglycerides (MCT) as oil phase and Tween 80 as the main emulsifier, and then incubated with chitosan (CS) acetic solution. The preparation process was optimized by central composite design-response surface methodology. Besides the particle size, zeta potential, entrapment efficiency and micromorphology were investigated, CS-PN/SE's precorneal residence properties and miotic effect were especially studied using New Zealand rabbits as the animal model. When CS-PN/SE was administered topically to rabbit eyes, the ocular clearance and the mean resident time (MRT) of pilocarpine nitrate were found to be dramatically improved (P < 0.05) compared with PN/SE and pilocarpine nitrate solution (PNs), since the K(CS-PN/SE) was declined to 0.006 4 +/- 0.000 3 min(-1) while MRT was prolonged up to 155.4 min. Pharmacodynamics results showed that the maximum miosis of CS-PN/SE was as high as 46.3%, while the miotic response lasted 480 min which is 255 min and 105 min longer than that of PNs and PN/SE, respectively. A larger area under the miotic percentage vs time curve (AUC) of CS-PN/SE was exhibited which is 1.6 folds and 1.2 folds as much as that of PNs and PN/SE, respectively (P < 0.05). Therefore, CS-PN/SE could enhance the duration of action and ocular bioavailability by improving the precorneal residence and ocular absorption significantly.
10.Determination of bisphenol A in water by truncated aptamer-fluorescence method

Objective:
To establish a fluorescence method based on turncated aptamer for the determination of bisphenol A in water.
Methods:
The bisphenol A truncated aptamer containing 38 bases was selected as a recognition module, and was modified with the fluorophore 6-FAM at the 5'end. The 3'end of the complementary sequence cDNA was modified with the quencher DABCYL. The standard solutions of bisphenol A and interfering compounds were configured. The detection system was established after optimizing the number of bases in cDNA, the concentration ratio of truncated aptamer to cDNA, the incubation temperature and time, and the pH of the buffer. The specificity and recovery experiments were carried out.
Results:
When the complementary sequence cDNA included 9 bases, the concentration ratio of the truncated aptamer to cDNA was 1:1.5, the pH value of the buffer solution was 7.5, the cDNA was incubated at 55 ℃ for 60 minutes, in the concentration range of 10-75 pmol/L, the linear regression equation was y=2 230.7x+110 825, the correlation coefficient was 0.926. The limits of detection was 3.3 pmol/L. The difference values of fluorescence intensity between tetrabromobisphenol A, estradiol, estriol, bisphenol S and bisphenol A were obviously different, so there was no significant interference to the test result. The recovery rates were 97.8%, 98.8% and 102.3% with the spiked concentrations of 20.0, 40.0 and 60.0 pmol/L. The relative standard deviations were 4.4%, 2.1% and 2.6% (n=5), respectively.
Conclusion
The fluorescence method based on turncated aptamer has the advantages of easy operation, high sensitivity and specificity, which can be used for the determination of bisphenol A in water.
