ObjectiveTo investigate the expression and effects of nitric oxide synthase in the Abeta transgenic drosophila.MethodsmRNA was obtained from 20 Abeta transgenic drosophila and 20 wild type drosophila,and cDNA was synthesized by reverse transcription.The expression of nitric oxide synthase mRNA was detected by RT-PCR.ResultsThe expression of nitric oxide synthase mRNA (0.020 ± 0.006)in the Abeta transgenic drosophila was significantly lower than that in the wild type drosophila ( 1.000 ±0.149),there had significant difference between Abeta transgenic drosophila and wild type drosophila ( P ＜ 0.05)..ConclusionsNitric oxide generated by nitric oxide synthase was an important messenger molecule,and it was closely related to Alzheimer's disease.

Objective To study the regulation of serum FSH and E2 in the treatment of menopausal syndrome with 17 beta estradiol combined with DRSP. Methods A total of 90 female patients with menopausal syndrome treated in our hospital from November 2013 to November 2016 were selected. The patients were randomly divided into three groups: 17β group (30 patients, 17β-estradiol alone), DRSP group (30 patients treated with DRSP alone) and 30 patients (17β-estradiol combined with DRSP). Kupperman's menopausal syndrome was used to evaluate the physical symptoms of the patients and to compare the patients after the treatment. The patients' treatment efficiency was observed. The items examined before treatment included liver and kidney function, hematuria routine , Fasting blood glucose, FSH, E2, LDL, blood TC, electrocardiogram, intracavitary Doppler ultrasound, breast ultrasound, HDL, Kupperman evaluation table in accordance with the evaluation before the end of each treatment measure again, And performs the above-mentioned inspection. Results In the combined group, there was no significant difference in the scores of other symptoms except the differences between the ants on the skin and the treatment before treatment. There was significant difference between the combined group and the other groups (P<0.05). The difference of TC, E2 and FSH in the combined group before and after treatment (P<0.05). There was no difference in FSH between the two groups before and after treatment. There was no difference between the two groups before and after treatment. Conclusion 17β-estradiol combined with DRSP in the treatment of menopausal syndrome patients achieved good clinical effect, the serum levels of FSH and E2 in the body is significant.

Objective To explore the evaluation value of modified USA National Institutes of Health Stroke Scale (mNIHSS) score combined with bedside transcranial Doppler ultrasonography (TCD) on the early prognosis of ischemic stroke (AIS).Methods One hundred and thirty-six anterior circulation AIS patients were collected,mNIHSS score was evaluated on admission,and emergency bedside TCD examination was performed to evaluate vascular anomalies.After active treatment after discharge,the modified Rankin scale (mRS) score was recorded to evaluate 90 d prognosis.The value of mNIHSS score combined with TCD examination analysis on the prognosis was analyzed.Results If the mNIHSS score ＞ 8 scores was judged to be poor prognosis,the sensitivity was 0.684,the specificity was 0.806,the positive predictive value was 0.578,the negative predictive value was 0.868.If related macro vascular abnormalities in bedside TCD examination were evaluated for clinical prognosis,the sensitivity was 0.736,the specificity was 0.643,the positive predictive value was 0.444,the negative predictive value was 0.863.If both,the specificity was 0.918,the positive predictive value was 0.714,the positive likelihood ratio was 6.41 ;and the two were not available,the negative predictive value was 0.928,negative likelihood ratio was 0.20.Conclusion mNIHSS score combined with TCD examination in evaluating the prognosis of AIS patients can improve the positive or negative predictive value.

Objective To investigate the effects of exercise on the expression of synapotophysin around hemotomas after intracerebral hemorrhage (ICH). Methods Ninety-five male Sprague-Dawley rats weighing 270 to 300 g were divided into 3 groups: a trial group (ICH-induction and exercise, n = 20), a control group ( ICH-in-duction only, n =20) and a sham-operation group (no ICH and no exercise, n = 20). The rats' brains were re-moved at 7, 14, 21, and 28 days after the operation. The other 35 rats were divided into 7 groups (6 h, 12 h, 24 h, 48 h, 72 h after ICH, no ICH and normal ). The activation of synapotophysin was measured by immunohis-tochemical techniques. The rats in the trial group begin cage-running exercise at 72 h after the operation. The oth-ers lived in standard cages. Results Synapotophysin-positive cells were seen in the tissues around the hematomas and in the cortex. There was no synapotophysin expression in the centres of the hematomas. The number of synapo-tophysin-positive cells in ICH group was significantly less than in the sham-operation and normal groups from 6 h to 24 h after the operation. There was then a gradual increase again from 48 h onwards until the 28th day after the op-eration. The trial group had the largest increases in expression. There was a significant difference compared with the control group. Compared with the sham operated group, the trial and control groups showed very significant differences. Conclusion The results suggest that synapotophysin is involved in neuron plasticity after ICH, but exercise training (cage-running) can accelerate the expression of synapotophysin, thereby improving functional re-covery.

Objective The aim of this study is production of organ specific animal model for studying reproductive toxicity in mice.Methods F1 generation was gotten by mating the Ddx4 -cre transgenic male mice with the Rosa26mT/mG transgenic female mice.F1 offspring and its parents phenotype was screened by molecular biological, histopathological and in vivo imaging technology.Results At molecular level, specific DNA fragment was only found in testis of F1 offspring; At the organ level, the expression of green fluorescent protein could only be observed in testis of F1 offspring; Testicular frozen sections and sperm fluorescence observation showed that green fluorescent protein were mainly expressed in the germ cell lineage such as secondary spermatocyte and spermatocyte and spermatozoon.Conclusions The production of the mice with specific germ cell expressed green fluorescent protein by Cre/loxP recombination system were built successfully.

AIM: To investigate the effect of digoxin on apoptosis and growth in human gastric carcinoma SGC7901 cells and its possible mechanism.METHODS: SGC7901 cells were incubated in the medium containing digoxin at different concentrations for 24 h.CCK-8 assay was employed to detect the anti-tumor effect of digoxin on SGC7901 cells, and IC50 value of digoxin was calculated.Flow cytometry was used to determine the apoptosis and the cell cycle.Western blot was used to analyze the protein levels of c-Src, p-c-Src(Tyr416), Akt, p-Akt(Ser473), ERK1/2 and p-ERK1/2 (Tyr204).The mRNA expression of c-Src was by RT-PCR.RESULTS: As the concentration of digoxin increased, the cell viability was reduced gradually starting at 50 nmol/L digoxin treatment (P<0.05).The cell viability was reduced to the lowest extent by exposure to 500 nmol/L digoxin (P<0.05).The IC50 value of digoxin was 191.45 nmol/L for 24 h.After treatment with 200 nmol/L digoxin for 24 h, the apoptotic rate and the proportion of G0/G1 phase were significantly increased (P<0.05), the phosphorylated levels of c-Src, ERK1/2 and Akt declined (P<0.05), the mRNA and protein levels of c-Src were down-regulated (P<0.05) and the ability of migration was weaker (P<0.05) than that in control group.CONCLUSION: Digoxin may suppress the growth and induce the apoptosis of human gastric carcinoma SGC7901 cells by inhibiting the expression of c-Src gene and down-regulating the phosphorylated levels of ERK1/2 and Akt.
