1.Expression and clinical significance of PKD1 in human endometrial carcinoma

Purpose To investigate the expression of protein kinase D1 (PKD1) in endometrial carcinoma and normal endometrium,and to investigate its relationship with the clinicopathological features of endometrial carcinoma.Methods Immunohistochemical SP and qRT-PCR was used to detect the mRNA and protein expression of PKD1 in 92 cases of endometrial cancer and 48 cases of normal endometrium,and to analyze the relationship of expression of PKD1 with the tumor differentiation and clinical stage of endometrial carcinoma tissue.Western blot method was used to detect the expression level of PKD1 protein in normal endometrial cell line and endometrial carcinorna cell lines with different degree of differentiation.Results mRNA and protein expression of PKD1 in endometrial carcinoma tissues was significantly higher than those in normal endometrial tissues (P < 0.01),which showing a correlation to the degree of tissue differentiation and clinical pathologic staging.While,the expression level of PKD1 protein in endometrial cancer cell lines was also much higher than that in normal endometrial cells (P < 0.01),and the lower differentiation,the higher level of PKD1 protein expression.Conclusion PKD1 is highly expressed in endometrial cancer patients.The level of PKD1 expression may be an important reference for predicting the malignant degree of endometrial cancer.
2.Short effect and adverse reaction of dog days plaster for allergic rhinitis.

OBJECTIVETo evaluate the short effect and safety of the acupoint application of dog days for allergic rhinitis.
METHODSTwo hundred and forty-nine patients were randomly divided into an application group (166 cases) and a placebo group (83 cases). On the first day of the first dog days, the first day of the second of the three ten-day periods of the hot season and the first day of the last of the three ten-day periods of the hot season, acupoint application was adopted mainly at Dazhui (GV 14), Feishu (BL 13), Pishu (BL 20) and Shenshu (BL 23) in the application group. The plaster was made of Chinese herbs, including Baijiezi (semen brassicae), Xixin (asarum), Yanhusuo (corydalis tuber) and Gansui (euphorbia kansui). Buckwheat plaster without medical ingredient was used in the placebo group. The application was pasted for 2-6 h every time. On the first three days of all the three periods of dog days, treatment was used continuously. The whole period of dog days was made into one course. The changes of symptom and syndrome scores, visual analogue scale (VAS) for symptom before and after treatment and the adverse reaction with the condition disposed were observed in the two groups.
RESULTSAfter treatment, the symptom and syndrome scores and VAS for symptom were improved compared with those before treatment in the two groups (all P < 0.05), and the improvements of the application group were more obvious than those of the placebo group (all P < 0.05). The short effective rate of the application group was 68.1% (113/166), which was better than 48.2% (40/83) of the placebo group (P < 0.05). The adverse reaction rates were 4.2% (7/166), 2.4% (2/83) respectively, with no statistical significance between the two groups (P > 0.05).
CONCLUSIONThe dog days plaster achieves obvious effect with good safety for allergic rhinitis.

3.Assessment of coronary stenosis by 256-slice spiral CT

Objective:To assess coronary stenosis using 256-slice spiral CT (CTA).Methods:The data of 126 pa-tients diagnosed as coronary heart disease (CHD)using CTA and coronary angiography (CAG)Were retrospectively analyzed.The CAG results for CHD Were regarded as “gold standard”diagnosing CHD.The CTA diagnostic value for CHD and consistency of CAG and CTA diagnosing coronary stenosis Were analyzed.Results:A total of 393 cor-onary segments Were assessed using CTA.According to CAG results for CHD ,the CTA diagnostic accuracy Was 89.3%,sensitivity Was 91.4%,specificity Was 84.8%,negative predictive value Was 82.2% and positive predictive value Was 92.8%;the consistency of CAG and CTA diagnosing coronary stenosis Was good (Kappa value =0.755, P<0.01).Conclusion:The 256-slice spiral CT can diagnosing coronary stenosis,it possesses high sensitivity in di-agnosing vascular stenosis;as a simple and noninvasive examination it can be used as a reliable method for early screening subhealthy population and reexamine of putients With coronary heart disease.
4.How to break through the spell of intensive care unit infection

Nosocomial infection is one of the most common diseases in the intensive care unit (ICU), which has received much concern due to its multi-type infection, more severe infection, difficulty in treatment and high mortality. However, the drug resistance of bacteria is increasingly serious with the wide use of the antibiotics which is no longer an ideal strategy. Instead, the control and prevention of infection from their sources is the key to reduce ICU infection. Especially for some particular nosocomial infections, such as hospital acquired pneumonia (HAP) and ventilator associated pneumonia(VAP), comprehensive measures are needed to take to break the spell of ICU infection.

6.Detection of food allergen-specific IgE and IgG in patients with chronic urticaria

Objective To evaluate the roles of food allergen detection in patients with chronic urticaria.Methods Food allergen-specific IgE and IgG were detected in 502 patients with chronic urticaria and 100 normal human controls.Double-blind,placebo-controlled food challenge (DBPCFC) test was performed in patients who were proved to be positive or negative for food-specific IgG or IgE.Results Food-specific IgE was detected in 37.45% (188/502) of the patients,with cashew,peanuts and soybeans as the most common allergens.The detection rate of food-specific IgG was 72.11% (361/502),with the major allergens being milk,egg,shrimp and crab.DBPCFC revealed false positivity and negativity in the detection of food-specific IgG and IgE.Conclusions Clinicians should pay attention to food allergen detection in the treatment of chronic urticaria,especially in the treatment of allergy mediated by IgG antibodies.
7.Hospital Infection:Analysis of 706 Patients

OBJECTIVE To understand the related factors,the situation and dynamics of hospital infection in order to effectively prevent and control it.METHODS From 12306 cases of inpatients who had investigated retrospectively,706 cases were with nosocomial infection monitored from Jan to Dec 2006.RESULTS The hospital infection rate was 5.74% in 2006.The hematological disease and,cancer patients and the elderly,were the high-risk population.The main infection site was the respiratory tract.Fungi accounted for 21.35%,the top was the Candida albicans.CONCLUSIONS To strengthen the training of medical staff,to monitor the key departments of hematology and cancer and respiratory diseases and to attend the disease surveillance and rationally use of antibiotics are the effective measures to reduce hospital-acquired infections.

OBJECTIVETo elucidate the effect of hydroxysafflor yellow A ( HYSA) on the proliferation of vascular smooth muscle cells (VSMCs) and the related mechanism.
METHODSVSMCs derived from SD rats were treated with DMEC culture medium (Control), 10 ng/ml PDGF (PDGF group), pretreatment with HYSA at different doses (1, 5, 10, 20, 40, 60 µmol/L) for 24 h then cotreatment with PDGF. After 24 h, MTT assay, Western blot and immunohistochemical staining were performed to evaluate the inhibitory effects of HYSA on VSMCs proliferation.
RESULTSHYSA inhibited PDGF induced VSMCs proliferation in a dose-dependent manner, dowregulated proliferating cell nuclear antigen (PCNA) expression and blocked PDGF activated PDGFR-MEK-ERK1/2 signaling pathway.
CONCLUSIONSHYSA inhibits VSMCs proliferation possibly via downregulating the expression of PCNA and blocking MEK-ERK1/2 signal transduction in VSMCs.

9.Expression of membrane CD14, Toll-like receptor 4 and nuclear factor-kappa B in peripheral blood leukocytes from patients with psoriasis vulgaris

Objective To investigate the expression of membrane CD14 (mCD14), Toll-like receptor (TLR4) and nuclear factor-kappa B (NF-kB) in peripheral blood ieukocytes from patients with psoriasis vulgaris (PV). Methods Blood samples were obtained from 29 normal controls and 32 patients with PV. Patient cohort consisted of 19 males and 13 females, with an average age of 30.28 ±13.11 years, disease duration of 8.21±10.21 years, and PASI score of 21.19±5.85. Flow cytometry was used to measure the expression of mCD14, TLR4 and activated NF-kB in peripheral blood leukocytes in these subjects. Results The expressions of mCD14 and TLR4 in peripheral blood leukocytes were significantly higher in patients than in the normal controls (5.454±2.78 vs 3.937±1.964, P < 0.05; 17.641±18.120 vs 7.527±8.574, P <0.01 ). Although the expression of activated NF-kB in total blood leukocytes in patients was higher than that in normal controls, no statistical difference was noticed (63.538±8.650 vs 62.236±6.72, P > 0.05). Conclusion There is an abnormal expression of mCD14 and TLR4 in patients with PV, suggesting that the abnormal activation of endotoxin signal transduction pathway may exert a role in the induction of psoriasis.
10.Semi-rational modification for improving bond selectivity of recombinant β-glucuronidase.

To improve bond selectivity of recombinant β-glucuronidase in Escherichia coli (PGUS-E), based on the PGUS-E structure guidance, three key points R329, T369 and N467 were identified to be responsible for the bond selectivity of PGUS-E, and further saturation mutagenesis was conducted. Two positive mutants R329K and T369V were obtained by a combined selection technique of thin-layer chromatography and high performance liquid chromatography. Compared to PGUS-E, the bond selectivity of mutants R329K and T369V increased by 26.9% and 34.3%, respectively; whereas the biochemical properties such as pH and temperature profile were unchanged. Nevertheless, the activity was decreased compared to PGUS-E. These results further confirmed that sites R329 and T369 played important roles for the bond selectivity and activity. In summary, this study significantly increased the bond selectivity of PGUS-E by structure guided saturation mutagenesis, providing experimental support for elucidating the relationship between the structure and function of PGUS-E.
