1.A retrospective analysis of laparoscope versus open repair on treating upper gastrointestinal ulcer perforation

ObjectiveTo evaluate the effect of laparoscope and open repair on treating upper gastrointestinal(UGI) ulcer perforation.MethodsRetrospective analysis was performed in consecutive series of 143 patients undergoing UGI ulcer perforation repairing.Among the total 143 patients,98 cases underwent laparoscope repair(laparoscope group),45 cases were with open repair (open group).The operation methods included either simple interrupted suture or pedicled omental patch.Postoperative management of triple therapy included proton pump inhibitor,β -lactam antibiotics and arilin was conducted.Therapeutic effects were compared between two groups.ResultsOperating time in laparoscope group was significantly longer than that in open group [ ( 83.88 ± 19.67 ) min vs.( 63.33 ± 12.06) min,P < 0.01 ].Time of passage of gas by anus,hospital stay,cost of hospitalization and postoperative dosage of opiates in laparoscope group were significantly lower than those in open group [ (29.45 ± 9.24) h vs.( 46.40 ± 49.21 )h,(6.92 ± 1.06) d vs.(7.71 ± 2.48 ) d,(6929.39 ± 832.40) yuan vs.(7546.67 ± 1393.92) yuan,(5.7 ±8.3 ) mg vs.( 10.9 ± 9.5 ) mg,P< 0.01 ].There was no significant difference between two groups in perforated diameter,perforated position and flushing dose of abdominal cavity (P> 0.05 ).The rate of complication was 14.29% (14/98) in laparoscope group,while 26.67% (12/45) in open group,there was no significant difference between two groups (P> 0.05 ).ConclusionsLaparoscope repair of UGI ulcer perforation is a safe and feasible procedure compared with open repair.Although operating time of laparoscope repair is longer than open repair,the rate of complication does not increase.
2.Clinical Efficacy of the Self-made TCM on Toxicity of PR Combined with CTX Treatment for Lupus Nephritis

OBJECTIVE:To explore optimal regime and less ADR for lupus nephritis(LN) by adopting self-made TCM prescription.METHODS:109 LN patients were assigned into control group(n=48) and therapy group(n=61).Both groups were treated with metacortandracin and cyclophosphamide.Control group were additionally given self-made TCM prescription.The clinical efficacy and adverse drug reaction(ADR) of 2 groups were compared.RESULTS:The curative rate and total response rate of therapy group were 77.05%(47/61) and 95.08%(58/61) respectively and those of control group 58.33%(28/48) and 77.08%(37/48).The curative rate and total response rate of therapy group were all higher than those of control group.There were significant difference between 2 groups(P
3.Effects of trichostatin A on HDAC8 expression, proliferation and cell cycle of molt-4 cells.

The effects of Trichostatin A (TSA) on histone deacetylase 8 (HDAC8) expression, proliferation and cell cycle arrest in T-lymphoblastic leukemia cell line Molt-4 cells in vitro were investigated. The effect of TSA on the growth of Molt-4 cells was studied by MTT assay. Flow cytometry was used to examine the cell cycle. The expression of HDAC8 was detected by using immunocytochemistry and Western blot. The results showed that proliferation of Molt-4 cells was inhibited in TSA-treated group in a time- and dose-dependent manner. The IC50 of TSA exposures for 24 h and 36 h were 254.3236 and 199.257 microg/L respectively. The cell cycle analysis revealed that Molt-4 was mostly in G0/G1 phase, and after treatment with TSA from 50 to 400 microg/L for 24 h, the percents of G0/G1 cells were decreased and cells were arrested in G2/M phase. Treatment of TSA for 24 h could significantly inhibit the expression of HDAC8 protein in Molt-4 cells (P<0.01). It was concluded that TSA could decrease the expression of HDAC8 in Molt-4 cells, which contributed to the inhibition of proliferation and induction of cell cycle arrest in Molt-4 cells.

Objective To establish chronic obstructive pulmonary disease(COPD)rat models,and to study the intervention of the formula of nourishing Qi,activating blood circulation and dispersing phlegm recipe on the morphology.Method To establish rat COPD models by intratracheal instillation of lipopolysaccharide and exposure to cigarette smoke.To instill intervention drug daily either the formula of nourishing Qi,activating blood circulation and dispersing phlegm recipe or the roxithromythin,starting on the 20th,30th and 40th day of the experiment respectively(the groups was named h 20,h 30,h 40,r 20, r 30 and r 40 for short),and to observe the effect on the morphology by means of collagen staining and image analyzer.Results The pathological changes and lung function in the model group were accorded with the human COPD.In drug intervention groups,airway inflammation and epithelial proliferation were alleviated to different degree compared to the model group.In model group,the collagen deposition was increased predominant type I collagen compared to the health comtrol group,and the deposition in the drug intervention groups were decreased compared to the model group,according to the Sirius redpolarizing microscopy morphometry method.The thickness of the airway wall in the model group was significantly increased compared to the health control group(P

Birth defects are the abnormalities in structure,function or metabolism of embryonic or fetal development.They are the main cause of early abortion,stillbirth,perinatal mortality,infant mortality and congenital disabilities.Birth defects affect not only the health and life quality of children,but also the population quality of the whole country.This paper reviews the current epidemic situation and possible causes and risk factors of birth defects.It also expounds on the importance of epidemiological study on birth defects,possibility of primary prevention of birth defects,and necessity of interdisciplinary and collaborative research.It may provide reference for prevention and control of birth defects.
6.Application of bioinformatics analysis in identifying a novel annexin subfamily-anx32

Objective: To investigate the structural characteristics and the primary functions of antigen gene cC1. Meth-ods: The primary and the secondary structures were analyzed using bioinformatics programs provided by Internet servers.The predicted 3D structure of cC1 was established by homology protein modeling method. Anticoagulant activity of GST-anx32 was assayed by modified kaolin partial thromboplastin time (KPTT). Results: cC1 had high homology to annexinsgenes both at nucleic acid and at amino acid level. It contained 4 homologous regions, and each region included the typical an-nexin motif "G-X-G-T (38 residues)-D/E". The results of KPTT assay showed that the recombinant protein GST-anx32 hadhigh anticoagulant activity. Conclusion: cC1 has the common structures of annexins but the homology to the extant annexinsis no more than 48%, cC1 is a member of a novel annexin subfamily and designed as annexin32.

AIM: To determine whether chronic hypercholesterolemia affects ionic currents on cardiac ventricular myocytes of rats. METHODS: Whole - cell patch - clamp technique was used to record the ionic currents in single cardiac myocytes isolated from normal cholesterolemia and hypercholesterolemia rats. Objective To study the dynamic changes and clinical significance of D-D, t-PA and PAI in patients with acute renal failure during the process of different methods of blood purification. Methods Thirty-seven ARF patients were divided into three groups: HD group, HDF group and HF group. Plasma D-D level, t-PA and PAI activity were determined 1 hour, 4 hours before and after treatment. Normal control group consisted of fourteen healthy people. 10.Anti-cancer effects of deguelin on human leukemia K562 and K562/ADM cells In Vitro.

In order to investigate the anti-cancer effects of deguelin and on K562 and K562/ADM cells in vitro and the underlying molecular mechanism and compare the cytotoxicity of deguelin on K562, K562/ADM cells and human peripheral blood mononuclear cells (PBMCs). The effects of deguelin on cell proliferation were assessed by MTT assay. Apoptosis were detected by Annexin V/PI double-labeled cytometry. The effects of deguelin on the cell cycle were studied by a propidium iodide method. Our study showed that deguelin inhibited the proliferation of K562 cell and K562/ADM cell in a time- and dose-dependent manner and had minimal effects on normal human peripheral blood mononuclear cells. The ratio of IC(50) value of deguelin of 24 h on K562/ADM cells to K562 cells was only 1.27, which was significantly lower than the ratio of IC(50) value of ADM (higher than 20). Deguelin could induce apoptosis of K562 cells and K562/ADM cells. K562 cells were arrested at G(2)/M phase while K562/ADM cells were arrested at G(0)/G(1) phase. Our results suggested that deguelin was a novel anti-leukemia agents with high efficacy and low toxicity and it is also a promising agent for reversing drug resistance.
