Objective To investigate the expression of S100A4 and hypoxia-induced factor 1α (HIF-1α) protein in type Ⅰ endometrial carcinoma,and to analyze their relationship.Methods The expressions of S100A4 and HIF-1α protein were detected in 65 cases of type Ⅰ endometrial carcinoma and 40 normal endometrial tissues.Results The positive rate of S100A4 in endometrial carcinoma was 47.7 ％ (31/65),the normal tissues was 0 (0/40),and there was significant difference (x2 =27.069,P =0.001).The positive rate of HIF-lα was significantly higher in endometrial carcinoma [63.1％ (41/65)] than that in the normal tissues [7.5 ％ (3/40)] (x2 =31.417,P =0.001).The expressions of S100A4 and HIF-1α were positively related with FIGO staging,pathological grades,myometrial invasion and lymphatic metastasis (all P ＜ 0.05).The expressions of S100A4 and HIF-1α were positively correlated in type Ⅰ endometrial carcinoma (r =0.348,P =0.005).Conclusion The expression levels of S100A4 and HIF-1α are closely related with the oncogenesis and progression of type Ⅰ endometrial carcinoma,and their co-detection may be useful in prognosis assessment.

OBJECTIVE To investigate the infection status and drug resistance of mycoplasma and chlamydia isolated from female genitourinary tract in order to provide reference for doctors′ reasonable use of medicine.METHODS The IST2 kit produced by Bio-Merieux was used to culture the mycoplasma strains and to do drug susceptibility test.The VEDALAB kit was used to detect chlamydia.RESULTS Among 376 suspected patients with nongonococcal urethritis(NGU),the number of the patients infected by Ureaplasma urealyticum(Uu),mixed strains of Uu and Mycoplasma hominis(Mh),and Chlamydia trachomatis(Ct) were 216,35 and 125,respectively.The positive rate was 57.4%,9.3% and 33.3%,respctively.The drug susceptibility test showed that drug resistance rates of the other six antibiotics in mixed infection of Uu and Mh were higher than that in simple infection of Uu except for josamycin,ofloxacin and doxycycline.The sensitivity of doxycycline,josamycin and platenomycin in mycoplasma was the most highest.CONCLUSIONS Infection in female genitourinary tract is mainly due to Uu.We should reasonably choose and use antibiotics according to drug susceptibility.

Objective To detect the expression of hypoxia-inducible factor 1α (HIF-1α) and bcl-2 ovarian serous carcinoma and their clinical significances. Methods Paraffin specimens including 61 cases of ovarian serous carcinoma and 50 normal ovarian tissues were selected. The expressions of HIF-1α and bcl-2 proteins were detected by immunohistochemical EnVision method and their relationship between them was analyzed. Results The positive rate of HIF-1α and bcl-2 proteins expression in 61 ovarian serous carcinoma was 68.9%and 54.1%, respectively. There was a significant difference between the two groups (χ2=55.381, P< 0.05; χ2= 38.493, P< 0.05). The clinical pathological parameters showed that the positive expression of HIF-1αand bcl-2 proteins were not related with the age (P>0.05). HIF-1αpositive expression was correlated with tumor grades, the state of lymph node metastasis and FIGO stages (χ2=4.931, 25.008, 5.610, P<0.05). Bcl-2 was significantly associated with tumor grades and lymph node metastasis (χ2= 6.956, 33.869, P<0.05), but not with FIGO stages (χ2=3.391, P>0.05). The expression of bcl-2 was positively correlated with HIF-1α in ovarian serous carcinoma (r= 0.304, P= 0.017). Conclusions The expressions of HIF-1α and bcl-2 play a synergic role in the progression of ovarian serous carcinoma. The combined detection of HIF-1αand bcl-2 is effective for patients'prognosis judgment.

OBJECTIVEThis study aims to evaluate the antimicrobial activity of decapeptide, a novel antimicrobial peptide, against several major cariogenic and periodontopathogenic bacteria in vitro. METHODS In this study, we investigated the antimicrobial activity of decapeptide against Streptococcus mutans (S. mutans), Streptococcus sobrinus, Lactobacillus acidophilus, Streptococcus sanguis, Streptococcus gordonii, Actinomyces viscosus, Actinomyces naeslundii, Porphyromonas gingivalis, Prevotella intermedia, Fusobacterium nucleatum, Actinobacillus actinomycetemcomitans, and Saccharomyces albicans in vitro using the agar diffusion method and broth dilution method. Furthermore, a time-kill kinetic study of decapeptide against S. mutans was performed.

RESULTSThe results showed that decapeptide exhibited antimicrobial activity against various oral bacteria and fungi. The minimum inhibitory concentration (MIC) of main cariogenic bacteria ranged from 62.5 μg · mL(-1) to 125 μg · mL(-1), and the MIC of periodontopathogenic bacteria tested ranged from 250 μg · mL(-1) to 1,000 μg · mL(-1). Among the bacteria tested, decapeptide had a strong inhibitory effect on cariogenic S. mutans. Results of the time-kill kinetic studies showed that decapeptide reduced the viable counts of S. mutans by more than one order of magnitude after 20 min of incubation, and thoroughly killed S. mutans after 30 min. No viable cells could be detected after 24 h of incubation.

CONCLUSIONThis study suggest that decapeptide might have potential clinical application in treating dental caries by killing S. mutans within dental plaque.

The recombinant plasmid of mChlL-18 prokaryotic expression was transformed into E.coli BL21(DE3) strain and then induced by IPTG at 37℃.After crushed and washed,the expressing inclusion bodies were thoroughly denatured with 6 mol/L guanidine hydrochloride.Then according to experiment design,the effects of rChlL-18 protein refolding yield at different densities were investigated by the systems of artificial chapercne at different densities.Experiment results indicate that there is a optimal condition on assiting rChIL-18 protein by using the artificial chaperone technique.The optimal condition can improve the refolding yield of rChIL-18 protein,and then the expressed product of fusion chicken IL-18 gene in E.coli has a relativity high bioactivity.

Objective To detect abnormal anisotropy on the splenium of the corpus callosum in patients with mild Alzheimer’s disease (AD).Methods Normalized hippocampal volume and fractional anisotropy on the splenium of the corpus callosum were measured in 13 patients with mild Alzheimer’s disease and 19 normally aging volunteers. Non-dependent samples t-tests were conducted to compare them between the two groups. In addition, the sensitivity in the diagnosis of mild Alzheimer’s disease was calculated.Results The splenium of the corpus callosum showed reduced anisotropy in mild Alzheimer’s disease. The normalized hippocampal volume also decreased in patients. The FA of the splenium of the corpus callosum was more sensitive in the diagnosis of mild Alzheimer’s disease. Conclusion The decreased anisotropy on the splenium of the corpus callosum reflects the degeneration of the fibers, and it is promising in the early diagnosis of Alzheimer’s disease.

Objective To investigate the expression and diagnostic significance of CK19,CD56 and p53 protein in papillary thyroid microcarcinoma and thyroid papillary hyperplasia.Methods The expressions of CK19、CD56 and p53 protein were detected in 52 cases of papillary thyroid microcarcinoma and 31 of thyroid papillary hyperplasia by iImmunohistochemical methods.Results The positive rate of CK19 expression was 100 ％ (52/52) of papillary thyroid microcarcinoma and 29.0 ％ (9/31) in 31 of thyroid papillary hyperplasia.There was significant difference between two groups (P ＜ 0.001).CK56 in 2 cases (3.8 ％) of papillary thyroid microcarcinoma appeared mild positive expression,and in 20 cases (66.7 ％) of thyroid papillary hyperplasia positive expression (P ＜ 0.001).The positive expression rates of p53 were 69.2 ％(36/52) in papillary thyroid microcarcinoma and 6.5 ％ (2/31) in thyroid papillary hyperplasia (P ＜ 0.001).Conclusion CK19,CD56 and p53 may be important value on differential diagnosis of papillary thyroid microcarcinoma from thyroid papillary hyperplasia,and they are the indispensable markers of differential diagnosis.

Objective To evaluate the relationship between airway obstruction and peripheral arterial stiffness in elderly patients with chronic obstructive pulmonary disease (COPD).Methods Ninetyeight elderly patients with COPD were enrolled.The patients were divided into two groups according to forced expired volume in 1 second (FEV1) percentage of predicted (FEV1 ％):mild-moderate group (FEV1 ％ ≥ 50％,62 cases) and severe group (FEV1％ ＜ 50％,36 cases).The brachium-ankle pulse wave velocity (baPWV),body mass index (BMI),24 h average heart rate,blood level of lipid,fasting blood glucose,arterial partial pressure of oxygen (PaO2) and high sensitivity C reactive protein (hs-CRP) level were compared between two groups.Results Compared with mild-moderate group,the BMI in sever group was lower:(22.9 ± 4.3) kg/m2 vs.(25.4 ± 3.3) kg/m2,24 h average heart rate was higher:(77.5 ± 12.8) times/rmin vs.(70.7 ± 9.5) times/min,PaO2 was lower:(71.7 ± 13.1) mmHg (1 mmHg =0.133 kPa) vs.(84.5 ± 13.2) mmHg,and there were statistical differences (P ＜ 0.05).The baPWV in mild-moderate group was (18.9 ± 4.2) m/s,and in sever group was (21.8 ± 3.6) m/s,and there was statistical difference (P ＜ 0.05).The hs-CRP in miid-moderate group was (6.62 ± 4.57) mg/L,and in sever group was (3.38 ± 2.13) mg/L,and there was statistical difference (P＜ 0.05).Conclusions The morbidity of peripheral arterial stiffness is increased in elderly patients with COPD.Progression of peripheral arterial stiffness is related to the severity of airway obstruction.

OBJECTIVETo compare the protein profiles between decapeptide-treated and untreated planktonic cells of Streptococcus mutans (S. mutans) by differential proteomic analysis to determine and identify the key proteins.

METHODSIn our previous study, we investigated decapeptide (KKVVFKVKFK-NH2), which was a novel adenosine monophosphate. Compared with other oral pathogens tested, decapeptide had a preferential antibacterial activity against S. mutans. It also inhibited S. mutans biofilm formation and reduced the one-day developed biofilm. In the present study, we first synthesized decapeptide, and then compared the protein profiles between decapeptide-treated and untreated planktonic cells of S. mutans by two-dimensional gel electrophoresis and matrix-assisted laser desorption ionization time-of-flight mass spectrometry. We also verified different expressions of key protein enolase in the protein level.

RESULTSThe results showed that decapeptide altered the protein expression of planktonic S. mutans. These proteins were functionally involved in carbohydrate degradation by glycolysis, protein folding, conjunction, transport, translation, adenosine triphosphate binding, protein binding, sequence-specific DNA binding, transcription factor activity, and two-component response regulator activity. Western blot results showed that enolase protein expression decreased obviously in decapeptide-treated cells of S. mutans.

CONCLUSIONThe protein expression of S. mutans significantly changed after synthetic antimicrobial decapeptide treatment, suggesting that decapeptide may present a preferential effect on oral caries by changing the expression of certain key proteins, such as enolase protein.

Objective To detect gray matter abnormalities of whole brain in patients with mild Alzheimer's disease(AD)by voxel-based morphometry(VBM).Methods Thirteen patients with mild Alzheimer's disease and sixteen normal aging volunteers underwent 3D SPGR scanning.For every subject,data was transferred to PC to be normalized,segmented and smoothed using SPM99.Non-dependent samples T-tests were conducted to compare gray matter density voxel to voxel between the two groups.Results Significant reductions in gray matter density were found in the bilateral hippocampi and nucleus amygdalae,bilateral insulae,bilateral medial thalami,bilateral rectus gyri,right superior temporal gyrus,right caudate nucleus,right prefrontal lobe,right basal forebrain and portions of right occipital lobe.Conclusion VBM reveals significant gray matter reductions of numeral cortices in mild Alzheimer's disease.It can be a useful method to evaluate the anatomical changes in the progress of the disease.
