1.Effect of estrogen on cardiac injury and cardiomyocyte apoptosis of rat induced by isoproterenol

Objective To study the effect of estrogen on cardiac injury and cardiomyocyte apoptosis of rat induced by isoprotere-nol by modeling cardiac inj ury induced by bilateral ovariectomized (OVX)and isoproterenol (ISO).Methods Fifty female SD rats with bilateral ovariectomy and sham operation (Sham)were randomly divided into 5 groups:sham operation group (Sham group), bilateral ovariectomy group (OVX group),cardiac injury group (OVX+ISO+Vehi group),low dose estrogen treatment group (OVX+ISO+E2 a group,4μg·kg-1 ·d-1 ),high dose estrogen treatment group (OVX+ISO+E2 b group,40μg·kg-1 ·d-1 ). these status were separately measured:rats′general features,hemodynamics parameters monitored of carotid artery,morphological observation and cardiomyocyte contraction change of single-cardiomyocyte separate cultured,cardiomyocyte apoptosis protein ex-pression were detected by immunoblotting.Results ISO significantly reduced myocardial pump function,increased hypertrophy and apoptosis of cardiomyocytes,reduced contractility of single cardiomyocytes (P<0.05).High-dose estrogen (40μg·kg-1 ·d-1 ) replacement therapy significantly improved ISO induced cardio inj ury and cardio functions decreasing,also inhibited Bax expression and caspas-3 activation and decreased myocardial hypertrophy and cardiomyocytes apoptosis through increasing Bcl-2 expression (P<0.05),significantly.while low dose estrogen (4μg·kg-1 ·d-1 )treatment showed marginally protection effects on ISO in-duced cardio inj ury with no statisticly significance.Conclusion Appropriate dose estrogen replacement therapy can decrease cardio-myocyte apoptosis,improve cardiomyocytes contractility,so as to protect ISO-induced cardiomyocyte apoptosis.

Objective To observe the DNA damage in the human lung fibroblasts(HLF)induced by insoluble nickel compounds(Ni3S2 and Ni2O3)and soluble nickel compound(NiSO4),and to study the mechanism of carcinogenesis.Methods The DNA damage of HLF cells treated with different concentrations of Ni3S2,Ni2O3 and NiSO4 were detected with the method of single cell gel electrophoresis(SCGE).Results All of the three kinds of nickel compounds significantly caused DNA damage in HLF cells,the average tail moments of all nickel-treated groups were larger than those in the control group(P

4.A screen assay of ?-secretase inhibitors in vitro

Aim To construct a screen assay of ?-secretase inhibitors.Methods A human ?-secretase cDNA was cloned by RT-PCR and then reconstructed in an expression vector.The recombinant ?-secretase fusion protein was expressed and purified from Escherichia coli.A screen assay of ?-secretase inhibitors was constructed in vitro using a recombinant A ?-fusion protein as substrate.The enzyme reaction was detected by SDS-PAGE and an ELISA method using metal chelating plates to catch the substrate.Results A human ?-secretaae cDNA was cloned and expressed in escherichia coli.The ?-secretase enzyme reaction can be detected by two kinds of method: SDS-PAGE and ELISA.The IC_(50) for an inhibitor statVal determined by this method was in good agreement with those reported in the literature.Seventy seven species of Chinese traditional medical plants were screened using this method and seventeen species show weak inhibitory activity.Conclusion A screen assay of ?-secretase inhibitors was successfully constructed and can be used to screen natural products.
5.Mental Status Analysis of Stomatology Graduates before Job Hunting

Objective To understand the mental status of 180 stomatology graduates(including undergraduates,masters and PhD students)in China Medical University,whom are hunting job in 2016. Methods A total of 180 stomatology graduates were tested by SCL?90 symptom check?list and self?questionnaire. Results Compared with the national norm,4 factors of SCL?90 in stomatology graduates shows significant differences (P<0.05),they were obsessive?compulsive symptoms,depression,anxiety and paranoid. Chi?square test showed that the 4 factors of masters and doctoral graduate students were significantly higher than undergraduates(P<0.05),and there was no difference between the former two(P>0.05). Multivariate logistic regression analysis showed that family economic conditions were positively correlated with postgraduate examinations;their will?ing to work in stomatology hospitals were closely related to anxiety,depression and other negative emotions. Statistical techniques with univariate re?gression analyses indicated that households’financial status,whether to study for graduate student and doctoral student further,job object for stoma?tological hospital,dental clinic,or department of stomatology in general hospital may be risk factors. Conclusion There are various degrees of men?tal health problems in stomatology graduates of 2016. We should actively guide them,and strengthen their mental health education.
6.Effect of Crownless Roots on Surrounding Alveolar Bone Tissues in SD Rats with Osteoporosis

Objective To study the adverse effects on periodontal tissues caused by crownless roots in elderly people with osteoporosis. Methods Totally 60 12?week SD rats(30 male and 30 female)were randomly divided into the control group(10 male and 10 female sham?operated rats)and the osteoporosis group(20 male and 20 female castrated rats). After 12 weeks,the bone mineral density of right femora was measured by dual energy X?ray for all rats to confirm osteoporosis. The right molar crown was pinched off with haemostatic forceps and the molar root was retained. The 10 con?trol rats and the 20 experimental rats were sacrificed respectively at postoperative 2 and 4 weeks. The periodontal tissues of right molar were sampled and the morphology of the parodontium and the alveolar bone was observed by haematoxylin and eosin(HE)staining and the expression of tumour necrosis factor?alpha(TNF?α)and inducible nitric oxide synthase?2(NOS2)was detected by immunohistochemical staining and real time fluores?cent quantitative PCR(qRT?PCR). Results The HE slices indicated that the periodontal tissues in the osteoporosis group were significantly im?paired compared to the control group as the structure of parodontium loosened. Bone impairment aggravated over time. TNF?αstaining showed as a few claybank spots on the parodontium in the control group,while the staining colour was darkened in the osteoporosis group. The expression of TNF?αmRNA was significantly higher in the osteoporosis group than in the control group(P<0.05). The result of NOS2 was contrary to that of TNF?α. The differences in TNF?αand NOS2 were not statistically significant at 2 and 4 weeks(P>0.05). Conclusion The crownless roots in rats with os?teoporosis cause adverse effects of periodontal tissues as the roots accelerate absorption of the surrounding alveolar tissues. It is indicated that crown?less roots in elderly people with osteoporosis should be removed as soon as possible.
7.STUDY ON THE ANTIOXIDANT ACTIVITY OF SWEET POTATO ANTHOCYANIN AND ITS INHIBITING EFFECT ON GROWTH OF CANCER S_(180)

Objective: To study the antixiodant activity of sweet-potato anthocyanin (SPAC) and its inhibiting effect on growth of cancer S180. Method: The scavenging capacity of ??O2 and?OH and the inhibitory effect of MDA were determined in vitro.The SOD activity of serum and skin, the effect on S180 growth, serum GSH-Px activity and MDA were tested in mice. Results: SPAC had antioxidant activity in vitro, with elimination rate of O2?? and?OH 30.63% and 20.57% higher than the same concerntration of VC respectively. Activity of SOD in the serum and skin of mice were raised 27.30% and 13.50% respectively after giving SPAC. The growth rate of cancer S180 in the mice was inhibited, and the highest inhibitory rate was 43.12%. The activity of serum GSH-Px, SOD of the mice was raised as compared to control, but MDA declined . Conclusion: SPAC is a perfect natural pigment with inhibitory effect on growth of cancer S180 probably through its antioxidant activity.

OBJECTIVE To discuss management in the prevention and treatment of infectious diseases and influenza A(H1N1) the according to law.METHODS The requirements of prevention and treatment for infectious diseases and influenza A(H1N1),were set out according to "Communicable Disease Prevention Act" and other relevant laws and regulations.RESULTS The prevention and control strategies and measures of influenza A(H1N1) still existed some problems.CONCLUSIONS The key doing a good job of prevention and control for infectious diseases and influenza A(H1N1) is according to the law scientificly and orderly.
9.EFFECTS OF SELENIUM AND VITAMIN E ON APOPTOSIS AND C-MYC EXPRESSION IN LEUKEMIC CELLS

Objective: To study the effects of vitamin E and selenium on apoptosis and c-Myc expression in human leukemia cells. Methods: DNA gel electrophoresis and Northern blotting hybridization were used to detect the apoptosis and the expression of c-Myc gene, respectively. Human leukemia cell line HL-60 and K562 were cultured in vitro. Results: The apoptosis of HL-60 and K562 cells were induced after being exposed to vitamin E (100 ?mol/L) and selenium (8 ?mol/L) for 24 hours, respectively. In HL-60 cell line, c-Myc mRNA was down-regulated significantly by vitamin E(100 ?mol/L),but not selenium(8 ?mol/L).In contrast, the expression of c-Myc gene was repressed by selenium(8 ?mol/L) and not by vitamin E(100 ?mol/L) in K562 cell. Conclusion: Our observations suggest that c-Myc down-regulation and induction of apoptosis by selenium and vitamin E are important pathways in repressing leukemia cell proliferation.The results suggest there are different mechanisms of repressing leukemia cell proliferation for selenium and vitamin E.
10.THE PROTECTIVE EFFECTS OF VITAMIN C ON OXIDATIVE DAMAGE OF ALEVEOLAR MACROPHAGE INDUCED BY Ni2O3 IN VITRO

Objective:To study the effects of vitamin C (VC) on the injury of alveolar macrophages exposed to Ni2O3 in vitro and the expression of inducible nitric oxide synthase (iNOS). Method:The alveolar macrophages were cultured with exposure to Ni2O3 in vitro. Meanwhile,VC with different concentrations (25,50 and 100 ?mol/L) were added to the medium, respectively. The cell activity,nitric oxide(NO),reactive oxygen species (ROS), malondialdehyde (MDA) and activities of SOD, GSH-Px,CAT and iNOS were detected. iNOS mRNA expression was detected by using RT-PCR. Results:VC could decrease mortality and increase survival activity of alveolar macrophages. VC could also decrease ROS,NO and NOS activity,increase SOD,GSH-Px, CAT activities and downregulate the expression of iNOS mRNA. Conclusion:The lipid peroxidation of alveolar macrophage could be induced by Ni2O3. VC could downregulate the expression of iNOS mRNA and reduce the production of ROS and NO through increasing the activity of antioxidative enzymes and antagonize the oxidative damage induced by Ni2O3.
