25% signified analgesic effect. Part II (the effect of agmatine on analgesic effect of morphine). Fifty-six animals with neuropathic pain were randomly divided into 4 groups ( n = 14 each) . Each group received NS or agmatine 20, 40 or 60 mg? kg-1 . 30 min later each group received either NS or morphine 1 mg?kg-1(n=7 each). PWT was measured and PMAP was calculated Part III (the effects of agmatine on the development of morphine tolerance). Morphine tolerance was incuced by morphine 10 mg?kg-1, 3 times a day for 5 days. Thirty-five animals with neuropathic pain were randomly divided into 5 groups ( n = 7 each): group A received NS: group B received NS + morphine; grouop C, D, E received agmatine 5, 10, 20 mg ? kg-1 + morphine 3 times a day for 5 days. PWT was measured on the 5th day and PMAP was calculated. Results PMAP of agmatine (40, 60 mg?kg-1 i.p.) and morphine (1 mg?kg-1 s.c.) were lower than 40% . Agmatine 60 mg?kg-1 increased the PMAP of morphine 1 mg?kg-1 to 84% compared with 34% for morphine alone (P

Underwater exercise and hydrotherapy have rapidly developed in recent years. As an noninvasive and convenient tool, surface electromyography (sEMG) has been applied in underwater research. In favour of experiment procedure, underwater sEMG needs localized waterproof seal or whole-body waterproof suit, and are mainly detected by telemetry system. The muscle activity pattern of underwater exercise is different from on land exercise, the sMEG amplitude is correlated with motion velocity. Compound underwater motion analysis includes sEMG, video and foot pressure will be more effective assessment tool for underwater exercise and hydrotherapy research.

Objective To conduct an empirical study for quantifying the anastomosis between two vessels after skin expanded technique by the method of angiography and to provide a precise basis for vascular study in skin flap. Methods Bilateral skin flaps based on deep iliac circumflex vessels were elevated from the abdominal wall including deep superior epigastric vessels. One was expanded at the boundary between two vessels and the other unexpanded. An X-ray image was obtained by carotid arterial injection of gelatin-lead oxide mixture.Three parallel lines with equal intervals perpendicular to long axis of the two vessels were designed at the communication area.Vessel anastomosis quantity was determined by counting the number of marks derived from the intersections of the lines and the vessels and statistical analysis was carried out. Results The marks of intersection in expanded group were more than unexpanded group with statistical significance. Conclusions The method for quantifying vessel anastomosis in skin flap is reliable. The principles of this procedure may also be applied to other experimental and elinical studies.

AIM: To study the effect of cocaine on caspase-3 in myocardiac cells of male rats in different age. METHODS: Three-week-old( n= 16), six-week-old( n= 16) and twelve-week-old( n= 16)male Sprague Dawley rats were all divided into control groups and experiment groups randomly, each group had eight animals, experiment groups were given cocaine hydrochloride (15 mg?kg -1 body weight) subcutaneously daily for four weeks. The ratio of heart weight to body weight (HW/BW,mg/g) were measured. DNA fragmentation of myocardia cells was determined by gel electrophoresis, and caspase-3 activity in myocardia cells was tested by flow cytometry (FCM). RESULTS: In three experiment groups, the DNA isolated from myocardial cells displayed clear ladder pattern. The HW/BW and the caspase-3 activity were increased significantly than those of control groups ( P

Objective To assess the relationship of CYP2C19 gene polymorphism with clopidogrel respon-siveness and the level of EETs in patients with ACS. Methods A total of 123 patients with ACS receiving aspirin combined with clopidogrel dual antiplatelet were enrolled. According to the results of CYP2C19 genotype, patients were divided into three groups:fast metabolic type ,medium metabolic type ,and slow metabolism type. The concentration of EETs and PAIR were compared between three groups. Logistic analysis was used to analyze the risk factors of LCR. Results There were differences statistically in level of EETs and PAIR among the three groups(P<0.05). Logistic analysis showed that the slow metabolism of CYP2C19 gene and lower EETs level were risk factors for LCR. The area under the ROC curve was 0.893(P < 0.05)by EETs level to predict the CYP2C19 genotype. Conclusion The slow metabolism of CYP2C19 gene was an independent risk factor for LCR,while the increase of plasma EETs level was a protective factor.

Objective: To explore the endothelial microparticle (EMP) level in elderly patients with acute coronary syndrome (ACS) combining diabetic mellitus (DM)) and to study the relationship between EMP level and ACS combining DM in elder patients.
Methods: A total of 208 patients≥65 years with coronary angiography in our hospital were summarized including 124 male with the age of (71.9 ± 5.2) years. The patients were divided into 3 groups, Control group, n=51 normal subjects, ACS without DM group, n=83 and ACS+DM group, n=74. Plasma EMP levels were measured by FACSCalibur lfow cytometry as CD31+/CD42b-EMPS and the vascular stenosis degree was quantitatively calculated with Gensini score.
Results: The CD31+/CD42b-EMPs level in ACS + DM group >ACS without DM group > Control group, all P<0.01. Compared with ACS without DM group, the ACS+DM group had more patients with multiple vascular lesions and less patients with single vascular lesions, all P<0.01. The Gensini score was positively related to CD31+/CD42b-EMP level in both ACS+DM group and ACS without DM group (r=0.637, P<0.01).
Conclusion: Plasma CD31+/CD42b-EMPs level increased in elderly ACS patients and the elevation level related to vascular lesion degree/combining with DM, which indicated the endothelial dysfunction in such patients.

Objective To investigate the effects of arsenic trioxide (AS2O3)on SOCS-1 gene methylation and expression of P-STAT3 in multiple myeloma (MM) cells. Methods MM cell lines U266 and CZ-1 were used as in vitro models.Methylation status of SOCS-1 gene was detected by the methylation specific PCR (MSP)while P-STAT3 protein expression was determined by Western blotting assay before and after AS2O3 treatment.Meanwhile growth inhibition and apoptosis of MM cells were determined by flow cytometry. Results Hypermethylation of SOCS-1 gene was observed in each MM cell line compared with wide type. After exposure to AS2O3,it was shown that SOCS-1 gene was demethylated obviously, meanwhile the expression level of P-STAT3 protein and cell proliferation was inhibited significantly in each cell line. The apoptosis rate was increased. When U266 and CZ-1 were treated with AS2O3 of 0,0.5,1.0,2.0 μmol/L respectively, the total cell apoptosisis ratio of U266 was 0.06%,0.56%,48.96%,61.07% (X2 =9.19,P < 0.05); and the total cell apoptosisis ratio of CZ-1 was 4.2%,,40.3%,,47.72%,,68.49% (X2 =8.96,P <0.05 ).Conclusion AS2O3 could inhibit JAK/STAT signal transduction pathway by inducing SOCS-1 gene demethylation in MM cells which might be related to cell apoptosis.
