1.The Thermodynamic Evidence of Two Native Conformations Coexisting in Solution For Apoazuirn: a DSC and CD Study

Whether protein could adopt multiple conformations coexisting in solution is disputable. In a previous report,the conformation heterogeneity of apoazurin mutant M121L had been identified. The thermal unfolding of wild type apoazurin from Pseudomonas aeruginosa is re-investigated with differential scanning calorimetry (DSC) and circular dichroism (CD) methods. The results show that unfolding in the pH range from 4 to 9 is associated with two heat capacity maxima. The low temperature transitions are reversible at all pH conditions used, while the high temperature transitions are irreversible. The two unfolding transitions were analyzed by the two-interchangeable-conformation model with the fraction for the first transition (N1) from 64% at pH 4.0 to 55% at pH 9.0. Temperature induced unfolding monitored at 219 nm shows also two separate transitions. The ratio of the signal changes is consistent with the fractions obtained from the corresponding DSC measurements. These results provide further support for the hypothesis that at least two conformations of apoazurin coexists in solution.

3.Morphology Observation of A Family of May-Hegglin Abnormal Cell

Objective The morphological changes were observed in a pedigree with May-Hegglin anomaly cells.Methods Blood routine detection of abnormal May-Hegglin family members,peripheral blood cell Wright-Giemsa staining,and obser-vation of the white blood cell and platelet morphology.Made bone marrow smears of the proband,and megakaryocyte counts and classification.The proband peripheral blood leukocyte TEM examination were collected and analyzed by PCR sequen-cing.Results To reduce the May-Hegglin abnormal family blood platelet count in different degree,peripheral blood eosino-phil,eosinophilic inclusion visible form was not an alkaline cells,monocytes,neutrophils in body.Conclusion It’s provided valuable clues to diagnosis of morphological examination for May-Hegglin disorders.
4.Two Conformations of Native Pseudomonas aeruginosa Apoazurin in Solution

The mechanism of complex unfolding process of Pseudomonas aeruginosa apoazurin is an arguing problem. Recent published results indicated that this problem could be resolved by hypothesizing two native conformations coexisting in solution.Urea-induced unfolding of apoazurin was investigated further using intrinsinc fluorescence and CD spectra. Equilibrium unfolding curves in urea could be depicted with an apparent two-state transition, but a biphase kinetic process. The fast unfolding process is finished within a few seconds as monitored by stopped-flow fluorescence intensity, whereas the slow process requires several hours for as well as the circular dichroism spectra just after manual mixing of protein and denaturant could be simulated by superposition of the spectra of the native and fully unfolded protein with the same coefficient of 0.37. This strongly suggests the three-state mechanism with a partially unfolded intermediate on its pathway is inadequate. The hypothesis of two native conformations coexistence is a reasonable selection.
5.Design of MRI interventional RF coil

Objective To design a new type of RF endorectal coil so as to increase signal to noise ratio(SNR) and improve the efficiency of MRI diagnosis.Methods Through circuit and construction optimization,a MRI model of coil in vivo was designed.Results The images and spectroscopy of a special phantom were acquired and the results were compared to those of the commercial TORSO coil.Conclusion This coil gives a significantly higher SNR at the region of interest(ROI).The achieved high local SNR and resulting high spatial resolution can add more anatomic and biochemical information to the diagnosis of prostate diseases.
6.A preliminary study on the correlation between gastric lymphoid follicles and Helicobacter pylori infection

Objective To investigate the correlation between H.pylori strain possessing cagA gene and the formation of gastric lymphoid follicles(GLF),and the effect of H.pylori infection on the occurrence of GLF.Methods Antral biopsy specimens from 655 patients (chronic gastritis:n=479,peptic ulcer:n=176)were used for detection of H.pylori infection and histological analysis. CagA gene was examined in 70 clinical isolates by means of PCR- amplification. Results The incidence of lymphoid follicles in gastric mucosa is significantly higher(60.14% ) in patients with H.pylori infection than those without infection(17.06% ).GLF is easier to be detected in patients with active gastritis than in those with inactive gastritis. There is no significant difference in the presence of GLF among H.pylori associated gastroduodenal diseases,such as chronic gastritis ,gastric ulcer. Moreover, there is also no significant relationship between H.pylori strains possessing cagA gene and GLF. Conclusion The presence of GLF might directly related with H.pylori infection, and can be observed as a constant morpholgical marker of H.pylori related gastroduodenal disease ;The formation of gastric lymphoid follicles is not related to the cytotoxin of cag A gene of H.pylori.

Objective:To investigate the expression of matrix metalloproteinase-9(MMP-9) and its correlation with cancer cell proliferation and metastasis in oral squamous cell carcinoma (OSCC). Methods: Immunohistochemical SP method was used to detect the expression of MMP-9 and Ki-67 in 40 cases of OSCC.Results:The expression of MMP-9 was closely correlated with lymph nodes metastasis and TNM stage of OSCC(P0.05). 8.Endovascular treatment of inferior vena cava syndrome

Objective To investigate the curative effect of endovascular treatment for inferior vena cava syndrome(IVCS).Methods A total of 17 cases of inferior vena cava syndrome was treated by endovascular dilatation and self-expandable metallic stent placement between June 2002 and November 2004.Routine anticoagulation therapy was given after the procedure.Results The pressure gradient across the inferior vena cava was reduced from 16.8?4.3 mmHg preoperatively to 2.6?0.6 mmHg postoperatively(t=13.280,P=0.001).The IVCS symptom scores were decreased from 4.4?1.6 preoperatively to 2.1?1.7 postoperatively(t=6.880,P=0.010).Symptoms of edema on lower limbs,scrotal or pubic dropsy,ascites,and anasarca subsided at 1~4 days after procedure.Conclusions Endovascular treatment of inferior vena cava syndrome is effective and reliable.
9.The role of apoptosis of ?-cells in the pathogenesis of Diabetis in OLETF, of obese type 2 diabetes rats

Objective To determine whether the ? cells apoptosis exits in the obesity associated type 2 diabetic rats OLETF and the relationship between the apoptosis and development of type 2 diabetes. Methods Twenty OLETFs and twenty LETOs as control were used. HE staining and TUNE were the way to examine apoptotic ? cell in pancreatic sections. Results (1) The incidence of diabetes in OLETF rats was 95%. None of diabetic rat was found among LETO rats. (2) Apoptotic cells only appeared in pancreatic sections from OLETF rats. (3) The percentage of islet ? cell apoptosis in OLETF rats was higher than that in LETO rats (P=1.26?10 -17 ). (4) In OLETF rats, the percentage of islet ? cells apoptosis was higher in the ages of 4 weeks after the onset of diabetes than that in the ages of four weeks before the onset of diabetes. Conclusion There was a high level of ? cell apoptosis in OLETF rats prior to the onset of diabetes and a further increase of apoptosis after diabetes.
10.Expression of estrogen receptor in human tongue cancer cells

Objective:To observe the expression of estrogen receptor alpha(ER?) and beta(ER?) in human tongue cancer cell line Tca8113 and its highly metastatic cell line Tb. Metheds:Immunocytochemistry, RT-PCR methods were used to observe the expression of ERs in the in vitro cultured human tongue cancer Tca8113 and Tb cells.Results:ER? and ER? mRNA were expressed in both cell lines,and the expression of ER? in Tb cells was stronger than that in Tca8113 cells, the expression of ER? was the same in the two cell lines. The ratio of ER? to ER? was higher in Tb cells than that in Tca8113 cells.Conclusion:The overexpression of ER? and ER?/ER? ratio may be associated with the higher potential of metastasis of Tb cells.
