1.Vitrified cryopreservation and regeneration of shoot-tips for Citrus medica var.sarcodactylis

Objective To provide a new approach of preservation for germplasm resources of Citrus medica var.sarcodactylis(CMS).Methods A micrografting test was made on CMS shoot-tips after vitrified cryopreservation,resulting in living shoot-tip.Results First the CMS shoot-tip is inoculated in medium consisting of MS,5%DMSO,and 5%sucrose for 48 h preincubation.Then the shoot-tips were cut off 3—4 mm long and fore-treated by 60%PVS_2 at room temperature(25℃) for 30 min.After that,they were treated by PVS_2 at 0℃for another 80 min and conserved in liquid nitrogen for 24 h.Next the shoot-tips were defrosted by aqueous bath at 40℃and cleaned twice at room temperature(25℃) by MT minimal medium with additive 1.2 mol/L sucrose,10 min once.Finally data collected recorded a higher survival rate of 81.25%by TTC inspectation and a better regeneration rate of 52.94%if the CMS shoot-tips had been grafted on darkly-cultured parental stock,growing,and differentiating normally afterwards.The plantlets detected by polymerase chain reaction(PCR) showed that the pathogens of Huanglong disease had been removed.Conclusion Therefore a conclusion is made that the approach of vitrified cryopreservation may be applied to the preservation of CMS germplasm resources.
2.Determination of ganoderic acid B in different parts of Ganoderma lucidum

To explore the distribution of the bitter principle ganoderic acid B in different parts of Ganoderma lucidum (Leyss. ex Fr.) Karst.. Methods Ganoderic acid B in epidermis, phellem, stipe, spore and pileus were determined by RP-HPLC. Results Ganoderic acid B was found to be mainly concentrated in the entire surface of the basidiocarp, other parts contained much less amount. Conclusion Result of the study may provide references for the search of such active principle from this fungus.
3.Factors Affecting on the Survival Rate for Shoot-tip Micro-grafting of Citrus medica L.var.sarcodactylis Swingle

Objective To lay a foundation for the cultivation of disease-free plantlet of Citrus medica L.var.sarcodactylis Swingle(CMS) by discussion of the factors affecting survival rate for micro-grafted shoot-tip of CMS.Methods With citrus limon seedlings cultured in the dark as the rootstock,shoot-tip of CMS was grafted to the inverse T-shaped cut in the cotyledon of rootstocks.Then an investigation was carried out on the factors which affected the survival rate for micrografted shoot-tip of CMS.The factors included age of rootstocks,scion size,culture medium with different concentrations of phytohormones,and rootstocks with or without cotyledon.Results The survival rate for micro-grafted shoot-tip of CMS was closely related to the factors mentioned above.A higher survival rate was obtained when CMS shoot-tip with 4 pieces of leaf primordia was grafted to the inverse "T" shaped cut in the cotyledon of parental stock(citrus limon seedlings) which was cultured in the dark for 14 days,and when the citrus limon seedlings were cultivated in MT-based medium with ?-naphthylacetic acid 0.1 mg?L-1 added.Conclusion The optimized micro-grafting conditions for CMS shoot-tips improve the propagation of micro-grafted plantlet and the survival rate of micro-grafted shoot-tip of CMS.
4.Psychosomatic Symptoms and Coping Styles and Their Influences on Quality of Life in Chronic Hepatitis B

Objective:To investigate the Psychosomatic symptoms and coping styles and their influences on quality of life in chronic Hepatitis B.Methods:The Chronic Hepatitis Disease Questionnair(CLDQ),Symptoms Checklist(SCL-90) and Medical Coping Modes Questionnair(MCMQ) were administered to 152 Chronic Hepatitis B.Results:The scores of SCL-90 of the patients were found to be different from the norm and the normal controls and negatively correlated with scores of CLDQ for all domains.The patients had a lower Confronce score and higher Avoidance and Acceptance-Resignation scores compared with the norm,and there was negative correlation between the scores of negative coping styles and the quality of life.Conclusion:The psychosomatic symptoms and the negative coping styles of chronic Hepatitis B can influence their quality of life.
5.Current status on informational self-management intervention of chronic obstructive pulmonary disease patients

Chronic obstructive pulmonary disease (COPD) is the leading death cause of pulmonary diseases, ranking the fourth of global death causes. Self-management program is able to gradually improve the Airways of COPD patients with ventilation, reduce the degree of lung failure, elevate the health status as well as promoting self-efficacy, which is the key process to effectively control COPD relapse. In recent years, more and more domestic and foreign academic employ the widely-used network technology and carry out a large number of information study on COPD self-management intervention. This article reviewed literatures about the intervention methods on self-management of patients suffered from COPD, aimed at providing a better basis for exploring more effective self-management.
6.Research of Field Seedling Technique on Seed Germination and Growth of Desmodium styracifolium(Osb.) Merr.

Objective To observe the influence of sowing time and planting density on seed germination percentage and growth of Desmodium styracifolium(Osb.) Merr..Methods After quality inspection of the seed,the seeds planting test for Desmodium styracifolium(Osb.) Merr.was carried out from March to October,and the influences of sowing time and planting density on the seed germination percentage and growth were observed.Results On the planting base of Pingyuan county in Guangdong province,the seeds of Desmodium styracifolium(Osb.) Merr.planted from March to October had a high germination percentage and the seedling grew well.With the increase of planting density,the seed germination percentage decreased,but was up to 80% when the planting density was 0.5~1.0 g /m2.Conclusion The optimum planting time for Desmodium styracifolium(Osb.) Merr.is in autumn and the optimum planting density is 0.5~1.0 g /m2.
7.DETECTION OF CAg IN CSF OF 231 CEREBRAL CYSTICERCOSIS PATIENTS

The results of detection of circulating antigen(CAg) in cerebrospinal fluids(CSF) of 231 cerebral cysticercosis patients with McAb(4F8)-based ELISA were reported and compared with the case histories , clinical manifestations and CT scans. No relationship was found between CAg detected in CSF of these cases and history of taeniasis. However, the positive rate of CAg in cerebral cysticercosis patients with subcutaneous nodules was found significantly higher than those in cases with simple cerebral cysticercosis and in cases with subcutaneous nodules disappeared after anti-Cysticercus therapy before CSF collection. The results of CAg detection in CSF were related with the CT findings as well as the stage of the disease. It is thus indicated that MeAb(4F8)-based ELISA might be useful not only for diagnosis of neurocysticercosis, but also for the evaluation of efficacy of anti-Cysticercus therapy.
8.Comparative Identification of Different Cultivated Varieties of Amomum villosum

[Objective] To compare and identify the different cultivated varieties of Amormun villosum for providing scientific evidence for the selection, breeding, standardization and industrialization. [Methods] Source appraisal, character identification and scanning electron microscope technique were applied to observe and compare the appearance of the original plant, the properties of the medicinal material and the outside characteristics of pollen of Amomum villosum in Yangchun, Guangdong province. [Results] The two cultivated varieties of Amomum villosum in this area, long-fruit and round-fruit types, are quite different in the appearance and the characteristics of pollen. [Conclusion] It is suggested that long-fruit Amomum villosum and round-fruit Amomum villosum should be the selected varieties for breeding.
9.Influence of Biological Pesticide on Fruting Rate and Yield of Amomum villosum Lour.

[Objective] To observe the influence of different kinds of biological pesticide on the fruiting rate and yield of Amomum villosum Lour. (AVL) . [Methods] Medicinal plant field experiment method was used to observe the influence of biological pesticide A (highly effective immuno-biologic bactericide, 75.76mg?m-2 for one time) and its 1000-fold, 800-fold and 500-fold diluent on the fruiting rate and yield of AVL. Meanwhile, the influence of biological pesticide B (edible oligosaccharide, 30.30mg?m-2 for one time) and biological pesticide C (deguelin emulsion, 1.89mg?2m-2 for one time) was also observed. [Results] The three biological pesticides, as well as the diluents of biological pesticide A increased the fruiting rate and yield of AVL, the influence of the moderate-concentration biological pesticide A being the greatest. [Conclusion] Proper application of biological pesticide is one of the important ways to raise the yield of AVL.
10.Identification of Fructus Citri Sarcodactylis from Three Different Production Areas by Fourier Transform Infrared Spectroscopy

[Objective] To establish a method for rapid identification of Fructus Citri Sarcodactylis (FCS) from different production areas. [Methods] Fourier transform infrared (FTIR) spectroscopy was adopted to identify FCS. The height and position of the characteristic absorption peaks, and I value (a quantitative parameter of peak height ratio) were compared in FCS from different production areas. [Results] Each kind of FCS had their fixed range in peak height ratio I: 0.7-0.9 in FCS from Guangdong, 1.3-1.6 in FCS from Sichuan, 0.9-1.1 in FCS from Zhejiang, indicating that I value can be used to identify FCS from different areas . [Conclusion] For the identification of FCS from different production areas, FTIR spectroscopy is an effective method, in which there is no need for isolation, purification and chemical treatment.
