Objective To estimate the effect of pedicle screw fixation of thoracolumbar fractures via paraspinal approach and compare it with the conventional posterior midline approach. Methods Forty-two cases of thoracolumbar monosegmental fractures subjected to single posterior pedicle screw fixation and reduction from December 2008 to May 2010 were included in the study.Among the patients,19 cases were operated through paraspinal muscular-sparing approach (paraspinal approach group) and 23 cases through posterior midline surgical approach (conventional approach group).Surgical incision length, operation time, intraoperative blood loss, postoperative drainage volume, postoperative hospital stay,pre-and post-operative VAS and other perioperative indices as well as fracture reduction outcome were compared between the two groups.Oswestry disability index (ODI) was assessed after operation. Results There were no statistical differences between the two groups in aspects of surgical incision length, operation time, postoperative hospital stay, height restoration of fractured vertebra (P > 0.05),but intraoperative blood loss (148.5 ± 26.5) ml,postoperative draining loss (72.9 ± 17.3) ml,postoperative VAS (1.1 ± 0.3) points and ODI (13.4 ± 2.7) points in paraspinal approach group showed statistical differences from those in conventional approach group (P < 0.05).Conclusion Paraspinal muscle-sparing approach is characterized by minor trauma, less bleeding, slight pain and quick recovery as compared with conventional posterior midline approach and hence may be the preferred choice for the treatment of thoracolumbar fracture without spinal canal decompression.

Objective To explore the role and significance of free radicle in the pathophsiological mechanisms of diabetic cystopathy(DCP).Methods The detrusor strip contraction experiments were performed and observed in the group of type 2 diabetes mellitus(T2DM) rats at different time stages and the control group. The bladder tissue samples was made into homogenate, and the activities of superoxide dismutase(SOD) and the contents of malondialdehyde(MDA) were determined. Results The minimum stretch forces to induce the detrusor contractions in T2DM group were larger than the control group.Compared with the control group, the detrusor contractive frequency was higher in the period of 0 to 16 weeks, but was lower after the 20th week. The maximum detrusor contractive forces in the T2DM group showed a descended tendency with the elapse of the experiment time.In the bladder homogenate of the control group, the activities of SOD was declined after reaching the peak at the stage of 8th week, and then escalated at the stage of 24th week.As for the T2DM group, the activities of SOD became higher in the 4th week, than descended at the stage of 8th and 24th weeks. The contents of MDA in both groups showed descended tendency. In T2DM group,MDA became even lower than the control group in the 4th week and became higher than the control group in the 8th week. The ratio of SOD/ MDA in T2DM group was lower than the control group. In details, the ratio of SOD/MDA in T2DM group rose significantly in the 4th week, while declined in the 8th week. There was a negative correlation between the maximum detrusor contractive force and detrusor contractive frequency, and a positive correlation between the maximum detrusor contractive force and the contents of MDA.Conclusion The destrusor functions are impaired by Diabetes Mellitus. In the initial stage of DCP,the bladder functions are normal, because the detrusor had a high ability to remove the free radicle. In the progression stage of DCP,the bladder functions become decompensation, because the organism is seriously injured by the free radicle. So the impairment by free radicle is one of important mechanisms of destrusor impairment of diabetic cystopathy.

Objective To investigate changes in the firing activities of nucleus accumbens (NAc)neurons and their response to 5-HT7 receptor stimulation in a rat model of Parkinson’s disease (PD).Methods The firing activities and response of NAc neurons to 5-HT7 receptor agonist in PD rats were recorded by in vivo electroneurophysiology and neuropharmacology and then were compared with those in the sham group. Results The mean firing rate of NAc neurons was (5.46 ±0.88)Hz in the sham rats and (3.77 ±0.48)Hz in the PD rats. The firing rate of NAc neurons increased significantly compared with that in the sham rats (P <0.05).In PD rats, 65% of NAc neurons fired in bursts and 35% fired irregularly. However,in the sham rats,57.5% of NAc neurons fired in bursts and 42.5% fired irregularly. There was no significant difference in the firing pattern of NAc neurons between the PD and sham rats (P >0.05 ).Systemic administration of 5-HT7 receptor agonist AS1 9 increased the firing rate of NAc neurons in the sham and PD rats. This excitation was significant at a high dose of 1 60 μg/kg for NAc neurons in the sham rats (P <0.05).However,the excitation produced by AS1 9 was significant at a high dose of 80 μg/kg in PD rats (P <0.05).The cumulative dose-produced excitation in the PD rats was lower than that in the sham rats. The effects induced by AS1 9 were reversed by the 5-HT7 receptor antagonist SB269970 in both groups. Conclusion The reinforced firing activity of NAc neurons might be mediated by 5-HT7 receptor in the neurons of PD rats.

Following Escherichia coli lysis with alkali, cetyltrimethylammonium bromide (CTAB) was directly titrated into the supernatant. An easy and feasible technology for plasmid purification was established with the optimized proportion between the quantity of CTAB and plasmid, combined with the specific solution for DNA release and TritonX-114 for endotoxin removal. Quality detection showed that the purified plasmid was free of contamination of host RNA. The host bacterial genomic DNA, endotoxin and bacterial protein were less than 10 microg, 50 EU and 10 microg per mg plasmids, respectively. The ratio of OD260/OD280 was between 1.75-1.85. Eighty percent of the prepared plasmids were presented in the supercoiled form. The plasmid purified with this technology can satisfy all criteria stipulated by FDA. The main advantages of the technology include the avoidance of animal-derived enzymes such as ribonucleases A, Proteinase K and toxic reagents like chloroform and phenol. In addition, the technology has low cost and no pollution.

Chinese Journal of Endocrine Surgery 2020;14(3):246-250
Objective:To investigate the expression changes of SKOV3 and MRTFA in ovarian cancer and their effects on tumor cell proliferation and invasion.Methods:91 cases of patients with ovarian cancer admitted to our hospital from Jun. 2017 to May. 2019 were enrolled in this study. The relationship between MRTFA mRNA expression and clinical characteristics in ovarian cancer tissues was analyzed. The ovarian cancer SKOV3 cells were cultured and grouped, and the proliferation activity of ovarian cancer SKOV3 cells was detected by MTT. The invasive ability of SKOV3 cells was determined by Transwell method, and the expression of MRTFA protein in SKOV3 cells was determined by Western blot. Results:The relative expression of MRTFA mRNA in ovarian cancer tissues was significantly higher than that in adjacent tissues ( P<0.05) . The relative expression of MRTFA mRNA in ovarian cancer tissues with tumor diameter <10 cm, FIGO stage I-II stage, moderately high differentiation and no lymph node metastasis was lower than that with tumor diameter≥10cm, FIGO stage III-IV stage, poor differentiation, lymph node metastasis in patients with ovarian cancer ( P<0.05) . The absorbance values of ovarian cancer SKOV3 cells in MRTFA intervention group were lower than those in the control group and the negative control group at 24 h, 48 h, 72 h and 96 h ( P<0.05) . Conclusions:The expression of MRTFA is abnormally increased in ovarian cancer. The specific inhibition of MRTFA protein expression can block the proliferation and invasion of SKOV3 cells. The mechanism of action may be related to the transformation process of SRF induced epithelial mesenchyme.
