Objective To determine the effect of transcription and translation in telomeric related proteins,and synergism of progressive telomere shortening and cell cycle alteration in human lung adenocarcinoma A549 cell line,which is induced by antisense tankyrase oligonucleotide(asTANKS) combinated with antisense human telomerase reverse transcriptase(ashTERT) oligonucleotide.Methods A549 cells were randomly assigned as 3 test groups: ashTERT,ashTERT + asTANKS and asTANKS,three control groups(shTERT,sTANKS and blank).With individual intervention for different hours,the effect of transcription in hTERT mRNA was evaluated by RT-PCR,and telomerase activity was tested by ELISA-PCR,tankyrase activity was tested by Western blot as well.Moreover,telomere average length was analyzed by Q-FISH,and duration of proliferation was observed by population double test.Results Transcription in hTERT mRNA and telomerase activity for 48 hrs was inhibited obviously by ashTERT,but not by asTANKS.Progressive telomere shortening in A549 cells for 48 hrs was induced by either asTANKS or ashTERT(vs control,P

In this article, an HPLC method for the contents determination of amino acids in Qihong Maitong injection was reported. In detailed, OPA-Fmoc pre-column derivatization was adopted, and related HPLC methods to determine the contents of amino acids was established. Linear relationship was well constructed for 17 amino acids through the method mentioned above. Briefly speaking, the optimized method was accurate and reproducible, and suitable for the determination of amino acids in Qihong Maitong injection and corresponding quality control.

4.Protective Effects of Proanthocyanidins on Intestinal Function after Cerebral Ischemia-reperfusion Injury

Objective To explore the protective effects of proanthocyanidins pretreatment on intestinal function after cerebral ischemia-reperfusion injury. Methods 24 Sprague-Dawley rats were randomly divided into sham group (group A, n=8), ischemia-reperfusion group (group B, n=8) and proanthocyanidins pretreatment group (group C, n=8). The model of cerebral ischemia-reperfusion injury in rats was established according to Longa's method. Group C was intraperitoneally injected with proanthocyanidins 10 mg/(kg ⋅ d), group A and group B were injected with normal saline for 5 consecutive days. 1 and 3 days after cerebral ischemia-reperfusion, ileum myoelectric slow wave and smooth muscle contractility, the activity of superoxide dismutase (SOD) and content of malondialdehyde (MDA) were measured, the content of the serum TNF-α was tested with ELISA kit, ileum tissues were tested with hematoxylin eosin (HE) staining and used for measuring the moisture content. Results Compared with group B 1 and 3 days after cerebral ischemia-reperfusion, the intestinal mucosa injury relieved, the intestinal mucosa score decreased (P<0.05) and the number of infiltrated inflammatory cell decreased in group C; the frequency of slow wave and contraction trended to increase (P>0.05), and the amplitude increased (P<0.05) in group C; the serum SOD activity increased (P<0.05), and the content of MDA and TNF-α decreased (P<0.01) in group C; the intestinal moisture content reduced (P<0.01) in group C. Conclusion Proanthocyanidins pretreatment can protect intestinal function from injury after cerebral ischemia-reperfusion.
5.Prognostic relevance of preoperative platelet-to-albumin ratio(PAR) in patients with upper urinary tract urothelial carcinoma

Objective:To assess the value of platelet-to-albumin ratio on overall survival(OS) and cancer specific survival(CSS) in patients with upper urinary tract urothelial carcinoma(UTUC).Methods:169 patients with UTUC in Qilu hospital of Shandong University from January 2006 to December 2013 were included in this study, retrospectively. 107(63.3%) were male and 62(36.7%) were female. Their age ranged from 36 to 87 years old, mean 65 years old. 73(43.2%)were less than 65 years old and 96(56.8%) were more than 65 years old. 38 patients had the history of smoking.15 patients had the history of bladder cancer. The single tumor was found in 159 cases (94.1%). 10 patients(5.9) owned more than one site of tumor. For the ECOG, 159 cases (94.1%) were 0 score and 10 cases(5.9%) were ≥1 score. The mean Platelet count was 217.0×10 9/L, ranging (68.0-772.0)×10 9/L. The average serum albumin was 42.0 g/L, ranging (27.3-52.7)g/L. The mean PAR was 5.40×10 9, ranging(0-17.31)×10 9. 147(87.0%) patients accepted radical nephroureterectomy and 22(13.0%) cases accepted segmental ureterectomy.155 cases(91.7%) used postoperative adjuvant chemoradiation. The patients were divided into two groups as low PAR or high according to the receiver operating curve (ROC)analysis of PAR. And the Kaplan-Meier analysis was used to assess the OS and CSS and test the equivalences of the curves by log-rank tests. Univariate and multivariate survival analyses were carried out using Cox proportional hazards regression model. Results:We set 6.67×10 9 was the optimal cutoff value of PAR. Therefore, the cohort was divided into 2 groups, 133(78.7%) patients had low PAR (<6.67×10 9) and 36 patients had high PAR (≥6.67×10 9). The 3-year OS and 5-year OS of high preoperative PAR group were 50.0% and 46.2%, respectively. And low preoperative PAR group were 77.6% and 66.7%, respectively. In univariate analysis, no difference was found in gender, age, tumor size, history of smoking and LVI. Meanwhile, tumor grade (G 1-2 or G 3), pathological T stage (pT a/T 1 or pT 2-4) and PAR (<6.67×10 9 or ≥6.67×10 9) were significantly associated with OS and CSS. Multivariate analysis with a Cox proportional hazards regression model were performed and showed that PAR was significantly associated with OS and CSS (OS: HR=1.850, 95% CI 1.095-3.127, P=0.022; CSS: HR=2.154, 95% CI 1. 242-3.736, P=0.006). Conclusions:Preoperative PAR is an independent prognostic factor in predicting patient with UTUC. And the platelet and serum albumin level are low cost and easy to obtain. PAR may be expected to become a biological factor to predict the prognostic factor in patients with UTUC.
6.Modification and innovation of in-situ full-left/full-right liver splitting technique

Objective:To explore the feasibility of technological modification and innovation of full-left/full-right liver splitting in situ for donors and examine the safety of clinical application for liver transplantation (LT).Methods:From March 2021 to June 2022, clinical and surgical data are retrospectively reviewed for 27 donors undergoing full-left/full-right liver splitting in situ and the corresponding 49 recipients undergoing full-left/full-right LT.According to the split liver technique used in donor liver surgery, they are divided into conventional split group(group A, 13 cases)and innovative split group(group B, 14 cases). The corresponding recipients are divided into two groups of recipient C(25 cases)and recipient D(24 cases). General profiles, intraoperative findings, type of vascular allocation and short-term outcomes in two groups are compared.After full-size split liver transplantation(fSLT), follow-ups continued until the end of September 2022.Results:There are 23 males and 4 females in donors.The causes of mortality for donors are traumatic head injury(12 cases)cerebrovascular accident(13 cases)and anoxia encephalopathy(2 cases). Baseline characteristics of two groups indicate that body weight and body mass index(BMI)are higher in group B and blood sodium level is lower than that in group A( P<0.05). No statistical differences exist for the others.Liver splitting time is significantly shorter in group B than that in group A(175 vs.230 min, P=0.022). No significant inter-group difference exists in type of vascular allocation.Retrohepatic inferior vena cava(IVC)is split in one case in group A and 10 cases in group B( P=0.001). Among 20 cases of right hemiliver requiring a reconstruction of segment Ⅴ/Ⅷ venous outflow, 12 cases in group A and 3 cases in group B are reconstructed with conventional independent bridging method(independent type)while another 5 cases in group B reconstruct with innovated technique by bridging Ⅴ/Ⅷ vein for splitting IVC with iliac vessel and molding all outflows as one for anastomosis(combined typ e). There is significant inter-group difference( P=0.004). No significant differences exist in operative duration, anhepatic phase or blood loss between groups C and B, except for T tube retaining in 7 cases of group A and 14 cases of group D( P=0.032). Twelve cases developed a total of 26 instances of≥Clavien-Dindo grade Ⅲ complications.Of which, 7 cases in group C and 5 cases in group D show no significant difference in postoperative morbidity.However, for serious biliary complications(≥Clavien Dindo grade Ⅲ), there are 6 cases in group C versus none in group D( P=0.016). Two cases died from postoperative complication with a postoperative mortality rate of 4.1%.Postoperative hospital stay is similar in two groups.And accumulates 6/12-month survivals were 95.9% and 87.7% for grafts and 95.9% and 92.4% for recipients respectively. Conclusions:Operative duration of full-left/full-right liver splitting in situ tends to shorten with an accumulation of a certain amount of cases.Technological modification and innovation in IVC splitting and segment Ⅴ/Ⅷ vein reconstruction should be further validated as both feasible and safe by short-term outcomes of the corresponding recipients.

Objective To explore the relationships of β2-microglobulin(β2-MG),retinol bind-ing protein(RBP)and aquaporin-2(AQP2)in serum and urine with the severity of hydronephrosis induced by ureteropelvic junction obstruction(UPJO)in children.Methods Sixty children with hy-dronephrosis who underwent surgery for UPJO at the Department of Urology of the Sixth People's Hos-pital of Nanning were selected as hydronephrosis group.According to the severity of hydronephrosis,they were divided into mild group(n=13),moderate group(n=22),and severe group(n=25).Meanwhile,60 healthy children in the same period were selected as the control group.The serum and urine levels of β2-MG,RBP and AQP2 were compared between control group and children with hydronephrosis of hydronephrosis group.The correlation between the levels of β2-MG,RBP,AQP2 and ultrasound parameters[anteroposterior diameter of renal pelvis(APD),parenchymal thickness(PT)]was analyzed.The receiver operating characteristic(ROC)curves were used to analyze the diagnostic efficacy of β2-MG,RBP and AQP2 in serum and urine for moderate to severe hydrone-phrosis.Results The levels of β2-MG and RBP in serum and urine of the hydronephrosis group were higher than those in the control group,and the levels of AQP2 in serum and urine were lower than those in the control group(P<0.05).The levels of β2-MG inserum and urine,RBP in urine,and APD from low to high in different groups were as follows:mild group,moderate group to severe group.Urine AQP2 level and PT from high to low in different groups was as follows:mild group,moderate group to severe group(P<0.05).Serum RBP level in the mild group was lower than those in the moderate group and the severe group(P<0.05).Urinary AQP2level was negatively correlated with APD and positively correlated with PT(P<0.05).ROC curve analysis results showed that serum β2-MG,urine β2-MG,urine RBP,urine AQP2 detection had certain diagnostic value in moderate and severe hydronephrosis,and the area under the curve(AUC)was 0.652,0.688,0.859,0.680,respectively.The AUC of urinary RBP in the diagnosis of moderate and severe hydronephrosis was significantly higher than that of other indexes(P<0.05).Conclusion β2-MG in serum and urine,urine RBP and urine AQP2 are related to the severity of hydronephrosis induced by UPJO in children.Urine RBP level has better diagnostic efficacy for moderate to severe hydrone-phrosis than other indicators.

9.Association between GPX-1 single nucleotide polymorphisms and susceptibility to noise-induced hearing loss among Chinese Han population.

OBJECTIVETo investigate the association between single nucleotide polymorphisms (SNPs) in glutathione peroxidase 1 (GPX-1) gene, rs3448, rs1050450, rs1800668, and rs1987628, and the susceptibility to noise-induced hearing loss (NIHL) among Chinese Han population.
METHODSA case-control study was conducted to investigate the threshold shift of the left ear at 3000 Hz among the workers of Chinese Han population who were exposed to the same level of sound pressure. Two hundred and one (10%) of the subjects with the highest level of threshold shift were recruited in susceptible group, while 202 of (10%) of the subjects with the lowest level of threshold shift were recruited in tolerant group. Targeted occupational health survey and questionnaire survey were performed among these people. For each individual, genome DNA was extracted from 5 ml of fasting peripheral venous blood. Four SNPs (GPX-1 rs3448, rs1050450, rs1800668, and rs1987628) were genotyped by the TaqMan SNP genotyping kit. The main effects of SNPs and the association between NIHL susceptibility and SNPs were analyzed by logistic regression.
RESULTSThe C allele of rs1987628 was a risk factor for NIHL, with an odds ratio (OR) of 2.531 (95%CI: 1.878-3.411) as compared with the T allele. The CC genotype of rs1987628 was more associated with NIHL than the TT genotype (OR = 3.500, 95% CI: 1.984-6.174; adjusted OR = 3.544, 95% CI: 1.974 ∼ 6.364).
CONCLUSIONAmong Chinese Han population, GPX-1 SNP rs1987628 may be associated with the susceptibility to NIHL.

As the backbone force of China's social and economic construction, the health status of workers is closely related to the nation's productivity and social development. Currently, cancers have become one of the major diseases threatening the health of workers. However, there are still many shortcomings in the cancer screening services for the workers. To standardize cancer screening services for workers, ensure the quality of screening services, and improve the overall screening effectiveness, 19 institutions, including Peking Union Medical College Hospital of the Chinese Academy of Medical Sciences, have jointly formulated the Group Standard "Specification for service of cancer screening for workers (T/CHAA 023-2023)". This standard follows the principles of "legality, scientific rigor, advancement, and feasibility" and combines the frontier scientific advances in cancer screening. It clarifies the relevant requirements for service principles, service design, service delivery, service management, service evaluation, and improving worker cancer screening. Implementing this group standard will help connect the common screening needs of workers, employers, and cancer screening service providers, standardize the screening process, improve screening quality, and ultimately increase the early diagnosis rate and survival rate of cancer patients. Consequently, this group standard will help safeguard workers' health rights and interests, ensure the labor force resources, promote the comprehensive coordinated and sustainable development of society, and contribute to realizing the "Healthy China 2030" strategic policy.
