1.Comparison of conventional and new mesh hydroxyapatite in repair of cleft lip and palate

BACKGROUND:Despite regular mesh hydroxyapatite has certain biocompatibility and bone conductivity, it cannot be absorbed by human body after transplantation, and wil affect the growth and eruption of maxila and fangs when using for filing the injured alveolar cleft. OBJECTIVE: To compare the effect of conventional and new mesh hydroxyapatite composites in repair of alveolar cleft defects. METHODS: Totaly 46 patients with cleft lip and palate were randomly and evenly divided into test and control groups, and al were arranged according to cleft lip and palate sequence treatment. After nasal soft tissue defect repair, new mesh hydroxyapatite composite bone material (hydroxyapatite and bone morphogenetic protein compound). Conventional mesh hydroxyapatite material was adopted in the test and control groups, respectively. Within 6 months after the repair, the vertical distance from the newly formed bone tissue plane to the top of alveolar ridge was observed. The clinical repair effect was comprehensively evaluated from the aspects of complications, alveolar cleft shape and size, calus formation and material absorption. RESULTS AND CONCLUSION: At 2, 4, 8, 12 and 24 weeks, the vertical distance from the newly formed bone tissue plane to the top of alveolar ridge in the test group was higher than that in the control group (P < 0.05). The clinical repairing significant efficiency and total effective rate were al higher than those in the control group (P< 0.05). These results show that new mesh hydroxyapatite composite bone materials in the repair of alveolar cleft defects have good bone osteoinductivity and biocompatibility, rare complications and rapid calus formation.
2.Effect of acupuncture Zusanli on brain functional magnetic resonance imaging

Objective To study the effect of acupuncture Zusanli on brain functional magnetic resonance imaging.Methods 96 healthy volunteers were randomly divided into acupoint group and non-acupoint group.The acupoint group received acupuncture Zusanli,and the non-acupoint group received manipulation,acupuncture and sham acupuncture.The functional brain imaging was determined by MRI,the brain functional images were compared between two groups.Results All subjects were matched with the end of the experiment,the acupoint group received acupoint stimulation,had obvious needling sensation,the non-acupoint group had mild needling sensation.The acupoint group activated 13 brain regions,non-acupoint group in 10 brain regions,the thalamus,pons,supramarginal gyrus in non-acupoint group without the activation.In the basal ganglia (28 cases vs.5cases) and temporal lobe (38cases vs.21cases) between the two groups were activated,the difference was statistically significant(x2 =36.250,P < 0.05).In the temporal lobe,the left hemisphere response rate(73.68%) was significantly higher than the right hemisphere (36.84%),the difference was statistically significant (x2 =8.265,P < 0.05).In the basal ganglia,left hemisphere activation (67.86%) was higher than that of the right hemisphere(42.86%),but the difference was not statistically significant (x2 =0.724,P > 0.05).Conclusion The research of acupuncture by brain functional magnetic resonance imaging showed that Zusanli acupoint can activate the brain function area through brain regulation and treatment of body organs,so as to prove the existence of its own and meridian sensing characteristics.

5.Expression of Twist and relation with epithelial-mesenchymal transition in oral squamous cell carcinoma.

OBJECTIVEThe objective of this paper was to study the expression of related protein and Twist transcription factor of epithelial-mesenchymal transition in oral squamous cell carcinoma (OSCC) tissue and the correlations of OSCC and oral squamous cell carcino-metastasis. The paper also investigated the clinical significance of expression on OSCC.
METHODSThe labels of epithelium materialization (E-cadherin and cytokeratin), stromal labels (N-cadherin), transcription factor Twist protein, and mRNA expression in 30 OSCC tissues were investigated via immunohistochemistry and in situ hybridization. The paper also conducted contrast analysis with clinicopathology.
RESULTSImmunization result showed that the expressions of Twist and N-cadherin in the OSCC group were more significant than those of the normal group (P<0.05). The expressions of E-cadherin and keratin in OSCC were significantly lower than those of the normal group (P<0.05). In the moderate- and low-differentiated group of OSCC, the expressions of Twist and N-cadherin were higher than those of the high-differentiated group (P<0.05). The expressions of E-cadherin and keratin were lower than those in the high-differentiated group (P<0.05). In the lymphatic metastasis group, the expressions of Twist and N-cadherin were higher than those of no-lymphatic metastasis group (P<0.05). The expressions of E-cadherin and keratin were lower than those of the no-lymphatic metastasis group (P< 0.05). Results of in situ hybridization showed that the expression of Twist mRNA in the moderate- and low-differentiated groups of OSCC, T3, and T4 groups as well as that of the lymphatic metastasis group were higher than those of the high-differentiated, T1 and T2 groups, and no-separate lymphatic metastasis group, and the differences were statistically significant (P<0.05).
CONCLUSIONEpithelium materialization exists in OSCC tissue. Twist can enhance the invasiveness of tumor cell and promote the infiltration and metastasis of OSCC. The combined detection of Twist, E-cadherin, and N-cadherin expressions can effectively predict and estimate OSCC metastasis.

6.Function of diffusion-weighted imaging-perfusion-weighted imaging mismatch for intravenous thrombolysis in patients with acute ischemic stroke

Objective To assess the relationship between diffusion-weighted imaging-perfusionweighted imaging (DWI-PWI) mismatch and the prognosis of patients with acute anterior circulation ischemic stroke receiving intravenous thrombolysis.Methods Patients with acute anterior circulation ischemic stroke who finished multimode MRI (T1 WI,T2 WI,DWI,PWI) within 4.5 hours after disease onset were recruited in this study.They were divided into DWI-PWI mismatched and matched groups.All patients received intravenous thrombolysis using recombinant tissue type plasminogen activator (rt-PA).Head CT was performed 24 hours later to exclude cerebral hemorrhage,and the patients were followed up for 3 months.The improvement of neurological function,the rate of death and symptomatic intracranial hemorrhage (sICH) were recorded.Results A total of 63 patients were recruited in this study,of which 41 patients presented DWI-PWI mismatch while 22 patients presented DWI-PWI match.Three months later,the rate of the improvement of neurological function in the DWI-PWI mismatched group was significantly higher than that in the DWI-PWI matched group (70.7% vs 40.9%,x2 =5.32,P =0.021).Of note,no significant difference in the rate of death and sICH was found between DWI-PWI mismatched and matched groups (9.8% vs 13.6%,x2 =0.22,P =0.640).Conclusions Compared with DWI-PWI matched group,the DWI-PWI mismatched group has better prognosis after receiving intravenous tbrombolysis.The results provide more direct evidence supporting the application of intravenous thrombolysis under the guidance of multimode MRI in acute anterior circulation ischemic stroke.
7.An experimental study on cytotoxicity of monocytes/macrophages against Tca8113 cells in hypoxic microenvironment

Objective; To investigate the cytotoxicity of monocytes/macrophages against Tca8113 cells in hypoxic microenvironment in vitro. Methods; Peripheral blood mononuclear cells were extracted from healthy person^ blood and cultivated to transform into monocytes/macrophages. The monocytes/macrophages were cultured with Tca8113 cells in hypoxic microenvironment (O_2 < 1%) and in normal microenvironment (20% O_2). The cytotoxicity of monocytes/macrophages against Tca8113 cell was examined by MTT assay. Results;The cytotoxicity of monocyles/macrophages against Tca8113 cells in hypoxic microenvironment was lower than that in normal environment(P<0.05). Conclusion; In hypoxic microenvironment, the cytotoxicity of monocytes/macrophages against Tca8113 cells is decreased. Bui the mechanism of monocytes/macrophages' function in tumors need further research.
8.Infiltration and effect of CD68+ and CD163+ macrophages in oral squamous cell carcinoma

Objective To investigate the correlation between the number of CD68+ and CD163+ macrophages infiltration and the clinical characteristics of oral squamous cell carcinoma (OSCC),and to investigate the role and significance of CD163+ and CD68+ macrophages in OSCC.Methods Forty-two cases of OSCC resected by operation and diagnosed by postoperative pathology with intact clinical data in our hospitals during 2013-2016 were selected.Other 12 cases of oral mamaxillofacial normal tissue served as controls.The number of CD68+ and CD163+ macrophages infiltration in OSCC was detected by immunohistochemistry.Its correlation with clinical characteristics was analyzed.Results The number of CD68+ and CD163+ macrophages infiltration in OSCC was significantly higher than that in the control group (P<0.05);the infiltration quantity of CD68+ and CD163+ macrophages in poor differentiation and moderate differentiation was significantly higher than that in high differentiation(P<0.05).The number of CD68+ macrophages in lymph node metastasis was significantly lower than that without lymph node metastasis (P<0.05).The number of CD163+ macrophages in the lymph node metastasis was significantly higher than that of CD68+ (P<0.05).The number of CD68+ and CD163+ macrophages had positive correlation in OSCC (r=0.48,P=0.00).Conclusion Tumor-associated macrophages (TAMs) and M2 macrophages may play an important role in OSCC,TAMs may inhibits lymph node metastasis in OSCC,while M2 marcophages may promote the lymphatic matastasis of OSCC.
9.Research into the antibody detection technology of mink plasmacytosis and its current applications.

Mink plasmacytosis, caused by Aleutian Mink Disease Virus (AMDV), poses a threat to the development of the animal fur industry. Neutralizing antibodies against AMDV may result in a persistent infection rather than providing protection for minks. To date,no specific methods to prevent or cure this disease have been developed. In order to eliminate mink plasmacytosis, antibody detection technology has been used globally as a dominant approach to screen for AMDV-positive minks. This paper introduces the classical technology, counterimmunoelectrophoresis and emerging technology in terms of AMDV antibody detection,and provides a glimpse into the future development of these technologies.

10.Effects of WWOX gene transfection on cell growth of epithelial ovarian cancer

Objective To study the effects of anti-oncogene WWOX on cell growth of epithelial ovarian cancer,in order to find a new approach of gene therapy for ovarian cancer.Methods A eukaryotic expression vector containing WWOX was transfected into ovarian cancer cell line HO8910 in vitro (recombinant plasmid group),and positive cell clones were selected and amplified.Expression of WWOX protein was detected by western blot. Untransfected cell(blank contrast group) and transfected empty plasmid cell(empty plasmid group)were served as control groups.In vitro,the biology effect of WWOX on HO8910 cell was analyzed throush the methyl thiazolyl tetrazolium test,transwell chamber cell invasion assay in vitro,agarose clony-formation and flow cytometry.In vivo,the cell of transfection was transplanted intraperitoneally in to BALB/c nude mice.The survival time and growth ability of nude mice were observed.Results (1)Recombinant plasmid group cell could steadily express WWOX protein,while in empty plasmid group and blank control group the expression of WWOX protein were not detected.(2)The growth rate of recombinant plasmid group cell was inhibited.(3)The agnrose clony-formation rate of recombinant plasmid group(19.8%)was significantly lower than that of the empty plasmid group(54.5%)and blank control group(56.0%,P<0.05).(4)Flow cytometry showed that(72.08±0.39)% of cells was arrested at G0/G1 stage in recombinant plasmid group, while in empty plasmid group and blank control group G0/G1 stage cells were at (41.02±1.08)% and (39.31±0.67)% (P<0.05). (5) In vitro invasion assay showed that invasion cell number in recombinant plasmid group (89.7±3. 1 ) was not significantly different from that of empty plasmid group(91.2±1.3) and blank control group(91.4±1.3, P >0. 05). (6) In vivo test in nude mice showed that WWOX gene could inhibit tumor growth of the HO8910 cells. Conclusions Tumor suppressor gene WWOX could interfere with the cell cycles of ovarian cancer cell and inhibit cell proliferation. As a new valuable tool,it premises to have application in the gene therapy of ovarian cancer.
